病毒病对植物的危害在近些年日益严重,培育抗病毒转基因植物是众多防治方法中较为有效的一种。转基因植物在不同机制介导下会产生抗性,尤其是病毒基因介导的抗性试验最为普遍。众多研究证明,抗性受多种因素作用且表现为不同水平。对插入序列来源、转录后hpRNA长度、茎环比和拷贝数等多个因素在近些年的研究进展进行了综述,重点对各自在转基因植物抗性产生中的特点及影响程度进行了分析,并对其他因素进行了展望。  相似文献

In the present study, separation of l-lysine from dilute aqueous solution by solid-phase extraction based on molecular imprinting technique using a polar porogen was investigated. l-Lysine imprinted polymer (LLIP) was prepared by free radical solution polymerization of methacrylic acid and ethylene glycol dimethacrylate as functional and cross-linking monomers, in the presence of l-lysine as an imprint molecule, mixture of water and methanol as solvent and AIBN as an initiator. Non-imprinted polymer (NIP) as control was also prepared by the same procedure in the absence of template molecules. LLIP particles were applied to determine the optimum operational condition for l-lysine separation from dilute aqueous solution. In adsorption step, optimum pH and retention time were 7.8 and 90 min, while corresponding values in extraction step were 12 and 50 min, respectively. l and d-Lysine recovery by LLIP at optimum condition were found to be 96 and 58% with corresponding distribution coefficients of 8000 and 460, respectively. The retention capacity of LLIP was 27.26 mg l-lys/g of polymer at optimum condition.  相似文献

Rapamycin is a 31-member ring macrolide produced by Streptomyces hygroscopicus and has many applications in clinical medicine. In the present work, several protoplasts-related techniques including protoplasts mutation, intraspecies and interspecies protoplasts fusion were tried to improve the rapamycin productivity in S. hygroscopicus. Although mutation and fusion of different protoplasts of S. hygroscopicus did not improve the productivity of rapamycin significantly, the interspecies fusion of protoplasts of S. hygroscopicus D7-804 and Streptomyces erythreus ZJU325 could have brought about one high-yield (345 mg/L) rapamycin producer with 23.6% higher than that of the parental strain. Then, with seven mutants of S. hygroscopicus with different features and rapamycin productivities as the parental strains, only one-round genome shuffling has generated a high-yield rapamycin-producing strain with an outstanding yield of 445 mg/L. The systematic research of protoplast-related techniques has established an applicable way to generate high-yield strains from original microorganisms which can only produce low amount of expected natural products, without information of target gene clusters and gene sequences.  相似文献

The purpose of this study was to determine the releasing behavior of copper in municipal solid waste (MSW) in landfill with respect to refuse and leachate as an inseparable system. Two simulated bioreactor landfills, one with leachate recirculation and the other without, were operated in room temperature for 320 days. Copper in refuse showed behaviors of staggered migration and retention, which corresponded with the degradation process of landfill obviously. The significant different amounts of Cu2+ leached out from refuse into leachate of two landfills were 24.74 mg and 118.53 mg after 320 days’ operation, respectively. It also reflected the releasing behavior of copper in landfill refuse at different stage accordingly. The results confirmed that the refuse in landfill had high potential of secondary pollution after closure.  相似文献

Polysaccharides production from Poria cocos was carried out using aqueous NaOH with the assistance of ultrasonic. Experimental design was used to investigate the effect of three parameters (extraction time, extraction concentration of NaOH, and ratio of aqueous NaOH to raw material) on polysaccharides yields. The ranges of the factors investigated were 1–3 min for extraction time (X1), 0.5–1.0 mol/L for extraction concentration of NaOH (X2), and 30–50 for ratio of aqueous NaOH to raw material (X3). The statistical analysis of the experiment indicated that extraction concentration of NaOH had significant effect on P. cocos polysaccharides yields. The central composite design showed that polynomial regression models were in good agreement with the experimental results with the coefficients of determination of 0.9935 for P. cocos polysaccharides yield. The optimal condition for P. cocos polysaccharides yield within the experimental range of the variables studied was at 2.44 min, 0.789 mol/L, and 53.0. At this condition, the predicted yield of polysaccharides extracted was 82.3%.  相似文献

转氨酶(ω-transaminase,ω-TA)作为一种天然的生物催化剂,在手性胺类化合物的合成中具有较好的应用前景。但ω-TA在催化非天然底物的反应过程中存在稳定性差、活性低的缺陷,大大限制了ω-TA的应用。为改善此缺陷,针对来源于土曲霉(Aspergillus terreus)的(R)-ω-TA(At TA),采用基于分子动力学模拟的计算机辅助设计与随机突变、组合突变相结合的策略进行酶的热稳定性改造,获得了热稳定性与活性同步提高的最佳突变酶At TA-E104D/A246V/R266Q (M3)。与At TA野生酶(wild-type, WT)相比,M3的半衰期t1/2 (35℃)由17.8 min提升至102.7 min,提升了4.8倍,半失活温度T5010比WT (38.1℃)提高2.2℃。最佳突变酶M3对丙酮酸和1-(R)-苯乙胺的催化效率分别是野生酶的1.59倍和1.56倍。分子动力学模拟与分子对接结果表明,分子内氢键与疏水相互作用的增加所导致α-螺旋的加固稳定是酶热稳定性提升的主要原因;底物分子与结合口袋氨...  相似文献

Coarse woody debris (CWD) characteristics are expected to reflect forest stand features. Few studies evaluated logging-induced stand characteristics of secondary coniferous forests by quantifying the quality and quantity in CWD. After selective logging, the form of secondary forest of Pinus tabulaeformis in the Qinling Mountains is inferior and the regeneration is poor. We measured the CWD characteristics of the forest which had an average CWD biomass amount of 12.56 t hm−2, and was predominated by abundant logs (65.68%), followed by snags (33.13%). The CWD biomass of P. tabulaeformis and Toxicodendron vernicifluum was significantly higher than that of other species, which took up 85.51% of the total. Although there was no significant difference among different diameter sizes (P > 0.05), the CWD biomass of diameter 30–40 cm occupied 46.26% of the total (5.81 t hm−2). Similarly, the CWD biomass of decay class I and II accounted for 39.89% (5.01 t hm−2) and 33.04% (4.15 t hm−2) of the total CWD biomass respectively, despite no significant difference among those 5 decay classes (P > 0.05). The results indicated that the combination of young forest developmental stage caused by past selective logging and natural and anthropogenic disturbances such as strong wind, tapping lacquer, firewood collection, and illegal tree felling played a crucial role in distribution characteristics of CWD in this secondary forest of P. tabulaeformis.  相似文献
