As an important opportunistic pulmonary pathogen, Pneumocystis carinii has been the focus of extensive research over the decades. The use of laboratory animal models has permitted a detailed understanding of the host-parasite interaction but an understanding of the basic biology of P. carinii has lagged due in large part to the inability of the organism to grow well in culture and to the lack of a tractable genetic system. Molecular techniques have demonstrated extensive heterogeneity among P. carinii organisms isolated from different host species. Characterization of the genes and genomes of the Pneumocystis family has supported the notion that the family comprises different species rather than strains within the genus Pneumocystis and contributed to the understanding of the pathophysiology of infection. Many of the technical obstacles in the study of the organisms have been overcome in the past decade and the pace of research into the basic biology of the organism has accelerated. Biochemical pathways have been inferred from the presence of key enzyme activities or gene sequences, and attempts to dissect cellular pathways have been initiated. The Pneumocystis genome project promises to be a rich source of information with regard to the functional activity of the organism and the presence of specific biochemical pathways. These advances in our understanding of the biology of this organism should provide for future studies leading to the control of this opportunistic pathogen.  相似文献

The total lipid content, neutral and polar lipid composition, and fatty acid profiles were determined for the different developmental stages of the entomopathogenic fungus, Nomuraea rileyi. In general, the lipid composition of N. rileyi was found to be very similar to that of other Fungi Imperfecti. Triacylglycerides and sterols were the major neutral lipids and phosphatidylethanolamine, phosphatidylinositol, and phosphatidylserine were the major polar lipids. Phosphatidylcholine, resolved in conidial and mycelial samples, was not detected in hyphal body samples. The major fatty acids were palmitic, oleic, and linoleic. Stearic acid was detected as a minor component of hyphal body samples. During conidiogenesis the overall lipid composition was altered in (1) a reduction in total lipid content, (2) an increase in the polar/neutral lipid ratio, and (3) an increase in the degree of fatty acid unsaturation.  相似文献

Metarhizium anisopliae was grown by solid-state fermentation using a medium based on a mixture of rice bran and rice husk (1:1 on a dry weight basis) where the initial water content was fixed at a value of 47%. The experiments were performed in glass column bioreactors of three different sizes. Moist saturated air was continuously passed through the bottom of each column at a flow rate of 0.34 l h−1 g−1 initial dry matter, and cultivation was performed for two weeks at 27 ± 1°C. The effect of medium packing density was studied in the columns of small size. For initial apparent densities of 0.270 and 0.357 g ml−1, no significant differences were observed either in total biomass production or spore yields; however, when the initial apparent density was increased up to a value of 0.496 g ml−1, both growth and sporulation were strongly affected.

Thermal gradients in the medium and larger columns packed at an apparent density of 0.357 g ml−1 were observed both in the radial and axial directions. Heat build-up effects were analyzed in the larger column. In this case, there were significant differences between different segments of the column both in the water content of the fermented matter and the sporulation pattern.  相似文献

白蚁菌圃存在于白蚁巢中,具有硬而脆的多孔结构,是特殊的真菌生存环境。当有白蚁在白蚁巢内活动时,蚁巢伞Termitomyces是菌圃上的优势菌;当白蚁巢被废弃,炭角菌Xylaria成为菌圃上的优势真菌。菌圃中还存在其他微生物如无性型真菌(anamorphic fungi)和酵母等。菌圃中的真菌很多具有潜在药用价值或其他经济价值。从蚁巢伞、炭角菌等主要真菌类群出发,结合分子生态学研究菌圃真菌多样性的方法,综述了白蚁菌圃真菌多样性的研究进展,揭示了目前的研究热点及存在的问题,并针对这些问题提出可能的发展方向。  相似文献

The present aeromycological investigation was undertaken to study atmospheric fungal spores in Timi?oara (western Romania). This study was carried out using a Hirst type volumetric sampler. The study revealed the existence of a rich airborne mycoflora. The atmospheric fungal spores were classified and evaluated into three groups (‘major’, ‘minor’ and ‘sporadic’) depending upon their catch percentage in the air. Cladosporium/Fusarium/Leptosphaeria-group, Alternaria, Helminthosporium airborne fungal spores and airborne fungal fragments regularly recorded (frequency 100% of days). Cladosporium accounted for 81.09% of the outdoor fungal spores. The airborne fungal fragments have been identified as abundant in our geographic area. Spearman’s correlations were applied to meteorological parameters and airborne fungal spore concentrations. In addition, correlations were calculated between the fungal spore concentrations and the meteorological variables from the previous day. A total of eleven weather factors were selected for this investigation. Following Spearman’s correlations, I identified two patterns of behaviour: most of the airborne fungal spores prefer cloudiness, lower near-surface soil temperature, lower atmospheric pressure, higher relative humidity and precipitation (pattern A) while other spore concentrations favour increased sunshine, higher near-surface soil temperature and dry conditions (pattern B). The behaviour of some fungal spores during the warm season has proven unclear (pattern C). This study demonstrates the need for investigations throughout the year and the evaluation with complementary statistical methods, regarding the correct interpretation of airborne mycoflora relationships with meteorological parameters.  相似文献

