试验拟对谷子重要农艺性状进行数量性状位点QTL分析。以表型差异较大的沈3/晋谷20F2作图群体为材料,观测其株高、穗长等性状,选用SSR做分子标记,利用完备区间作图法(BASTEN C J)进行QTL分析。结果显示,表型数据在作图群体中呈现连续分布,表现为多基因控制的数量性状,被整合的54个SSR标记构建10个连锁群,LOD阈值设置为2.0,检测到与株高相关的主效QTL2个,联合贡献率45.9637%,穗长主效QTL1个,贡献率14.9647%,与穗重、粒重相关的主效QTL为同一位点,贡献率分别为11.9601%和10.1879%。有6组QTL位点之间存在基因互作效应,大小范围为-0.4986-16.6407,对性状的贡献率在2.2716%至6.7478%之间。谷子表型控制复杂,相关QTL的检测受环境影响较大,不同连锁群QTL间互作明显。  相似文献

以遗传性状差异较大的甜瓜材料日本安农二号与新疆哈密瓜K413杂交产生的143个F2单株为作图群体,以AFLP与SSR分子标记为主构建了包含12个连锁群、142个遗传标记位点的甜瓜遗传图谱,其中包括121个AFLP标记、16个SSR标记、3个STS标记、2个性状标记,连锁群总长度为1 014.2 cM。应用复合区间作图法对甜瓜果实的大小、长宽比、糖度、硬度以及甜瓜种子的长、宽、形状、重量等性状进行遗传定位与分析。基因定位结果显示控制果肉颜色的基因位于C9连锁群AFLP分子标记NDAA与NCFA之间。其他性状表现为数量性状控制,共检测到25个数量性状基因座,不同性状基因座位有重叠分布的特点。其中C5连锁群标记NCA-N73C区间检测到QTLs Sl5.1、Sw5.1和Swt5.1分别控制种子长、宽和千粒重,分别可解释表型变异的17%、19%和23%。该区域包含的来自母本安农二号的基因位点对甜瓜种子的长、宽、千粒重均有明显的抑制作用;位于C8连锁群标记N73A与NFDA间的QTL通过影响种子的宽度从而影响种子的形状与重量;同样位于C8连锁群的果实长宽比QTL Fs8.1在F2和F3中均检测到,分别解释表型变异的25%和19%,表现为部分显性,来自安农二号的等位基因抑制甜瓜果实伸长,生成圆形甜瓜;还发现控制甜瓜果实心糖、边糖、果实硬度的QTL各一个。  相似文献

油分含量和百粒重是大豆中两个重要的性状。本研究利用东农46和L-100衍生的重组自交系(RIL)群体,经过两年3个地点种植,通过分子标记技术定位与大豆油分含量和百粒重相关的QTL(quantitative trait locus)。结果表明,检测到6个与油分含量相关的QTL,分别位于E、H、G和I连锁群上,可解释的表型贡献率范围为2.12%~2.77%;检测到5个与百粒重相关的QTL,分别位于K、H、B2和G连锁群上,可解释的表型贡献率范围为2.30%~7.59%,在H连锁群上有2个QTL两年均被检测到,标记区间分别为Satt279-Sat_122和Satt192-Satt568。在H连锁群上Satt192-Satt568标记区间内同时检测到与油分含量和百粒重相关的QTL。研究结果为大豆油分含量和百粒重等性状的分子辅助育种提供了理论依据。  相似文献

利用苹果栽培品种‘红富士’和新疆野苹果优系‘红肉苹果’杂交的110个F1株系为作图群体,构建了苹果的分子遗传图谱,采用区间作图法对苹果9个叶片相关性状(叶片长度、叶片宽度、叶片厚度、叶柄长度、叶片面积、总叶绿素含量、叶绿素a含量、叶绿素b含量和类胡萝卜素含量)进行了QTL定位分析。结果显示:从110个F1株系中共检测到20个控制叶片相关性状的QTL位点,分布在第1、2、3、4、5、7、8、10、11、12、16、17连锁群上;各QTL位点的LOD值在2.58~3.55之间,其中主效QTL位点2个(LOD≥3.5),可解释11.63%~16.36%的表型变异。获得紧密连锁的特异标记(CH05d11-435m、CH04c06-201m)为进一步进行QTL精细定位提供了参考。  相似文献

