In this work, carbon dots (CDs) was easily synthesized from aspartic acid through a pyrolysis method. Based on their favourable fluorescence properties, CDs were utilized to design a metal ion-mediated fluorescent probe for N-acetyl-l -cysteine (NAC) detection. The fluorescence intensity of CDs was firstly quenched by manganese ions (Mn2+) through static quenching effect and subsequently restored by NAC via the combination with Mn2+ due to the coordination effect. Therefore, the fluorescent turn-on sensing of NAC was actuated based on the fluorescence quenching stimulated by Mn2+ and recovery induced by coordination. The fluorescence recovery efficiencies showed a proportional range to the concentration of NAC in the range 0.04–5 mmol L−1 and the detection limit was 0.03 mmol L−1. Furthermore, this metal ion-mediated fluorescent nanoprobe was applied to human urine sample detection and the standard recovery rates were located in the range 97.62–102.34%. This was the first time that Mn2+ was used to construct a fluorescent nanoprobe for NAC. Compared with other heavy metal ions, Mn2+ with good biosecurity prevented the risk of application, which made the nanoprobe green and biopractical. The facile synthesis of CDs and novel metal ion-mediated sensing mode made it a promising method for pharmaceutical analysis.  相似文献

Phasins play an important role in the formation of poly(3-hydroxybutyrate) [PHB] granules and affect their size and number in the cells. Recent studies on the PHB granule proteome and analysis of the complete genomic DNA sequence of Ralstonia eutropha H16 have identified three homologues of the phasin protein PhaP1. In this study, mutants of R. eutropha deficient in the expression of the phasin genes phaP1, phaP2, phaP3, phaP4, phaP12, phaP123, and phaP1234 were examined by gas chromatography. In addition, the nanostructures of the PHB granules of the wild-type and of the mutants were imaged by atomic force microscopy (AFM), and the molecular masses of the accumulated PHB were analyzed by gel permeation chromatography. For this, cells were cultivated under conditions permissive for accumulation of PHB and were then cultivated under conditions permissive for degradation of PHB. Mutants deficient in the expression of phaP2, phaP3, or phaP4 genes mobilized the stored PHB only slowly like the wild-type, whereas degradation occurred much earlier and faster in the phaP1 single mutant as well as in all multiple mutants defective in the phaP1 gene plus one or more other phasin genes. This indicated that the presence of the major phasin PhaP1 on the granule surface is important for PHB degradation and that this phasin is therefore of particular relevance for PHB accumulation. It was also shown that the molecular weights of the accumulated PHB were identical in all examined strains; phasins have therefore no influence on the molecular weight of PHB. The AFM images obtained in this study showed that the PHB granules of R. eutropha H16 form a single interconnected system inside the wild-type cells.  相似文献

Using recent pooled data from the World Health Organization Global Infobase and the World Factbook compiled by the Central Intelligence Agency of the United States, this study assesses the relation between income inequality and obesity prevalence among 31 OECD countries through a series of bivariate and multivariate linear regressions. The United States and Mexico well lead OECD countries in both obesity prevalence and income inequality. A sensitivity analysis suggests that the inclusion or exclusion of these two extreme cases can fundamentally change the findings. When the two countries are included, the results reveal a positive correlation between income inequality and obesity prevalence. This correlation is more salient among females than among males. Income inequality alone is associated with 16% and 35% of the variations in male and female obesity rates, respectively, across OECD countries in 2010. Higher levels of income inequality in the 2005-2010 period were associated with a more rapid increase in obesity prevalence from 2002 to 2010. These associations, however, virtually disappear when the US and Mexico have been excluded from the analysis. Findings from this study underscore the importance of assessing the impact of extreme cases on the relation between income inequality and health outcomes. The potential pathways from income inequality to the alarmingly high rates of obesity in the cases of the US and Mexico warrant further research.  相似文献

Domestic ducks in southern China act as an important reservoir for influenza viruses and have also facilitated the establishment of multiple H6 influenza virus lineages. To understand the continuing evolution of these established lineages, 297 H6 viruses isolated from domestic ducks during 2006 and 2007 were genetically and antigenically analyzed. Phylogenetic analyses showed that group II duck H6 viruses had replaced the previously predominant group I lineage and extended their geographic distribution from coastal to inland regions. Group II H6 virus showed that the genesis and development of multiple types of deletions in the neuraminidase (NA) stalk region could occur in the influenza viruses from domestic ducks. A gradual replacement of the N2 NA subtype with N6 was observed. Significant antigenic changes occurred within group II H6 viruses so that they became antigenically distinguishable from group I and gene pool viruses. Gene exchange between group II H6 viruses and the established H5N1, H9N2, or H6N1 virus lineages in poultry in the region was very limited. These findings suggest that domestic ducks can facilitate significant genetic and antigenic changes in viruses established in this host and highlight gaps in our knowledge of influenza virus ecology and even the evolutionary behavior of this virus family in its aquatic avian reservoirs.  相似文献

To prove that the peptidic HIV-1 fusion inhibitors containing the pocket-binding domain (PBD) mainly target the hydrophobic pocket in the gp41 N-terminal heptad repeat (NHR), we constructed pseudoviruses by replacement of Q64 in the gp41 pocket region with Ala (Q64A) or Leu (Q64L). These viruses were highly resistant to C34 and CP32M containing the PBD, while they were susceptible to T20 (enfuvirtide) lacking the PBD but containing the GIV-motif-binding domain (GBD) and lipid-binding domain (LBD). They were also sensitive to C52L, which contains the PBD, GBD, and LBD. Those mutations may disrupt the hydrophilic interaction between Q64 in the NHR and N113 in the peptides containing the PBD. This report provides insights into the mechanisms of drug resistance, with implications for the design of novel HIV fusion and entry inhibitors.  相似文献

