为了研究许氏平鲉(Sebastes schlegelii)雌鱼体内受精后仔鱼开口前和仔鱼开口后两个阶段氨基酸和脂肪酸的组成变化规律, 采用生化常规方法定量测定并分析了许氏平鲉发育早期的受精卵(FE)、胚胎期(ES)、初产仔鱼(PL1)、前仔鱼期(PL2)、后仔鱼期(PL3)和稚鱼期(J)6个阶段的氨基酸和脂肪酸组成特点及含量变动。结果表明: 总氨基酸含量从FE至PL1显著下降, 至PL3显著上升, 至J又显著下降(P<0.05); 游离氨基酸含量以FE最低(12.77 mg/g), 从FE到PL1显著上升(P<0.05), 并在PL1含量达到最高值(92.19 mg/g), PL1发育到J呈现先下降后上升再下降的变化趋势(P<0.05), 游离氨基酸与总氨基酸的比值范围在2.37%—19.66%。在各发育阶段干样中检出碳链长度在C14-C24的29种脂肪酸, 分别为9种饱和脂肪酸(SFA)、9种单不饱和脂肪酸(MUFA)和11种多不饱和脂肪酸为(PUFA), 受精卵中主要脂肪酸依次为C22:6n-3(DHA)、C18:ln-9c、C16:0和C20:5n-3(EPA)。胚胎期(FE-ES)的脂肪酸利用率顺序为SFA、MUFA、n-6PUFA、n-3PUFA, 主要以C18:3n-3、C18:0、C16:1n-7及C20:5n-3(EPA)作为胚胎期的能量来源, C22:6n-3(DHA)的实际利用率最低(9.71%), 被优先保存下来, C16:0的实际利用量最高(10.94mg/g); 仔鱼内源营养阶段(ES-PL1)脂肪酸利用率顺序为MUFA、n-6PUFA、SFA、n-3PUFA, 主要以C16:1n-7、C18:0、C20:4n-6(ARA)及C18:1n-9c作为开口前仔鱼的主要能量来源, 其中仔鱼对DHA实际利用量最高(18.23 mg/g)。PL1-PL3阶段DHA相对于EPA和ARA被选择性利用; PL3至J阶段ARA相对于EPA和DHA被选择性利用。研究表明: 许氏平鲉仔鱼开口前阶段总氨基酸含量与游离氨基酸含量的变化趋势截然相反, 胚胎期与仔鱼内源营养阶段脂肪酸利用率和利用量均有所不同, 仔鱼期DHA优先被利用, 过渡至稚鱼期ARA优先被利用。建议在仔鱼开口后添加富含DHA生物性饵料, 仔鱼过渡到稚鱼期在配合饵料中添加ARA营养物质, 防止苗种营养不足, 保证成活率。  相似文献

通过采用GC/MS法研究了条石鲷(Oplegnathus fasciatus)仔鱼、稚鱼及幼鱼阶段的脂肪酸组成和变化特点。共检测到28种脂肪酸,其中饱和脂肪酸(SFA)13种,单不饱和脂肪酸(MUFA)7种,多不饱和脂肪酸(PUFA)8种。结果表明:条石鲷鱼苗内源性营养阶段以饱和脂肪酸C16∶0、C20∶0及单不饱和脂肪酸C16∶1、C18∶1作为能量代谢的主要来源;必需脂肪酸C20∶4(n-6)(AA)、C22∶5(n-3)(DPA)和C22∶6(n-3)(DHA)在稚鱼期含量较低,∑EPA+DHA仅为6.89%,认为是发生稚鱼"胀鳔病"的主要原因;仔鱼开口前体内的DHA和EPA是由母体卵黄提供的。  相似文献

