外源有机酸对镉胁迫下秋华柳镉积累特征的影响   总被引:2,自引:0,他引:2

为探究外源有机酸在加强秋华柳(Salix variegata Franch.)镉积累效率中的应用潜力,采用营养液培养方法,研究了5种100μmol/L外源有机酸(草酸、柠檬酸、酒石酸、苹果酸和琥珀酸)对50μmol/L Cd胁迫下秋华柳生长适应性及Cd积累特征的影响,并通过化学平衡程序VISUAL MINTEQ v3.0计算溶液中不同化学形态Cd(游离态和螯合态Cd)的含量。结果表明:柠檬酸、酒石酸、苹果酸和琥珀酸的添加有效缓解了Cd对秋华柳的毒害,明显促进了秋华柳的生长。除草酸处理组外,其余4个有机酸处理组培养液中游离Cd~(2+)含量均得到较大提升,显著促进了秋华柳对Cd的吸收。柠檬酸、酒石酸、琥珀酸和苹果酸的添加显著提高了秋华柳植株的Cd积累量,分别是未加入有机酸Cd处理组的210%、190%、190%和178%。外源有机酸的施加提高了介质中游离Cd~(2+)的含量,并通过与重金属的络合等作用提升了植株对Cd的吸收和积累能力。但不同有机酸对秋华柳Cd积累特征的影响差异明显,柠檬酸加入后通过提高秋华柳根生物量和根中Cd含量,显著增加了根系Cd积累量;酒石酸、苹果酸和琥珀酸的应用则明显提高了秋华柳地上部分Cd积累量。因此,外源酒石酸、琥珀酸和苹果酸的应用可提高秋华柳地上部分的Cd积累效率,更有利于对Cd污染土壤的修复。  相似文献

为探讨从紫色小白菜(Brassica campestris L.ssp.chinensis(L.)Makino var.communis Tsen et Lee)叶片中提取花青苷的最佳工艺条件,在超声功率为420 W、40%乙醇和两次超声提取条件下,采用Box-Behnken的中心组合试验设计原理,设计4因素3水平试验,研究了pH、料液比、超声温度、超声时间对花青苷得率的影响。结果表明,紫色小白菜叶片花青苷提取的优化工艺参数为料液比1∶16(pH 2.76),在超声功率为420 W、温度为58℃下超声12 min,花青苷的提取得率可达4.11 mg g–1 DW。  相似文献

分别通过不同有机酸为惟一碳源,研究了产朊假丝酵母(Candida utilis)能够利用的有机酸的种类,旨在为微生物的基础研究和应用基础研究提供部分依据。分别通过对培养时间和菌体在不同有机酸合成培养基中OD值进行统计分析,发现培养时间和柠檬酸、乳酸、琥珀酸、L-苹果酸培养基的OD值极显著相关(P〈0.01);培养时间和乙酸、酒石酸、富马酸和草酸培养基的OD值无相关性。通过对比菌体培养前后有机酸合成培养基pH值的变化发现乙酸、酒石酸、富马酸和草酸合成培养基的pH值没有明显变化,而苹果酸、乳酸、琥珀酸和柠檬酸为碳源的合成培养基的pH值均明显增大。从而说明产朊假丝酵母能利用L-苹果酸、乳酸、琥珀酸、柠檬酸为碳源,而不能利用乙酸、酒石酸、草酸、富马酸为碳源。  相似文献

菠萝叶片中有机酸含量、转氨酶和脱氢酶活性的昼夜变化   总被引:2,自引:0,他引:2  
研究了菠萝叶片中苹果酸、柠檬酸、草酰乙酸、α-酮戊二酸和草酸等有机酸和转氨酶、脱氢酶活性的昼夜变化。结果表明,除了草酸外,其他4种有机酸的含量都呈白天降低晚上增高的趋势。有机酸的含量在上午9∶00时有最高值,下午18∶00时有最低值。苹果酸、柠檬酸、草酰乙酸、α-酮戊二酸和草酸的昼夜最大差值分别为140、61、31.8、8.2和6.7mmol·kg-1FW。异柠檬酸脱氢酶和琥珀酸脱氢酶活性的昼夜变化与有机酸的趋势一致,白天降低晚上增高。可溶性蛋白和游离氨基酸含量以及谷草转氨酸、谷丙转氨酶活性在下午18∶00时有最高值,凝胶电泳显示菠萝叶片中有新的蛋白带出现。  相似文献

