百年来关于扬子鳄的分类学位置存在着很多争议,本测得扬子鳄、暹罗鳄和湾鳄的mtDNA ND4和Cytb基因,并从GenBank中获得密西西比鳄和海龟的DN4基因和Cytb基因相应片段。用Clustal X1.8进行对位排列,以海龟为外群构建分子进化系统树。结果显示,在鳄类动物中,扬子鳄与密西西比鳄的亲缘关系最近,两ND4基因序列碱基差异的20.68%,而Cytb基因序列碱基差异为14.43%,但扬子鳄与密西西相比与鳄的分类问题仍将有待进一步探讨。  相似文献

Aimed to evaluate the phylogenetic position of the recently described Protobothrops dabieshanensis Huang et al. (2012), phylogenic relationships of 12 species within Protobothrops based on four mtDNA gene fragments (12S RNA, 16S RNA, ND4 and Cyt b) were reconstructed in our study. The result indicates a clade composed ofP dabiesha- nensis, P. jerdonii and P xiangchengsis with strong support. The genetic distance among P dabieshanensis, P jerdonii and P xiangchengsis was much lower than other congeners. Based on the data from the phylogenetic analysis and pre- viously described morphological differences, we conclude that P dabieshanensis is a valid species with close affinities to P jerdonii and P xiangchengsis.  相似文献

The sequences of the mitochondrial 16S rRNA gene of 16 Oedipodidae species were amplified and sequenced. All sequences were aligned and analyzed and the phylogenetic relationships were inferred. The properties of 16S gene in Oedipodidae showed typical patterns of many insects such as a high A+T content and variable distance-dependent transition/transversion ratios. The 0.2 weight for sites of loops may be advisable for phylogeny reconstruction using the maximum parsimony method. The phylogenetic analysis results do not support the current subfamily classification systems of Oedipodidae. Bryodemellinae and Bryodeminae are closely related and should be merged as one subfamily. The status of Oedipodinae and Locustinae is also problematic.  相似文献

In this study,both long PCR and conserved primers walking sequencing methods were used to determine the complete sequence of the of Pyrgilauda ruficollis mitochondrial genome(KC836121).The results showed that the complete mitochondrial genome of P.ruficollis is 16909 bp in length with 55.0%A+T content,harboring the typical 37 genes.The mitogenome had the same gene order with that of Podoces hendersoni.All protein coding genes started with ATG codon,except ND3 with GTG.For the stop codon usage,most genes terminate with codons TAA or TAG,but ND5 terminated with AGA,while ND1 and COI genes with AGG,and both the genes COIII and ND4 have an incomplete termination codon(T).The secondary structures of 22 tRNA genes were also predicted,showing that all tRNAs can form typical clover-leaf secondary structures,except for the tRNASer(AGN)which loses the DHU arm,while tRNAPhe harbor an extra nucleotide inserted in the TψC arm.The predicted secondary structures of 12S rRNA and16S rRNA exhibit 47 helices in 4 domains and 60 helices in 6 domains respectively.The control region of P.ruficollis with the length of 1 305 bp was located between tRNAGlu and tRNAPhe,and typical domains of which could be found as other bird groups.Using the data from 13 mitochondrial protein-coding genes,results of a final phylogenetic analysis strongly supports the traditional view that P.ruficollis is closely related with Passeridae and Fringillidae.  相似文献

从1 2S rRNA基因序列探讨8种鳄类的系统学关系   总被引:8,自引:0,他引:8  
测得扬子鳄(Alligator sinensis)和暹罗鳄(Crocodylus siamensis),的mtDNA12SrRNA基因片段的部分序列,与GenBank中的2种鳄及文献中4种鳄的12SrRNA基因相应片段,经比对后构建系统树。其结果显示,现存鳄类为单系起源,可划分为3个科,即:鳄科、食鱼鳄科和假食鱼鳄科。食鱼鳄与假食鱼鳄亲缘关系较近,支持将假食鱼鳄作为食鱼鳄科的一个属,与以形态学为基础的研究结果不同。在钝吻鳄科中,扬子鳄与凯门鳄亲缘关系较远,而与密西西比鳄的亲缘关系虽较近,但它们的确存在很多差异。两者12SrRNA基因序列差异达12.12%,碱基的颠换数为9。在基于碱基转换和颠换的NJ系统树及仅基于碱基颠换的NJ系统树中,前者支持扬子鳄与密河鳄间的亲缘关系较近,而后者支持扬子鳄与凯门鳄间的亲缘关系较近,说明扬子鳄与密河鳄间亲缘关系是值得研究的问题。因本文仅根据12SrRNA基因部分序列来进行分析的,尚需进一步研究。  相似文献

