Evaluation of a crown-of-thorns starfish (Acanthaster planci) control program at Grub Reef (central Great Barrier Reef)

A crown-of-thorns starfish control program was conducted at Grub Reef (central Great Barrier Reef) in an area (0.64 km²) which encompassed 53 individual patch reefs. During a two week period, 15 divers injected 3175 starfish with copper sulphate. The program was considered unsuccessful. Although starfish abundance had declined significantly after the control efforts, biological surveys indicated that a relatively large number of starfish remained. The surveys also indicated a general decline in the number of starfish along the reef perimeter, outside the control area. The total cost of the control program was $35 per starfish. These results have important implications for the implementation of future control programs and highlight the need to undertake before and after biological surveys to assess the effectiveness of the control efforts.     

Genetic variation of liverleaf (Hepatica nobilis Schreb.) in Bavaria against the background of seed transfer guidelines in forestry and restoration

The liver leaf, Hepatica nobilis (Ranunculaceae), is a perennial forest understory herb with specific habitat requirements, which occurs on calcareous soils in beech and oak forests. In Bavaria it can be found, therefore, only in four geographic regions, which are Franconia, the Franconian-Swabian Jura, the prealpine transitions region and the prealpine moraine belt with the Alps. In German forestry, provenance delineations are used since 1987. Similarly for herbs seed provenances and production areas are applied in commercial seed production and restoration. In this study we analyzed whether the genetic variation of H. nobilis reflects the geographic distribution of the species in Bavaria or the provenance delineations used for seed production.We applied AFLPs to study genetic variation within and between 24 populations of H. nobilis.Our analysis revealed high levels of genetic variation within and moderate variation between populations. Variation between seed production areas and seed provenances was low and comparable to the variation between geographic regions. Genetic variation within populations or rare fragments did not differ between production areas, provenances or geographic regions. A significant positive relation of genetic and geographic distances was present within 100 km.Pollination and seed dispersal seem mainly to happen within populations of the myrmecochorous H. nobilis, whereas long-distance dispersal is presumably occasional and random. Our study supports the relevance of dispersal traits for seed collections used in conservation. The protection of historically old beech and oak forests is exceedingly important to preserve the genetic variation of H. nobilis in Bavaria.     

Natural product pectolinarigenin exhibits potent anti-metastatic activity in colorectal carcinoma cells in vitro and in vivo

Colorectal carcinoma (CRC) is one of the most common cancers with high metastatic potential, explaining why identifying new drug candidates that inhibit tumour metastasis is an urgent need. The aim of this study was to evaluate the biological activities of pectolinarigenin (PEC, a natural flavonoid present in Cirsium chanroenicum) in CRC in vitro and in vivo and to determine its underlying mechanism of action. Here, we observed that treatment with PEC could inhibit cell viability and induce apoptosis in cancer cells in a concentration- and time-dependent manner. The occurrence of apoptosis was associated with activation of caspase-3 and Bax and decreased expression of Bcl-2. In addition, PEC markedly impaired CRC cell migration and invasion by downregulating the expression of matrix metalloproteinase (MMP-9) and phosphorylated-Stat3^Tyr705. Moreover, our studies showed that PEC inhibited abdominal metastasis models of murine colorectal cancer. In addition, histological and immunohistochemical analyses revealed a decrease in Ki67-positive cells, MMP9-positive cells and p-Stat3^Tyr705 cells upon treatment with PEC compared to control samples. Furthermore, PEC reduced the number of myeloid-derived suppressor cells (MDSCs) in the blood and tumours, which was accompanied by the increased infiltration of CD8⁺T cells in the blood. Taken together, our findings suggested that PEC could be used as a natural drug to inhibit CRC metastasis.     