为掌握刀鲚(Coilia nasus)胚胎及胚后发育早期的脂肪酸变化规律,采用生化分析手段对刀鲚的胚胎(原肠期,受精后7~9 h)、0日龄仔鱼(初孵仔鱼)、3日龄仔鱼和5日龄仔鱼(开口前)的脂肪酸组成和含量进行了检测分析。结果显示,刀鲚发育早期的总脂占干物质的相对含量均较高(53.10%~60.97%),干物质的总脂相对含量随个体发育显著降低,单个个体的总脂含量随个体发育呈现剧烈下降趋势,数值从胚胎的43.62μg/ind剧烈下降到5日龄仔鱼的16.27μg/ind;水分含量随个体发育而升高。刀鲚发育早期上述4个时期的干样中检出6种饱和脂肪酸(SFA)、4种单不饱和脂肪酸(MUFA)和8种多不饱和脂肪酸(PUFA)。4个发育时期脂肪酸相对含量,C18:1n9c占绝对优势(50.39%~57.00%),C16:1丰富且稳定(13.77%~14.24%),C16:0也较丰富(7.45%~9.15%)。单不饱和脂肪酸(MUFA)比例占绝对优势(65.14%~72.26%),n-3与n-6系列多不饱和脂肪酸含量的比值(∑n3PUFA/∑n6PUFA)较小(1.78~2.38)。刀鲚胚胎孵化出膜期间,单个个体单不饱和脂肪酸(MUFA)实际减少程度较高,尤其是C18:1n9c(减少量为13.21μg/ind,减少比例达到55.49%)和C16:1(减少量和比例分别为3.30μg/ind和53.12%),而二十碳五烯酸(EPA)加二十二碳六烯酸(DHA)的减少程度较低(1.44μg/ind和38.41%),尤其是DHA(0.95μg/ind和36.52%)。然而,出膜后,仔鱼对单不饱和脂肪酸(MUFA)的利用相对较低(1.94μg/ind和14.17%),尤其是C18:1n9c(13.21μg/ind和12.41%)和C16:1(0.63μg/ind和21.81%);而对EPA+DHA利用相对较高(1.04μg/ind和45.10%),尤其是DHA(0.71μg/ind和42.61%)。研究表明,刀鲚胚胎优先蓄留EPA和DHA,仔鱼在摄食前大量利用EPA+DHA(特别是DHA),呈现出海水鱼类脂肪酸的利用特点。因此,建议在刀鲚亲本强化培育及产后培育中,增加投喂富含单不饱和脂肪酸(MUFA)(特别是C18:1和C16:1)的饵料,以加强刀鲚亲本的营养积累和产卵后亲本生理机能的恢复;在刀鲚育苗前期,要及时补充富含DHA和EPA的饵料,如,单胞藻、蛋黄等,以提高刀鲚仔鱼开口期间的成活率。  相似文献

鱼类的受精卵在完成胚胎发育的正常过程后经过脱膜孵化成为仔鱼。此时,初孵仔鱼(newly hatched larvae)一般消化道尚未与外界相通,均有较大的卵黄囊作为孵化后一段时间内继续发育的营养来源。但随着卵黄物质的逐渐消耗,消化管与口腔接通以后,仔鱼必需从外界环境中摄取必要的营养物质,以便进一步发育与生长,这就是仔鱼的开口摄食期。    相似文献

研究对人工繁殖的北极茴鱼(Thymallus arcticus grubei)胚胎发育开展系统观察,记录分析其胚胎及仔鱼发育各时期的形态特征,旨在为北极茴鱼的人工繁育和种质资源保护提供必要的基础数据。结果显示,北极茴鱼受精卵呈圆球形,金黄色,沉性卵,未吸水卵径(2.46±0.14) mm,吸水卵径(3.14±0.18) mm,卵黄质内有多个油球。油球的数量和空间分布在胚胎发育过程中发生了规律性变化。在孵化水温(11.06±0.72)℃,溶氧8.3—9.8 mg/L条件下,历时301h完成整个胚胎发育过程,所需积温为3384.84h·℃,经历合子期、卵裂期、囊胚期、原肠胚期、神经胚期、器官形成期和孵化出膜7个阶段,共26个时期。北极茴鱼仔鱼尾鳍、胸鳍的分化和眼色素沉积在受精卵胚胎发育后期就已经完成,背鳍、腹鳍、臀鳍和脂鳍等在胚后发育过程中相继分化。其初孵仔鱼平均全长为(9.33±0.35) mm,仔鱼卵黄囊呈圆球形, 18日龄时卵黄囊和油球被完全消耗。其早期发育阶段(0—16日龄)的生长特性符合公式:y=0.0005x4–0.0201x3+0....  相似文献