以抗病性不同的一对水稻近等基因系——感病CO39和抗病C101LAC(Pi-1)为实验材料,在水培条件下研究了施硅和稻瘟病接菌对水稻根系和叶片抗性物质有机酸含量的影响,揭示硅提高水稻对稻瘟病抗性的机理。结果表明:接菌条件下硅处理显著降低了2个基因型材料的叶片反丁烯二酸、柠檬酸的含量,而增加了叶片中草酸、顺丁烯二酸的含量;加硅显著降低接菌后第3天的CO39叶片酒石酸含量,但增加了C101LAC(Pi-1)接菌后第7天的叶片酒石酸含量;硅处理还显著增加了根系中苹果酸和草酸的含量;各种有机酸在水稻植株体内的分布也不尽相同,如柠檬酸主要分布在叶片中,苹果酸主要分布在根系中,顺丁烯二酸、反丁烯二酸、酒石酸和草酸在叶片和根系中都有分布。研究表明,硅可能通过影响植株体内的有机酸代谢而增强稻瘟病的抗性。  相似文献

比较洗根法、层析纸法和琼脂膜法收集土培条件下生长的菌根化和非菌根化三叶草根分泌物的效果。试验采用三室根箱装置 ,将根系与菌丝生长空间分开 ,三叶草生长 5 6d后 ,打开三室根箱装置 ,由于尼龙网的阻挡作用使根系均匀垫积在尼龙网内侧并形成根垫。分别采用洗根法、层析纸法和琼脂膜法 3种方法收集三叶草根系分泌物 ,并通过高效液相色谱方法测定分泌物中草酸、酒石酸、苹果酸、乳酸、乙酸、顺丁烯二酸、反丁烯二酸、柠檬酸、丁二酸等有机酸的含量。结果表明 :3种收集方法收集的三叶草分泌的有机酸无论在种类上还是在数量上都存在相当大的差别。从检测到的有机酸种类来看 ,琼脂膜法收集检测到苹果酸、乙酸、顺丁烯二酸、柠檬酸、丁二酸和乳酸 6种有机酸 ;洗根法收集的分泌物检测到酒石酸、苹果酸、乙酸、顺丁烯二酸、柠檬酸、丁二酸和乳酸 7种有机酸 ;层析纸法收集的分泌物检测到酒石酸、苹果酸、柠檬酸和乳酸 4种有机酸。从收集到的有机酸数量来看 ,洗根法收集到的有机酸总量为 2 9.97~ 2 32 .7μg/( gfw· 2 h) ;琼脂膜法集到的有机酸总量为 1 .5~ 7.3μg/( cm2· 2 h) ;层析纸法收集的有机酸总量为0 .2 3~ 6.5 8μg/( cm2 · 2 h)。丛枝菌根真菌侵染对三叶草根系分泌的有机酸的组分和含量都有一  相似文献

采用室内溶液培养法,运用气相色谱-质谱联用(Gas chromatography-mass spectrometry,GC-MS)技术测定在As(Ⅲ)、As(Ⅴ)和二甲基胂酸(Dimethylarsinic acid,DMA)3种形态砷(浓度0—5.0 mg/L)胁迫下,砷超富集植物黑藻(Hydrilla verticillata(Linn.f.)Royle.)和非砷超富集植物竹叶眼子菜(Potamogeton malaianus Miq.)体内生成和体外分泌的7种有机酸的含量。结果表明,黑藻体内草酸、丙二酸、柠檬酸、苹果酸和棕榈酸都显著高于竹叶眼子菜(P<0.05)。在As(Ⅲ)处理时,黑藻体内柠檬酸、苹果酸、棕榈酸、亚麻酸和总有机酸显著下降,竹叶眼子菜棕榈酸显著下降;在As(Ⅴ)处理时,黑藻棕榈酸和总有机酸显著下降,竹叶眼子菜棕榈酸显著下降;在DMA处理时,黑藻草酸、柠檬酸和苹果酸显著下降,竹叶眼子菜棕榈酸、亚麻酸和总有机酸显著下降。黑藻在As(Ⅲ)处理下棕榈酸、亚麻酸、总有机酸和As(Ⅴ)处理下草酸含量与体内As含量呈显著负相关(P<0.05),而竹叶眼子菜在As(Ⅲ)或DMA处理下棕榈酸、亚麻酸、总有机酸含量以及在DMA处理下柠檬酸含量与体内As含量均呈显著负相关(P<0.05)。两种植物根系分泌物中均检测出草酸、丙二酸、琥珀酸和棕榈酸,但只有在As(Ⅲ)或As(Ⅴ)处理时黑藻分泌的草酸与体内As含量呈显著正相关。本研究表明As(Ⅲ)或As(Ⅴ)胁迫下黑藻向体外分泌草酸是其超富集砷的一种重要机制。  相似文献

