啤酒酵母的自溶会影响啤酒的风味和品质,对酵母自溶的调控是工业啤酒生产的迫切需求。前期在啤酒酵母自溶的研究中发现柠檬酸循环相关基因对酵母自溶有较大影响。为探究柠檬酸循环中异柠檬酸脱氢酶基因调控对自溶的影响,在典型拉格啤酒酵母(Saccharomyces pastorianus H.)Pilsner中对IDP1和IDP2基因进行了破坏和过表达。结果发现IDP1基因的破坏能提高酵母的抗自溶能力,自溶96 h抗自溶指数为8.40,比原始菌提高了1.5倍。IDP1基因的破坏提高还原型辅酶Ⅱ(nicotinamide adenine dinucleotide phosphate,NADPH)的供应,NADPH/NADP^(+)的比值为1.94,发酵结束时胞内ATP水平比原始菌提高1.8倍,活性氧(reactive oxygen species,ROS)相比原始菌减少10%。IDP2基因的缺失造成酵母快速自溶和NADPH供应的减少,自溶96 h抗自溶指数为4.03,NADPH/NADP^(+)的比值为0.89。发酵结束时胞内ATP水平相比原始菌减少8%,ROS是原始菌的1.3倍。本研究进一步阐释了柠檬酸循环相关基因对酵母自溶的调控机理,为选育自溶性能可控的优良酵母提供了理论基础。  相似文献

酵母被誉为啤酒酿造的灵魂,然而随着啤酒高浓酿造技术的发展,酿造过程中渗透压增加、乙醇含量升高及营养平衡改变等会加快酵母的自溶,对啤酒的风味品质产生不利的影响。为提高酵母的抗自溶能力,本研究构建了以酿酒酵母18S r DNA序列为同源位点的FKS1过表达菌株。结果表明,过表达菌株细胞壁葡聚糖含量较原菌高62%;通过平板耐受性分析可知,FKS1过表达菌株在8%的乙醇浓度、0.4 mol/L Na Cl的渗透压冲击以及24 h饥饿培养的条件下,其胁迫耐受性均高于原始菌株;模拟自溶实验结果显示FKS1过表达菌株自溶速度缓慢,抗自溶能力明显优于原始菌株。该结果有助于探究酵母自溶的机理,同时也对提高啤酒风味品质和稳定性有着重要的意义。  相似文献

拉格啤酒酵母是我国啤酒酿造的主要菌种。细胞絮凝是啤酒酵母重要的生产性状,在不影响发酵性能的情况下适度提高酵母的絮凝能力,有助于发酵结束时细胞和产物的分离,有利于工业化啤酒生产,具有较高的经济价值。前期在对一株工业用拉格啤酒酵母G03及其絮凝突变株的研究中,挖掘到一个可能影响啤酒酵母絮凝性的候选基因RIM21。为了验证该基因的作用,文中在G03中对RIM21进行了敲除,发现RIM21敲除后,酵母在11 ℃发酵条件下的絮凝性能增强,基因FLO5、Lg-FLO1及细胞壁完整性途径中的部分基因表达上调。同时,CO2失重、酒精度、发酵度等发酵指标未有明显变化。另外,发现RIM21的缺失增强了啤酒酵母对细胞壁抑制剂的耐性。研究结果为阐释低温发酵条件下啤酒酵母的絮凝调控机理及菌株絮凝性的改善提供了基础。  相似文献

构建一株酿酒酵母(Saccharomyces cerevisiae)NST1基因缺失菌株并研究其对抗氧能力的影响。以酿酒酵母BY4741中的NST1基因为研究对象,利用Cre-LoxP基因敲除系统将NST1基因与G418抗性基因(kan~r)相替换,实现目的基因的敲除。通过稀释点样实验、荧光电子显微镜检测和荧光定量PCR等技术分析NST1基因在酿酒酵母抗氧化体系中的作用。经过G418抗性筛选和基因组PCR鉴定,成功获得了NST1基因缺失菌株nst1Δ,实验数据显示nst1Δ重组菌胞内ROS增多,并且降低了细胞壁完整性信号通路(CWI)途径下游基因RLM1的表达水平,表明NST1基因的敲除对酿酒酵母BY4741的抗氧化性能有影响。  相似文献

