A review of in vitro mutagenesis assessment of metal compounds in mammalian and nonmammalian test systems has been compiled. Prokaryotic assays are ineffective or inconsistent in their detection of most metals as mutagens, with the notable exception of hexavalent chromium. Mammalian assay systems appear to be similarly inappropriate for the screening of metal compounds based upon the limited number of studies that have employed those compounds having known carcinogenic activity. Although of limited value as screening tests for the detection of potentially carcinogenic metal compounds, the well-characterized in vitro mutagenesis systems may prove to be of significant value as a means to elucidate mechanisms of metal genotoxicity.  相似文献

Concentrations of soluble aluminum (Al) and manganese (Mn) frequently reach phytotoxic levels in acid soils. While dose response relationships for these metals are well documented, the effects of combined exposure have received less attention. We have examined the effect of combinations of Al and Mn on growth and metal accumulation in Vigna unguiculata (L.) Walp. grown in solution culture under conditions of low ionic strength (conductivities typically < 100 µS cm−1). The nature of interaction between these metals varied with the specific physiological response, the part of the plant investigated, and the relative amount of stress imposed. Analysis of growth data provided evidence for amelioration of metal toxicity (antagonistic effects), although this effect was dose dependent. Analysis of metal content data provided evidence for antagonistic and synergistic (exacerbation of toxicity) effects, again depending on dose. Analysis of foliar symptoms also provided evidence for antagonisms and synergisms, with the nature of the response dependent on the specific physiological response and specific plant part investigated. In contrast with previous reports, evidence for antagonistic, synergistic, and multiplicative effects on growth, metal uptake, and expression of foliar symptoms have been obtained under physiologically and environmentally relevant conditions. These results suggest a more detailed analysis of the potential for interactions between metals in the environment is required.  相似文献

The effect of linolenic acid (18:3) on release of the 43 kDa polypeptide and manganese from photosystem II ( PS II ) membranes depleted of extrinsic polypeptides was studied. In both control and NaCl-washed particles which were depleted of the extrinsic 23 and 16 kDa polypeptides, the 18:3 treatment caused a 20% release of the 33 and 43 kDa polypeptides. In CaCl2, (or urea + NaCl)-washed particles, which were depleted of the 33 kDa polypeptide in addition to the 23 and 16 kDa polypeptides, the release of the 43 kDa polypeptide increased to 70%, whereas only 25% of the 47 kDa polypeptide was removed. These findings suggest (i) that the 33 and the 43 kDa polypeptides are neighbows in the photosynthetic membrane and (ii) that the 33 kDa polypeptide shields the 43 kDa polypeptide against the action of 18:3. Incubation of CaCl2, or (urea + NaCI)-treated PSII particles in the presence or absence of 18:3 resulted in the loss of only 2 of the 4 Mn atoms present per reaction center. this indicates that the 2 Mn atoms more firmly associated with PSII are not affected by the removal of the extrinsic 16, 23 and 33 kDa polypeptides, and the intrinsic 43 kDa polypeptide. nor by the treatment with linolenic acid.  相似文献

Summary Application of a granular formulation of hexachlorocyclohexane (HCH) to the potted soil at flooding decreased the concentration of iron and. to some extent, manganese in rice plants, especially at concentrations above 3 ppm active ingredient (a.i.) Likewise, HCH, applied to rice fields at transplanting (several days after submergence) caused a significant decrease in the concentration of iron, and not manganese, in the rice plant but only at concentrations above 12.5 kg a.i./ha despite high levels of reduced iron in the soil. Inhibition of iron reduction by HCH was more pronounced when applied at flooding than at several days after flooding.  相似文献

Addition of manganese, at levels of 50 ppm, to a liquid growth medium simulating adverse silage conditions had no effect on the growth or on the fermentation pattern of Enterobacter cloacae and Proteus vulgaris. Yet, the manganese strongly enhanced the growth of Lactobacillus plantarum. Co-cultures of L. plantarum and E. cloacae or P. vulgaris were, by addition of manganese ions, significantly altered in the favour of the former. This finding can be of use in mixed cultures where Enterobacteriaceae act as spoiler microorganisms.  相似文献

