利用免疫组化法检测c-Fos 蛋白在不同季节刺猬嗅球各层次的表达差异,探讨c-Fos、嗅觉、冬眠三者的关系。分别选取春、夏、秋、冬四个季节各6 只野生健康刺猬,固定剥离嗅球,石蜡切片,免疫组化显色,拍片,载入Motic Images Advanced 3.2 软件,测量四个季节刺猬嗅球各层次c-Fos 的表达率,将结果载入GraphPadPrism4 软件分析,Microsoft Excel 作图。结果表明:c-Fos 蛋白在成年刺猬嗅球各层均有不同程度的表达,阴性对照不着色,且表现出明显的季节性差异。1)与秋季相比,冬眠期c-Fos 蛋白在刺猬嗅球各层次的表达均有极显著的降低(P <0.01);2)与夏季相比,冬眠期c-Fos 蛋白在外网丛层、僧帽细胞层、颗粒细胞层的表达有极显著降低(P < 0. 01),在嗅神经层、嗅小球层、室管膜层的表达也有显著降低(P < 0. 05);3)与冬眠期相比,春季c-Fos 蛋白在嗅小球层、僧帽细胞层、颗粒细胞层的表达有极显著的升高(P <0. 01),在嗅神经层、外网丛层、室管膜层的表达却没有显著变化(P ﹥ 0.05);4)嗅神经层c-Fos 的表达在春季显著低于秋季,夏季与秋季没有显著差异。颗粒细胞层夏季显著低于秋季(P < 0.05)。秋季c-Fos 在其余各层次的表达与春季、夏季相比都有极显著的提高(P <0.01)。结论:秋季刺猬嗅球神经元最活跃,嗅觉最灵敏,冬眠期刺猬嗅球活跃性大大降低,嗅觉系统最迟钝。c-Fos 在刺猬嗅球中的强表达表明其在嗅觉信息的传递中可能发挥一定作用,c-Fos 表达率的显著季节性差异揭示了刺猬嗅球的活跃性与其冬眠具有一定的相关性。  相似文献

植被恢复一直是恢复生态学研究的核心问题和首要解决目标。该文拟评估山东蒙山森林植被恢复与重建的现状和程度, 评价不同造林树种对植物多样性的影响, 筛选一批造林工具种, 为今后沂蒙山区森林植被恢复与重建的调控和预测提供依据。采用样地法和样方法, 选择林龄超过40年的6种主要人工林为样地, 采用典型取样法进行林内调查。根据乔木层、灌木层和草本层的物种丰富度、Shannon-Wiener多样性指数、Simpson多样性指数和Pielou均匀度指数评价物种多样性, 分析乔木径级分布, 判断植物发展类型。结果表明: 乔木层、灌木层和草本层的4种多样性指数较为一致地显示赤松(Pinus densiflora)、栓皮栎(Quercus variabilis)、黑松(Pinus thunbergii)、日本落叶松(Larix kaempferi)和刺槐(Robinia pseudoacacia)的成林效果较好, 油松(Pinus tabuliformis)相对较差。6种人工林群落灌木层和草本层的4种多样性指数基本无显著差异, 而乔木层中, 油松人工林群落的4种多样性指数与黑松人工林、刺槐人工林、栓皮栎人工林和日本落叶松人工林存在部分显著差异(p < 0.05)。从整体上看, 人工林群落灌木层发育最好, 4种多样性指数均为灌木层>乔木层>草本层, 且差异大多极显著(p < 0.01)。研究发现: 乔木扩展种29种、隐退种3种、稳定侵入种9种和随机侵入种11种, 表明人工林群落正处于向森林演替的过程中。数据显示: 3种隐退种赤松、油松和日本落叶松均为群落局部优势种, 赤松和油松种群更新困难, 日本落叶松完全没有更新幼苗。作者认为林下发达的灌草层可能是阻碍针叶林天然更新的关键因素。评估筛选出15种造林工具种: 早期开荒工具种(A类) 7种和后期顶级工具种(B类) 8种。其中A1类(建群种) 2种: 赤松和油松, A2类(伴生种) 5种: 黄檀(Dalbergia hupeana)、花曲柳(Fraxinus rhynchophylla)、君迁子(Cerasus serrulata)、山合欢(Albizia kalkora)和鹅耳枥(Carpinus turczaninowii); B1类(建群种) 2种: 栓皮栎和麻栎(Quercus acutissima), B2类(伴生种) 6种: 水榆花楸(Sorbus alnifolia)、槲树(Quercus dentata)、山樱花(Diospyros lotus)、小叶朴(Celtis bungeana)、大叶朴(Celtis koraiensis)和三桠乌药(Lindera obtusiloba)。  相似文献

