目的:探讨血卟啉单甲醚(hematoporphyrin monomethyl ether,HMME)介导的声动力疗法(sonodynamic therapy,SDT)对牙龈卟啉单胞菌(Porphyromonas gingivalis,Pg)生物膜中脂质过氧化物丙二醛(malondialdehyde,MDA)含量的影响。方法:羟基磷灰石片培养Pg生物膜厌氧培养3天,将生物膜随机分为4组(对照组、HMME组、超声组、SDT组),分别与无菌生理盐水或HMME进行避光孵育,然后进行声动力处理。采用平板计数法计算细菌存活率,MDA含量使用MDA检测试剂盒在可见光分光光度计下进行检测。结果:当超声强度为3 W/cm~2,超声时间为5 min时,SDT组的生物膜细菌存活率仅为40%,与对照组相比显著降低(P0.05),超声组细菌存活率为62%,与对照组相比亦显著降低(P0.05)。相同超声参数下,SDT组Pg生物膜中MDA含量最高,达17.3±1.2 nmol/mL(P0.05),超声组生物膜中MDA含量为7±0.8 nmol/mL,与对照组相比差异并无统计学意义(P0.05)。结论:HMME介导的SDT对Pg生物膜有一定杀伤效果,并且在杀伤过程中,可引发脂质过氧化反应,导致MDA释放。  相似文献

研究了冬凌草提取物对金黄色葡萄球菌生物被膜形成的影响。采用结晶紫染色法评价了冬凌草提取物在不同质量浓度和不同添加时间下干预金黄色葡萄球菌生物被膜的作用。实验结果表明,冬凌草提取物对金黄色葡萄球菌的MIC和MBC分别为125μg/mL和250μg/mL,在亚抑菌浓度,冬凌草提取物对金黄色葡萄球菌生物膜形成的干预作用不明显;当冬凌草提取物质量浓度大于1倍MIC时,其对金黄色葡萄球菌生物膜形成的有明显的抑制作用,在生物膜形成的初始时间(0 h)添加药物抑制效果最佳,2MIC冬凌草提取物对金黄色葡萄球菌成膜的抑制率达到79%以上;24 h后添加抑制作用较弱,抑制率仅有60%左右。采用银染法和荧光染色法研究了冬凌草提取物对金黄色葡萄球菌生物膜的清除效果和抑制胞外多糖的合成,实验结果也表明,在亚抑菌浓度,冬凌草提取物对金黄色葡萄球菌生物膜的清除和胞外多糖合成的抑制作用很弱,当冬凌草提取物质量浓度大于1 MIC时,在生物膜形成的初始时间(0 h)添加药物,对金黄色葡萄球菌生物膜清除效果明显,胞外多糖的合成也受到显著的抑制。本研究为冬凌草提取物在食品防腐和保鲜,以及无抗饲料的广泛应用提供理论基础。  相似文献

目的:探讨血卟啉单甲醚(Hematoporphyrin monomethyl ether,HMME)在A549肺癌细胞内亚细胞分布的动态变化。方法:传代培养A549肺癌细胞,分别与光敏剂HMME孵育2 h和12 h。应用由荧光显微镜及电感耦合器材(Charge-coupled device,CCD)组成的高分辨率荧光显微成像系统,结合荧光探针标记技术,采用细胞器-细胞荧光强度比值法研究HMME在不同时间的亚细胞分布情况。结果:在2 h和12 h两个孵育时间点高尔基体的平均荧光强度比值(J1/J2)值都最高;随着孵育时间延长,A549细胞的四种细胞器J1/J2值都升高且溶酶体幅度最大。结论:孵育时间是影响HMME亚细胞分布的一个重要因素。随着孵育时间的延长,A549肺癌细胞各细胞器吸收HMME能力逐渐增强,尤以溶酶体显著。  相似文献

原子力显微镜观测血卟啉单甲醚对细菌光动力杀伤作用   总被引:2,自引:0,他引:2  
[目的]探讨血卟啉单甲醚(Hematoporyrin monomethyl Ether,HMME)对革兰氏阳性(G )、阴性(G-)菌的光动力杀伤作用.[方法]通过平板菌落计数法和原子力显微镜(AFM),观察细菌与HMME作用前后形貌的变化.[结论]当HMME浓度为50 μg/mL,可见光(光功密度为200 mW/cm2)光照30min时90%以上的金黄色葡萄球菌(Staphyrlococcus aureus)能被杀死,无光照时对S.aureus杀灭效果显著.同等条件下,无论光照还是无光照,HMME对大肠杆菌(E.coli)无明显的杀伤作用.AFM图像显示,S.aureus细菌表面破坏严重,完全碎裂成鱼鳞状的片状堆积.对HMME作用后的E.coli扫描可见,菌体原来光滑的表面变成网格状的裂纹排列.[讨论]HMME对G 有明显的光失活效应,而对G-效果不明显.AFM的超微图像显示HMME对细菌细胞的攻击位点主要在细胞膜上.AFM为我们研究光敏剂对细菌的光动力损伤作用机制的可视化提供了依据.  相似文献

