介绍一种简便实用的DNA片段回收方法,与以前所报道的DEAE-纤维素膜电泳法、透析袋电洗脱法、低融点琼脂糖凝胶法、凝胶冻融法等相比,所需器材简单、操作简便、回收率高、成本低。回收的DNA片段在进一步克隆和测序中表现出较好的效果,是一种适合于科研和教学的实验方法。  相似文献

 采用PXY104(含化学合成的HIV-env26肽基因4重串联体结构)为材料。温和裂解法提取质粒DNA,制备电泳纯化,Hind Ⅲ和EcoR Ⅰ酶解,低融点琼脂糖凝胶电泳回收HIV-DNA片段作为核酸探针;用[α-~(32)P]dATP通过缺口移位法标记,比放射性为4.05—6.69×10~7cpm/μgDNA通过分子杂交能测出lPg的靶DNA,初步试验结果表明,在本文试验条件下,可用该探针检测与之互补的核酸分子。  相似文献

为了简化琼脂糖凝胶中DNA片段的回收,该文报道一种新的挤压回收法。将含有DNA片段的凝胶块放在折叠的封口膜之间,然后用一小塑料平板将凝胶块中的DNA溶液用力挤出,用移液枪把所有挤压出的DNA溶液放入effendorf管中,然后用常规的苯酚抽提法进行纯化。DNA回收率达到40-60%(w/w)。该方法简单而有效,且回收的DNA能够直接应用于酶切、连接及PCR等各种分子生物学操作。  相似文献

用两只离心管制成的凝胶过滤装置,从电泳后的琼脂糖凝胶中回收DNA片段的简易方法。它依次包括以下步骤:凝胶过滤装置的制作、凝胶切割、凝胶低温冷冻、低温高速离心、ddH20洗胶、DNA纯化和回收效果检测等。用此方法回收的DNA片段产率高、质量纯,可直接用于分子生物学实验的后续操作,如载体连接、PCR模板获得、DNA探针制备、基因测序等。其优点是:DNA片段的回收率高(90%以上),质量好;操作简便,耗时短;回收装置简单,成本低廉,可进行商品化开发。  相似文献

介绍一种从琼脂糖凝胶同步回收DNA和琼脂糖的方法。利用0.25 mol/L异硫氰酸胍溶液(p H 8.0)溶解含有目的 DNA片段的的凝胶条,胶条溶解后,静置冰上10 min再加入预冷的异丙醇,琼脂糖呈颗粒状析出,通过离心即可初步分离DNA和琼脂糖。上清液用异丙醇沉淀回收DNA片段,利用50%PEG溶液沉淀琼脂糖。分别对0.2 kb、1 kb和10 kb长度的DNA片段进行回收,回收率分别为19.44%、36.40%、13.49%,回收的DNA纯度高,电泳条带清晰。琼脂糖均回收率为62.52%,回收琼脂糖脱水后的状态为白色颗粒。该方法切实可行,回收成本低廉,回收的DNA和琼脂糖可用于后续实验。  相似文献

介绍了一种有效回收小于200bp DNA片段的方法。用改进的冻融-离心回收法对155bp的DNA片段进行回收,并且与常规冻融-离心回收、TaKaRa回收试剂盒的结果做对比,用琼脂糖凝胶电泳检测回收结果,紫外吸收法定量分析。结果证明:改进的方法是一种经济、方便、可靠的回收小分子DNA片段的方法。  相似文献

