Luminescence characteristics of gramine, dimethyltryptamine and related indoles on Si gel TLC are described, chromatographic conditions are devised which enhance the luminescence, and a fluorometric TLC scanning procedure is developed for the quantitative determination of gramine in Phalaris arundinacea. 5-Methoxy-and 5-hydroxyindoles are readily distinguished from the indoles lacking a polar substituent at position 5.  相似文献

The grass Festuca arundinacea is often planted for slope stabilisation in South Korea, and is spreading widely beyond the introduction sites. This study used a functional group approach to examine the resistance of plant combinations to invasion by F. arundinacea based on the limiting similarity and diversity-resistance hypotheses, and to elucidate the process of colonisation. The study simulated the environment of construction sites and surrounding areas that might be encountered by expanding populations of F. arundinacea. The role of nutrient condition in the ranking of functional group competitive ability was also examined. Twelve native plant species were categorised into three functional groups using combinations of functional traits. Pairwise (one-to-one competition), multiple (four different neighbouring species) and monoculture experimental settings were designed using two nutrient levels. The Relative Competition Index was used to interpret the competitive effect of neighbouring species on F. arundinacea. Species of the same functional group as F. arundinacea were unable to resist invasion, but annual plants with niche preemption ability could outcompete it. Competitive relationships between native plants and F. arundinacea were explained partially by functional group identity but were inconsistent with the limiting similarity hypothesis and the diversity-interaction. Unforeseen interactions within the artificial communities also produced unexpected effects. In designing artificial plant communities, it is necessary to consider functional traits that reflect the species characteristic of particular periods and indirect effects that modify the interaction between other species.  相似文献

We investigated the responses of growth and non-structural carbohydrates to submergence and de-submergence in three wetland macrophyte species. Survival rate, recovery ability, and soluble sugar and starch contents of flood-tolerant Polygonum hydropiper and of flood-sensitive Phalaris arundinacea and Carex argyi from Dongting Lake wetlands were investigated after 20, 40, and 60 days of complete submergence without light and 10 days after de-submergence. Plant dry weight and soluble sugar and starch contents decreased in all species during the submergence period. The decreases were slowest in P. hydropiper, intermediate in C. argyi, and most rapid in P. arundinacea. After 60 days of submergence, survival rates were 100, 50, and 0 % in P. hydropiper, C. argyi, and P. arundinacea, respectively. After recovery, plant dry weight increased in P. hydropiper and in C. argyi, but decreased in P. arundinacea. Compared to pre-submergence, soluble sugar contents generally increased and then remained relatively constant after recovery in all species, while starch content increased in P. hydropiper and decreased in P. arundinacea with increasing submergence time. For C. argyi, starch content decreased after recovery from the 20-day submergence, but increased after recovery from the 40- and 60-day submergences. These data illustrate mechanisms behind the flood tolerance of P. hydropiper and the sensitivity to flooding in P. arundinacea and C. argyi. These mechanisms include lower consumption and quicker accumulation of non-structural carbohydrates in flood-tolerant plants.  相似文献

Mouse chromosomes, with the exception of the Y chromosome, are telocentric. The telomere at the p-arm is separated from the centromere by the tL1 sequence and TLC tandem repeats. A previous report showed that the TLC array was also conserved in other strains of the subgenus Mus. These results suggest that the TLC arrays promote the stable evolutionary maintenance of a telocentric karyotype in the subgenus Mus. In this study, we investigated the degree of conservation of TLC arrays among a variety of wild-derived inbred strains, all of which are descendants of wild mice captured in several areas of the world. Genomic PCR analysis indicates that the sequential order of telomere-tL1 is highly conserved in all strains, whereas tL1-TLC is not. Next, Southern blot analysis of DNAs isolated from a panel of mouse subspecies showed both Mus musculus domesticus and Mus musculus castaneus subspecies possess TLC arrays. Unexpectedly, this repeat appears to be lost in almost all Mus musculus musculus and Mus musculus molossinus subspecies, which show a clear geographic divide. These results indicate that either other unknown sequences were replaced by the TLC repeat or almost all M. m. musculus and M. m. molossinus subspecies do not have any sequence between the telomere and minor satellites. Our observation suggests that the TLC array might be evolutionarily unstable and not essential for murine chromosomal conformation. This is the first example of the subspecies-specific large genome alterations in mice.  相似文献