The complete sequence of the 18 S ribosomal RNA gene(18S rRNA) from Lymantria dispar was cloned and analysed here. 18 S rRNA and mitochondrial cytochrome c oxidasel(cox1) gene sequences of Lymantria dispar were compared with homologous sequences of other nine insects from different orders. Analytic results showed that 18 S rRNA of these insects had two conserved domains and the second domain was an even more conserved region. The phylogenetic trees based on the full-length sequence and the second domain fragment of 18 S rRNA as well as sequence of cox1 from different orders indicated that Lepidoptera and Trichoptera, which belongs to Amphiesmenoptera, had a closer phylogenetic relationship and fewer differences were observed comparing with traditional taxonomic results.  相似文献

To investigate genetic mechanisms of high altitude adaptations of native mammals on the Tibetan Plateau, we compared mitochondrial sequences of the endangered Pantholops hodgsonii with its lowland distant relatives Ovis ames and Capra hircus, as well as other mammals. The complete mitochondrial genome of P. hodgsonii (16,498 bp) revealed a similar gene order as of other mammals. Because of tandem duplications, the control region of P. hodgsonii mitochondrial genome is shorter than those of O. ames and C. hircus, but longer than those of Bos species. Phylogenetic analysis based on alignments of the entire cytochrome b genes suggested that P. hodgsonii is more closely related to O. ames and C. hircus, rather than to species of the Antilopinae subfamily. The estimated divergence time between P. hodgsonii and O. ames is about 2.25 million years ago. Eutther analysis on natural selection indicated that the COXI (cytochrome c oxidase subunit I) gene was under positive selection in P. hodgsonii and Bos grunniens. Considering the same climates and environments shared by these two mammalian species, we proposed that the mitochondrial COXI gene is probably relevant for these native mammals to adapt the high altitude environment unique to the Tibetan Plateau.  相似文献

DNA barcoding is a powerful approach for characterizing species of organisms, especially those with almost identical morphological features, thereby helping to to establish phylogenetic relationships and reveal evolutionary histories. In this study, we chose a 648-bp segment of the mitochondrial gene, cytochrome c oxidase subunit 1 (COI), as a standard barcode region to establish phylogenetic relationships among brine shrimp (Artemia) species from major habitats around the world and further focused on the biodiversity of Artemia species in China, especially in the Tibetan Plateau. Samples from five major salt lakes of the Tibetan Plateau located at altitudes over 4,000 m showed clear differences from other Artemia populations in China. We also observed two consistent amino acid changes, 153A/V and 183L/F, in the COI gene between the high and low altitude species in China. Moreover, indels in the COI sequence were identified in cyst and adult samples unique to the Co Qen population from the Tibetan Plateau, demonstrating the need for additional investigations of the mitochondrial genome among Tibetan Artemia populations.  相似文献

This study focuses on taxonomy of Uromastyx species. Morphologically, U. ocellata and U. ornata were identified and cytogenetically, 2n = 36 chromosome. 16S, cyt b and 12S genes fragmentamplification shows 597, 399 and 450 base pair respectively and more A-T than G-C base pair andmore A/C than T/G base contents. While, sequences blasting in GenBank, followed by construction of phylogenetic tree revealed, 16S and cry b sequences were similar to U. ocellata sequences, andclustered together in phylogenetic tree, in contrast, 12S sequences from same species were related tosubspecies U. ornata ornata sequences and clustered together in the phylogenetic tree. Therefore, the results of this study suggest more studies on Uromastyx species in Sudan.  相似文献

Classification of the Asian snake genera Lycodon and Oligodon has proven challenging. We conducted a molecular phylogenetic analysis to estimate the phylogenetic relationships in the genus of Lycodon and clarify the taxonomic status of Oligodon multizonatum using mitochondrial(cyt b, ND4) and nuclear(c-mos) genes. Phylogenetic trees estimated using Maximum Likelihood and Bayesian Inference indicated that O. multizonatum is actually a species of Lycodon. Comparing morphological data from O. multizonatum and its closest relatives also supported this conclusion. Our results imply that a thorough review of the evolutionary relationships in the genus of Lycodon is strong suggested.  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献
