病毒病是危害萝卜作物的主要病害之一,芜菁花叶病毒(TuMV)作为萝卜病毒病的主要毒源,给萝卜生产造成严重经济损失。由于缺乏精准鉴定,萝卜育种中缺乏稳定可靠的抗TuMV材料。本研究就国家蔬菜种质资源中期库提供的来自中国25个省份125个县市和其他3个国家的150份代表性萝卜种质资源对TuMV的抗性进行了苗期鉴定和ELISA检测,筛选出23份抗病材料,其中1份表现免疫。这些材料对萝卜抗病毒病基因定位和新品种培育具有重要意义。  相似文献

烟草花叶病是广东省产烟区的主要病害之一。我们在南雄等八个县进行调查,1983年至1984年的一般发病率为2~20%。根据血清学反应、病毒粒体形态、鉴别寄主反应及寄主范围、媒介昆虫种类、物理性质、交互保护反应等各项检验结果,鉴定广东省烟草花叶病病原是:三个黄瓜花叶病毒可能株系(普通株系CMV-C,烟草坏死株系CMV-TN,烟草黄色坏死株系(CMV-TYN),烟草花叶病毒(TMV),马铃薯病毒Y(PVY),烟草脉带花叶病毒(TVBMV)和烟草褪绿斑驳病毒(TCMV)(暂定)。  相似文献

利用基因工程方法培育抗病毒植物新品种的途径之一,是在植株中建立一个产生病毒基因组功能片段的反义RNA的系统。本工作设计并合成了一段烟草花叶病毒(TMV)装配起始位点反义RNA的基因,再以pBR 325为基本质粒,构建了包含带有花椰菜花叶病毒(CaMV)35S启动子和PolyA信号的反义RNA表达单元,以及为筛选转基因植株所必须的NPT-Ⅱ表达单元的中间载体,为以后经土壤农杆菌而获得转基因烟草植株打下了基础。  相似文献

本文报道了高产量提取芜菁花叶病毒(TuMV)、莴苣花叶病毒(LMV)、芋花叶病毒(DMV)和大豆花叶病毒(SMV)的提取方法。本方法通过使用高盐浓度的磷酸盐缓冲液以及在缓冲液中加入氯化镁和脲,并用TritonX-100作为澄清剂,替代常规使用的氯仿和正丁醇,成功的提取到了大量病毒粒子,上述四种病毒提取的得率分别是TuMV为173.3mg/kg病叶,LMV为96mg/kg病叶,SMV为199.2mg/kg病叶,DMV为176.6mg/kg病叶。  相似文献

【背景】黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)是严重威胁葫芦科作物生产的毁灭性病原之一,该病毒已入侵我国十多个省份,危害西瓜、黄瓜等作物并造成严重的经济损失。早在2009年广东即发现CGMMV为害西瓜和黄瓜,但黄瓜等葫芦科作物对其抗性情况尚不清楚。【方法】采用人工机械摩擦接种方法,测定了14份黄瓜种质资源对CGMMV广东分离物的抗性水平。【结果】从广东葫芦病样中分离获得CGMMV,该病毒分离物MP基因序列与国内报道的各分离物同源率均在99%以上;14份黄瓜种质资源对该病毒分离物均表现为感病。【结论与意义】广东主要黄瓜资源对CGMMV均表现为感病,这为我省防控该病毒病提供了科学依据,也为黄瓜抗病育种提供了指导。  相似文献

Cupressus sempervirens L. var. pyramidalis and C. sempervirens var. horizontalis were examined for their pollen morphometry and ultrastructure to verify whether, in addition to phenotype, pollen could also show characteristics useful to discriminate the two infraspecific taxa. C. sempervirens var. pyramidalis compared to C. sempervirens var. horizontalis has mainly larger, subcircular pollen with a higher percentage of apolar and germinating grains, and a wall with thicker intine; the cytoplasm has a greater number of callose-containing vesicles. C. sempervirens var. horizontalis has pollen distributed in five classes of shape. In addition it is smaller and shows a higher percentage of ruptured grains, and a lower percentage of germination in vitro. The cytoplasm contains vesicles rich in callose and also a higher amount of osmophilic bodies. Pollen grains in both varietas contain only one cell at dispersal. The reported differences in pollen morphometry, structure and biology, together with the phenotypic characteristics of the tree, support the validity of two infraspecific taxa.  相似文献

Two names are here validated at the rank of variety, with the specimens designated as types. 取荷兰鸢尾(IrisxiphiumL.var.hybridum)鳞茎片不同部位外植体块,接种于附加不同激素配比的基本培养基上,其中取自鳞茎片基部外植体块2mm×2mm×2mm,培养基为MS+BA1.0mg/L+NAA0.2mg/L的不定芽诱导率为最高(70%);最理想的增殖培养基为MS+BA2.0mg/L+NAA0.2mg/L。不定芽直径4~5mm,培养基为MS+BA0.2mg/L+NAA0.5mg/L有利于不定根的发生,诱导生根率达833%。试管苗不经练苗可直接出瓶,移栽于泥炭∶田土∶河沙=1∶1∶1(V/V)的基质中。  相似文献

Abstract

Embryological researches on SENECIO VULGARIS L. var. THYRRENUS Fiori. — Male gametophyte, development of tapetal cells and female gametophyte have been studied in Senecio vulgaris L. var thyrrenus Fiori.

1) The development of male gametophyte is normal. Divisions of the microspore mother cells are of the simultaneous type. The division of the generative nucleus has never been observed till the pollen grain was in the anther.

2) The tapetal cells follow a very simple development. The nucleus of each cell divides only twice starting at the same time with the meiotic divisions of pollen mother cells but ending much earlier; subsequently, as usually happens with the Asteraceae, the ameboid involution of the tapetum begins. Endomitosis or any other process which leads to a polyploidy not due to nuclear fusion, has never been observed.

3) The female gametophyte is eight nucleate of the normal type (Polygonum). At maturity it shows only three antipodal cells whose nucleus undergoes at first, two or three divisions. Only later these new nuclei, always within the antipodal cell, may fuse in a polyvalent one.  相似文献

A main pungent amide, spilanthol (1), and three alkamides, (2E)-N-(2-methylbutyl)-2-undecene-8,10-diynamide (2), (2E,7Z)-N-isobutyl-2,7-tridecadiene-10,12-diynamide (3), and (7Z)-N-isobutyl-7-tridecene-10,12-diynamide (4) were isolated from the flower heads of Spilanthes acmella L. var. oleracea Clarke. Their structures were established by spectroscopic methods. Compounds 2 and 4 were new and 3 was found for the first time in Spilanthes species. Chemotaxonomic aspects are discussed.  相似文献

Mango (>Mangifera indica) belonging to Anacardiaceae family is a fruit that grows in tropical regions. It is considered as the King of fruits. The present work was taken up to identify a tool in identifying the mango species at the molecular level. The chloroplast trnL-F region was amplified from extracted total genomic DNA using the polymerase chain reaction (PCR) and sequenced. Sequence of the dominant DGGE band revealed that Mangifera indica in tested leaves was Mangifera indica (100% similarity to the ITS sequences of Mangifera indica). This sequence was deposited in NCBI with the accession no. GQ927757.