Simple-sequence repeats (SSRs) comprising three tetranucleotide repeat sequences with two-base ’anchors’, namely 5′-(AGAC)4GC, 5′-AC(GACA)4 and 5′-(GACA)4GT, were used in PCR reactions as primers to develop inter-SSR DNA fingerprints of the outbreeding grass species Lolium multiflorum, L. perenne, Festuca pratensis and F. arundinacea. Each species was represented by DNA samples from 3 to 6 varieties. In all four species distinctive species-specific DNA profiles were produced that were common across a number of varieties despite their diverse origin. While the fingerprints of the two ryegrasses, L. multiflorum and L. perenne, were the most similar, a number of inter-SSR DNA markers were generated that enabled them to be distinguished from each other. Some slight variations were found between varieties, which provided putative variety-specific markers for cultivar identification. In addition, variations in the DNA profiles of the genotypes of L. multiflorum and F. pratensis were examined, and the results showed that variety-specific fingerprints are integrated patterns made up from the profiles of individual genotypes. Amongst the primers used, AC(GACA)4 generated the best distinction between Lolium and Festuca individuals and provides an effective new tool for genome identification. A number of species-discriminating sequences, ranging in size between 550 bp and 1,600 bp, were cloned: three clones for F. pratensis, one clone for L. multiflorum and one clone for F. arundinacea. A F. pratensis fragment pFp 78H582 was sequenced. Southern hybridization confirmed the presence of this fragment in F. arundinacea (which contains one genome of F. pratensis), but no homology was found with L. multiflorum. However, a F. arundinacea clone amplified with (GACA)4GT, pFa 104H1350, was found to be unique to the F. arundinacea genome. Received: 23 June 1999 / Accepted: 27 August 1999  相似文献

Vascular endothelial growth factor (VEGF) is up-regulated during mesothelial to mesenchymal transition (MMT) and has been associated with peritoneal membrane dysfunction in peritoneal dialysis (PD) patients. It has been shown that normal and malignant mesothelial cells (MCs) express VEGF receptors (VEGFRs) and co-receptors and that VEGF is an autocrine growth factor for mesothelioma. Hence, we evaluated the expression patterns and the functional relevance of the VEGF/VEGFRs/co-receptors axis during the mesenchymal conversion of MCs induced by peritoneal dialysis. Omentum-derived MCs treated with TGF-β1 plus IL-1β (in vitro MMT) and PD effluent-derived MCs with non-epithelioid phenotype (ex vivo MMT) showed down-regulated expression of the two main receptors Flt-1/VEGFR-1 and KDR/VEGFR-2, whereas the co-receptor neuropilin-1 (Nrp-1) was up-regulated. The expression of the Nrp-1 ligand semaphorin-3A (Sema-3A), a functional VEGF competitor, was repressed throughout the MMT process. These expression pattern changes were accompanied by a reduction of the proliferation capacity and by a parallel induction of the invasive capacity of MCs that had undergone an in vitro or ex vivo MMT. Treatment with neutralizing anti-VEGF or anti-Nrp-1 antibodies showed that these molecules played a relevant role in cellular proliferation only in naïve omentum-derived MCs. Conversely, treatment with these blocking antibodies, as well as with recombinant Sema-3A, indicated that the switched VEGF/VEGFRs/co-receptors axis drove the enhanced invasion capacity of MCs undergoing MMT. In conclusion, the expression patterns of VEGFRs and co-receptors change in MCs during MMT, which in turn would determine their behaviour in terms of proliferation and invasion in response to VEGF.  相似文献

通过采用两处不同浓度富营养化水体对高羊茅进行灌溉,研究了高羊茅生理生态特征响应。结果表明:稀释后的富营养化水体能显著促进高羊茅根系生长,当稀释浓度大于50%时,不利于株高的增长。富营养化水体的各个浓度对高羊茅叶绿素各项指标无显著影响。各富营养化水体稀释后灌溉高羊茅,对其游离脯氨酸、可溶性糖、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性的影响与对照相比差异不显著,外环水体能显著促进高羊茅过氧化物酶(POD)活性增加,稀释浓度为75%能使高羊茅过氧化氢酶(CAT)活性显著增加。综上分析表明,富营养化水体稀释后灌溉高羊茅是可行的,且生活区污水更适合用于高羊茅的灌溉。  相似文献

