目前认为,1968年甲3型流感发生时,至少有二种H3N2亚型的变异株同时存在。一种是1968年7月在香港分离的A/HK/1/68;另一种是1968年8月在日本分离的A/Aichi/2/68。Webster等用一组抗A/Mem/1/77(H3N2)血凝素(HA)的单克隆抗体证实,二种变异株至少有一个反应部位不同。这可能是H3N2亚型有二种不同的世系(lineage),  相似文献

2009年A(H1N1)pdm09亚型流感病毒在墨西哥暴发,之后在全世界流行。为了解海南省2016-2018年A(H1N1)pdm09亚型流感病毒流行态势,分析血凝素(HA)与神经氨酸酶(NA)基因遗传进化特征与变异情况,本研究从中国流感监测信息系统获取海南省2016-2018年流感病毒病原学监测数据,选取5家流感监测网络实验室分离鉴定的37株A(H1N1)pdm09亚型流感毒株进行HA与NA基因测序,利用MEGA 10.1.8构建HA与NA基因种系进化树,并分析其氨基酸变异情况。结果显示,2016-2018年共出现3次A(H1N1)pdm09亚型流感病毒活动高峰。2017年10月份以后的分离株(4/8)与2018年大部分分离株(21/22)独立于疫苗株A/Michigan/45/2015聚为一个小支,发生20余处HA与NA氨基酸位点变异。与疫苗株A/California/7/2009(2010-2016)相比,2016-2018年流感病毒分离株在HA基因抗原决定簇上发生7处氨基酸变异并有一个潜在糖基化位点,未发现HA基因受体结合位点变异与NA基因耐药性变异。本研究提示,2016-2018年,A(H1N1)pdm09亚型流感病毒逐步发生规律性进化,氨基酸变异频率有增加趋势,今后应持续加强流感病毒病原学监测,密切追踪A(H1N1)pdm09亚型流感病毒基因变异情况,为科学防控提供理论依据。  相似文献

选择一个于1998年开始发生H9亚型禽流感的封闭式大型养鸡场,连续5年内分离到22株H9N2亚型病毒,对其中9株与1998年分离株进行HA基因序列和病毒抗原性的比较结果表明,这些分离株均与1998年的具有较高的序列同源性,且在本研究期内HA基因的这些变化尚未产生引起交叉保护性改变。初步推断这些分离株均系1998年分离株在场内循环传播变化得来,其HA基因的变异可能与频繁的疫苗免疫选择压力有关。这为进一步研究禽流感病毒变异的规律和制定正确的禽流感防治对策具有重要意义。  相似文献

提纯猪链球菌 2型江苏分离株HA980 1的毒力相关蛋白溶菌酶释放蛋白 (muramini dase releasedprotein ,MRP)和胞外因子 (extracellularfacter,EF) ,制备其抗体供免疫转印之用。另提取猪源链球菌 1 7株国内分离株、1株德国分离株及 1株猪链球菌 2型人分离株的胞壁和胞外蛋白 ,经SDS PAGE ,与HA980 1株的MRP和EF的抗体作免疫转印。 1 1株MRP阳性 ,1 0株EF及EF 阳性 ,MRP及EF的分布存在 4种表型 :MPR+ EF+ (8 1 9) ,MRP+ EF (1 1 9) ,MRP+ EF- (1 1 9) ,MRP- EF- (1 0 1 9)。  相似文献

从过去10年由中国中部分离的具有不同致病力的25株H9N2亚型禽流感选出6株(3#、12#、25#、14#、4#、22#)代表性毒株,利用RT-PCR扩增它们的HA基因,并比较分析该基因的序列,旨在探讨HA基因的变异对AIV毒力、抗原性变化的影响。结果表明:6株H9N2 AIV亚型分离株的HA基因在HA1和HA2的氨基酸裂解位点上没有出现高致病性禽流感病毒所特有的R-X-R/K-R模式,它们均为弱毒力毒株。HA上潜在糖基化位点除了3#和12#分离株多出一个之外,其余均为8个。3#和12#所表现出较强的致病性可能与其在HA的头部(HA1)的A抗原位点上多了一个糖基化位点(145~147aa),改变了HA基因空间构型有关,空间构型的改变导致抗HA抗体作用位点的变异或缺失并影响其较近的受体结合位点,从而改变该毒株的抗原性。研究结果提示需要持续跟踪H9N2 AIV在中国鸡群中的传播和进化,以便及时掌握疫情,有效防控禽流感。  相似文献

