双波长分光光度法测定肝素钠效价

本文介绍了在室温条件下,以天青作显色剂,用双波长分光光度法测定肝素钠的效价。本法快速准确,测定浓度0—20u/ml,相对误差在±4％以内,标准偏差0.19,变异系数2.5‰,与羊血浆法测定结果相差±4u/ml以内。     

流行性出血热地鼠肾细胞灭活疫苗人体试用细胞免疫反应观察

观察了20位志愿者在接种试验性流行性出血热(EHF)地鼠肾细胞(GHKC)双价灭活疫苗后的细胞免疫反应,并以其体液免疫反应作对照观察。用淋巴细胞转化试验测定细胞免疫水平。结果首针疫苗接种后42天和56天,特异性刺激指数(SSI)和非特异性刺激指数(NSI)均较免疫前显著增高(P_(SSI)<0.001;P_(NSI)<0.02),免疫后SSI累计阳转率为100％,NSI累计阳转率为60％,免疫后6个月二者均降低至正常水平。免疫后56天测定抗体,荧光抗体阳转率为100％;微量感染性中和试验表明,针对家鼠型病毒L99株中和抗体阳转率为95％,而针对野鼠型病毒JR株阳转率为65％。     

浙江产蝮蛇毒(Agkistrodon halys pallas venom)凝血酶样酶的性质及其对血凝和血脂的影响

从浙江产蝮蛇毒中分离出一种凝血酶样酶,该酶能明显地延长全血凝固时间,白陶土部分凝血活酶时间(KPTT),并降低了血浆中的纤维蛋白原水平,同时全血的比粘度和血浆的比粘度及血清中的胆固醇和β脂蛋白均有所下降.该酶不激活凝血因子ⅩⅢ.     

光钳操纵生物活体的动态监测技术的研究

采用摄像、录像和视屏监控系统及显色偏振装置与光钳系统耦合,从空间分辨、色分辨和时间分辨多方面改善系统品质,实现了光钳捕获与操纵生物活体的动态监测、实时记录、资料保存和屏幕再现的功能,并能测量光钳操纵细胞的位移量和由此计算操纵速度,提高了光钳的自我调整和光钳操纵细胞的精细度。本研究为激光光钳技术在细胞工程等方面的应用研究提供了行之有效的技术手段。     

染色体辐射效应的微机检测 Ⅱ断片率 微核率与细胞畸变率间的相关性检测

本文继前文后,按照设计的线性回归程序在“IBM—PC/XT”微型计算机上,进一步检测了断片率、微核率与细胞畸变率之间的相关性,肯定了微核测定法,断片测定法可以替代染色体畸变分析法。     

Changes of carp myosin subfragment-1 induced by temperature acclimation

Summary Heavy meromyosin subfragment-1 (S1) was prepared by -chymotrypsin from myosin of carp acclimated to either 10°C or 30°C for a minimum of 5 weeks. The objective of these studies was to document thermally-induced changes in the myosin molecule and to extend previous observations. Ca²⁺- and K⁺ (EDTA)-ATPase activities of cold-acclimated carp S1 were 1.1 and 0.8 mol P_i·min^-1·mg^-1, respectively, and these values did not differ significantly from those of warm-acclimated carp. The inactivation rate constant (K_D) of S1 from cold-acclimated carp was 32.1x10^-4· s^-1, compared to 13.2x10^-4·s^-1 for warm-acclimated carp. The maximum initial velocity of acto-S1 Mg²⁺-ATPase activity at pH 7.0 in 0.05 M KCl was 9.3 s^-1 with cold-acclimated carp, about 3.7 times higher than that for warm-acclimated carp. However, no significant difference was observed in the apparent affinity of S1 to actin. 氟中毒抑制大鼠泌鼠的实验研究

The effect of fluorosis on lactation, lactotroph function and ultrastructure were studied in lactating rats. The results were as follows: 1) Inhibition of lactation in lactating rats with chronic fluorosis was assessed by stunting growth of pups and decrease in the amount of milk suckled by pups in 30 min. Metoclopramide, a blocker of dopamine receptor, could improve lactation in these rats. 2) During chronic fluorosis serum PRL level was decreased, however, PRL content in pituitary was increased. Electronmicroscopic examination showed accumulation of large mature secretory granules and appearance of extremely large abnormal secretory granules in lactotroph cytoplasma. These findings indicate that hormone release of pituitary lactotrophs is obstructed in lactating rats with fluorosis, and the toxic effect of fluoride is mediated by an enhanced function of dopaminergic system in hypothalamus.     

The refined 2.3 A crystal structure of human leukocyte elastase in a complex with a valine chloromethyl ketone inhibitor

The stoichiometric complex formed between human leukocyte elastase and a synthetic MeO-Suc-Ala-Ala-Pro-Val chloromethyl ketone inhibitor was co-crystallized and its X-ray structure determined, using Patterson search methods. Its structure has been crystallographically refined to a final R value of 0.145 (8.0 and 2.3 A). The enzyme structure is very similar to that recently observed in a complex formed with the ovomucoid third domain from turkey [(1986) EMBO J. 5,2453-2458]. The rms deviation of all alpha-carbon atoms is 0.32 A. The peptidic inhibitor is bound in a similar overall conformation as the ovomucoid binding segment. Covalent bonds are formed between Val-P1 of the inhibitor and His-57 NE2 and Ser-195 OG of the enzyme. The carbonyl carbon is tetrahedrally deformed to a hemiketal. The valine side chain is arranged in the S1 pocket in the g-conformation.