Joint multiple quantitative trait loci mapping for allometries of body compositions and metabolic traits to body weights in broiler

In order to map quantitative trait loci (QTLs) for allometries of body compositions and metabolic traits in chicken, we phenotypically characterize the allometric growths of multiple body components and metabolic traits relative to BWs using joint allometric scaling models and then establish random regression models (RRMs) to fit genetic effects of markers and minor polygenes derived from the pedigree on the allometric scalings. Prior to statistically inferring the QTLs for the allometric scalings by solving the RRMs, the LASSO technique is adopted to rapidly shrink most of marker genetic effects to zero. Computer simulation analysis confirms the reliability and adaptability of the so-called LASSO-RRM mapping method. In the F₂ population constructed by multiple families, we formulate two joint allometric scaling models of body compositions and metabolic traits, in which six of nine body compositions are tested as significant, while six of eight metabolic traits are as significant. For body compositions, a total of 14 QTLs, of which 9 dominant, were detected to be associated with the allometric scalings of drumstick, fat, heart, shank, liver and spleen to BWs; while for metabolic traits, a total of 19 QTLs also including 9 dominant be responsible for the allometries of T4, IGFI, IGFII, GLC, INS, IGR to BWs. The detectable QTLs or highly linked markers can be used to regulate relative growths of the body components and metabolic traits to BWs in marker-assisted breeding of chickens.     

Reply to the first systematic response by the Global Footprint Network to criticism: A real debate finally?

We respond to a reaction of the Global Footprint Network/GFN on our 8-point criticism of the ecological footprint. We also refer to, and comment on, an associated debate in this journal between Giampietro and Saltelli, 2014a, Giampietro and Saltelli, 2014b, on the one hand, and Goldfinger et al. (2014), on the other. We conclude that criticism on the footprint is accumulating and coherent across the various studies and disciplines and among the different authors. This was the first time that Wackernagel/GFN systematically responded to our criticisms. Hence, our response contains several original elements and the resulting exchange can be seen to add value to the existing literature. It ultimately allows readers to better make up their mind about the different viewpoints on the ecological footprint.     

EPR study of iron status in human body during intensive physical activity

The iron metabolism was studied in serum blood samples collected from 26 professional sportsmen undergoing intensive physical exercises using EPR combined with hematological and biochemical laboratory tests. Only 23% of EPR spectra (n = 6) were practically normal while in the rest spectra additional abnormal absorption lines were detected. Presumably, the significant portion of new signals may be caused by different cytochromes. Moreover, the anisotropic signals with g ₁ ? 2.02; g ₂ ? 1.94 and g ₃ ? 1.86 registered in some spectra pointed to the sulfur-iron centers. There was nearly linear correlation between the concentration of Fe³⁺ in transferrin (Fe³⁺-Tf) obtained from the EPR spectra and the serum iron concentration measured by absorption photometry both for sportsmen and controls (healthy individuals and patients with different diseases). At equal serum iron concentrations the Fe³⁺-Tf level was higher in sportsmen than that in controls. The Pearson correlation coefficient (r) for Fe³⁺-Tf and serum iron values was equal to 0.89 in sportsmen versus r = 0.97 in controls. Additional new lines in serum EPR spectra of professional sportsmen prove the suitability of EPR assay for scheduled medical exams since routine biochemical and hematological tests are insufficient to discover all abnormalities in iron metabolism under intensive physical exercises.     

Low mtDNA Cytb diversity and shallow population structure of Eleutheronema tetradactylum in the East China Sea and the South China Sea

Eleutheronema tetradactylum is an economically important fish species in China water. To investigate the genetic diversity and describe population structure of it, an 1151 base pair (bp) fragment of the mitochondrial DNA Cytb sequence was analyzed in 120 individuals from four populations in the East China Sea and the South China Sea. A total of 16 haplotypes were defined by 24 variable nucleotide sites. High level of haplotype diversity and low nucleotide diversity were observed in all populations. The results of AMOVA detected that 89.44% of the genetic variation occurred within populations. Significant genetic differentiations were detected among populations (0.05097, P < 0.05), but no large-scale regional differences were detected. Analysis of neutral evolution and mismatch distribution suggested no recent population expansion happened. The present results provided new information for genetic assessment, fishery management and conservation of this species.