比较加入神经氨酸酶抑制剂奥司他韦(Oseltamivir)对我国A(H3N2)亚型流感病毒红细胞凝集(Hemagglutination,HA)和凝集抑制(Hemagglutinin inhibition,HI)试验结果的影响,以期获得病毒更真实的HA和抗原性变异分析结果。选择2014年10月-2015年5月在中国大陆分离的395株A(H3N2)亚型流感病毒,在HA及HI试验中加入神经氨酸酶抑制剂Oseltamivir,对实验结果进行比较分析,根据HA试验,挑选其中部分毒株进行基因组测序,比较NA氨基酸位点变异情况。在HA试验中加入神经氨酸酶抑制剂Oseltamivir后,44.8%的毒株HA滴度未改变;43.8%的毒株HA滴度下降,仅有11.4%的毒株HA滴度升高。加入Oseltamivir后,与A/TX/50/2012鸡胚株抗原性类似的毒株的比例高于未加入Oseltamivir时,与A/SZ/9715293/13细胞株抗原性类似的毒株的比例低于未加入Oseltamivir时类似株的比例,统计学分析有显著性差异。以A/TX/50/2012细胞分离株和A/SZ/9715293/133鸡胚分离株作为参考病毒,Oseltamivir对实验结果的影响无显著性差异。挑选19株A(H3N2)亚型流感毒株进行基因组测序,进行NA蛋白氨基酸位点分析,与A/TX/50/2012鸡胚分离株相比加入20nM Oseltamivir后,滴度降低超过4倍的5株毒株,没有共同氨基酸位点变异,但A/山东莱城/119/2015流感毒株有D151G氨基酸位点突变;A/吉林铁西/1194/2015流感毒株有V412I和T434A氨基酸位点变异。加入20nM Oseltamivir后,滴度降低为2~4倍的毒株,具有I26T、G93S、V149I、N234D、T267K、S416G等位点突变。在对我国近年流行的A(H3N2)亚型流感病毒进行血凝滴度和抗原性分析中,加入Oseltamivir可获得更为真实的HA和HI结果,分析病毒的变异情况,评价疫苗的匹配性。  相似文献

以两株甲2型流感病毒株贵57-1、甘66-3(H2N2)和两株甲3型流感病毒株京科68-1、穗防74-315(H3N2)为代表株,测定温度、pH及金属离子对感染的鸡胚尿囊液病毒神经氨酸酶(NA)活性的影响,并用NA交互抑制试验检查贵57-1NA与穗防74-135NA之间的N抗原变异情况。 结果表明,在50℃加热30分钟4株病毒的NA活性仅下降原活性的10%以下;但56°C30分钟加热,两株甲2型病毒(H2N2)的NA活性明显下降,仅为原活性的12%以下;而两株甲3型(H3N2)则分别为原活性的34%及37%。根据Paniker和Tams等的报道,N2的活性对45℃及56℃加温比N1更稳定。似乎表明,变异着的NA抗原有越来越耐热的趋势。  相似文献

2009年6月12日,江苏确诊首例甲型H1N1(2009)病例。通过细胞和鸡胚分离系统,我们分离到一株具有较高血凝活性的病毒,命名为A/Jiangsu/1/2009。为了跟踪病毒的变异情况,我们开展了病毒的全基因组测序工作,在此基础上对其血凝素基因(Haemagglutinin,HA)的遗传特性进行了详细研究。分离株HA蛋白不具有多碱基HA裂解位点,具有低致病性流感病毒特点。与参考株A/California/04/2009相比,分离株A/Jiangsu/1/2009HA蛋白的有4个氨基酸发生了突变,但都不在已知的抗原位点上。分离株有5个潜在糖基化位点,这与近年来古典猪H1N1和北美三源重配猪H1病毒完全一致,保留了古典猪H1的特点。与禽流感H1病毒相比,分离株HA蛋白受体结合位点上的E190D和G225D发生突变,这可能成为新甲型H1N1(2009)在人际间传播的一个重要分子基础。此外,其它受体结合位点上相关氨基酸同时具有人和猪流感病毒的特点。本研究首次对早期流行的甲型H1N1(2009)流感病毒的HA蛋白的分子遗传特征进行了详细研究,对进一步监测病原变异具有重要指导意义。  相似文献

