Regeneration of the arterial wall in microporous,compliant, biodegradable vascular grafts after implantation into the rat abdominal aorta

Summary The ultrastructure of a new type of vascular graft, prepared from a mixture of polyurethane (95 weight %) and poly-L-lactic acid (5 weight %), was examined six weeks after implantation into the abdominal aorta of rats. These microporous, compliant, biodegradable, vascular grafts function as temporary scaffolds for the regeneration of the arterial wall.Smooth muscle cells, covering the grafts, regenerated a neo-media underneath an almost completely regenerated endothelial layer (neo-intima). These smooth muscle cells varied in morphology from normal smooth muscle cells to myofibroblasts. They were surrounded by elastic laminae and collagen fibers.Macrophages, epithelioid cells, multinucleated giant cells, fibroblasts and capillaries were present in the disintegrating graft lattices. The epithelioid cells and multinucleated giant cells engulfed polymer particles of the disintegrating grafts.The regeneration of the endothelial and smooth muscle cells is similar to the natural response of arterial tissue upon injury. The presence of macrophages, epithelioid cells, multinucleated giant cells, fibroblasts and capillaries in the graft lattices resembles the natural response of tissue against foreign body implants. Both of these responses result in the formation of a neo-artery that possesses sufficient strength, compliance and thromboresistance to function as a small caliber arterial substitute.Supported by Grant nr. 82.042 from the Dutch Heart Foundation     

Cloning of the pectate lyase genes from Erwinia carotovora and their expression in Escherichia coli

A hybrid cosmid coding for pectate lyase (PL) activity was identified from an Erwinia carotovora genomic library by an immunological screening method. A 7-kb DNA fragment was identified which codes for three proteins identical in size to proteins with PL activity purified from E. carotovora culture supernatants. The three proteins had apparent Mrs of 41, 44 and 44 X 10(3) as estimated by SDS-PAGE. None of the PLs were exported from Escherichia coli strain HB101 but all were found in the periplasmic space. Plant tissue was macerated by the PLs made in E. coli.     

The araBAD operon of Salmonella typhimurium LT2. I. Nucleotide sequence of araB and primary structure of its product, ribulokinase

Hybrid plasmids containing the araBAD operon of Salmonella typhimurium LT2 were characterized by Southern blot and genetic analyses. The nucleotide sequence of araB was determined. The araB gene product, ribulokinase (EC 2.7.1.16), was purified and the results of amino acid composition analysis and partial amino acid sequence are in agreement with predictions from the DNA sequence. Ribulokinase is 569 amino acid residues long and has a calculated Mr of 61 793. Ribulokinase shares significant homology with xylulose kinase from Escherichia coli. Codon usage in the araB gene does not favor those codons which have intermediate codon-anticodon binding energy.     

细菌(Pseudomonas maltophilia)之绒毛膜促性腺激素(hCG)结合物质之研究

 细菌(Pseudomonas moltophilia)与hCG及LH有特异的亲和力,实验发现,细菌之生长曲线与hCG结合活性成平行关系,96小时达高峰,细菌之培养液中含有可溶性结合蛋白,该蛋白经硫酸铵沉淀(80％饱和度)、Sephadex G-100柱层析、DEAE-纤维素柱0.5mol/L NaCl梯度洗脱,再过Sepharose CL-AB柱,收集之活性部分经SDS电泳测得其分子量为70,000,凝胶层析测Stokes radius为41A,Schiff氏染色未见着色带。     

贮藏温度和气体状况对苹果果胶、多聚半乳糖醛酸酶变化的影响

在苹果贮藏前期温度和CO_2浓度由高到低同步变动形成双维变动气调(TDCA)。 前期高温期间,果实予以高浓度CO_2处理,可抑制高温促使早熟的不利效应,其保鲜效果优于冷库,接近传统气调贮藏,表现在果肉硬度下降平缓;原果胶转化为水溶性果胶的速度减慢;多聚半乳糖醛酸酶活性显著受抑。低浓度CO_2抑制高温效应的作用大为减弱。低温下,不同浓度CO_2作用差异不大,均优于对照。     

酵母鉴定程序在酵母分类鉴定中的应用

本文介绍了Barnett等1985年编制并由剑桥大学发行的计算机软件《酵母鉴定程序》,以及如何使用该程序在IBM PC DOS操作系统上进行酵母菌的分类鉴定。我们使用该程序对从云南鸡足山和紫金山两地森林土壤中分离到的82株酵母菌进行了分类鉴定,其中:鉴定到种的有47株,占总株数的57.3％;鉴定到属但未能直接定到种的有21株,占总株数的25.6％;暂未定名的有14株,占总株数的17.1％。     