Reed canary grass (Phalaris arundinacea, L.) invasion of wetlands is an ecological issue that has received attention, but its impact on soil microbial diversity is not well documented. The present study assessed the size (substrate-induced respiration), catabolic diversity (CLPP, community level physiological profiles) and composition (selective inhibition) of the soil microbial community in invaded (>95% P. arundinacea cover) and in non-invaded areas of a wetland occupied by native species grown either as a mixed assemblage (22 species) or as quasi-monotypic stands of Scirpus cyperinus (74% cover). The study also tested the hypothesis that decomposition of lignin- and phenolics-rich plant tissues would be fastest in soils exhibiting high catabolic diversity. Results showed that soil respiration, microbial biomass and diversity were significantly higher (P?<?0.03; 1.5 to 3 fold) in P. arundinacea-invaded soils than in soils supporting native plant species. Fungal to bacterial ratios were also higher in invaded (0.6) than in non-invaded (0.4) plots. Further, canonical discriminant analysis of CLPP data showed distinct communities of soil decomposers associated with each plant community. However, these differences in microbial attributes had no effect on decomposition of plant biomass which was primarily controlled by its chemical composition. While P. arundinacea invasion has substantially reduced plant diversity, this study found no parallel decline in the size and diversity of the soil microbial community in the invaded areas.  相似文献

SSR primers specific to Lolium perenne generated a total of 96 alleles and 124 genotypes within Festuca arundinacea and Lolium perenne accessions. Their highly transferability (100 %) across genera was evidenced. Six alleles specific to loci H01F02, H02C11 and K01A03 and only 5/96 common alleles between both species (60, 140, 144, 190 and 192) expressed the differentiation between species. Besides, based on the Wrights fixation indices, the genetic variation within each species was attributable to differences within populations with a significant deficiency of heterozygous. The unweighted pair group method with arithmetic averaging dendrogram based on the Nei’s distances and the principal coordinate analysis based on Jaccard coefficient similarity distinguished each genus independently of the geographical origin. However, typically continuous genetic diversity and a low level of gene flow (Nm: 0.29–2.47) expressed the relatively closely relationships of both genera and suggest a possible hybridization in nature.  相似文献

Shoot status, such as orientation and connection to the root system, and sediment burial depth after flooding disturbances have important ecological consequences on the post-flooding growth and vegetative reproduction of emergent macrophytes in wetlands. In the present study, we investigated the effect of shoot status (vertical, prostrate, or detached) and sediment burial depth (0.5 or 10 cm) on biomass accumulation and propagule production in Phalaris arundinacea (Poaceae) using an outdoor mesocosm system. In contrast to our prediction that shallow sediment burial would activate the axillary buds on prostrate shoots and regenerate more ramets, significantly fewer new ramets, rhizomes, buds, and biomass accumulation formed in P. arundinacea as the shoots changed from vertical to prostrate. Deeper sediment burial resulted in lower biomass and propagule production in plants with prostrate shoots, whereas vertical shoots increased the number of ramets. P arundinacea with detached shoots also produced a number of propagules after shallow or deep sediment burial, which might be important for the long-distance dispersal of P. arundinacea. These results suggest that P. arundinacea is a potentially invasive species in many lacustrine wetlands, particularly those with a high sedimentation rate, due to its high capacity for vegetative propagation.  相似文献

Clonality as a plant growth strategy has been a successful adaptation contributing to clonal plants being the dominant vegetation in many ecosystems and has been implicated as a significant factor contributing to invasiveness. The objective of this study was to determine if Phalaris arundinacea, an invasive wetland plant, modifies its clonal growth behavior when grown in high resource conditions. When grown in ideal conditions (high soil-N and moisture), we hypothesized that along with an increase in tiller production and robustness (biomass per tiller), P. arundinacea would increase the spatial spread of tiller placement (distance from parent and daughters). To test this we conducted a greenhouse study in which we grew P. arundinacea seedlings under two soil-N levels (no nitrate addition or 40 g N m?2 year?1) at two soil moisture levels (dry or saturated) for 10 weeks and recorded the placement (angle and distance from the parent plant) of each tiller produced. Total aboveground and belowground biomass, shoot/root ratio, and biomass per tiller were measured at the conclusion of the experiment. Plants grown in saturated conditions produced significantly more tillers that were more widely dispersed. Surprisingly, soil-N did not significantly affect most characteristics of spatial pattern, though soil-N did affect biomass production, shoot/root ratio, and biomass per tiller. These results indicate soil moisture and soil-N affect different aspects of the clonal growth behavior of P. arundinacea in the early stage of colonization. This new information provides a mechanism to explain how P. arundinacea aggressively competes for space in wetland habitats.  相似文献