为分析福建省输入性D8基因型麻疹病毒分子流行病学特征,采集咽拭子标本采用实时荧光定量反转录-聚合酶链反应(Real-time RT-PCR)筛查麻疹病毒核酸.用反转录-聚合酶链反应(RT-PCR)扩增麻疹病毒核酸筛查阳性咽拭子及Vero/Slam细胞培养阳性产物,对麻疹病毒核蛋白(Nucleoprotein,N)羧基(COOH)端634个核苷酸(Nucleotides,nt)片段进行测序分析,构建系统进化树.最终分离获得1株麻疹病毒株,26条麻疹病毒N蛋白羧基末端450个nt序列.亲缘性分析发现,所有福建麻疹毒株与WHOD8基因型参考株(MVi/Manchester.GBR/30.94)在亲缘关系树上同属一个大分支,两者核苷酸序列和氨基酸(Amino acid,aa)序列同源性分别为96.4%~99.1%和96.7%~98.0%.其中2014年的福建毒株 MVs/Fujian.CHN/28.14和 MVs/Fujian.CHN/30.14与越南胡志明市2014年分离株MVs/HoChiMinh.VNM/11.14及美国纽约2013年分离株MVs/New.York.USA/19.13的nt同源性为 100%;2019年的毒株MVs/Fujian.CHN/25.19与泰国龙仔厝府2018年分离株MVs/Samut.Sakhon.THA/8.18的nt同源性为100%;而剩余的23个监测毒株则与日本神户2019年分离株MVs/KobeC.JPN/28.19的核苷酸同源性和氨基酸同源性最高,分别为99.6%~100.0%和99.3%~100.0%.在病毒N蛋白羧基端150个氨基酸位点上,福建株与WHO D8参考株存在3~5个氨基酸位点差异.而与现用的疫苗株(Shanghai-191)相比,存在17~19个氨基酸变异位点,其中有14个氨基酸位点为所有福建株共有的变异位点,这些位点的变异总体上未对编码蛋白的氨基酸造成明显改变.结论是福建省成功分离获得1株D8基因型麻疹毒株.D8基因型为福建省发现的输入性麻疹基因型,病毒N蛋白羧基端氨基酸位点上与疫苗株相比均出现了差异位点.  相似文献

In order to clarify the effect of an accumulation of amino acid substitutions on the hemadsorption character of the influenza AH3 virus hemagglutinin (HA) protein, we introduced single-point amino acid changes into the HA1 domain of the HA proteins of influenza viruses isolated in 1968 (A/Aichi/2/68) and 1997 (A/Sydney/5/97) by using PCR-based random mutation or site-directed mutagenesis. These substitutions were classified as positive or negative according to their effects on the hemadsorption activity. The rate of positive substitutions was about 50% for both strains. Of 44 amino acid changes that were identical in the two strains with regard to both the substituted amino acids and their positions in the HA1 domain, 22% of the changes that were positive in A/Aichi/2/68 were negative in A/Sydney/5/97 and 27% of the changes that were negative in A/Aichi/2/68 were positive in A/Sydney/5/97. A similar discordance rate was also seen for the antigenic sites. These results suggest that the accumulation of amino acid substitutions in the HA protein during evolution promoted irreversible structural changes and therefore that antigenic changes in the H3HA protein may not be limited.  相似文献

摘要:目的 了解2016?2017年辽宁省H3N2亚型流感病毒基因变异情况及流行株与疫苗株的匹配情况。方法 采用逆转录聚合酶链反应(RT-PCR)对分离得到的H3N2亚型流感毒株的HA1基因进行扩增,扩增片段经测序与近年来WHO推荐的北半球疫苗株进行比对和基因特征分析。结果 进化分析表明,2016?2017年H3N2亚型流感病毒与近三年的疫苗株均不在同一分支上;基因特性分析中,所有病毒均在A、B抗原决定簇上发生了两处以上的变异;19株病毒的受体结合位点131位氨基酸发生了新的变异;20株病毒中有1株突变产生了新的半胱氨酸,提示可能有新的二硫键产生;糖基化位点并未检测到新的突变。结论 2016?2017年辽宁省H3N2亚型流感病毒的抗原性及基因特性均发生了一定的变化,但变异程度不大,应密切关注疫苗株对流感病毒的免疫效果及流感毒株的变异情况。  相似文献

