BACKGROUND AND AIMS: In spatially heterogeneous environments, a trade-off between seedling survival and relative growth rate may promote the coexistence of plant species. In temperate forests, however, little support for this hypothesis has been found under field conditions, as compared with shade-house experiments. Performance trade-offs were examined over a large resource gradient in a temperate hardwood forest. METHODS: The relationship between seedling survival and seedling relative growth rate in mass (RGR(M)) or height (RGR(H)) was examined at three levels of canopy cover (forest understorey, FU; small gap, SG; and large gap, LG) and at two microsites within each level of canopy cover (presence or absence of leaf litter) for five deciduous broad-leaved tree species with different seed sizes. KEY RESULTS: Within each species, both RGR(M) and RGR(H) usually increased with increasing light levels (in the order FU < SG < LG), whereas little difference was observed based on the presence or absence of litter. Seedling survival in FU was negatively correlated with both RGR(M) and RGR(H) in both LG and SG. The trade-off between high-light growth and low-light survival was more evident in the relationship with LG as compared with SG. An intraspecific trade-off between survival and RGR was observed along environmental gradients in Acer mono, whereas seedlings of Betula platyphylla var. japonica survived and grew better in LG. CONCLUSIONS: The results presented here strongly support the idea of light gradient partitioning (i.e. species coexistence) in spatially heterogeneous light environments in temperate forests, and that further species diversity would be promoted by increased spatial heterogeneity. The intraspecific trade-off between survival and RGR in Acer suggests that it has broad habitat requirements, whereas Betula has narrow habitat requirements and specializes in high-light environments.  相似文献

Transgenic hybrid aspen (Populus tremula L. x P. tremuloides Michx.) plants expressing a high-isoelectric-point superoxide dismutase (hipI-SOD) gene in antisense orientation were generated to investigate its function. Immunolocalization studies showed the enzyme to be localized extracellularly, in the secondary cell wall of xylem vessels and phloem fibers. The antisense lines of hipI-SOD exhibited a distinct phenotype; growth rate was reduced, stems were thinner and leaves smaller than in wild-type (WT) plants. The abundance of hipI-SOD was reduced in the bark and xylem of plants from these antisense lines. The vascular tissue of transgenic lines became lignified earlier than in WT plants and also showed an increased accumulation of reactive oxygen species (ROS). Xylem fibers and vessels were shorter and thinner in the transgenic lines than in WT plants. The total phenolic content was enhanced in the antisense lines. Furthermore, microarray analysis indicated that several enzymes involved in cell signaling, lignin biosynthesis and stress responses were upregulated in apical vascular tissues of transgenic plants. The upregulation of selected genes involved in lignin biosynthesis was also verified by real-time PCR. The results suggest that, in the transgenic plants, a premature transition into maturation occurs and the process is discussed in terms of the effects of increased accumulation of ROS due to reduced expression of hipI-SOD during development and differentiation.  相似文献

Background and Aims

Silicon (Si) has been shown to ameliorate the negative influence of cadmium (Cd) on plant growth and development. However, the mechanism of this phenomenon is not fully understood. Here we describe the effect of Si on growth, and uptake and subcellular distribution of Cd in maize plants in relation to the development of root tissues.

Methods

Young maize plants (Zea mays) were cultivated for 10 d hydroponically with 5 or 50 µm Cd and/or 5 mm Si. Growth parameters and the concentrations of Cd and Si were determined in root and shoot by atomic absorption spectrometry or inductively coupled plasma mass spectroscopy. The development of apoplasmic barriers (Casparian bands and suberin lamellae) and vascular tissues in roots were analysed, and the influence of Si on apoplasmic and symplasmic distribution of 109Cd applied at 34 nm was investigated between root and shoot.

Key Results

Si stimulated the growth of young maize plants exposed to Cd and influenced the development of Casparian bands and suberin lamellae as well as vascular tissues in root. Si did not affect the distribution of apoplasmic and symplasmic Cd in maize roots, but considerably decreased symplasmic and increased apoplasmic concentration of Cd in maize shoots.