Fungi are a highly diverse group of heterotrophic eukaryotes characterized by the absence of phagotrophy and the presence of a chitinous cell wall. While unicellular fungi are far from rare, part of the evolutionary success of the group resides in their ability to grow indefinitely as a cylindrical multinucleated cell (hypha). Armed with these morphological traits and with an extremely high metabolical diversity, fungi have conquered numerous ecological niches and have shaped a whole world of interactions with other living organisms. Herein we survey the main evolutionary and ecological processes that have guided fungal diversity. We will first review the ecology and evolution of the zoosporic lineages and the process of terrestrialization, as one of the major evolutionary transitions in this kingdom. Several plausible scenarios have been proposed for fungal terrestralization and we here propose a new scenario, which considers icy environments as a transitory niche between water and emerged land. We then focus on exploring the main ecological relationships of Fungi with other organisms (other fungi, protozoans, animals and plants), as well as the origin of adaptations to certain specialized ecological niches within the group (lichens, black fungi and yeasts). Throughout this review we use an evolutionary and comparative‐genomics perspective to understand fungal ecological diversity. Finally, we highlight the importance of genome‐enabled inferences to envision plausible narratives and scenarios for important transitions.  相似文献

The morphological effects of a fungal sex hormone, trisporic acid (TA), and a synthetic insect juvenile hormone (JH) on mycelial cultures derived from protoplasts of the fungus Entomophthora egressa were determined. In comparison with the control treatment (no added hormone), only one treatment (JH and TA both added at 40 μg/50 ml) produced all of the control cell types. Under both experimental conditions, normal hypha (without swollen tips), spherical hyphal bodies, irregularly shaped hyphal bodies, and thick-walled spheres were present. This JH plus TA treatment differed from the control in that normal rod-shaped hyphal bodies were also present and the thick-walled structures tended to be variable in shape. Hyphal tip swelling which did not lead to conidium production was common to all treatments in which only JH was added. The results of the addition of various concentrations and combinations of JH and TA with regard to the production of thick-walled cell types and three forms of hyphal bodies are given and discussed.  相似文献

Fungi are a megadiverse group of organisms, they play major roles in ecosystem functioning and are important for human health, food production and nature conservation. Our knowledge on fungal diversity and fungal ecology is however still very limited, in part because surveying and identifying fungi is time demanding and requires expert knowledge. We present a method that allows anyone to generate a list of fungal species likely to occur in a region of interest, with minimal effort and without requiring taxonomical expertise. The method consists of using a cyclone sampler to acquire fungal spores directly from the air to an Eppendorf tube, and applying DNA barcoding with probabilistic species identification to generate a list of species from the sample. We tested the feasibility of the method by acquiring replicate air samples from different geographical regions within Finland. Our results show that air sampling is adequate for regional‐level surveys, with samples collected >100 km apart varying but samples collected <10 km apart not varying in their species composition. The data show marked phenology, and thus obtaining a representative species list requires aerial sampling that covers the entire fruiting season. In sum, aerial sampling combined with probabilistic molecular species identification offers a highly effective method for generating a species list of air‐dispersing fungi. The method presented here has the potential to revolutionize fungal surveys, as it provides a highly cost‐efficient way to include fungi as a part of large‐scale biodiversity assessments and monitoring programs.  相似文献

Nowadays, artificial intelligence solutions such as digital image processing and artificial neural networks (ANN) have become important applicable techniques in phytomonitoring and plant health detection systems. In this research, an autonomous device was designed and developed for detecting two types of fungi (Pseudoperonospora cubensis, Sphaerotheca fuliginea) that infect the cucumber (Cucumis sativus L.) plant leaves. This device was able to recognise the fungal diseases of plants by detecting their symptoms on plant leaves (downy mildew and powdery mildew). For leaves of cucumber inoculated with different spores of the fungi, it was possible to estimate the amount of hour post inoculation (HPI) by extracting leaves’ image parameters. Device included a dark chamber, a CCD digital camera, a thermal camera, a light dependent resistor lightening module and a personal computer. The proposed programme for precise disease detection was based on an image processing algorithm and ANN. Three textural features and two thermal parameters from the obtained images were measured and normalised. Performance of ANN model was tested successfully for disease recognition and detecting HPI in images using back-propagation supervised learning method and inspection data. Such this machine vision system can be used in robotic intelligent systems to achieve a modern farmer’s assistant in agricultural crop fields.  相似文献

Liposomal amphotericin B (L-AmB) has been a key cornerstone for the management of invasive fungal infections (IFI) caused by a wide array of molds and yeasts during the last three decades. Multiple studies performed over this period have generated a large body of evidence on its efficacy and safety, becoming the main antifungal agent in the management of IFI in patients with hematologic malignancies in several not mutually exclusive clinical settings. First, L-AmB is the most commonly used antifungal agent in patients undergoing intensive chemotherapy for acute leukemia and high-risk myelodysplastic syndrome, as well as in hematopoietic stem cell transplant recipients. Additionally, due to the administration of newer targeted therapies (such as monoclonal antibodies or small molecule inhibitors), opportunistic mold infections are increasingly being reported in patients with hematologic malignancies usually considered low-risk for IFI. These agents usually have a high drug-drug interaction potential, being triazoles, commonly used for antifungal prophylaxis, included. Finally, patients developing breakthrough IFI because of either subtherapeutic concentrations of antifungal prophylactic drugs in blood or selection of resistant strains, require broad spectrum antifungal therapy, usually with an antifungal of a different class. In both situations, L-AmB remains as the best option for early antifungal therapy.  相似文献