通过观察日本七鳃鳗Lampetra japonica (Martens, 1868)胚胎外部形态和内部组织结构变化, 描述受精卵从卵裂至器官形成以及仔鱼孵出的发育阶段, 采用实验生态学方法研究卵黄囊期仔鱼的异速生长模式。结果表明: 日本七鳃鳗卵子为乳白色, 呈卵圆形; 受精卵卵裂方式为全卵裂; 胚胎发育过程主要包括卵裂期、囊胚期、原肠胚期、神经胚期、头凸期、孵出前期和孵出期, 历时11—12d; 初孵仔鱼为乳白色, 全长约(3.41±0.24) mm, 体质量约为0.0006 g。日本七鳃鳗胚胎发育研究可为了解七鳃鳗胚胎发育过程, 早期脊椎动物的起源和发育进化研究提供参考。卵黄囊期内仔鱼身体各部分中, 头长和尾长均表现出快速生长, 同在7 日龄出现生长拐点, 且生长拐点后的生长速率都大于生长拐点前的生长速率; 而仔鱼体长在卵黄囊期内表现出慢速生长。在头部器官中, 吻长、鳃前长和鳃长均表现出快速生长现象, 吻长和鳃长分别在9日龄和8日龄出现生长拐点; 口笠长在3日龄时出现生长拐点, 在生长拐点前为等速生长, 而在生长拐点后表现出快速生长; 眼径和眼鳃间距则分别表现出等速生长和慢速生长。泄殖孔在卵黄囊期内未出现生长拐点, 生长速率相对于全长生长速率表现出快速生长现象。七鳃鳗卵黄囊期仔鱼的异速生长是在长期进化过程中, 适应早期生活环境和独特的生活方式而形成特有的发育机制。  相似文献

本文研究和测定了江鲽( Platichthys flesus L.)在卵和卵黄囊期仔鱼发育阶段水分、钠、钾、脂肪和蛋白质的含量变化。含水量在卵和仔鱼发育期基本保持在90—92%,在仔鱼进入初次摄食期时可降到89%左右。初孵仔鱼具一大卵黄囊,含水量高达93.3%,起“浮力器官”(buoyancy organ)的作用。钠、钾离子含量在卵受精和仔鱼出膜后波动剧烈,呈“上升—下降—再上升”式型。脂肪和蛋白质含量在卵和仔鱼早期发育和饥饿期均呈明显线性下降。同时,本文还就生化成分变化和海洋浮性鱼卵及其仔鱼的生态习性的关系作了初步探讨。  相似文献

在胚胎发育中期,半滑舌鳎胚胎由胚体、卵黄囊和卵周液构成.对半滑舌鳎胚胎发育中后期的卵黄囊进行超微结构观察.结果表明,卵黄囊是由卵黄囊膜和包裹其内的卵黄物质组成.在半滑舌鳎胚胎发育过程中,卵黄囊内的卵黄物质逐渐消耗产生低分子量的卵黄磷蛋白分裂小泡.分裂小泡转移到卵黄囊内部消黄细胞中,在消黄细胞的作用下分裂小泡转化成卵黄颗粒.随后卵黄颗粒在卵黄囊内表面聚集成囊状结构并转移运输到卵黄囊膜内部,最后把卵黄物质从卵黄囊膜转移并释放到卵周液中,为胚胎发育提供营养.  相似文献