Conclusions

Differences in Cd uptake of roots and shoots are probably related to the development of apoplasmic barriers and maturation of vascular tissues in roots. Alleviation of Cd toxicity by Si might be attributed to enhanced binding of Cd to the apoplasmic fraction in maize shoots.  相似文献

为了解NAC类转录因子与桃果实内果皮发育木质化的关系,本文以桃内果皮为试材,采用同源基因克隆法获得了两个NAC类转录因子的全长编码区,分别命名为PpNST1和PpSND1。序列分析表明2个基因编码区全长均为1188bp,可编码396个氨基酸,其预测蛋白均包含1个NAC结构域和2个特征结构域(LP—box和WQ-box)。氨基酸序列系统进化分析显示PpNST1和PpSND1与苹果、葡萄、毛果杨和拟南芥等参与调控次生细胞壁形成和木质化的NST/SND1类转录因子具有较高同源性。荧光实时定量PCR分析表明,PpNST1和PpSND1的表达水平随内果皮的木质化程度加强而不断升高,分别于盛花期后59和52d达到最高表达量,且它们在内果皮中的表达水平远高于中果皮。对PpNST1和PpSND1下游可能的作用靶标进行进一步的表达分析表明,PpNST1和PpSND1可能在内果皮发育木质化过程中扮演重要角色。  相似文献

Plants, animals and some fungi undergo processes of cell specialization such that specific groups of cells are adapted to carry out particular functions. One of the more remarkable examples of cellular development in higher plants is the formation of water-conducting cells that are capable of supporting a column of water from the roots to tens of metres in the air for some trees. The Zinnia mesophyll cell system is a remarkable tool with which to study this entire developmental pathway in vitro. We have recently applied an RNA fingerprinting technology, to allow the detection of DNA fragments derived from RNA using cDNA synthesis and subsequent PCR-amplified fragment length polymorphisms (cDNA-AFLP), to systematically characterize hundreds of the genes involved in the process of tracheary element formation. Building hoops of secondary wall material is the key structural event in forming functional tracheary elements and we have identified over 50 partial sequences related to cell walls out of 600 differentially expressed cDNA fragments. The Zinnia system is an engine of gene discovery which is allowing us to identify and characterize candidate genes involved in cell wall biosynthesis and assembly.  相似文献

Symptoms of infection by the fungal pathogen Gaeumannomyces graminis were studied in roots of wheat seedlings grown in sand culture in order to assess resistance. Three sets of wheat were studied, 10 cultivars previously thought to differ in infection, and 44 and 54 segregating families from two crosses between four of these wheats, Aus1080 × Condor and Termu89–72 × Bayonet respectively. The isolates of G. graminis employed had previously been selected for differences in virulence. The interpretive value of symptoms of infection were considered, by comparison with direct measurement of the spread of hyphae from a fixed point of inoculation, and by repeatability of measurements over experiments. Two symptoms of infection, cortical browning and stelar blackening, are thought to be host mechanisms to resist hyphal invasion and were suppressed by rapid colonisation of root tissues. A third symptom, general discolouring of tissue, appeared to be a necrotic symptom of aggressive colonisation. Since only root segments distal to the point of infection were studied, stelar blackening appeared the least flexible measure of severity of infection. Cortical browning appeared to be influenced by nutritive status of seedlings, and was negatively associated with the incidence of necrotic discolorations. Wheats differed consistently and mostly in cortical browning, and while also differing in extent of necrotic discoloration, difficulty in quantifying the latter character limited its value in assessing resistance to G. graminis in this study.  相似文献

Water content, peroxidase activity and isoperoxidases, phenylalanine ammonia-lyase activity and phenolic content were comparatively analyzed in tissues of normal and vitreous plants cultured in vitro. The release of ethylene in flask atmospheres by normal and vitrifying plants was also measured. On the basis of the results, it is hypothesized that vitrification results from a burst of ethylene controlled by the peroxidase-IAA-oxidase system. An initiating stress (e.g. excess of cytokinins or of NH4+ ions) would mediate the enhancement of the activity of soluble and membrane-bound peroxidases through a rapid modification of the phenolic level. The excess of ethylene in the atmosphere of stressed plants would retroinhibit its own biosynthesis and as a consequence decrease the activities of PAL and acidic peroxidases, thus hindering lignification processes. A parallel decrease in cellulose synthesis due to a diverted conversion of sugars to amino acids is expected (from data in the literature). Deficiency of both cellulose and lignin would allow more water uptake due to reduced wall pressure and bring about the hyperhydric malformations.  相似文献

Lignin is a major structural polymer of secondarily thickended plant vascular tissue and fibres, imparting mechanical strength to stems and trunks and hydrophobicity to conducting vessels. Constitutive expression of a lucerne caffeic acid 3-O-methyltransferase antisense RNA in transgenic tobacco leads to a significant reduction in lignin content, particularly in the younger parts of the stems, without apparent alterations in lignin monomer composition. These observations open up the possibility of genetically manipulating plants with reduced lignin for improved processing and biomass digestibility.  相似文献

Vitrification of internodes of carnation was brought about by culturing in liquid medium. Cell wall extensibility of these internodes was kinetically followed in comparison to that of normal plants using the constant stress method. Liquid culture induced increased immediate and total deformation capacities of the walls from the second day. Measurements indicated that these deformation capacities involved plastic properties rather than elastic ones. These changes were paralleled by decreased relative levels of cellulose and lignin.  相似文献