为了建立一种直接从普通琼脂糖凝胶中回收DNA片段的简便实用的方法,采用聚合酶链式反应扩增人P53基因外显子7、8和其间的内含子7序列,用普通琼脂糖凝胶电泳,直接从凝胶中切下产物带,用加热熔化法回收DNA;紫外比色法测定回收率;用测序法鉴定回收产物质量。并用QIAquick Spin纯化柱对照。结果表明,本法回收的产物质量明显优于用QIAquick Spin柱回收,本法回收的产物用于测序效果极佳,回收率达80%,用QIAquick Spin柱回收率不到20%,差异非常显著(P<0.01)。证明这种方法回收PCR产物质量可靠,能代替低熔点胶回收DNA,有较大的实用价值。 Abstract: In order to find a simple and efficient method to isolate single or double?strand DNA fragment amplified by polymerase chain reaction (PCR),we used PCR method to amplify exon 7,exon 8 and intron 7 of human P53 gene, electrophoresis to identify products,fusion and phenol-chlorofom extraction (FPC) to isolate specific DNA from agarose gel,ultraviolet colorimetry to deteminate collected rate,and direct sequencing to identify the quality of recollected DNA. A control test was also made by using QIAquick Spin Colum.The results showed that the quality of PCR products recollected by using FPC method was very good.When the recollected DNA was used in sequencing,no matter what was single or double-strand DNA,the sequence data was clear and even,with low noise.The recollected rate of using FPC,which was over 80 per cent, was higher than that of using colum (lessthan 20 per cent), there were statistical significances (P<0.01).In the control test, it had a little non-specific DNA in the collected products,and the sequencing experiment of using double-strand products was failure.All above mentioned suggested that general agarose gelis efficient in place of low melting-temperature for isolating DNA fragment.  相似文献

本文介绍一种快速获取高纯度质粒DNA的方法。本法对煮沸提取法进行改良后,快速提取质粒DNA粗制品,然后用国产滤纸从琼脂糖凝胶中回收纯化质粒DNA。同时对本法所提取的质粒DNA的回收率、浓度、纯度及可能的用途进行验证和讨论,证明此法简易,所得样品纯度高,可直接用于转化、酶切和基因克隆。  相似文献

利用高分辨率的非变性聚丙烯酰胺凝胶电泳是分离、纯化特异性小片段DNA的首选方法,也是开展后续多种分子生物学试验的前提。本试验比较了改良煮沸法(液氮研磨+煮沸+低温浓缩)、煮沸法及直取法回收小片段DNA,以回收样品模板进行第二次PCR扩增,结果表明:改良方法回收的DNA的纯度高,特异性好,回收率与经典煮沸法相当。在保证回收产物的特异性时,用低温浓缩法替代乙醇/醋酸钠共沉淀也具有可行性及一定的创新性。  相似文献

目的显微注射用DNA的纯度是影响转基因动物制备成功与否的重要因素,本文建立一种可行的适用于普通实验室的纯化DNA方法,替代普遍使用的试剂盒纯化方法。方法分别使用酚-氯仿多次抽提法及常规的凝胶提取试剂盒纯化含有蚓激酶基因的DNA片段,通过显微注射技术将纯化的DNA片段导入小鼠受精卵的原核,制备转基因小鼠。根据转基因实验的结果对两种方法进行比较。结果使用两种方法纯化DNA均能获得转基因小鼠。在DNA纯度及注射卵的存活率上,两种方法无明显差别;在移植卵的出生率及转基因阳性率上,抽提法优于试剂盒法。结论本实验建立的抽提方法可以替代试剂盒方法纯化显微注射用DNA片段,在降低实验成本、简化实验条件及提高转基因阳性率方面具有优势。  相似文献

为筛选和建立风沙土中总DNA的提取和纯化方法,选取了5种直接提取法、1种间接提取法和2种纯化法分别对风沙土中总DNA进行了提取和纯化,并对其质量和产量进行了比较.结果表明:6种方法均可从风沙土中提取到大小为23 kb左右的总DNA,其中改进后的高盐提取法(用40%聚乙二醇8000和4 mol·L-1 NaCl沉淀DNA)效果最好,纯化后总DNA的纯度最高,可进行16S rDNA的PCR扩增,且产量仅稍低于试剂盒提取法;电泳加柱回收纯化法的纯化效果较好,经该方法纯化后的总DNA大部分可进行PCR扩增,可满足后续分子操作对DNA纯度的要求.  相似文献