在水温23.4-24.0℃、盐度33.00、pH7.78-8.02的培养条件下,半滑舌鳎初孵仔鱼全长为3.55±0.161mm(n=60),依照公式4/3π.R/2.(r/2)2计算其卵黄囊体积为0.606±0.106mm3(n=60)。1日龄仔鱼,全长为4.99±0.211mm(n=10),卵黄囊体积较初孵仔鱼缩小近10倍,约为0.066±0.008mm3(n=10);2日龄,仔鱼的巡游模式建立,仔鱼全长为5.61±0.069mm,卵黄囊体积为0.030±0.002mm3(n=10),口初开;2.5日龄,口完全裂开,口裂0.24±0.024mm(n=10),仔鱼进入摄食期;3日龄,仔鱼的外源性摄食关系初步建立;4日龄,仔鱼的摄食率达100%,完成了内源性营养向外源性营养的转换;5日龄,仔鱼的卵黄完全被吸收,仅剩聚成一团的小油球,仔鱼的混合营养期持续2.5天时间;21日龄,稚鱼全长为12.96±0.611mm(n=11),仍有40%的个体残余的油球还没有完全被吸收,其体积仅为0.0000005±0.000003mm3(n=11)。仔鱼发育过程中,其长度的生长存在内源性营养阶段的快速生长、混合营养阶段的慢速生长以及饥饿期间的负生长三个生长期相,平均增长率为0.45mm/d,依照TL=aD3 bD2 cD d方程式对仔鱼的全长与日龄进行回归,其生长模型为TL(mm)=0.0026D3-0.0704D2 0.7993D 3.55(R2=0.9811,n=324)。仔鱼耐受饥饿的时间临界点发生在孵化后第10天(即9日龄)。仔鱼具有摄食能力的时间约6天,不可逆转饥饿期的时间约3天。残余的油球较长时间的存在,相对地延长了仔鱼混合营养期的时间,对仔鱼的发育、生长和存活有着至关重要的作用。5-20日龄的个体都具有胸角这个明显的形态学特征,只是饥饿个体和不可逆转饥饿期个体的胸角比摄食个体更为明显和尖锐,胸角不能作为区分健康仔鱼和饥饿仔鱼的形态学依据之一[动物学报51(6)1023-1033,2005]。  相似文献

用气相色谱仪分析了鲢受精卵和仔鱼的脂肪酸组成及其变化。受精卵中共检测到19种脂肪酸;SFA含量最高,其次为PUFA,MUFA最少。SFA中以16C:0为主,MUFA中以18C:1为主,PUFA中以DHA、AA和EPA为主;n-3/n-6为2.45。初孵仔鱼中HUFA因卵膜的脱落而减少,以后逐渐积累;鲢仔鱼具有将18C PUFA转化为长链HUFA的能力;在早期发育阶段,鲢仔鱼主要消耗16C:0作为能量代谢的基质。  相似文献

Fermentation at low pH is an efficient way to improve the competitiveness of biological succinic acid-producing process. Actinobacillus succinogenes shows good performance of succinic acid production under anaerobic conditions, but its succinic acid production capability at the low-pH is inefficient due to the poor acid resistance. Herein, a mutant A. succinogenes BC-4 with improved cell growth and succinic acid production under weak acid conditions was obtained by adaptive evolution. The specific growth rate and succinic acid production of BC-4 reached 0.13 g/L/h and 20.77 g/L, which were increased by 3.25- and 2.95- fold, respectively compared with the parent strain under anaerobic condition at pH 5.8. The activities of specific enzymes with ATP generation were significantly enhanced under weak acidic conditions, resulting in 1.28-fold increase in the maximum ATP level. Membrane fatty acid composition analysis demonstrated that the ratio of saturated to unsaturated fatty acids was decreased from 1.62 to 1.44 in mutant BC-4, leading to improved intracellular pH homeostasis. Furthermore, the change from long-chain to median-chain fatty acid might lower the permeability of H+ into cytoplasm for survival under acid stress. These results indicated that A. succinogenes BC-4 is a promising candidate for succinic acid production under weak acid condition.  相似文献