Antigenic drift forces us to frequently update influenza vaccines; however, the genetic basis for antigenic variation remains largely unknown. In this study, we used clade 7.2 H5 viruses as models to explore the molecular determinants of influenza virus antigenic variation. We generated eight monoclonal antibodies(MAbs) targeted to the hemagglutinin(HA) protein of the index virus A/chicken/Shanxi/2/2006 and found that two representative antigenically drifted clade 7.2 viruses did not react with six of the eight MAbs. The E131 N mutation and insertion of leucine at position 134 in the HA protein of the antigenically drifted strains eliminated the reactivity of the virus with the MAbs. We also found that the amino acid N131 in the H5 HA protein is glycosylated. Our results provide experimental evidence that glycosylation and an amino acid insertion or deletion in HA influence antigenic variation.  相似文献

Avian influenza viruses of subtype H5N1 circulating in animals continue to pose threats to human health. The binding preference of the viral surface protein hemagglutinin (HA) to sialosaccharides of receptors is an important area for understanding mutations in the receptor binding site that could be the cause for avian-to-human transmission. In the present work, we studied the effect of two receptor binding site mutations, S221P singly and in combination with another mutation K216E in the HA protein of influenza A H5N1 viruses. Docking of sialic acid ligands corresponding to both avian and human receptors and molecular dynamics simulations of the complexes for wild and mutant strains of H5N1 viruses were carried out. The H5N1 strain possessing the S221P mutation indicated decreased binding to α2,3-linked sialic acids (avian receptor, SAα2,3Gal) when compared to the binding of the wild-type strain that did not possess the HA-221 mutation. The binding to α2,6-linked sialic acids (human receptor, SAα2,6Gal) was found to be comparable, indicating that the mutant strain shows limited dual receptor specificity. On the other hand, the S221P mutation in synergism with the K216E mutation in the binding site, resulted in increased binding affinity for SAα2,6Gal when compared to SAα2,3Gal, indicative of enhanced binding to human receptors. The in-depth study of the molecular interactions in the docked complexes could explain how co-occurring mutations in the HA viral protein can aid in providing fitness advantage to the virus, in the context of host receptor specificity in emerging variants of H5N1 influenza viruses.  相似文献

Virus growth during influenza vaccine manufacture can lead to mutations that alter antigenic properties of the virus, and thus may affect protective potency of the vaccine. Different reassortants of pandemic "swine" H1N1 influenza A vaccine (121XP, X-179A and X-181) viruses as well as wild type A/California/07/2009(H1N1) and A/PR/8/34 strains were propagated in embryonated eggs and used for DNA/RNA Illumina HiSeq and MiSeq sequencing. The RNA sequences of these viruses published in NCBI were used as references for alignment of the sequencing reads generated in this study. Consensus sequences of these viruses differed from the NCBI-deposited sequences at several nucleotides. 121XP stock derived by reverse genetics was more heterogeneous than X-179A and X-181 stocks prepared by conventional reassortant technology. Passaged 121XP virus contained four non-synonymous mutations in the HA gene. One of these mutations (Lys226Glu) was located in the Ca antigenic site of HA (present in 18% of the population). Two non-synonymous mutations were present in HA of viruses derived from X-179A: Pro314Gln (18%) and Asn146Asp (78%). The latter mutation located in the Sa antigenic site was also detected at a low level (11%) in the wild-type A/California/07/2009(H1N1) virus, and was present as a complete substitution in X-181 viruses derived from X-179A virus. In the passaged X-181 viruses, two mutations emerged in HA: a silent mutation A1398G (31%) in one batch and G756T (Glu252Asp, 47%) in another batch. The latter mutation was located in the conservative region of the antigenic site Ca. The protocol for RNA sequencing was found to be robust, reproducible, and suitable for monitoring genetic consistency of influenza vaccine seed stocks.  相似文献

We have succeeded in engineering changes into the genome of influenza B virus. First, model RNAs containing the chloramphenicol acetyltransferase gene flanked by the noncoding sequences of the HA or NS genes of influenza B virus were transfected into cells which were previously infected with an influenza B helper virus. Like those of the influenza A viruses, the termini of influenza B virus genes contain cis-acting signals which are sufficient to direct replication, expression, and packaging of the RNA. Next, a full-length copy of the HA gene from influenza B/Maryland/59 virus was cloned. Following transfection of this RNA, we rescued transfectant influenza B viruses which contain a point mutation introduced into the original cDNA. A series of mutants which bear deletions or changes in the 5' noncoding region of the influenza B/Maryland/59 virus HA gene were constructed. We were able to rescue viruses which contained deletions of 10 or 33 nucleotides at the 5' noncoding region of the HA gene. The viability of these viruses implies that this region of the genome is flexible in sequence and length.  相似文献