改进了根系分泌有机酸的前处理和高效液相色谱检测方法.将DEAE纤维素层析法直接用于根系分泌物中有机酸的富集浓缩,并与旋转蒸发浓缩的回收率进行了比较.结果表明,DEAE纤维素柱层析有较高的回收率,可用于根系分泌物中有机酸的富集浓缩,特别适用于大批量样品的前处理.确定了反向高效液相色谱同时检测草酸、酒石酸、马来酸、苹果酸、乙酸、柠檬酸、琥珀酸等7种有机酸的条件.玉米根系分泌物样品中可检出苹果酸等5种有机酸.  相似文献

该研究建立了植物根茎叶及根系分泌物中有机酸的离子色谱分析测定方法,并测定了4种不同植物幼苗根茎叶及根系分泌物中低分子量有机酸的组成,为揭示逆境胁迫下植物体内有机酸的作用提供依据。结果表明:离子色谱分析法对植物有机酸的加标回收率为91.10%~105.42%,检测限为0.12~0.36mg/L,方法线性关系良好(R2=0.965 3~0.998 8);4种植物根茎叶及根系分泌物中都可以检测出草酸、柠檬酸、苹果酸、丁二酸和酒石酸,其中草酸、柠檬酸和苹果酸为优势酸;有机酸的组成和含量具有物种以及器官的差异性;根系分泌物中的有机酸与根茎叶中有机酸的相关性也因种属差异而不同。这为研究逆境胁迫下植物器官及根系分泌物提供了可靠方法。  相似文献

以12个烤烟品种为材料,通过测定烤烟中部叶有机酸总量、苹果酸含量、柠檬酸含量、代谢相关酶活性的动态变化以及烤后烟碱含量和感官质量,探讨烤烟生长过程中叶片主要非挥发性有机酸含量及代谢相关酶活性变化规律。结果表明:(1)随着烟株的生长发育,烤烟中部叶有机酸总量呈先升高后降低趋势,于叶龄50~60d时达到最高值;烤烟品种间中部叶有机酸总量在各叶龄时期存在极显著差异,而在调制前后变化不大。(2)苹果酸含量呈"逐渐升高"趋势;柠檬酸含量呈缓慢"增-减"的趋势;苹果酸含量与苹果酸脱氢酶(NAD-MDH)活性之间极显著负相关;柠檬酸含量与柠檬酸合成酶(CS)活性之间极显著正相关,而与异柠檬酸脱氢酶(NAD-IDH)活性相关性不大。(3)当有机酸总量大于32%,同时烟碱含量介于1.5%~2.6%时,"酸碱比"较高的烟叶感官质量较好。研究认为,烤烟叶片苹果酸和柠檬酸的积累受其代谢相关酶活性变化的调控,"酸碱比"影响烤烟烟气的醇和度,可通过调控NAD-MDH和CS活性来调节苹果酸和柠檬酸的含量,协调"酸碱比",提高烟叶质量。  相似文献

Effects of low-molecular-weight organic acids on the accumulation and transportation of gadolinium (Gd) in tomato plants were studied under hydroponic condition. The results indicated that changes of organic acids occurred in the processes of Gd accumulation and transportation in tomato plants which were treated with extraneous Gd solutions. Malic, citric, and succinic acids contributed to both Gd accumulation in roots and transportation in xylem vessels. When Gd was unloaded from the xylem to the leaf cells, formic, lactic, citric, and succinic acids played important roles in Gd accumulation in leaves. When tomato plants were cultured in the uptake solution of Gd-containing malic, citric, or succinic acid for 48 h, the succinic acid in roots and leaves and the malic acid in xylem saps both increased obviously. From the results above, we can conclude that succinic acid had the most important role in Gd accumulation in tomato roots and leaves, while malic acid transported Gd via xylem vessels more effectively.  相似文献

The biosynthesis of macrotetrolides by Actinomyces chrysomalus var. carotenoides was stimulated by acetic, succinic, propionic, oxalic, malic, tartaric, citric, pyruvic, alpha-ketoglutaric and fumaric acids. Incorporation of 14C-acetate into the molecule of the antibiotic and the data on dependence of the stimulating effect upon the quantitative ratio and time of the organic acid addition were indicative of the role of acetic, succinic and propionic acids as precursors of macrotetrolides. The other organic acids increased the biosynthesis of macrotetolides when added to the culture within wide time ranges of the culture development and prolonged the period of the mycelium productive state.  相似文献