【目的】探究一株红酵母对Mn(Ⅱ)的去除效率及其作用机制。【方法】从酸性矿山废水中分离出一株耐酸酵母菌,通过形态和26S rRNA基因测序对菌种进行鉴定,研究不同pH和Mn(Ⅱ)浓度对该菌除Mn(Ⅱ)效果的影响。采用扫描电镜、X射线衍射分析和X射线光电子能谱仪进行产物表征。【结果】分离得到的酵母菌经鉴定为台湾红酵母(Rhodotorula taiwanensis),其在pH 2.0、2 000 mg/L Mn(Ⅱ)条件下仍能生长较好。在初始pH 6.0、Mn(Ⅱ) 300 mg/L条件下培养144 h后,对Mn(Ⅱ)的去除率能达到98.52%;然而较高浓度的Mn(Ⅱ) (≥500 mg/L)会对细胞产生毒性,从而降低去除效果。R. taiwanensis MF4在去除Mn(Ⅱ)的过程中可以将Mn(Ⅱ)氧化成锰氧化物(主要为无定型的MnO2、Mn2O3、MnO),形成层状物质在细胞表面积累,而且能产生碱度,提升环境pH值,最高可达8.4 [初始pH 7.0,Mn(II) 100 mg/L,144 h]。【结论】R. taiwanensis MF4具有耐受低pH和高浓度Mn(II)、有效去除Mn(II)以及产碱的作用,研究结果对酸性矿山废水修复与治理的末端工艺设计具有参考价值。  相似文献

Sediment pore water concentrations of Fe2+, Mn2+, NH inf4 sup+ and CH4 were analyzed from both diver-collected cores and anin situ equilibration device (peeper) in Lake Erie's central basin. Sediment oxygen demand (SOD) was measured at the same station with a hemispheric chamber (including DO probe and recorder) subtending a known area of sediments. The average SOD was 9.4 mM m−2 day−1 (0.3 g m−2 day−1). From pore water gradients within the near-surface zone, the chemical flux across the interface was calculated indirectly using Fick's first law modified for sediments. These calculations, using core and peeper gradients, always showed sediment loss to overlying waters, and variations between the two techniques differed by less than an order of magnitude for Fe2+ and CH4. The transport of these reduced constituents can represent a sizeable oxygen demand, ranging from less than 1% for Fe2+ and Mn2+ to as high as 26% for NH inf4 sup+ , and 30% for CH4. The average flux of these constituents could account for about a third of the SOD at the sediment-water interface of this station.  相似文献

The addition of manganese oxides to iron-limited medium promoted the formation of the pyoverdin siderophore azotobactin by Azotobacter vinelandii. When active-MnO2 was used, there was greatly decreased iron uptake into the cells, hyperproduction of azotobactin and the abiotic, chemical destruction or adsorbtion of the catechol siderophores azotochelin and aminochelin by this strong oxidizing agent. Although the iron content of the cells was the same as iron-limited cells, the growth of cells in medium with active-MnO2 was increased 1.5- to 2.5-fold over iron-limited controls. This growth promotion was not caused by iron contaminating the oxide or by manganese solubilized from the oxide. Soluble 0.05–4 mm Mn2+ inhibited the growth of iron-limited cells and had a minimal effect on iron uptake, but caused hyperproduction of azotobactin. This later effect was caused by the inhibition of soluble ferric reductase, in a manner identical to that previously observed for Zn2+. These results suggest that active-MnO2 may interfere with a surface-localized iron uptake site, possibly another ferric reductase. The reason for the growth promotion by active-MnO2 remains unknown, but is most likely related to decreased oxygen toxicity.  相似文献

Micro-element deficiencies have been implicated in the development of poor stem form in Pinus radiata plantations. Cu, Mn and B have been implicated in previous studies as influencing tree form and/or the process of lignification in plants. Therefore an experiment was initiated to examine the individual and interactive effects exhibited by these trace elements on stem form and lignification.The investigation showed that Cu deficiency reduced both lignification and stem form in seedlings. Mn, competing with Cu for uptake, enhanced the symptoms of Cu deficiency. Boron addition did not alleviate the expression of deformity or increase levels of lignification. There was a clear family influence on the development of seedling stem form in response to variations in mineral nutrition.  相似文献

This study was carried out to assess manganese (Mn) status after an acute episode of myocardial infarction. Plasma and erythrocyte Mn concentrations were measured from admission to hospital to day 15 postadmission in 21 patients suffering from acute myocardial infarction and in three control groups. The determination of Mn in these biological fluids was performed by electrothermal atomic absorption spectrometry. Plasma Mn was higher (p<0.01) and erythrocyte Mn was similar in the acute myocardial infarction group compared to healthy age-matched control group. Plasma and erythrocyte Mn remained unchanged during the 2 wk after acute myocardial infarction and were not correlated to enzyme activities. A decrease of erythrocyte Mn with age, expressed in nmol/L, was noted (p<0.02). These results suggest that plasma and erythrocyte Mn do not provide an indication of myocardial damage. Nonetheless, Mn status in elderly merits further attention.  相似文献