将分属12个科的17种宿主植物分别种植于石灰土和紫色土上,120 d后收获取样,通过CTAB法提取共生菌根内AM真菌的DNA,用特异引物U1/U2扩增编码核糖体28S大亚基的rDNA部分序列,并利用聚丙烯酰胺凝胶电泳(PAGE)和银染显影技术显色, 比较了石灰土和紫色土上草本植物根系内丛枝菌根真菌群落的物种多样性.结果表明:生长于石灰土上的17种宿主植物根系内共发现AM真菌条带29种,平均每种宿主植物内8.29种;生长于紫色土上的17种宿主植物根系内发现条带24种,平均每种宿主植物内9.47种.根系内的AM真菌条带包括特异条带和共有条带.经聚类分析发现,AM真菌对宿主植物的侵染有一定的科属专一性,且受土壤环境等外在条件的影响.同时探讨了AM真菌应用于石灰岩地区生态恢复的可能性.  相似文献

从土壤的理化因子出发,利用综合评价的方法,对渝东地区城口县坪坝区大巴山南坡山地黄壤的土壤肥力变化特征与地上植被类型之间的关系进行了研究.结果表明,马尾松林、杉木林、落叶栎类林、茶树林和常绿阔叶林下土壤肥力的综合指标值分别为0.1256、0.2085、0.351、0.2479、0.9329.阔叶林(落叶栎类林、茶树林和常绿阔叶林)下山地黄壤的土壤肥力综合指标值均大于针叶林(马尾松林、杉木林)下山地黄壤的土壤肥力综合指标值.即使在同一植被类型下,由于建群种的不同,其土壤肥力综合指标值也不相同.根据植物群落演替的过程可以断定土壤的发育与植物群落演替是两个密不可分、相辅相成的过程.  相似文献

对曲阜孔林土壤动物进行了调查研究。共获标本5456只,隶属于5门11纲24目,其中优势类群3类,常见类群8类,占总捕量的95.12%.4种生境土壤动物种类组成极为相似,在垂直分布上具有明显的表聚性。在多样性分析中,H、E指标在各生境表现为杂木林>侧柏林>麻栎林>林缘草地;在整个孔林中大型土壤动物H、E远大于中小型土壤动物.  相似文献

The genetic structure of the locally threatened cyprinid Osteochilus melanopleurus, collected from four locations (Gerik, Manong, Bernam and Kahang) in Peninsular Malaysia River systems, was assessed based on partial sequences of the control region (D-loop) mitochondrial DNA. A partial D-loop of 364 base pair gene was extracted from 88 individuals through PCR amplification and direct sequencing. The results show that haplotype diversity (hd = 0.7956–0.9485) was high, with low nucleotide (π = 0.0045–0.0080) variation for each of the populations examined. A molecular variance analysis (AMOVA) revealed few differences between the four populations. A Tajima D test and Fu Fs test were used to investigate the neutrality of each population, revealing that the all examined populations might experience recent population expansion or the bottleneck effect. A phylogeny tree and mismatch distribution analysis provided further evidence that three of four populations had recently expanded, as well as indicating that the Kahang population had in contrast remained stable. Overall, this study concludes that the populations of O. melanopleurus distributed throughout the river systems of Peninsular Malaysia probably originated from a single ancestral source during the last glaciation period, and should therefore be treated as a single evolutionary unit in any conservation efforts.  相似文献

