Despite the improvement in acute myeloid leukemia (AML) treatments, most patients had a poor prognosis and suffered from chemoresistance and disease relapse. Therefore, there is an urgent need for elucidation of mechanism(s) underlying drug resistance in AML. In the present study, we found that AML cells showed less susceptibility to adriamycin (ADR) in the presence of hypoxia, while inhibition of hypoxia‐inducible factor 1α (HIF‐1α) by CdCl2 can make AML cells re‐susceptibile to ADR even under hypoxia. Moreover, HIF‐1α is overexpressed and plays an important role in ADR‐resistance maintenance in resistant AML cells. We further found hypoxia or induction of HIF‐1α can significantly upregulate yes‐associated protein (YAP) expression in AML cells, and resistant cells express a high level of YAP. Finally, we found that YAP may not only enhance HIF‐1α stability but also promote HIF‐1α's activity on the target gene pyruvate kinase M2. In conclusion, our data indicate that HIF‐1α or YAP may represent a therapeutic target for overcoming resistance toward adriamycin‐based chemotherapy in AML.  相似文献

ObjectiveThe objective was to investigate the anti-inflammatory effects of salidroside through the PI3K/Akt signaling pathway and its protective effects on acute hypoxia-induced myocardial injury in rats.MethodsA total of 24 healthy Sprague-Dawley male rats were selected as the experimental subjects. All rats were divided into 4 groups by using the random number table method, with 6 rats in each group. The groups included the normal control group, the salidroside group, the hypobaric hypoxia group, and the hypobaric hypoxia + salidroside group. Rats in the salidroside group were fed in the original animal laboratory and were intragastrically administered with salidroside every morning at a dosage of 35 mg/kg. Rats in the normal control group were intragastrically administered with an equal dosage of saline. Rats in the hypobaric hypoxia + salidroside group were intragastrically administered with salidroside every morning at a dosage of 35 mg/kg, who were fed in the hypoxic experiment module for animals. The altitude was increased to 4000 m, and the rats were kept in the module for 24 h. Rats in the hypobaric hypoxia group were intragastrically administered with an equal dosage of saline in the same environment, and the altitude was increased to 4000 m after administration. Parameters of blood gas analysis, histopathological changes in cardiac tissues, cardiac indexes, and inflammatory factors IL-6 and TNF-α levels of rats in groups were compared.Results1. The cardiac indexes of rats in groups were compared. The differences between the hypobaric hypoxia group and the hypobaric hypoxia + salidroside group were statistically significant (P < 0.05). 2. The results of blood gas analysis of rats in groups were compared. The differences between the hypobaric hypoxia group and the hypobaric hypoxia + salidroside group were significantly different (P < 0.05). 3. In the hypobaric hypoxia group, the myocardial cells of rats were arranged disorderly and shaped differently, with cases such as edema, degeneration, necrosis, nucleus pyknosis, and massive infiltration of inflammatory cells. In the hypobaric hypoxia + salidroside group, the above-mentioned pathological changes in myocardial cells were relieved. 4. Compared with the hypobaric hypoxia group, in the hypobaric hypoxia + salidroside group, the concentrations of IL-6 and TNF-α in rats decreased apparently, and the differences were statistically significant (P < 0.05).ConclusionSalidroside had the repairing and protective effects on the hypobaric hypoxia-induced myocardial injuries in rats. The application of salidroside could reduce the inflammatory responses of rats with hypobaric hypoxia-induced myocardial injuries through PI3K/Akt signaling pathway, thereby protecting the myocardial cells.  相似文献

目的研究儿童反复呼吸道感染与患儿肠道微生态平衡紊乱的关系。方法选择102例反复呼吸道感染患儿为研究组,167例急性肺炎患儿为肺炎对照组,142例健康体检儿童为正常对照组。采用16S rRNA荧光定量PCR检测3组对象肠道双歧杆菌及大肠埃希菌数量,计算B/E值,并比较3组研究对象细胞免疫功能。结果正常对照组、肺炎对照组以及研究组儿童肠道双歧杆菌数量依次降低,大肠埃希菌依次增多,B/E值依次降低,差异均有统计学意义(均P0.05)。正常对照组、肺炎对照组以及研究组儿童血液中CD3~+、CD4~+细胞水平以及CD4~+/CD8~+依次降低,CD8~+细胞水平依次增高,差异均有统计学意义(均P0.05)。结论儿童反复呼吸道感染与肠道微生态失衡具有一定相关性,肠道微生态稳态的维持可为反复呼吸道感染的防治提供新思路。  相似文献