为了解2006年广州地区流行的乙型流感病毒株血凝素(HA)和神经氨酸酶(NA)的基因特性,选择病原学监测病毒株和暴发性疫情病毒株,提取病毒RNA并逆转录为cDNA,通过PCR方法扩增乙型流感病毒HA和NA全长基因,将扩增的DNA片段接入T-A克隆载体进行测序,并使用DNAStar软件对测序结果进行分析。结果显示:不同来源的流感病毒株HA的同源性为99%以上,都属于Victoria系;不同来源的病毒株NA同源性为98%以上。HA和NA的种系发生树分析表明:病原学监测毒株同源性更接近,而暴发性疫情毒株的同源性则相对较为分散。所有毒株与WHO推荐的2005~2006年度疫苗株B/Shanghai/361/2002的同源性只有88.9%~89.7%,说明该年度的流感疫苗对乙型流感不能提供最佳的保护。  相似文献

In this study, the effect of innate serum inhibitors on influenza virus infection was addressed. Seasonal influenza A(H1N1) and A(H3N2), 2009 pandemic A(H1N1) (H1N1pdm) and highly pathogenic avian influenza (HPAI) A(H5N1) viruses were tested with guinea pig sera negative for antibodies against all of these viruses as evaluated by hemagglutination-inhibition and microneutralization assays. In the presence of serum inhibitors, the infection by each virus was inhibited differently as measured by the amount of viral nucleoprotein produced in Madin-Darby canine kidney cells. The serum inhibitors inhibited seasonal influenza A(H3N2) virus the most, while the effect was less in seasonal influenza A(H1N1) and H1N1pdm viruses. The suppression by serum inhibitors could be reduced by heat inactivation or treatment with receptor destroying enzyme. In contrast, all H5N1 strains tested were resistant to serum inhibitors. To determine which structure (hemagglutinin (HA) and/or neuraminidase (NA)) on the virus particles that provided the resistance, reverse genetics (rg) was applied to construct chimeric recombinant viruses from A/Puerto Rico/8/1934(H1N1) (PR8) plasmid vectors. rgPR8-H5 HA and rgPR8-H5 HANA were resistant to serum inhibitors while rgPR8-H5 NA and PR8 A(H1N1) parental viruses were sensitive, suggesting that HA of HPAI H5N1 viruses bestowed viral resistance to serum inhibition. These results suggested that the ability to resist serum inhibition might enable the viremic H5N1 viruses to disseminate to distal end organs. The present study also analyzed for correlation between susceptibility to serum inhibitors and number of glycosylation sites present on the globular heads of HA and NA. H3N2 viruses, the subtype with highest susceptibility to serum inhibitors, harbored the highest number of glycosylation sites on the HA globular head. However, this positive correlation cannot be drawn for the other influenza subtypes.  相似文献

从GenBank上获得194株不同来源的H5N1亚型禽流感病毒HA基因核酸序列,利用MEGA3分析了HA基N核酸序列碱基突变的特点。并通过比较序列同源性,构建NJ系统进化树,探讨了不同来源的H5N1病毒的系统进化关系。序列分析结果表明,被研究的194条H5N1亚型禽流感病毒HA基因核酸序列,碱基长度大约在1700bp左右,共发现了757个可变位点,其中Parsimony—informative sites有537个,Singleton sites有220个;病毒的变异速率很快,平均变异率为3.23%;病毒的序列变异具有显著的地区特点和时间特点;同时,全球化的贸易以及候鸟的迁徙在传播病毒过程中起一定作用。  相似文献

从GenBank上获得我国人(Homo sapiens)、家禽和野鸟42株H5N1亚型禽流感病毒的HA基因核酸序列,利用DNAStar分析HA蛋白关键位点氨基酸残基的变化,比较HA基因核苷酸序列同源性,构建遗传进化树.探讨我国部分人、家禽和野鸟H5N1病毒基因的遗传进化关系.序列分析结果表明:禽流感病毒H5N1亚型的HA基因持续地发生着变异,但并非以均一速度进行,时间间隔愈长,核苷酸同源性愈低;我国同一地区或临近地区,当年或前后两年发生的人及家禽感染的禽流感病毒高度同源.推测我国部分人发生的禽流感可能是通过家禽感染的;候鸟的迁徙在传播病毒过程中所起的作用有待深入探讨.  相似文献