啤酒酿造过程中正常情况下的酵母不会发生自溶,但如果细胞衰老死亡或操作不当则会出现酵母自溶现象.虽然自溶的细胞在生产中占的比例较小,但自溶后的酵母将胞内物质释放到酒液中,产生酵母味,严重影响了啤酒的口感和外观质量.早期研究认为酵母细胞壁在酵母自溶过程中不发生水解,即自溶后的细胞仅剩下空的细胞外壳.现在研究发现酵母在自溶时其细胞壁也会发生水解作用,葡聚糖酶将构成细胞壁成分的葡聚糖进行分解,分解产物最终释放到酒液里和细胞质内物质共同影响啤酒的质量.  相似文献

SOD1基因编码的铜锌超氧化物歧化酶是酵母细胞中最重要的抗氧化酶.前期研究发现,SOD1基因缺失(sod1Δ)导致酵母细胞对真菌细胞壁抑制剂刚果红(Congo red,CR)的敏感性增加,提示细胞抗氧化能力与细胞壁稳定性相关.本研究采用酵母全基因组表达谱芯片,比较了CR胁迫条件下,野生型酵母细胞和sod1Δ酵母细胞的转录表达谱.结果表明,与野生型酵母细胞相比,sod1Δ酵母细胞中260个基因发生了显著差异表达(140个基因表达上调、120个基因表达下调).随机选取12个差异表达基因采用定量PCR验证,结果与芯片分析结果一致.差异表达基因功能主要涉及细胞壁(几丁质合成)、细胞代谢、细胞防御(抗氧化和热冲击蛋白)、蛋白质合成以及大量功能未知基因.进一步研究发现,CR处理后,细胞壁几丁质含量和细胞内氧化应激指标丙二醛(MDA)含量在sod1Δ酵母细胞中显著升高,而在野生型酵母细胞中无明显变化,与芯片筛选差异表达基因的生物学功能分析结果一致.本研究提供了在全基因组水平上对SOD1基因与细胞壁应激反应之间关联的新认识.  相似文献

【目的】在不影响酵母正常代谢前提下,构建亚硫酸盐分泌量提高的基因工程菌株,增加二氧化硫生成量,有效地解决啤酒老化问题。【方法】以适量高产二氧化硫工业啤酒酵母突变株M8总DNA为模板,PCR方法得到带有不同长度5′端非编码区的基因片段SSU1-1、SSU1-2,以大肠杆菌-酿酒酵母穿梭质粒YEp352构建表达载体pSU1和pSU2,转化实验室酵母YS58,验证SSU1多克隆表达对其二氧化硫生成量的影响。进而将pSU2转化工业啤酒酵母M8,利用亚硫酸盐抗性筛选转化子,并对其二氧化硫和硫化氢生成量及其啤酒抗老化性能进行测定和分析。【结果】实验室酵母转化子pSU1-4和pSU2-3二氧化硫生成量较原株明显提高而硫化氢生成量基本不变,工业啤酒酵母转化子Y2二氧化硫生成量比原株M8提高74.4%,TBA值下降14.9%,DPPH自由基清除率提高38.2%,硫化氢生成量基本不变。【结论】SSU1基因的多拷贝表达有效提高了亚硫酸盐转运蛋白Ssu1p表达量,增加了亚硫酸盐分泌量,啤酒抗氧化能力得到明显增强,而对酵母硫代谢途径中亚硫酸盐还原为硫化物代谢过程没有影响。  相似文献

该文鉴定了一个来自黄酒酵母菌株D2的新基因g5170,并研究了其对黄酒酵母环境胁迫耐受性与发酵性能的影响。生物信息学分析发现,此基因编码的蛋白质含有酰胺酶(amidase)保守功能域,并在YPD培养条件下有较高的基因表达量。利用Cre/lox P系统构建了基因g5170的敲除菌株,通过扩增带有g5170基因上下游同源序列和kanr筛选标记的基因敲除组件,并通过醋酸锂法转化到黄酒酵母菌株D2获得阳性克隆子,然后将质粒p SH65转到阳性克隆子中,半乳糖诱导p SH65表达Cre酶切除kanr筛选标记。重复此实验过程,最终获得两个g5170基因拷贝完全缺失菌株D2?g5170。梯度生长实验显示,与原始菌株相比,敲除菌株对高糖浓度、高温耐受性无明显变化,但对乙醇胁迫的耐受性显著降低,而过表达g5170的重组酵母菌株对乙醇的耐受性明显比对照菌株强。模拟黄酒发酵实验结果表明,敲除菌株D2?g5170的生长速率、乙醇产量及理化指标与原始菌株相似。因此,g5170基因可能与黄酒酵母适应乙醇胁迫有关,并有助于提高酵母的黄酒发酵性能。  相似文献