目的分析急性脑梗死并发肺部感染患者病原菌分布、病原菌耐药性、患者用药特征及其危险因素。方法选取2015年3月至2017年3月在我院急诊科进行住院治疗,且年龄60岁的急性脑梗死并发肺部感染患者60例为研究对象,对患者的一般资料及病原菌分布情况、耐药情况和患者用药情况进行分析。结果 60例患者中有30例出现肺部感染,感染率为50.00%。肺部感染患者中死亡4例。肺部感染患者痰液中共培养出28株病原菌,其中革兰阴性菌21株,革兰阳性菌5株,真菌2株。30例肺部感染患者共使用6种抗感染药物,其中哌拉西林/舒巴坦的使用频率最高,其次为依替米星及美罗培南。Logistic回归分析显示,病原菌分布、耐药情况和患者用药情况与患者的预后存在相关性。结论急性脑梗死并发肺部感染患者的病原菌分布、耐药情况及患者用药情况与患者的预后密切相关,应针对上述因素给予患者合理的治疗。  相似文献

Excessive pulmonary inflammatory response is critical in the development of acute lung injury (ALI). Previously, microRNAs (miRNAs) have been recognized as an important regulator of inflammation in various diseases. However, the effects and mechanisms of miRNAs on inflammatory response in ALI remain unclear. Herein, we tried to screen miRNAs in the processes of ALI and elucidate the potential mechanism. Using a microarray assay, microRNA let-7e (let-7e) was chose as our target for its reported suppressive roles in several inflammatory diseases. Down-regulation of let-7e by antagomiR-let-7e injection attenuated LPS-induced acute lung injury. We also found that antagomiR-let-7e could obviously improve the survival rate in ALI mice. Moreover, antagomiR-let-7e treatment reduced the production of proinflammatory cytokines (i.e., TNF-α, IL-1β and IL-6) in bronchoalveolar lavage fluid (BALF) of LPS-induced ALI mice. Luciferase reporter assays confirmed that suppressor of cytokine signaling 1 (SOCS1), a powerful attenuator of nuclear factor kappa B (NF-κB) signaling pathway, was directly targeted and suppressed by let-7e in RAW264.7 cells. In addition, it was further observed that SOCS1 was down-regulated, and inversely correlated with let-7e expression levels in lung tissues of ALI mice. Finally, down-regulation of let-7e suppressed the activation of NF-κB pathway, as evidenced by the reduction of p-IκBα, and nuclear p-p65 expressions in ALI mice. Collectively, our findings indicate that let-7e antagomir protects mice against LPS-induced lung injury via repressing the pulmonary inflammation though regulation of SOCS1/NF-κB pathway, and let-7e may act as a potential therapeutic target for ALI.  相似文献

Acute leukemia is a hematological malignant tumor. Long non-coding RNA urothelial cancer-associated 1 (UCA1) is involved in the chemo-resistance of diverse cancers, but it is unclear whether UCA1 is associated with the sensitivity of acute leukemia cells to daunorubicin (DNR). DNR (100 nM) was selected for functional analysis. The viability, cell cycle progression, apoptosis, and invasion of treated acute leukemia cells (HL-60 and U-937) were evaluated by cell counting kit-8 (CCK-8) assay, flow cytometry assay, or transwell assay. Protein levels were detected with Western blot analysis. Expression patterns of UCA1 and miR-613 were assessed by quantitative real-time polymerase chain reaction (qRT-PCR). The relationship between UCA1 and microRNA-613 (miR-613) was verified by dual-luciferase reporter assay. We observed that UCA1 expression was elevated in HL-60 and U-937cells. DNR constrained viability, cell cycle progression, invasion, and facilitated apoptosis of HL-60 and U-937 cells in a dose-dependent manner, but these impacts mediated by DNR were reverted after UCA1 overexpression. MiR-613 was down-regulated in HL-60 and U-937 cells, and UCA1 was verified as a miR-613 sponge. MiR-613 inhibitor reversed DNR treatment-mediated effects on viability, cell cycle progression, apoptosis, and invasion of HL-60 and U-937 cells, but these impacts mediated by miR-613 inhibitor were counteracted after UCA1 inhibition. Notably, the inactivation of the PI3K/AKT pathway caused by DNR treatment was reversed after miR-613 inhibitor introduction, but this influence mediated by miR-613 inhibitor was offset after UCA1 knockdown. In conclusion, UCA1 up-regulation facilitated the resistance of acute leukemia cells to DNR via the PI3K/AKT pathway by sponging miR-613.  相似文献

急性出血性结膜炎(Acute hemorrhagic conjunctivitis,AHC)是目前人类最常见的眼病之一,柯萨奇病毒A组24型变异株(Coxsackievirus A24 variant,CV-A24v)是近年来报道引起该病的主要病原体。本研究选取10株来自江西省2010年AHC暴发疫情的CV-A24v,采用特异性引物扩增并测定其全基因组序列。对该10条CV-A24v的全基因组序列进行系统发育分析以及重组分析,计算本研究测定的江西10条以及GenBank中所有22条CV-A24v的全基因组序列的氨基酸置换熵值,并预测其正向选择位点。结果表明,在江西10条CV-A24v基因组序列中未检测到重组。基于全基因组序列构建的最大似然树表明江西10株CV-A24v属于GIV基因型,且分处于两条传播链。对上述32条CV-A24v序列的氨基酸置换熵值计算,共得到25个易突变位点(熵值>0.6),易突变概率最高的区段为2A区。基于Datamonkey中FUBAR和FEL模型分析,发现位于结构蛋白VP2区的234位氨基酸为两种模型共同获得的CV-A24v的正向选择位点。本研究分析了江西10株CV-A24v的全基因组序列特征,为CV-A24v引起的AHC防控工作提供了基础资料。  相似文献