该研究利用油菜双单倍体株系(348份)群体和已构建的遗传连锁图谱,采用复合区间作图法,对2009~2013年连续5年的千粒重性状表型数据进行QTL初步定位和分析,结果共获得46个显著性千粒重QTL,主要分布在A7、C1和C6等11条染色体上;其中qTSW-09 DL11-1的表型变异最高(19.63%),qTSW-11 DL9的表型变异最小(2.73%)。通过元分析方法将所获得的46个QTL进行整合,结果显示:cqTSW-C1-2的表型变异最大(10.64%),并发现多个整合后的一致性QTL能够在连续多年试验中被检测到,其中cqTSW-C1-3连续5年被检测到,表明控制千粒重的QTL在种植环境中能够稳定表达;同时,新发现位于C1染色体上的千粒重主效QTL cqTSW-C1-2,解释表型变异达到10.64%。油菜千粒重性状的QTL分析和主效QTL的获得,为进一步实现油菜大籽粒的分子育种和高产新品种的培育提供了重要的理论指导。  相似文献

富硒功能水稻具有富硒、优质、保健、防病、安全等特点,已成为当前研究的热点。子粒硒含量的QTL定位对研究富硒功能水稻的遗传育种具有重要的意义。以籼稻亲本奉新红米和明恢100杂交的145个株系的F2群体构建遗传连锁图谱,图谱拟合92个SSR标记位点,覆盖水稻基因组2187.5 cM,标记间平均遗传距离为23.7 cM,占水稻全基因组的49.2%。采用复合区间作图法,对水稻子粒硒含量进行QTL分析,在第5染色体上共检测到2个新的水稻子粒硒的QTL,对表型变异的贡献率分别为6.39%、8.01%。  相似文献

利用鸭梨和京白梨杂交得到的F1(145株)实生苗为作图群体,通过对AFLP和SSR两种分子标记的遗传连锁分析,应用Joinmap 3.0作图软件,368个AFLP标记、34个SSR标记构建了分属18个连锁群的梨分子遗传连锁图谱,各连锁群的LOD值在4.0~7.0范围之间,图谱总长度覆盖梨基因组1395.9cM,平均图距为3.8cM.采用区间作图法,对该群体与生长性状相关的调查数据进行QTL分析,检测到与新梢生长量、新梢茎粗、节间长度、节间数量、树干径、树高及皮孔密度7个农艺性状连锁的QTL位点35个,其中主效QTL位点11个(LOD≥3.5).与生长性状相关的农艺性状QTL位点多集中在LG16连锁群上.  相似文献

为了全面了解亚麻产量和品质相关性状的遗传基础,为亚麻基因克隆和分子标记辅助育种提供理论依据,在已构建SNP连锁遗传图谱的基础上,以LH-89为父本,R43为母本构建F2:3家系QTL定位群体,用R/QTL软件采用复合区间作图法对13个农艺和品质性状进行QTL定位。结果表明:(1)该研究共检测出35个QTL位点,与粗脂肪及其组成成分相关的QTL有20个,与农艺性状相关的QTL有15个;其中:亚油酸和粗脂肪各5个,亚麻酸、千粒重各4个,棕榈酸、株高、工艺长度各3个,硬脂酸、分枝数各2个,单株果数、果粒数、单株粒重、油酸各1个。(2)共有18个QTL的表型贡献率超10%(主效基因),其中农艺性状定位8个主效基因,品质性状定位10个主效基因。  相似文献

采用常规品系04-1139与高产多角果品系05-1054构建的F2代群体为作图群体, 运用SSR(Simple sequence repeat)和SRAP(Sequence-related amplified polymorphism)构建分子标记遗传图谱并对甘蓝型油菜单株产量构成因素进行QTL分析。遗传图谱包含200个分子标记, 分布于19个连锁群上, 总长度1 700.23 cM, 标记间的平均距离8.50 cM。采用复合区间作图法(Composite interval mapping, CIM)对单株产量构成因素(单株有效角果数、每果粒数和千粒重)进行QTL分析, 共检测到12个QTL: 其中单株有效角果数4个QTL, 分别解释表型变异为35.64%、12.96%、28.71%和34.02%; 每果粒数获得5个QTL, 分别解释表型变异为8.41%、7.87%、24.37%、8.57%和14.31%; 千粒重获得3个QTL, 分别解释表型变异为2.33%、1.81%和1.86%。结果表明: 同一性状的等位基因增效作用可以同时来自高值亲本和低值亲本; 文章中与主效QTL连锁的标记可用于油菜产量性状的分子标记辅助选择和聚合育种。  相似文献

以印度南瓜纯系大粒材料‘0515-1’和小粒材料‘0460-1-1’为亲本,获得193个南瓜F2单株群体,应用AFLP和SSR分子标记技术进行多态性筛选,构建了含84个标记位点的遗传连锁图谱。结果表明,整个图谱包含12个连锁群,全长683.50cM,标记平均间距为8.13cM。采用复合区间定位分析,共检测到控制南瓜籽粒宽度的4个数量性状位点(QTL),分别位于3个连锁群上,各QTL的贡献率在2.87%~29.68%之间。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
