Despite recent progress in the molecular characterization of high-conductance Ca2+-activated K+ (maxi-K) channels, the molecular identities of intermediate conductance Ca2+-activated K+ channels, including that of mature erythrocytes, remains unknown. We have used various peptide toxins to characterize the intermediate conductance Ca2+-activated K+ channels (Gardos pathway) of human and rabbit red cells. With studies on K+ transport and on binding of 125I-charybdotoxin (ChTX) and 125I-kaliotoxin (KTX) binding in red cells, we provide evidence for the distinct nature of the red cell Gardos channel among described Ca2+-activated K+ channels based on (i) the characteristic inhibition and binding patterns produced by ChTX analogues, iberiotoxin (IbTX) and IbTX-like ChTX mutants, and KTX (1–37 and 1–38 variants); (ii) the presence of some properties heretofore attributed only to voltage-gated channels, including inhibition of K transport by margatoxin (MgTX) and by stichodactyla toxin (StK); (iii) and the ability of scyllatoxin (ScyTX) and apamin to displace bound 125I-charybdotoxin, a novel property for K+ channels. Chaetoceros muelleri (Schütt) was cultured on a thermal gradient plate, subjected to two media types with a range of specific
conductances, and evaluated for growth and neutral lipid accumulation. Growth was measured directly by daily changes in cell
numbers and indirectly by changes in optical density at 750 nm. C. muelleri exhibited a growth rate of at least two doublings
day-1 over broad temperature (20 to 35 °C) and conductance ranges (10 to over 60 mS cm-1) and the optimum growth rate approached
4.0 doublings day-1 at 30 °C and a conductance of 25 mS cm-1. Intracellular neutral lipid storage was evaluated with fluorometry
and epifluorescent microscopy using the fluorochrome Nile Red. Gravimetric analysis revealed a total lipid content in nitrogen-depleted
cultures of C. muelleri of over 400 mg L-1, five to seven times that observed in nitrogen-replete cultures. Based on its high
growth rate, tolerance to a broad range of temperatures and specific conductances, and large quantity of intracellular lipid,
C. muelleri may have potential for exploitation as a renewable precursor to liquid fuels or as a lipid source.

Singuonensis)、奥利亚罗非鱼(Sarotherodon aurea)、 鳙(Aristichthys nobilis R.)和团头鲂(Megalobrama amblycephala Yin.)红细胞的DNA含量,四种鱼与鸡红细胞DNA含量的比值分别为1·65、0.96、0.91、1·15,以鸡红细胞DNA含量2.3pg/N计算,四种鱼二倍体体细胞的 DNA含量分别为3.80 pg/N, 2. 22pg/N、2. 08pg/N、 2. 66pg/N。另外,四种鱼外周血红细胞分别有26%、 26%、 23%、 24%的细胞处于 DNA合成期(S),DNA合成后期(G2)和分裂期(M),这表明这四种鱼类的外周血红细胞并非失去分裂能力的特化细胞群,它们表现了较强的细胞周期现象。这有可能是这几种硬骨鱼类外周血仍具有造血功能的缘故。 相似文献

Synopsis Diet, habitat use, diel and seasonal activity and a number of population parameters were studied on ruffe,Gymnocephalus cernuus, introduced to Mildevatn, western Norway. This lake is sited outside the natural range of the ruffe and has a lower fish
diversity and a different fish species composition than within its native range. From June through September the ruffe was
planktivorous and mainly caught at 4 to 6 m depth in the benthic zone. At other times of year ruffe was feeding on zoobenthos
and caught deeper in the benthic zone. Ruffe was mainly day active. Zooplankton feeding during summer is the clearest difference
compared to ruffe populations living within its natural range. Presence of large zooplankton organisms available for ruffe
is suggested as the main reason for the difference found in food choice. The availability of large zooplankton is probably
due to community structure caused by a predator and lack of interspecific competition for zooplankton in the deeper parts
of the lake. Piscivorous brown trout.Salmo trutta, restrict the habitat of threespined stickleback,Gasterosteus aculeatus, to the zone of littoral vegetation, allowing high densities of larger zooplankton species likeBythotrephes longimanus to be present in the lake. - 1. (1) VO3− combines with high affinity to the Ca2+-ATPase and fully inhibits Ca2+-ATPase and Ca2+-phosphatase activities. Inhibition is associated with a parallel decrease in the steady-state level of the Ca2+-dependent phosphoenzyme.
- 2. (2) VO3− blocks hydrolysis of ATP at the catalytic site. The sites for VO3− also exhibit negative interactions in affinity with the regulatory sites for ATP of the Ca2+-ATPase.
- 3. (3) The sites for VO3− show positive interactions in affinity with sites for Mg2+ and K+. This accounts for the dependence on Mg2+ and K+ of the inhibition by VO3−. Although, with less effectiveness, Na+ substitutes for K+ whereas Li+ does not. The apparent affinities for Mg2+ and K+ for inhibition by VO3− seem to be less than those for activation of the Ca2+-ATPase.
- 4. (4) Inhibition by VO3− is independent of Ca2+ at concentrations up to 50 μM. Higher concentrations of Ca2+ lead to a progressive release of the inhibitory effect of VO3−.