N-Feruloylglycyl-l-phenylalanine was obtained from barley globulins by partial hydrolysis with 4 N HCl. It was isolated by means of preparative and ‘multiple elimination’ TLC (METC) and further identified by TLC comparison with a synthetic sample. Additional proof for its identity was obtained by UV, fluorescence and IR spectroscopy and by the action of carboxypeptidase A. C-Glycosylation of 5,7-dihydroxy-3′,4′,5′-trimethoxyflavone was carried out with acetobromo-α-d-glucose, -α-d-galactose, -α-d-xylose, -β-l-arabinose and -αt-L-rhamnose. The respective 6-C-glycosides and 6, 8-di-C-glycosides (excepted for galactose) were isolated and permethylated. 通过室内盆栽试验,在生活垃圾堆肥中按质量比添加1%氧化石墨烯(GO)和不同比例的纳米铁(nZVI,1%、3%、5%),研究了GO和nZVI对高羊茅生长和生理特性的影响。结果表明:单独添加GO及共同添加GO和nZVI促进了高羊茅种子萌发,发芽率和发芽指数均在GO+5%nZVI处理达到最大。除1%nZVI处理外,其他添加剂处理显著提高了高羊茅地上生物量和叶绿素含量,在GO+5%nZVI处理达到最大,较对照分别提高25%和31%。添加3%、5%nZVI显著提高了超氧化物歧化酶(SOD)和过氧化物酶(POD)活性,且随nZVI添加比例的增加而升高。与对照相比,单独添加nZVI以及联合GO处理过氧化氢酶(CAT)与丙二醛(MDA)含量显著降低,随nZVI添加比例的增加而减少,分别在GO+5%nZVI和GO+3%nZVI处理中达到最低,较对照分别降低52%和48%。高羊茅地上部重金属含量随nZVI添加比例的增加而降低,在共同添加GO和nZVI的处理中效果显著,均显著低于对照。可见,nZVI和GO两种材料对堆肥重金属具有固定作用,减少了植物对重金属的吸收,促进了草坪植物的生长。  相似文献

PsbO, the manganese-stabilizing protein, plays a crucial role in oxygen-evolving complex functioning and stabilization, by maintaining optimal manganese, calcium and chloride concentrations at the active state of PSII. In this paper we present a study focused on recognizing the relationship between psbO gene activity and acclimation of the photosynthetic apparatus under abiotic stresses in the grasses Festuca arundinacea and F. pratensis. PsbO expression was compared between two distinct genotypes within each species which differed in their levels of stress tolerance (drought and frost, respectively) during drought treatment (F. arundinacea) and cold acclimation (F. pratensis). The research involved: (1) the analyses of psbO gene expression profiles using real-time PCR, and (2) the analyses of PsbO protein accumulation profiles using protein gel blot hybridization. The results indicate that PsbO plays a protective function with respect to the photosynthetic apparatus during abiotic stresses. In cold-treated F. pratensis plants the accumulation of PsbO seems to be responsible for differences in the PSII photochemical efficiency. Higher stability of PSII during drought, observed in the high-drought tolerant F. arundinacea genotype, is not associated with PsbO accumulation, although the degradation of this protein affects destabilization of the oxygen-evolving complex in drought.  相似文献

A facile two steps extrusion processing conditions are used to prepare thermoplastic starch (TPS)/glycerol modified-montmorillonite (GMMT) nanocomposites. X-ray diffraction (XRD) and transmission electron microscopy (TEM) demonstrate glycerol can enlarge the d-spacing and destruct the multilayer structure of montmorillonite (MMT) effectively using high speed emulsifying machine (HSEM) in the first modification step. So the enlarged d-spacing and destructed platelets of MMT are favorable to form intercalated or exfoliated TPS/GMMT nanocomposites in the second melt extrusion processing. However, scanning electron microscopy (SEM) and XRD show the possible competition between TPS matrix and plasticizer for the intercalation between MMT layers can deteriorate the plasticization of TPS. In addition, citric acid (CA) can increase the plasticization of TPS and dispersion of MMT in nanocomposites effectively detected by fourier transform infrared (FT-IR) spectroscopy and SEM. At the same time, this facile processing conditions and CA can improve the mechanical properties and water vapor permeability (WVP) of TPS/GMMT nanocomposites obviously.  相似文献
