利用电子显微镜术,对蒜休眠进程中鳞片薄壁细胞间胞间联络的特征进行了实验观察,发现不同时期胞间联络具有随细胞间生理关系密切程度而呈现相应结构变化的特点。并观察到萌芽期鳞片中衰败细胞与存活细胞之间有类外连丝型胞间连丝的存在;以激光共聚焦荧光显微镜结合荧光标记物示踪检测,发现不透膜荧光物质分子量为457Da的萤黄(Lucifer yellow,LYCH),可以共质体运输方式进入存活细胞内,论证了类外连丝这一胞间连丝特定修饰态的存在,并可在一段时间内继续保持生理活性,起到进行物质共质运输的功能。  相似文献

胞间连丝是植物体内连接两个相邻细胞原生质体的共质运输通道,在胞间物质的转运和通讯联络上发挥重要作用。胞间连丝的功能在生理上主要体现在它对胞间转运物质的通透性(permeability),通透性的变化和调节影响到许多生理过程的进展与协调。在植物的不同组织及其发育的不同阶段,不同的物化因素,不同的逆境胁迫以及病原物的侵染均可导致胞间连丝的通透性呈现相应的变动。胞间连丝存在形式的多样性以及对其不同程度和不同方式的修饰,均可对胞间连丝的生理功能及其通透性有明显的影响。[第一段]  相似文献

以蒜 (AlliumsativumL .)瓣鞘外表皮为材料 ,利用荧光特异探针与共焦镜检术 ,结合透射电镜与免疫金标记对表皮细胞间胞间联络的性质、结构进行了系统观察。结果表明 ,加厚壁上的通道是由狭长的管状胞质和初生纹孔场上成束的胞间连丝衔接组成 ,前者实为原生质体的一部分。单个胞间连丝的孔径为 6 0~ 70nm ,属正常胞间连丝范围 ,它们乃相邻细胞间共质联系的所在。荧光探针TRITC_Phalloidin (TRITC_Ph)标记的结果显示 ,整个通道上呈现红色荧光的纤索在接近初生纹孔场处明显变窄 ,与超微结构观察中所见的结构特点相吻合 ,共焦镜下观察到的初生壁上的荧光亮斑乃初生纹孔场中成束胞间连丝被标记的反映 ,从而有效地证实了F肌动蛋白在常态胞间连丝上的存在。免疫金标记实验显示在管状胞质中和胞间连丝上有金颗粒分布 ,这一结果为证实荧光标记物具肌动蛋白性质提供了有说服力的补充。  相似文献

外连丝作为特化的通道结构存在于植物的表皮细胞外侧壁中,而在植物局部组织衰败过程中衰亡细胞与存活细胞分界壁上有类似外连丝的半胞间连丝存在,可称之为类外连丝。本对外连丝和类外连丝的结构、功能方面的研究结果与近年进展予以综述,并讨论了植物细胞胞壁上类外连丝的出现在共质运输和细胞内含物再分配中的可能意义。  相似文献

以贮存蒜苔为材料,研究了在茎苔衰退与顶端珠蒜成长过程中物质的再分配,及其在显微、亚显微结构上的反映。观察到类 P-蛋白丝状物在衰退细胞中的形成及其经胞间连丝在薄壁细胞间、薄壁细胞与韧皮部之间进行运转的多种迹象,提出了大分子物质以集装囊泡的形式进行运输的新论点。进一步发现大分子物质在质外体(导管与细胞间隙)中存在与迁移状态;初步论证了它们与导管堵塞物在质上的差异;指出在特定生理状态下,质外体提供的“自由空间”作为大分子物质主动迁移途径的可能性。  相似文献