目的观察柠檬提取物对细菌生物膜的消除作用,对柠檬提取物抑制多重耐药金黄色葡萄球菌的机制进行初步研究,以及对环境中富集的病毒消除作用的研究。方法采用高分子滤膜法制备金黄色葡萄球菌生物膜后,加入柠檬提取物分别作用1、2和3h,通过细菌菌落计数判定不同生物膜的细菌存活数;采用SDS—PAGE电泳法,观察在柠檬提取物作用下菌体蛋白质的变化;EcoRⅠ酶切金黄色葡萄球菌DNA,观察酶切图谱的改变;在低渗环境下动态观察柠檬提取物在不同作用时间对细菌细胞壁的影响;通过柠檬提取物对病毒作用后RNA量的变化,说明柠檬提取物具有杀灭病毒的作用。结果柠檬提取物对生物膜中的细菌具有杀菌作用,且随作用时间的延长而逐渐增强;柠檬提取物对细菌蛋白质的组成和表达量均有一定影响,同时细菌DNA的EcoRⅠ酶切图谱发生改变;柠檬提取物作用15min时细菌呈现明显的胞壁缺损现象,即细菌个体胀大,呈现大球形;经柠檬提取物作用后,RNA病毒被杀灭。结论结果显示柠檬提取物对耐药金黄色葡萄球菌所形成的生物膜具有明显抑制作用;在柠檬提取物作用下,菌体蛋白合成量发生改变,同时对细菌的DNA和细胞壁有明显的影响。柠檬提取物处理病毒悬液后,病毒RNA量显著减少,说明病毒RNA结构被破坏。  相似文献

目的:探讨不同声强超声对孕鼠子宫收缩及胎鼠神经损伤的影响及机制。方法:将32只孕鼠随机分为A(0)组、B(2)组、C(4)组、D(8)组,每组各8只,分别接受声强为0 W cm~(-2)、2 W cm~(-2)、4 W cm~(-2)和8 W cm~(-2)的超声辐照20 min。记录孕鼠子宫收缩及子宫平滑肌ATP酶的活力,检测胎鼠大脑皮层与海马神经元凋亡及胆碱能神经系统相关酶的活力。结果:超声增加孕鼠子宫收缩频率、收缩幅度、收缩张力和子宫活动度(P均0.05),降低孕鼠子宫平滑肌中钠钾ATP酶、钙ATP酶和钙镁ATP酶的活性(P均0.05)。超声降低胎鼠大脑皮层和海马中Bcl~(-2)水平(P0.05),增加Bax和Caspase-3水平(P均0.05)。以及促进乙酰胆碱酯酶活力,降低乙酰胆碱和乙酰胆碱转移酶水平(P均0.05)。且4 W cm~(-2)和8 W cm~(-2)的超声比2 W cm~(-2)超声对这些指标的作用更强。结论:4 W cm~(-2)和8 W cm~(-2)超声可能通过降低ATP酶的活性促进孕鼠子宫平滑肌收缩,并可引起胎鼠大脑皮层和海马神经元损伤,机制可能与胆碱能神经元系统失衡有关,2 W cm~(-2)声强的超声对胎鼠神经元损伤甚小。  相似文献

目的:探讨血卟啉单甲醚(HMME)介导的光动力疗法(PDT)对人食管癌细胞杀伤效应的最佳作用参数,为HMME-PDT的临床应用提供一定的参考依据。方法:以4μg·m L-1HMME与人食管癌Eca-109细胞孵育不同时间后,利用形态学软件分析其荧光强度;进一步予不同浓度(0.5μg·m L-1~8μg·m L-1)HMME与细胞孵育1.5小时后,在特定波长下以不同能量的激光(2、4、8 J·cm-2)照射,采用CCK8法检测细胞的存活率。结果:HMME在细胞内的荧光强度于给药1.5 h后达到高峰。在一定范围内,随着HMME浓度与激光剂量的增加,细胞存活率逐渐下降,当HMME浓度增高到4μg·m L-1时,光照强度增高到4 J·cm-2,进一步提高药物浓度与激光剂量,细胞存活率并不随之降低。结论:不同的孵育浓度、不同的孵育时间及不同的光照剂量密度可以显著的影响HMME-PDT对人食管癌Eca-109细胞的体外效应。  相似文献