The hydrolysis of cellobiose by β-glucodisases is an important step of cellulose biodegradation. However, the interactive mechanism between cellobiose and β-glucosidases is still unclear until now. Thus, in this study, we explored the binding modes between cellobiose and three β-glucosidases from glycoside hydrolase family 1 by means of molecular docking. The three β-glucosidases were named as TmGH1 (from bacterium Thermotoga), SsGH1 (from archaea Sulfolobus solfataricus) and TrGH1 (from fungus Trichoderma reesei) respectively, according to the monophyletic groups they belong to. Molecular dockings were performed between cellobiose and the three β-glucosidases, resulting in three optimum docking complexes, that is TmGH1-cellobiose, SsGH1-cellobiose and TrGh1-cellobiose complexes. Our docking results indicated that there were non-bonded interactions between cellobiose and the three β-glucosidases. The binding affinities of the three complexes were -13.6669kJ/mol, -13.2973kJ/mol and -18.6492kJ/mol, respectively. Then the detailed interactions were investigated, which revealed the key amino acid residues interacted with cellobiose by hydrogen bonds (H-bonds) or hydrophobic interactions. It was observed that most of the key residues involved in the non-bonded interactions were equivalent and conserved for the three complexes, and these residues were a glutamine, a histidine, a tyrosine, a phenylalanine, three glutamics, and four tryptophans. This information is of great importance for designing β-glucosidase with higher cellobiose-hydrolyzing efficiency.  相似文献

精氨酸脱亚胺酶(ADI)是一种针对精氨酸缺陷型癌症(如:肝癌、黑素瘤)的新药,目前处于临床三期试验。文中通过定点突变技术分析了精氨酸脱亚胺酶的特定氨基酸位点对酶活力的影响机制。针对已报道的关键氨基酸残基A128、H404、I410,采用QuikChange法进行定点突变,获得ADI突变株M1(A128T)、M2(H404R)、M3(I410L)和M4(A128T/H404R)。将突变株在大肠杆菌BL21(DE3)中进行重组表达,并对纯化获得的突变蛋白进行酶学性质研究。结果表明,突变位点A128T和H404R对ADI最适pH的提高,生理中性(pH 7.4)条件下的酶活力和稳定性的提高,以及Km值的降低均具有显著的作用。研究结果为阐明ADI的酶活力影响机制和蛋白质的理性改造提供了一定的依据。  相似文献

基于"临水垂直插管法"采集赣江流域32个采样点的底泥样品,经索氏提取方法(Soxhlet Extraction,SE)前处理及气相色谱法(GC-ECD)检测样品中有机氯农药(OCPs)含量。结合多元统计学和ArcGIS9.3,研究了8种有机氯农药的残留状况及空间分布情况。结果表明,所检测8种OCPs中,除β-HCH有2个点未检出,其它7种OCPs检出率为100%。以DDTs残留含量最高,DDTs平均值为10.40μg/kg,其异构体以p,p’-DDT为主;HCHs的平均值为8.24μg/kg,其异构体以β-HCH为主。它们除了来自环境中的早期残留外,仍然具有新的外源HCHs和DDTs的输入。不同OCPs在研究区分布存在很大差异,HCHs总量分布较高的地区位于流域的中下游,DDTs总量分布较高的地区位于流域的中上游。  相似文献

南方小花蝽是多种猎物的天敌,为评价西花蓟马、二斑叶螨、蚕豆蚜和腐食酪螨饲养南方小花蝽的效果,研究了南方小花蝽取食四种猎物时的生长发育和繁殖情况。结果表明南方小花蝽取食腐食酪螨时若虫只能发育到5龄,4龄若虫累计存活率仅为6.8%。其它3种猎物均可使南方小花蝽正常生长发育和繁殖,其中南方小花蝽取食西花蓟马时生长发育时间最短,繁殖率最高;取食二斑叶螨时生长发育时间最长,繁殖率最低。取食西花蓟马的南方小花蝽净生殖率、内禀增长率和周限增长率明显高于其它两种猎物。用3种猎物连续饲养南方小花蝽两代,以蚕豆蚜和二斑叶螨为猎物时南方小花蝽的未成熟期、成虫的寿命、繁殖情况和生命参数在第一代和第二代之间都有所不同,但以西花蓟马为食料时南方小花蝽第一代和第二代之间生长发育和繁殖情况没有明显的差异。以上结果表明在供试猎物中西花蓟马对南方小花蝽实验种群的增长最有利。  相似文献

Phosphorylated modification of a polysaccharide obtained from Radix Hedysari (RHP) was studied. Three phosphorylated polysaccharides (RHPP) with variable degrees of substitution (DS(p)) were obtained with 4-dimethylaminopyridine (DMAP) and N, N' Dicyclocarbodiimide (DCC) as catalyst. The structures of RHPP were characterized by FT-IR spectra and (13)C NMR spectra. Depending on different reaction time, RHPP showed different DS(p) ranging from 0.30 to 0.66, and different Mw ranging from 86.6 to 89.7?KDa. Compared with RHP, RHPP exhibited superior antioxidant activities in vitro, which indicated that phosphorylated modification could enhance antioxidant activities of RHP. Furthermore, it was obvious that the DS(p) had a significant effect on the antioxidant activity.  相似文献