摘要 目的:探讨高频振荡通气(HFOV)联合肺表面活性物质(PS)对治疗新生儿急性肺损伤/急性呼吸窘迫综合征(ALI/ARDS)的疗效及对肺动态顺应性的影响。方法:选择2018年1月至2020年12月我院新生儿科收治的160例ALI/ARDS患儿进行研究,按照随机数表法分为观察组和对照组,每组80例。对照组患儿给予常频通气(CMV)模式联合PS治疗,观察组患儿给予HFOV模式联合PS治疗。比较两组患儿一般治疗情况、治疗前后肺动态顺应性、动脉血氧分压(PaO2)、动脉二氧化氮分压(PaCO2)、氧合指数(OI)、血清肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-6、IL-10的变化,以及治疗期间并发症发生情况。结果:观察组胸片恢复正常时间、机械通气时间、氧暴露时间、ICU停留时间、住院时间结果均明显短于对照组(P<0.05),两组患儿病死率相比较,无统计学意义(P>0.05);治疗后12 h、24 h、48 h时,观察组肺动态顺应性及PaO2、OI结果明显高于对照组,PaCO2明显比对照组低,差异有统计学意义(P<0.05);治疗后48 h时,观察组血清TNF-α、IL-6水平均明显低于对照组,IL-10明显比对照组高,差异有统计学意义(P<0.05);两组治疗期间,呼吸机相关性肺损伤、颅内出血、气漏、呼吸道感染的总发生率比较,无统计学意义(P>0.05)。结论:HFOV联合PS治疗新生儿ALI/ARDS疗效明显,可有效改善患儿肺动态顺应性,促进血气分析指标恢复,且可降低炎症因子的表达,值得推广应用。  相似文献

摘要 目的:探讨与研究Aurora-A激酶对急性胰腺炎大鼠肺脏损伤的修复作用。方法:36只雄性SD大鼠均分为三组:对照组、模型组与Aurora-A组。对照组进行假手术操作,模型组建立急性胰腺炎模型后给予注射等量生理盐水治疗,Aurora-A组建立急性胰腺炎模型后给予阴茎背静脉注射鼠Aurora-A类因子-MLN8054 10 mg/kg治疗,记录大鼠肺脏损伤的修复情况。结果:造模过程中无大鼠死亡情况发生,模型组与Aurora-A组造模后2 w与4 w的肺组织病理评分、血清中性粒细胞弹性蛋白酶(neutrophil elastase,NE)与髓过氧化物酶(myeloperoxidase,MPO)含量、肺组织W/D、肺组织蛋白激酶B(AKT)、细胞外信号调节激酶1(ERK1)蛋白相对表达水平都高于对照组(P<0.05),Aurora-A组少于模型组(P<0.05)。结论:Aurora-A激酶在急性胰腺炎大鼠的应用能抑制Akt/ERK信号通路激活,减少血清NE与MPO的表达,从而促进肺脏损伤修复。  相似文献

目的:研究参麦注射液联合西药对急性心肌梗死患者的疗效及机制。方法:选择我院2016年3月~2019年12月收治的101例急性心肌梗死患者,根据随机数字表法分为对照组和观察组。对照组口服替格瑞洛治疗,观察组联用参麦注射液治疗,观察两组治疗的疗效及治疗前后血浆乳酸、脑钠肽(Brain natriuretic peptide,BNP)和血清降钙素原(Procalcitonin,PCT)、超敏C反应蛋白(Hypersensitive C-reactive protein,hs-CRP)水平及心功能指标变化。结果:观察组的有效率明显高于对照组(P<0.05);两组治疗前的血浆乳酸、BNP和PCT、hs-CRP水平无明显差异(P>0.05),治疗后,两组的上述指标明显降低(P<0.05),且观察组低于对照组(P<0.05);两组治疗前的左心室射血分数(Left ventricular ejection fraction,LVEF)、左心室舒张期末内径(End diastolic diameter of left ventricle,LVEDd)、CO和左心室收缩末期直径(leftventricular end systolic diameter,LVESd)无明显差异(P>0.05),治疗后,两组的上述指标明显改善(P<0.05),且观察组优于对照组(P<0.05);观察组的药物不良反应率为14.00%(7/50),与对照组的11.76%(6/51)相比无明显差异(P>0.05)。结论:参麦注射液联合替格瑞洛能减轻急性心肌梗死患者体内的炎症反应,改善心功能,可能与其机制有关。  相似文献