【背景】金黄色葡萄球菌是一种常见的食源性致病菌,易在食品及加工器具表面形成生物膜,引起食品腐败和疾病的传播,威胁食品安全。【目的】研究冬凌草甲素抑制金黄色葡萄球菌生物膜形成的作用机制。【方法】使用结晶紫染色法和扫描电镜观察冬凌草甲素对金黄色葡萄球菌生物膜形成的抑制作用,刚果红平板法定性检测冬凌草甲素对细胞间多糖黏附素(polysaccharideintercellular adhesion,PIA)合成的影响,分光光度法测定冬凌草甲素对供试菌株胞外DNA (eDNA)释放量的影响,RT-PCR技术检测冬凌草甲素对供试菌株ica A、cid A、agr A和sar A基因表达量的影响。【结果】冬凌草甲素对金黄色葡萄球菌生物膜形成有较强的抑制作用;冬凌草甲素能显著抑制PIA的合成,且呈浓度剂量依赖;冬凌草甲素能抑制供试菌株e DNA的释放量,其中1/4最小抑菌浓度(minimum inhibitory concentration,MIC)的冬凌草甲素作用金黄色葡萄球菌16 h后,与对照组相比,e DNA的释放量降低了48.62%;冬凌草甲素可显著抑制金黄色葡萄球菌生物膜形成相关基因的表达,其中1/2MIC的冬凌草甲素作用金黄色葡萄球菌16 h后,ica A、cid A、agr A和sar A基因的表达量分别比对照降低了91.6%、94.7%、77.6%和70.4%。【结论】冬凌草甲素通过抑制ica A和cid A基因的表达,影响PIA的合成和eDNA的释放,进而干预生物膜的形成。  相似文献

评价黑大蒜提取物分别与头孢唑林或庆大霉素联合应用,对金黄色葡萄球菌和大肠埃希菌的体外抗菌效应。采用液体稀释法分别测定黑大蒜提取物对金黄色葡萄球菌和大肠埃希菌的最低抑菌浓度(MIC)。采用棋盘法设计,微量肉汤稀释法测定黑大蒜提取物联合头孢唑林或庆大霉素对金黄色葡萄球菌和大肠埃希菌的MIC,并计算部分抑菌浓度(FIC指数)。测定黑大蒜提取物对金黄色葡萄球菌和大肠埃希菌的时间-杀菌曲线。黑大蒜提取物对金黄色葡萄球菌的MIC为256μg/mL,黑大蒜提取物对大肠埃希菌的MIC为256μg/mL。时间-杀菌曲线结果显示黑大蒜提取物对金黄色葡萄球菌和大肠埃希菌的抑菌作用呈现较强的浓度依赖性。黑大蒜提取物联合头孢唑林后对金黄色葡萄球菌的FIC指数为0.75;黑大蒜提取物联合庆大霉素后对大肠埃希菌的FIC指数为0.5。黑大蒜提取物与头孢唑林或庆大霉素联合用药,可明显降低抗生素对金黄色葡萄球菌和大肠埃希菌的MIC,表现为相加和协同效应。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Fluid collected from hatching eggs of Haemonchus contortus contained a lipase which hydrolysed 2-naphthyl laurate (about 0·7 μmol naphthol freed /h/106 eggs). The fluid also hydrolysed l-leucinamide (about 2·3 μmol leucine freed/h/106 eggs). The fluid when added to normal or heated eggs caused ‘hatching’. ‘Hatching’ also occurred in exsheathing fluid from infective juveniles and in a preparation of pancreatic lipase containing leucine aminopeptidase. A purified mammalian leucine aminopeptidase in combination with several different lipases did not attack egg shells.The ‘spontaneous’ hatching of eggs of H. contortus was strongly inhibited by 1,10-phenanthroline, 10?3M, and this inhibition was reversed by Zn2+. However, the inhibition of ‘hatching’ of eggs in externally applied hatching fluid, or the hydrolysis of leucinamide in hatching fluid was generally less marked.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献