应用透射电镜技术研究了宁夏枸杞果实韧皮部细胞的超微结构变化。结果表明:(1)随着枸杞果实的发育成熟,果实维管组织中的韧皮部筛分子筛域逐渐变宽,筛孔大而多,通过筛孔的物质运输十分活跃;筛分子和伴胞间有胞间连丝联系,伴胞属传递细胞类型,与其相邻韧皮薄壁细胞和果肉薄壁细胞连接处的细胞界面发生质膜内突,整个筛分子/伴胞复合体与韧皮薄壁细胞之间形成共质体隔离,韧皮部糖分的卸载方式主要以质外体途径进行。(2)韧皮薄壁细胞间的胞间连丝较多,而韧皮薄壁细胞与果肉薄壁细胞的胞间连丝相对较少,但果肉薄壁细胞间几乎无胞间连丝;果肉薄壁细胞之间胞间隙较大,细胞壁和质膜内突间形成较大的质外体空间,为质外体的糖分运输创造了条件。(3)筛管、伴胞、韧皮薄壁细胞和果肉薄壁细胞中丰富的囊泡以及活跃的囊泡运输现象,暗示囊泡也参与了果实糖分的运输过程。研究推测,枸杞果实韧皮部同化物的卸载方式以及卸载后的同化物运输主要以质外体途径为主。  相似文献

Summary Earlier work in our laboratory indicated that protoplasmic constituents can migrate from one cell to another in certain tissues of higher plants. Further investigations have been conducted using garlic bulbs and wheat nucellus for microscopic observation of intercellular protoplasmic movement in vivo. These gave preliminary indications of the dynamic characteristics of migrating nuclei and cytoplasm. The present paper gives recent results providing new evidence for intercellular protoplasmic movement that is neither hindered by the presence of cell wall nor the narrowness of channels of intercellular connection. By careful manipulation, intact endosperm sacs could be taken from developing caryopses (6–8 days after fertilization) without apparent injury to constituent cells. Shortly after the living specimen is mounted on the microscopic stage, asynchronous intercellular protoplasmic movement can be observed here and there. It can be seen that protoplasm extrudes in rapid but intermittent movements from one cell to the next by vigorous contraction. Although various cell constituents may move together, they can also be quite independent of each other. The moving units, though undergoing violent deformation, resume their normal shape and structure following intercellular migration. Evidently this kind of movement is a naturally occurring and active phenomenon closely related to the physiological state of the tissue. Electron microscopic studies reveal that a limited number of plasmodesmatal channels undergo modification and enlarge to 100–400 nm, through which the protoplasmic constituents pass.  相似文献

The oocytes of saturniid moths take up proteins selectively from the blood. The distribution of blood proteins in the ovary during protein uptake was investigated by staining 2 µ sections of freeze-dried ovaries with fluorescein-labeled antibodies. The results indicate that blood proteins occur primarily in the intercellular spaces of the follicle cell layer, in association with a brush border at the surface of the oocyte, and within the oocyte in the yolk spheres. That proteins derived from the blood are associated with the yolk spheres was confirmed by isolating these bodies and showing that lysis, which can be induced by any of a number of mechanical means, causes them to release immunologically defined proteins known to be derived from the blood. That the level of blood proteins in the cytoplasm is low relatively to that in the yolk spheres was confirmed by the observation that the yellow pigments associated with several blood proteins, although conspicuous in the yolk spheres, are not visible in the translucent layer of centrifuged oocytes. From these and previous physiological observations, it is proposed that blood proteins reach the surface of the oocyte by an intercellular route, that they combine with some component of the brush border, and that they are transformed into yolk spheres by a process akin to pinocytosis.  相似文献

在亲本受孕时生理状态与代性比的关系上存在着多种理论。在1988-1992年期间,作者用实验对其中的营养理论、氧化还原理论、酸碱理论和激素假说的部分观点进行了检验。结果表明:这些理论在某种速度上均具有合理性。作者还发现:这些理论之间是相互联系的。用一系列生理指标来描述双亲受孕时影响子代性比的生理状态,对于通过调节配种时双亲的生理状态来控制子代性比具有重要意义。  相似文献