在温室沙培灭菌条件下,以Al-P为磷源、枳为试材、Glomus mosseae (G.m)和G.versiforme (G.v)为菌剂,研究低磷胁迫下AM真菌对枳实生苗干物重、吸磷效应及根系分泌有机酸的影响。结果表明,接种AM真菌显著增加枳地上部、地下部干物重,增幅16.79%~135.25%;同时显著增加其吸磷量,菌丝对植株的吸磷贡献率为17.04%~71.95%(G.m>G.v),施Al-P显著提高菌丝吸磷贡献率。接种AM真菌的根系分泌的有机酸种类与对照有所不同,未接种处理枳分泌的有机酸有草酸、苹果酸、乳酸、乙酸、顺丁烯二酸和柠檬酸等6种,而接种G.m的则检测到草酸、酒石酸、苹果酸、乳酸、乙酸、柠檬酸、丁二酸等7种,G.v处理的检测到酒石酸,接种处理均未检测到顺丁烯二酸;接种丛枝菌根真菌增加了枳根系分泌有机酸的量(比未接种处理增加19.80~56.87 mg/kg,且施用AlPO4后有机酸含量显著增加(增加20.06~21.84 mg/kg);未接种植株根系仅分泌少量有机酸;接种植株根系分泌的有机酸以苹果酸(42.87%)、柠檬酸(39.22%)和草酸(12.06%)为主。  相似文献

A pot experiment confirmed that pigeonpea could efficiently utilize various sources of phosphorus (P) (aluminium phosphate, iron phosphate and apatite), irrespective of genotype. A qualitative assay method for iron (Fe)-P solubilizing activity showed that root exudates collected from P-deficient pigeonpea contained Fe-P solubilizing substances and that they were released mainly from root tips. Citric, malic, malonic, succinic and piscidic acids were identified in root exudates. Citric and piscidic acids release from roots was increased by low-P treatment in all the genotypes tested. The release rates of citric and piscidic acids were affected by the P concentration of shoots rather than that of roots. The pigeonpea roots released approximately 5–100 times more piscidic acid than citric acid depending on P stress status, plant age and genotype. When organic acids were added to Alfisols, citric acid was most capable of mobilizing P from the soil, followed by piscidic acid and malic acid. No correlation was found between genotypic variability in the release rates of citric and piscidic acids from the roots under low-P treatment at hydroponic culture and in the growth and P uptake of plants on Alfisols. Although citric and piscidic acids released from pigeonpea roots may play a partial role in solubilizing unavailable insoluble P in soils, the releases were thought to be an unsatisfactory strategy for explaining genotypic variation in low P availability of pigeonpea.  相似文献

Synthesis of carotenes by Actinomyces chrysomallus var. carotenoides was stimulated by citric, acetic, oxalacetic, fumaric, succinic, malic, alpha-ketoglutaric, tartaric, pyruvic, and propionic acids. Acetic acid acts as a precursor of carotene synthesis and also has another stimulating mechanism of action on carotenogenesis of the actinomycete. Acetic, furmaric, malic, succinic, and alpha-ketoglutaric acids stimulate cyclization of lycopene yielding beta-carotene.  相似文献

Roles of organic acids and nitrate in the long-distance transport of cobalt (Co) in xylem saps of hyperaccumulator Alyssum murale and non-hyperaccumulator Trifolium subterraneum were studied under hydroponic conditions. Organic acids (oxalic, malic, malonic, citric, and fumaric) and nitrate in xylem sap samples were separated and determined simultaneously by reversed-phase high performance liquid chromatography after solid-phase extraction with nanosized hydroxyapatite. Results indicated that Co treatment significantly increased the concentrations of xylem oxalic and malic acids for the hyperaccumulator A. murale compared to the control but significantly decreased the concentrations of xylem nitrate and malonic acid; concentrations of citric acid in xylem sap samples did not show significant difference between the control and Co treatments. By analyzing the relationship between the concentrations of organic acids, nitrate, and concentrations of Co in xylem saps, it could be concluded that oxalic and malic acids in xylem saps seemed to participate in the long-distance Co translocation process, and citric acid did not relate to the xylem Co transport of A. murale and T. subterraneum. Our work might be very useful for understanding the mechanism of long-distance transport of heavy metals in hyperaccumulator.  相似文献

以鄂南、赣北两红壤样品为材料,加入不同有机酸并经室温培养后,测定不同P组分、pH及活性Al含量的变化。结果表明,供试有机酸均使土壤Ca2-P含量增高,增幅大小依次为柠檬酸>苹果酸>琥珀酸>乙酸;2种土壤的Ca8-P和Ca10-P含量无明显变化规律,Fe-P、Al-P和O-P含量有所下降,除乙酸处理的土壤pH值无显著变化外,其它有机酸的加入使pH下降0.65-1.96;有机酸引起活性Al量增多,除乙酸处理的变化较小外,其它有机酸或混合物的加入使土壤中0.02mol.L^-1CaCl2提取Al增加4.7-50.3倍,1mol.L^-1提取Al增加4.0-67.3倍。可见,有机酸具有双重作用,既增加P的有效性,又增加土壤酸度和Al毒。  相似文献