啤酒酵母是啤酒酿造的灵魂,可以直接影响啤酒品质。在啤酒酿造过程中,由于啤酒酵母被多次传代和保藏,造成优良菌种发酵性能衰退等问题,导致发酵不彻底,影响最后啤酒的风味质量。为此以8株Lager型啤酒酵母为出发菌株,通过平板分离纯化获得80株分离菌株,再经过三角瓶发酵初筛和复筛、发酵罐中试发酵实验最终获得了8株发酵性能优良的啤酒酵母。其中,6株酵母可应用于酿造双乙酰含量低于0.1 mg/L的啤酒;3株酵母发酵度高于70%,适合酿造干啤酒;1株酵母发酵度低于50%,适合酿造低醇啤酒。在风味方面:1株酵母酿造的啤酒醇酯比为3.3,啤酒酯香味较突出;另1株酵母酿造的啤酒醇酯比为4.5,啤酒高级醇含量较高。8株经过选育的啤酒酵母发酵特征明显,便于精酿啤酒厂实际应用。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Seven strains of aerobic carbon monoxide-oxidizing bacteria (carboxydebacteria) when growing on CO as sole source of carbon and energy had doubling times which ranged from 12–42 h. The activity profiles obtained after discontinuous sucrose density gradient centrifugation indicated that the CO-oxidizing enzymes are soluble and the hydrogenases are membrane-bound in all strains examined. The CO-oxidizing enzymes of Pseudomonas carboxydohydrogena, Pseudomonas carboxydoflava, Comamonas compransoris, and the so far unidentified strains OM2, OM3, and OM4 had a molecular weight of 230,000; that of Achromobacter carboxydus amounted to 170,000. The molecular weights of the CO-oxidizing and H2-oxidizing enzymes turned out to be identical. The cell sonicates were shown to catalyze the oxidation of both CO and H2 with methylene blue, thionine, phenazine methosulfate, toluylene blue, dichlorophenolindophenol, cytochrome c or ferricyanide as electron acceptors. Methyl viologen, benzyl viologen, FAD+, FMN+, and NAD(P)+ were not reduced. The spectrum of electron acceptors was identical for all strains tested. Neither free formate, hydrogen nor oxygen gas were involved in the CO-oxidation reaction. Methylene blue was reduced by CO at a 1:1 molar ratio. The results indicate that CO-oxidation by carboxydobacteria is catalyzed by identical or similar enzymes and that the reaction obeys the equation CO+H2OCO2+2H++2e- as previously shown for Pseudomonas carboxydovorans.Dedicated to Otto Kandler remembering almost three decades of enjoyable cooperation  相似文献

Since the introduction of the concepts of allostery about four decades ago, much advancement has been made in elucidating the structure-function correlation in allostery. However, there are still a number of issues that remain unresolved. In this review we used mammalian pyruvate kinase (PK) as a model system to understand the role of protein dynamics in modulating cooperativity. PK has a triosephosphate isomerase (TIM)(α/β)8 barrel structural motif. PK is an ideal system to address basic questions regarding regulatory mechanisms about this common (α/β)8 structural motif. The simplest model accounting for all of the solution thermodynamic and kinetic data on ligand-enzyme interactions involves two conformational states, inactive ET and active ER. These conformational states are represented by domain movements. Further studies provide the first evidence for a differential effect of ligand binding on the dynamics of the structural elements, not major secondary structural changes. These data are consistent with our model that allosteric regulation of PK is the consequence of perturbation of the distribution of an ensemble of states in which the inactive ET and active ER represent the two extreme end states. Sequence differences and ligands can modulate the distribution of states leading to alterations of functions. The future work includes: defining the network of functionally connected residues; elucidating the chemical principles governing the sequence differences which affect functions; and probing the nature of mutations on the stability of the secondary structural elements, which in turn modulate allostery.  相似文献

对我国花型Hemibarbuspattern鱼类作了骨骼系统比较,结果表明,此类型鱼类脑颅较长,副蝶骨平直或稍弯曲,眶蝶骨腹纵嵴发达(铜鱼Coreius septentrionalis例外),下颞窝和咽突中等大,基枕骨后突发达;脑颅中的上筛骨的后突、侧筛骨的外筛突,蝶耳骨的外突、上耳骨的后突、围眶骨和后颞窝等均有明显的差异;咽颅中的舌颌骨、尾舌骨、鳃盖骨和下咽齿的列数等又有显著的区别;附肢骨骼中的腰带骨、脊椎骨中的复合神经骨和第4椎骨腹侧的悬器等也有不同之处。据此,这些差异和区别可作为属间或种间的分类依据。  相似文献