Tolbert , Robert J. (West Virginia U., Morgantown.) A seasonal study of the vegetative shoot apex and the pattern of pith development in Hibiscus syriacus. Amer. Jour. Bot. 48(3): 249–255. Illus. 1961.—The shoot apex of Hibiscus syriacus L. is described as having a cytohistological zonation superimposed on a tunica-corpus configuration. The apex is flat-topped or may have a saddle-back or concave appearance as seen in median longitudinal section. The metrameristem, consisting of the tunica and corpus initials, is comprised of large, light-staining, vacuolate cells that have thick cell walls and exhibit much dark-staining intercellular substance. Surrounding the metrameristem is the flanking meristem, which is responsible for the outer layers of the shoot, and from which the leaf primordia arise. The pith rib meristem lies below the metrameristem and consists of files of cells that are responsible for the pith. There are no major seasonal changes in the structure of the apex during the yearly cycle. The pith displays a long-shoot type of development with the cells remaining in distinct files during the first flush of growth in the spring. As growth slows and internode elongation is gradually reduced, the pith displays the characteristic short-shoot type of development, consisting of a spongy tissue of rounded cells with many intercellular spaces and no distinct files of cells. A crown is differentiated across the top of the pith at the end of the growth period. This consists of a band of cells with thick, dark-staining cell walls, which separates by the apex from the last year's growth. In contrast to many gymnosperms, this crown is dispersed by renewed cell activity the following spring.  相似文献

宁夏枸杞(Lycium barbarurn L.)花药发育过程中,淀粉粒和脂滴两种营养物质的积累和分布具有一定的特点:在造孢细胞时期,药隔薄壁细胞,表皮和药室内壁细胞中开始积累淀粉粒,而造孢细胞、绒毡层细胞和中层细胞中则没有淀粉粒。在四分体时期,绒毡层细胞开始积累脂滴并且数量逐渐增加。到小孢子晚期,绒毡层细胞降解,内含脂滴流入药室中。在小孢子发育过程中既没有淀粉粒也没有脂滴积累,直到二胞花粉的大液泡消失后花粉粒中才开始积累脂滴,然后又开始出现淀粉粒。枸杞成熟花粉中的营养储存物是脂滴和淀粉粒。  相似文献

气候室中研究了根温对苋菜幼苗生长及光合等生理特性的影响。结果表明,苋菜幼苗生长曲线呈S形, 30d苗龄时生长最快, 40d苗龄时每株干、鲜重分别为0.45g和6.79g。20~25℃根温时40d苗龄苋菜生长最快,高根温对生长的危害大于低根温, 40℃根温苋菜根系生长、代谢受害严重。苋菜净光合速率(Pn)、蒸腾速率(Tr)、气孔导度(Cs)和胞间CO2分压(Pi)随根温的变化趋势基本一致, 20~25℃根温时,Cs、Pi、Pn和Tr均高,不良根温影响了Cs,使Tr降低, Pi降低,进而影响到Pn。回归分析表明, Pn与Cs、Pn与Pi、Tr与Cs、Pi与Cs均呈显著正相关。说明Cs下降是不良根温影响Pn的主要原因。  相似文献

Rat leukemia cells IRC 741 in suspension culture form single cytoplasmic protrusions by which the cells preferentially adhere to one another. The induction and/or maintenance of these protrusions is sensitive to changes in intercellular contact, pH, temperature, and nutritional conditions. The protrusions are stable, rigid structures which take part in intercellular adhesion but not in adhesion to substrata. Movement on substrata occurs by means of ruffling membranes formed on the main cell body. This asymmetry in cellular form and function is associated with specialized cell surface regions. Ultrastructurally, the cell surface over the protrusions lacks microvilli, and is covered with a 3,000–4,000-Å thick cell coat consisting of 200–500-Å electron-dense particles in an amorphous matrix. In contrast, the surface over the main cell body has microvilli and a 200-Å wide cell coat which lacks particles. The extracellular particles overlying the protrusions have electron-lucent cores, are protease- and pepsin-resistant, and do not stain with colloidal iron, while the matrix in which they are embedded is sensitive to proteolytic enzymes and contains acidic moieties. The negative surface charge density over the protrusions is higher than that over the main cell body, as shown by the orientation of the cells in an electric field. The unexpected observation that a region of higher charge density is one of increased intercellular adhesiveness might be explained, in part, by the rigidity of the protrusions and by the very small radius of curvature of the overlying extracellular particles. The protrusions permit the observation of discrete regions, differing in charge density, on the surface of living leukemia cells.  相似文献