The differences on DNA yield and purity of three different DNA extraction protocols were compared with regard to the use for PCR and other molecular analyses. Total DNA was extracted from compost by the three protocols, and then was purified by spin-bind cartridges after being precipitated by PEG8000. The detection performed on a nucleic acid and protein analyzer showed that all three methods produced high DNA yields. The agarose gel electrophoresis showed that the fragments of crude and purified DNA had a length of about 23 kb. A eubacterial 16S rRNA gene-targeted primer pair was used for PCR amplification, and full length 16S rDNAs were amplified from all the purified DNA samples. After being digested by restriction endonucleases, the restriction map of amplified rDNA showed identical genetic diversity. The products of PCR using primer pair GC341F and 907R were also used for denaturing gradient gel electrophoresis analysis. The results indicated that high-quality DNA was extracted from compost by the three protocols, and each of the protocols is adapted to extract microbial genome DNA from compost expediently and cheaply.  相似文献

Purified DNA fragments are used for different purposes in Molecular Biology and they can be prepared by several procedures. Most of them require a previous electrophoresis of the DNA fragments in order to separate the band of interest. Then, this band is excised out from an agarose or acrylamide gel and purified by using either: binding and elution from glass or silica particles, DEAE-cellulose membranes, "crush and soak method", electroelution or very often expensive commercial purification kits. Thus, selecting a method will depend mostly of what is available in the laboratory. The electroelution procedure allows one to purify very clean DNA to be used in a large number of applications (sequencing, radiolabeling, enzymatic restriction, enzymatic modification, cloning etc). This procedure consists in placing DNA band-containing agarose or acrylamide slices into sample wells of the electroeluter, then applying current will make the DNA fragment to leave the agarose and thus be trapped in a cushion salt to be recovered later by ethanol precipitation.  相似文献

Agarose gel electrophoresis is commonly used to separate different species of nucleic acids. We compare four different methods of extraction which are commonly used. These methods include buffer extraction, electroelution, glass bead extraction and extraction of DNA from low-melting agarose. The results show that DNA extracted by these four methods is comparable in their ligability to the PMT 21 vectors and the plasmids with insert can be used for subsequent transfections of competent bacteria. There is a higher yield for buffer extraction and electroelution when compared with glass bead extraction and low melting agarose (p less than 0.05). To conclude, the four commonly used methods for DNA isolation are comparable qualitatively. But the simplest method, namely buffer extraction, has the highest yield.  相似文献

Four different DNA extraction methods were used to extract genomic DNA of the medicinal mushroom Lingzhi from its developing stage materials, such as mycelium, dry fruiting body, or sliced and spore powder or sporoderm‐broken spore powder. The DNA samples were analyzed using agarose gel electrophoresis, UV spectrophotometer, and PCR amplification. According to the average yields and purity of DNA, high salt concentrations and low pH methods were the best for DNA extraction. The mycelia and sporoderm‐broken spore powder yielded higher and purer DNA. The method developed could effectively eliminate the influence of the secondary metabolites to DNA extraction. The DNA samples extracted from the developed method could be successfully used for PCR applications.  相似文献

提取海南产桶形芋螺线粒体基因组完整DNA (mtDNA),并对提取条件进行优化。以桶形芋螺腹足肌肉、毒腺和肝胰脏三个不同组织为材料,分别采用改进高盐沉淀法、细胞器/磁珠法和试剂盒提取三种方法,提取桶形芋螺mtDNA,并利用琼脂糖凝胶电泳和紫外分光光度计对提取mtDNA的纯度和浓度进行测定。以coxⅠ-rRNA小亚基基因和α-芋螺毒素基因设计引物,通过PCR反应来确证所提取的DNA确实是mtDNA。试剂盒法提取肝胰脏、高盐沉淀法提取肝胰脏和腹足肌肉组织这三种方法的产率很高,分别为44.4μg/mg、43.3μg/mg和32.6μg/mg。A260/280比值表明,改进高盐沉淀法提取毒腺和腹足肌肉组织,细胞器磁珠法提取腹足肌肉组织的mtDNA纯度很高。综合比较,采用改进高盐沉淀法,利用桶形芋螺腹足肌肉组织所提取的mtDNA产率高、质量好、纯度高。高质量芋螺mtDNA的获取为利用分子生物学方法对芋螺进行遗传进化分析和系统分类提供了基础。  相似文献