Predicting the cofactors of oxidoreductases plays an important role in inferring their catalytic mechanism. Feature extraction is a critical part in the prediction systems, requiring raw sequence data to be transformed into appropriate numerical feature vectors while minimizing information loss. In this paper, we present an amino acid composition distribution method for extracting useful features from primary sequence, and the k-nearest neighbor was used as the classifier. The overall prediction accuracy evaluated by the 10-fold cross-validation reached 90.74%. Comparing our method with other eight feature extraction methods, the improvement of the overall prediction accuracy ranged from 3.49% to 15.74%. Our experimental results confirm that the method we proposed is very useful and may be used for other bioinformatical predictions. Interestingly, when features extracted by our method and Chou's amphiphilic pseudo-amino acid composition were combined, the overall accuracy could reach 92.53%.  相似文献

基于最近邻居算法,从蛋白质一级序列出发,利用蛋白质序列氨基酸组成、二肤组成以及混合组成方法对蛋白质单聚体、二聚体、三聚体、四聚体、五聚体、六聚体和八聚体进行分类研究。结果表明:采用二肽组成编码方法的预洲效果最好,Jackknife检验和独立测试集检验的总体预测精度分别达到90.83%和95.48%,比相同数据集上基于伪氨基酸组成和组分耦合预测的方法提高了12和15个百分点;特别是对于五聚体蛋白,预测精度分别提高了90和50个百分点;说明二肽组成对于蛋白质四级结构分类研究是一种非常有效的特征提取方法。  相似文献

Because a priori knowledge of a protein structural class can provide useful information about its overall structure, the determination of protein structural class is a quite meaningful topic in protein science. However, with the rapid increase in newly found protein sequences entering into databanks, it is both time-consuming and expensive to do so based solely on experimental techniques. Therefore, it is vitally important to develop a computational method for predicting the protein structural class quickly and accurately. To deal with the challenge, this article presents a dual-layer support vector machine (SVM) fusion network that is featured by using a different pseudo-amino acid composition (PseAA). The PseAA here contains much information that is related to the sequence order of a protein and the distribution of the hydrophobic amino acids along its chain. As a showcase, the rigorous jackknife cross-validation test was performed on the two benchmark data sets constructed by Zhou. A significant enhancement in success rates was observed, indicating that the current approach may serve as a powerful complementary tool to other existing methods in this area.  相似文献

The objective of this study was to identify single nucleotide polymorphisms (SNPs) in the thioesterase (TE) domain of the bovine fatty acid synthase (FASN) gene and to evaluate the extent to which they were associated with beef fatty acid composition. The four exons in FASN that encode for the TE domain were sequenced, and three SNPs, AF285607:g.17924A>G, g.18663T>C and g.18727C>T, were identified. Purebred Angus bulls (n = 331) were classified into three genotype groups, g.17924AA (n = 121), g.17924AG (n = 168) and g.17924GG (n = 42). The g.17924A>G genotype was significantly associated with fatty acid composition of longissimus dorsi muscle of Angus bulls. Cattle with the g.17924GG genotype had lower myristic acid (C14:0; P < 0.0001), palmitic acid (C16:0, P < 0.05) and total saturated fatty acid contents (P < 0.01), greater health index (P < 0.001), oleic acid content (C18:1; P < 0.001) and total monounsaturated fatty acid concentration (P < 0.01) in the total lipids and triacylglycerols fraction than did those with the g.17924AA genotype. Because of the linkage disequilibrium between SNPs g.17924A>G and g.18663T>C, similar significant associations of fatty acid contents with the g.18663T>C genotypes were observed. In conclusion, the SNPs g.17924A>G and g.18663T>C may be used as DNA markers to select breeding stock that have a healthier fatty acid composition.  相似文献

