一边是水资源的严重短缺，一边是干旱导致的农业减产，无疑使世界粮食危机雪上加霜。水资源正成为世界多数国家制约农业发展的重要因素，培育“不怕渴”农作物品种成为摆在世界育种学家面前的共同难题，也是节水农业中最有效的技术手段。     

Mitosis and microtubule organizational changes in rice root-tip cells

The pattern of change of the microtubule cytoskeleton of the root-tip cells of rice during mitosis was studied using immunofluorescence technic and confocal laser scanning microscopy. All the major stages of ceil division including preprophase, prophase, metaphase, anaphase and telophase were observed. The most significant finding was that in the preprophase cells microtubules radiating from the nuclear surface to the cortex were frequently seen. During development these microtubules became closely associated with the preprophase band and prophase spindie indicating that the microtubules radiating from the nuclear surface, the preprophase band and the prophazc spindle were structurally and functionally closely related to each other. Granule-like anchorage sites for the radiating microtubules at the muclear surface were often seen and the possibility that these gramle-like anchorage sites might represent the microtubule organizing centres was discussed.

Changes of intracellular Na+ concentration in erythrocytes caused by pulsed electrical field

Changes of sodium ionic concentration of human erythrocytes applied to pulsed electrical field (PEF) were studied by using shift reagent and NMR spectroscopy. The results show that the concentration of intracellular Na⁺ increases with the increasing intensity of PEF when the erythrocytes are applied to PEF with higher intensities. The relationship between intracellular Na concentrations and the intensities of PEF does not follow linear or exponen-tial behavior. As the intensities increase, the intracellular Na⁺concentrations increase even faster by an exponential curve. However under effects of PEF at lower intensities, intracellular Na⁺ concentration decreases. Ouabain can in-hibit the decrease of intracellular Na concentration, and the inhibition increases with the increasing concentration of ouabain, suggesting that Na⁺ , K⁺ -ATPase on cell membrane can be activated by PEF at lower intensities. Direct measurement of activities of the enzyme by using Malachite green method has confirmed this observation. Cell perme-abilities to ions, activation of enzymes by electrical fields and transmission of physical signals like PEF across cell mem-branes are discussed.

琼脂糖的制备、鉴定及应用——对流免疫电泳检测肝炎相关抗原和甲胎蛋白灵敏度的观察

琼脂糖是由D-半乳糖和3,6-脱水-L-半乳糖相间结合的链状多糖,存在于多种海藻的琼脂胶质中,应不含硫和灰分,可作为电泳支持物和亲和层析载体。琼脂中另一主要组成成分是果胶,含硫和羧基。如琼脂糖中含有较多的果胶,作电泳支持物使用时,电渗较大;用于亲和层析,非特异性吸附蛋白质能力较强,均影响分离效果。1971年S.Hjerten曾将每克琼脂干粉以150毫升pH6.8,0.03M磷酸缓冲液抽提,每天更换一次缓冲液,共抽四天,可得到含硫量0.25％的产物。进一步用DEAE-Sephadex A-50(每克     

乳酸菌D—80葡萄糖异构酶的研究

采用乳酸菌D-80为试验用菌种,经试验找出了适合产葡萄糖异构酶的培养条件。培养基含有：玉米芯水解液还原物1．2—1．4％,米糠饼2一4％,麸皮0.2～0.4％(二者经1398蛋白酶水解)及MnSO．0.05％。微量通风,开罐发酵,35—37℃培养。培养前期6—7小时,用NaoH调节发酵液pH为4．9—5．1。总的发酵时间为10一14小时。成熟醪中的菌体,经分离后,加入30—40％的葡萄糖浆,pH 6．4—6．7,MnSO4的含量10-4M,60—65℃的条件下转化,果糖的转化率为40％左右。经扩大试验后,进行技术鉴定,认为工艺路线基本上是合理的,经济上也是合算的,可进一步扩大试生产。

黑龙江省辖区内“120”急救中心调度员工作量及职业压力现状研究

目的:调查黑龙江省辖区内的120急救调度中心调度员的工作量与职业压力情况,为在120急救中心进行人力资源优化配置提供支持。方法:通过基础工作量问卷与工作内容量表(JCQ)问卷对黑龙江省内8家"120"急救调度中心内共计62名急救中心调度员进行调查和统计分析,并对调度科室主任进行访谈。结果:调度员的通话量呈现明显的地区差异性并异常地高于调度量,调度员的工作属于积极型的工作状态,但是决策自主度的分数较低,而技术自主度则与通话量和调度量具有明显线性关系(P0.05),调度时间与上司支持和同事支持有显著相关性(P0.05)。结论:黑龙江省辖区内的急救调度中心依旧处于组织结构复杂化、资源分配不均的情况,需要对调度员缩短调度时间进行训练以及对市民进行急救知识教育以减少无用通话的数量。