为探讨外源硫化氢(H2S)对盐碱胁迫下植物有机酸和激素水平的调控效应,以裸燕麦(Avena nuda)为材料,研究喷施50 μmol·L-1 H2S供体硫氢化钠(NaHS)溶液对3.00 g·kg-1盐碱胁迫下叶片有机酸、激素含量和产量性状的影响。结果表明:盐碱胁迫显著提高了琥珀酸、丁烯二酸、苹果酸、葡萄糖醛酸和总有机酸含量,显著降低了焦谷氨酸、茉莉酸-异亮氨酸(JA-Ile)、反式-玉米素(tZ)和N6-(Δ2-异戊烯)腺嘌呤(iP)含量。喷施NaHS溶液显著提高盐碱胁迫下裸燕麦叶片中3-羟基-3-甲基谷氨酸、吲哚乙酸(IAA)、赤霉素A7(GA7)、茉莉酸甲酯(MJA)、iP含量和IAA/ABA比值,显著降低葡萄糖醛酸、赤霉素A3(GA3)、赤霉素A4(GA4)、总赤霉素(GAS)、1-氨基环丙烷羧酸(ACC)含量和ACC/ABA比值,而对琥珀酸、丁烯二酸、苹果酸、柠檬酸、丙二酸、泛酸、烟酸、焦谷氨酸、辛二酸、苯丙酮酸、总有机酸、赤霉素A1(GA1)、茉莉酸(JA)、JA-Ile、脱落酸(ABA)、tZ、反式-玉米素核苷(tZR)、N6-(Δ2-异戊烯基)腺苷(iPA)、总茉莉酸类(JAs)、细胞分裂素(CTK)含量、GAs/ABA、JAs/ABA、CTK/ABA比值无显著影响。主成分分析表明,喷施NaHS溶液使盐碱胁迫下裸燕麦叶片中有机酸3-羟基-3-甲基谷氨酸和激素MJA、GA7、tZ、IAA含量分别显著上调14.31%和41.83%、50.00%、22.97%、13.02%;而有机酸烟酸、葡萄糖醛酸和激素GA4、ACC、tZR、GA3含量分别显著下调16.00%、23.87%和73.53%、32.72%、50.00%、33.91%。另外,喷施NaHS溶液使盐碱胁迫下裸燕麦千粒质量下降了5.91%,而穗数量、穗铃数量、穗粒数量和籽粒产量分别提高了2.19%、9.70%、61.60%和52.83%。表明外源H2S参与盐碱胁迫下裸燕麦有机酸和激素水平的调控,能够增强裸燕麦适应盐碱胁迫的能力。  相似文献

Better understanding of copper uptake and accumulation regulation in plants is critical to the phytoremediation of copper contaminated soil. This study employed a 30-day pot experiment to assess the relationship between organic ligands and copper accumulation in plants. Hyperaccumulator and nonaccumulator varieties of Commelina communis were used, different organic ligands were applied, and the data of copper accumulation in shoots were collected. The six organic ligands included ethylenediaminetetraacetic acid and organic acids (formic acid, citric acid, malic acid, tartaric acid, and succinic acid). The results showed that organic ligands added to culture increased the copper accumulation both varieties. The results of the copper accumulation in shoots agreed with the study of the root uptake kinetics of copper influx. The addition of organic acids could increase copper accumulation in shoots because the copper influx in roots was increased. The results also indicated that the copper influx of hyperaccumulator roots was higher than that of nonaccumulator roots. This is one of the mechanisms by which a hyperaccumulator could amass large amounts of copper in its shoots. In this accumulation process, little effect on the leaf relative water content was in the hyperaccumulator and nonaccumulator of leaves and normal physiological condition of plants.  相似文献

A GC and an HPLC method for the quantification of organic acids OAs in coffee have been compared. The GC procedure, employing trimethylsilyl derivatives, was found to be very tedious. The HPLC method, which employed an ion exchange column using a flow gradient of water containing 1% phosphoric acid and UV detection (210 nm), was found to be much simpler for the quantification of eight organic acids (oxalic, succinic, fumaric, malic, tartaric, citric, quinic and fumaric acids) in four representative coffee samples. The HPLC procedure was more convenient than that described in the literature since no pre-purification was required for quantification of the OAs.  相似文献