白芷是我国常用中药材的一种,关于白芷镇痛的有效成分,主要认为是异欧前胡素、欧前胡素等香豆素类具有解痉、镇痛等作用的挥发油类化学成分,但白芷镇痛的成分及其机理尚不十分清楚,是否有其他物质也具有一定的药理作用也不得而知,比如说某些药效性氨基酸。该研究采用自动氨基酸分析仪测定了湖南道地茶陵白芷的蛋白质类氨基酸和游离氨基酸的含量,并分析其氨基酸组成。结果表明:总氨基酸检出除Asn以外的其他16种蛋白质氨基酸,含量最高的Arg占总氨基酸的31.21%,接近1/3;必需氨基酸总量达27.01%,其中含量最高的是Leu,占总必需氨基酸的24.14%;药效氨基酸比例较高,在酸水解产物总氨基酸和游离氨基酸中分别达到73.89%和85.78%,其中Arg的含量最高,达1.383g·100g-1,占比为42.24%,游离氨基酸中γ-氨基丁酸、鸟氨酸含量也较高。此外,还含有少量的高半胱氨酸和鹅肌肽等游离非蛋白质氨基酸和短肽;必需氨基酸的组成接近WHO/FAO的建议摄入值,但Met+Cys的RC值最小,为第一限制性氨基酸。这些药效性氨基酸和游离氨基酸可能是茶陵白芷具有良好药效的一个因素,而且茶陵白芷的氨基酸组成和配比较合理、符合人体需要,在强化含硫氨基酸的基础上具有开发成为新型药食同源保健性食品的潜力。研究结果可为进一步研究白芷的药理作用提供依据。  相似文献

The objectives of the experiment were to evaluate growth and carcass characteristics, organ weight, meat quality and intramuscular fatty acid (FA) and amino acid composition between the Chinese indigenous pig breed Dahe and the crossbred Dawu. The Dahe pigs had lower average daily gain (P < 0.001) and a higher feed conversion ratio (P < 0.001) compared with the Dawu pigs. The Dahe pigs contained less lean meat percentage (P < 0.001) and more carcass fat percentage (P < 0.001) compared with the Dawu pigs. For organ weight, the Dahe pigs had lower relative heart weight and small intestine weight, respectively, compared with that of the Dawu pigs (P < 0.001). In addition, the Dahe pigs showed higher pH values (at 45 min and 24 h, P < 0.001 and P < 0.001, respectively), higher Marbling score (P < 0.05), lower Minolta L values (at 45 min and 24 h, P < 0.001 and P < 0.05, respectively) and lower muscle fiber area (P < 0.05) than did the Dawu pigs. C18:1, C16:0, C18:0 and C18:2 were the main FAs and nine essential amino acids were found in the Longissimus dorsi of the two breeds.  相似文献

The fatty acids of the triacylglycerol fraction of the latex of the rubber plant consists of 97% of a C18 furanoid fatty acid, 10,13-epoxy-11-methyloctadeca-10,12-dienoic. The free fatty acid fraction is composed of a more equally distributed mixture of 16:0, 18:0, 18:1, 18:2 and the furanoid acid. A novel dioxo fatty acid, 10,13-dioxo-11-methyloctadecanoic, was also isolated and characterized.  相似文献

The amino acid compositions of proteins from halophilic archaea were compared with those from non-halophilic mesophiles and thermophiles, in terms of the protein surface and interior, on a genome-wide scale. As we previously reported for proteins from thermophiles, a biased amino acid composition also exists in halophiles, in which an abundance of acidic residues was found on the protein surface as compared to the interior. This general feature did not seem to depend on the individual protein structures, but was applicable to all proteins encoded within the entire genome. Unique protein surface compositions are common in both halophiles and thermophiles. Statistical tests have shown that significant surface compositional differences exist among halophiles, non-halophiles, and thermophiles, while the interior composition within each of the three types of organisms does not significantly differ. Although thermophilic proteins have an almost equal abundance of both acidic and basic residues, a large excess of acidic residues in halophilic proteins seems to be compensated by fewer basic residues. Aspartic acid, lysine, asparagine, alanine, and threonine significantly contributed to the compositional differences of halophiles from meso- and thermophiles. Among them, however, only aspartic acid deviated largely from the expected amount estimated from the dinucleotide composition of the genomic DNA sequence of the halophile, which has an extremely high G+C content (68%). Thus, the other residues with large deviations (Lys, Ala, etc.) from their non-halophilic frequencies could have arisen merely as "dragging effects" caused by the compositional shift of the DNA, which would have changed to increase principally the fraction of aspartic acid alone.  相似文献