We investigated the molecular basis for the remarkably different survival outcomes of mice expressing different alloforms of the pro-apoptotic serine protease granzyme B to mouse cytomegalovirus infection. Whereas C57BL/6 mice homozygous for granzyme BP (GzmBP/P) raise cytotoxic T lymphocytes that efficiently kill infected cells, those of C57BL/6 mice congenic for the outbred allele (GzmBW/W) fail to kill MCMV-infected cells and died from uncontrolled hepatocyte infection and acute liver failure. We identified subtle differences in how GzmBP and GzmBW activate cell death signalling - both alloforms predominantly activated pro-caspases directly, and cleaved pro-apoptotic Bid poorly. Consequently, neither alloform initiated mitochondrial outer membrane permeabilization, or was blocked by Bcl-2, Bcl-XL or co-expression of MCMV proteins M38.5/M41.1, which together stabilize mitochondria by sequestering Bak/Bax. Remarkably, mass spectrometric analysis of proteins from MCMV-infected primary mouse embryonic fibroblasts identified 13 cleavage sites in nine viral proteins (M18, M25, M28, M45, M80, M98, M102, M155, M164) that were cleaved >20-fold more efficiently by either GzmBP or GzmBW. Notably, M18, M28, M45, M80, M98, M102 and M164 were cleaved 20- >100-fold more efficiently by GzmBW, and so, would persist in infected cells targeted by CTLs from GzmBP/P mice. Conversely, M155 was cleaved >100-fold more efficiently by GzmBP, and would persist in cells targeted by CTLs of GzmBW/W mice. M25 was cleaved efficiently by both proteases, but at different sites. We conclude that different susceptibility to MCMV does not result from skewed endogenous cell death pathways, but rather, to as yet uncharacterised MCMV-intrinsic pathways that ultimately inhibit granzyme B-induced cell death.  相似文献

The DAD1 (defender against apoptotic cell death) gene is a negative regulator of programmed cell death. It plays important roles in plant growth, development, environmental adaptation, and aging. We examined whether and how the expression of DAD1 gene is affected by the promoter changes in the allotetraploid upland cotton (Gossypium hirsutum L.). We compared the expression status of the genes in the leaves of an elite cultivar and a wild variety of G. hirsutum, and cloned and analyzed a 1.3-kb promoter region of the genes in two cotton types. Results revealed that the expression of a subgenomic homoeolog of DAD1 (DAD1 At ) was significantly upregulated in leaves of the cultivated cotton when compared with that in a wild cotton. In addition, we identified 18 variations, including 14 single nucleotide polymorphisms and four indels between the promoters of the two cotton types. We detected at least 10 of 18 variations in the promoters related to possible cis-regulatory elements. Further transient tobacco assays using heterogeneous DAD1 At promoter::GUS fusion showed that the activity of the DAD1 At promoter in the cultivated cotton was stronger than that in the wild cotton, which suggested a dynamic change in the DAD1 At promoter during genetic divergence of the two cottons. This study provides an example of how gene expression could be affected by specific changes in the gene promoter.  相似文献

关于长尾姬鼠(Apodemus orestes)的分类地位,一直没有确定。有的认为它是中华姬鼠(A.draco)的一个亚种,而有的认为是一个独立的种。分别从成体长尾姬鼠、中华姬鼠和高山姬鼠(A.chevrieri)(对照)各5只的胡须、头部、背部、腹部、前肢取毛样,清洗和处理后,在倒置显微镜下观察,用目镜测微尺分别测量和计算出其5个部位毛发的毛髓质指数。结果表明:长尾姬鼠与中华姬鼠5个部位及混合毛发的毛髓质指数无显著差异;二者5个部位及混合毛发的毛髓质指数与高山姬鼠均有显著差异。不支持长尾姬鼠作为一个独立种的观点。  相似文献