阿胶冲剂与阿胶的化学成分对比分析研究

阿胶在我国有着悠久的历史,是常用滋补中药,具有许多特殊的功效,国内外享有盛誉。据《本草纲目》及《本草纲目拾遗》等介绍,阿胶在明代以前系用(沙牛)牛,水牛,驴皮或猪、马、驼皮等杂皮熬制而成。自清代始一律采用黑驴皮熬制。以往服用阿胶必须将阿胶先用水浸泡后,然后加热溶解后才能服用.为了克服这一缺点,山东化工学院研制了一种用开水冲服的阿胶新制剂——阿胶冲剂.为了弄清阿胶经物理方法处理后的化学组成成份与原阿谱的化学组成成分的不同变化。我们对其所含微量元素、氨基酸进行了对比分析.另外,本文首次报道了阿胶及阿胶冲剂的红外光谱分析结果.     

Geometric design improvements for femoral graft-artery junctions mitigating restenosis

The present study is based on the hypothesis that nonuniform hemodynamics, represented by large time-averaged wall shear stress gradients, trigger abnormal biological processes leading to rapid restenosis, i.e. excessive tissue overgrowth and renewed plaque formation, and hence early graft failure. It implies that this problem may be significantly mitigated by finding graft-artery bypass configurations for which the wall shear stress gradient is approximately zero and hence nearly uniform hemodynamics is achieved. These fluid flow and geometric design considerations are applied to four different end-to-side anastomoses for the distal end of a femoral artery bypass with an appropriate test input pulse and a typical 20–80 flow division. A validated finite-volume code has been used to compute the transient three-dimensional velocity vector fields, wall shear stress distributions and surface contours of the wall shear stress gradients. It is shown that large anastomotic flow areas, small continuously changing bifurcation angles, and smooth junction wall curvatures reduce local time-averaged wall shear stress gradients significantly and hence should mitigate restenosis.     

Kinetics of tert-[35S]Butylbicyclophosphorothionate Binding in the Cerebral Cortex of Newborn and Adult Rats: Effects of GABA and Receptor Desensitization

Abstract: The effects of GABA on the kinetics of tert -[³⁵S]butylbicyclophosphorothionate ([³⁵S]TBPS) binding to the convulsant site of GABA_A receptors were studied in membrane suspensions from the cerebral cortex of newborn (1-day-old) and adult (90-day-old) rats. TBPS dissociation was biphasic in neonates and adults, indicating that more than one interconvertible state of [³⁵S]TBPS binding sites may be present in the cerebral cortex. In the absence of GABA, the fast ( t _1/2, 11 min) and slow ( t _1/2, 77 min) components of TBPS dissociation in newborn rats were approximately fourfold slower than in adults. The acceleration of the dissociation rates caused by 2 µ M GABA, however, was more robust in neonates than in adults (six- to ninefold vs. twofold increase, respectively). Moreover, the dissociation rates of TBPS in membranes preincubated with 2 µ M GABA (dissociation started by adding 40 µ M picrotoxin) were two- to fourfold slower than in membranes preincubated without GABA (dissociation started by adding 40 µ M picrotoxin plus 2 µ M GABA). Taken together, these results suggest that (1) the closed state of GABA_A receptors is associated with a more effective steric barrier for the binding of TBPS in neonates compared with adults, (2) GABA produces a larger acceleration of the binding kinetics of TBPS in neonates than in adults, and (3) long incubations with GABA may cause receptor desensitization, which in turn slows down the dissociation rates of TBPS.     

几种昆虫生长调节剂对家白蚁的毒效试验

在室内条件下测定了卡死克、抑太保、灭幼豚3号、爱力螨克和扑虱灵五种昆虫生长调节剂对家白蚁的毒杀效果。初步筛选结果表明：卡死克、抑太保和爱力螨克对家白蚁的毒杀效果均较好,家白蚁对爱力螨克尤其敏感。2．30pm。yL爱力螨克、327．36pm0VL、卡死克和369．80V＊wL抑太保处理白蚁5～6天后,其死亡率可达100％。忌避性试验表明：卡死克、抑太保和爱力螨克对家白蚁均无明显的驱避作用。
