研究了氨基糖甙类抗生素JI-20A分批发酵培养基和发酵工艺的优化。结果表明发酵培养基的优化组成为玉米淀粉60g/L,花生饼粉30g/L,麦芽糖10g/L,玉米浆8g/L,复合无机盐适量,淀粉酶适量,蛋氨酸1g/L和氯化钴6μg/mL。控制消后培养基pH值和氧的供应条件有助于抗生素JI-20A产生菌的菌体生长和产物合成。  相似文献

目的:研究赭曲霉高密度培养的发酵培养基及条件,实现坎利酮的高转化.方法:选取廉价易得的培养基成分并进行优化,同时对发酵条件进行优化,得到了最优发酵培养基配方及培养条件.结果:发酵培养基最优配方为:葡萄糖20g/L,玉米浆20g/L,酵母膏20g/L,K2HPO4 2.5g/L.种子液最佳培养时间为24h,发酵培养基初始pH 5.8,接种量为8%,装液量200mL/1000mL,摇床转速为180 r/min,28℃,底物投料时间24h,发酵结束时间72 h.结论:将该工艺在7L发酵罐中放大,菌体密度达到25.36g/L,11α羟基坎利酮的转化率为86.1%.  相似文献

对实验室构建的产琥珀酸大肠杆菌工程菌株(E.coliQZ1111)进行发酵工艺条件研究。以AM1低盐培养基为基础,研究不同C、N源及其质量浓度,培养基初始pH和发酵温度等因素对琥珀酸的影响,并在5L发酵罐中进行了补料-分批发酵实验。优化后的发酵条件为葡萄糖20g/L,玉米浆10g/L,pH6.4,发酵温度37℃。在5L发酵罐中培养,琥珀酸产量达到47.9g/L。  相似文献

研究了几种物质对D-核糖产量的影响,研究表明,当发酵培养基中玉米浆的含量为25g/L,D-核糖产量达到最高,为65g/L。发酵培养基中添加适量的粉及山梨醇亦有利于D-核糖的积累,当粉与山梨醇的添加量分别为10g/L及40g/L时,D-核糖产率分别增加7.8%、4.7%,而在发酵培养基中加入丙二酸可抑制D-核的分泌,在40ml发酵培养基中添加1ml丙二酸后,D-核糖的产率下降73.8%。甲醇也可抑制D-核糖的积累,当发酵培养基中的甲醇添加量为16g/L时,D-核糖产率下降66.2%。  相似文献

【目的】提高重组谷氨酸棒杆菌发酵L-苯丙氨酸(L-phenylalanine,L-Phe)的产量。【方法】使用正交试验设计以及响应面优化法分别对种子培养基及发酵培养基进行优化,确定了重组谷氨酸棒杆菌发酵L-Phe的最佳种子培养基及最佳发酵培养基。【结果】重组谷氨酸棒杆菌发酵L-Phe最佳种子培养基(g/L):葡萄糖25.0,玉米浆25.0,硫酸铵15.0,硫酸镁1.0,磷酸二氢钾2.0,尿素2.0,p H 6.8-7.0;最佳发酵培养基(g/L):葡萄糖110.0,玉米浆7.0,硫酸铵25.0,硫酸镁1.0,磷酸二氢钾1.0,柠檬酸钠2.0,谷氨酸1.0,碳酸钙25.0,p H 6.8-7.0;在最佳培养基条件下L-Phe产量最高达到9.14 g/L,较优化前的7.46 g/L提高了22.5%。【结论】通过正交试验和响应面分析对重组谷氨酸棒杆菌发酵L-Phe培养基进行优化,明显提高了L-Phe的产量,并确定了葡萄糖、玉米浆和硫酸铵为发酵培养基中影响L-Phe产量的3个关键因子。研究结果为L-Phe的发酵放大提供了依据。  相似文献

对本公司自主分离、优化得到的新型农用抗生素吩嗪-1-羧酸的产生菌STH16#生产吩嗪-1-羧酸的发酵培养基配方的初步确定。经实验确定吩嗪-1-羧酸的发酵培养基配方为:甘油15.0g/L、蛋白胨20.0g/L、磷酸氢二钾1.5g/L、硫酸镁2g/L。前体饲喂:在接种后8小时加入质量分数为2.0g/L的MCS。发酵产量可达2500mg/L。  相似文献

目的:为提高CpG基序重组菌的发酵水平,降低CpG重组质粒的生产成本,运用响应面法优化CpG重组菌发酵培养基。方法:利用Plackett-Burman试验筛选出影响发酵水平的3个最显著因素:糖蜜、玉米浆和工业蛋白胨。然后用响应面设计试验并优化得到最显著因素的最佳浓度,并进一步通过发酵试验验证优化后的培养基。结果:得到一组具有较高菌浓及质粒产量且价格低廉的发酵用培养基:糖蜜4.5g/L,玉米浆8.5g/L,工业蛋白胨8.5g/L,甘油10mL/L,KH2PO41.5g/L,K2HPO42.3g/L,MgSO4.7H2O 0.25g/L,在此培养条件下,OD600实测值为0.6771,理论值为0.6643,两者接近,与标准LB培养基相比,质粒产量提高了15%左右。结论:最终筛选到的培养基具有较高的性价比。  相似文献

【目的】研究内生真菌菌株YCEF005、YCEF053固态发酵培养基。【方法】通过单因素试验和正交试验设计优化固态发酵培养基基质组成;通过单因素试验和均匀试验设计及神经网络结合遗传算法优化基质外加组分和接种量。【结果】优化的YCEF005培养基基质组成(质量比)为麸皮50%、豆饼粉10%、米糠20%、玉米碎粒20%;YCEF053培养基基质组成为麸皮60%、豆饼粉10%、米糠10%、玉米碎粒20%。YCEF005基质外加组分和接种量(/kg基质)为蔗糖12.96 g、蛋白胨12.70 g、NH4NO3 4.00 g、KH2PO4 1.50 g、Ca SO4 10.00 g、Mg SO4 0.48 g、含水量500 g、接种量24 m L。YCEF053基质外加组分和接种量(/kg基质)为蔗糖13.37 g、蛋白胨14.02 g、Na NO33.85 g、KH2PO4 1.23 g、Ca SO4 10.89 g、Mg SO4 0.52 g、含水量480 g、接种量20 m L。在优化培养基上发酵7-9 d获得最大发酵生物产量。【结论】通过对固态发酵培养基的优化提高了发酵的生物产量。  相似文献

研究C、N源对里氏木霉(Trichoderma reesei)生产纤维素酶的影响,采用单因素实验方法和中心复合方法对发酵培养基进行优化。单因素实验表明:黄豆饼粉、玉米芯、玉米浆对纤维素酶的影响显著。通过响应面优化,得到最优培养基C、N源的组成:黄豆饼粉32.21 g/L,玉米芯42.29 g/L,玉米浆4.45 g/L。优化条件下,摇瓶发酵7 d的比酶活达到(10.65±0.50)U/mL。  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

肝癌中HBV和HCV基因和抗原的分布及意义   总被引:1,自引:0,他引:1  
采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.
