从树Qu肾细胞培养物中分离出3株病毒,嗜在对数分裂期的TL细胞上复制,产生细胞病变和血凝素抗原,能凝集豚鼠和小鼠红细胞。交互血凝抑制实验表明3株病毒同属一个血清型。免疫酶染色显示抗原首先出现于细胞株,电镜观察负染标本病毒形态近似圆形和六角形,无胞膜,直径约30nm。血清学检查大多数树Qu血清中含有该病毒抗体,证明是树Qu的一种潜在病毒。初步鉴定为类-细小病毒。  相似文献

建立共表达猪瘟病毒(Classic swine fever virus,CSFV)E2蛋白主要中和性抗原表位基因的猪繁殖与呼吸综合征病毒(PRRSV)反向遗传操作平台,为进一步研究PRRSV作为病毒载体提供实验数据。本研究在PRRSVHuN4-F112疫苗株感染性分子克隆的基础上,采用突变PCR技术将CSFV E2蛋白的主要中和性抗原表位基因插入HuN4-F112弱毒株PRRSV已鉴定出的两个NSP2蛋白的复制非必需区,经基因片段的克隆、拼接,构建了两株含有CSFV E2蛋白的主要中和性抗原表位基因的感染性PRRSV cDNA克隆psk-HuN4-F112-Δ508-532+E2和psk-HuN4-F112-Δ480-532+E2。用限制性内切酶SwaΙ分别将两株共表达的感染性克隆线性化,之后通过细胞外转录获得病毒RNA,用脂质体法分别将病毒RNA转染BHK-21细胞包装出病毒粒子,再分别将其转接到MARC-145细胞传代拯救出两株病毒。分别对两株拯救病毒进行RT-PCR扩增、酶切和序列分析鉴定。结果表明,两株拯救病毒含有不同于亲本病毒的分子标记(拯救病毒基因组的14 667位因同义突变产生的MluⅠ酶切位点)和插入基因序列。间接免疫荧光试验表明,CSFV E2蛋白主要中和性抗原表位基因均能在两株拯救的PRRSV中表达,且能够在其传代过程中稳定遗传。病毒生长特性结果比较显示两株拯救病毒与亲本病毒在MARC-145细胞上具有相似的增殖特性。本研究构建了两株共表达CSFV E2蛋白主要中和性抗原表位基因的PRRSV感染性克隆并获得了拯救病毒,且外源基因能在拯救病毒中稳定表达,为进一步开发新型PRRSV二联疫苗提供了有效的反向遗传操作平台。  相似文献

本文应用15株分别抗肾综合征出血热(HFRS)病毒糖蛋白(Glycoprotein Ⅱ G2),核蛋白(Nuclcocapsid,NP)及血凝素(Hemagglutinin,HA)抗原的单克隆抗体免疫细胞化学方法对19例HFRS尸检病例的16种组织中的病毒抗原进行了定位和分布的研究及抗原分析。结果表明,死于早期HFRS人体组织内病毒抗原量大,分布广泛,主要以可溶性和颗粒性两种形式存在,前者存在于细胞内和细胞外,呈G2,NP及HA抗原阳性,是参与形成免疫复合物的主要抗原形式;后者是以单纯病毒NP或HA抗原阳性的病毒包涵体(IB)形式存在于细胞内,是病毒直接作用所致细胞病变的标志,IB的广泛分布但只有极个别阳性细胞发生坏死,说明该病毒具有泛嗜性感染和弱致细胞病变能力的特性。抗原分析结果显示,组织细胞中病毒抗原的表达及其抗原量受宿主细胞的种属,组织结构特点及病期的影响,也存在个体差异以及因感染病毒株或血清型不同产生差异的可能性。病毒抗原染色形态学证实,NP上具有HA抗原位点,其抗原决定簇有三类,其中的某些抗原决定簇可因病毒宿主动物或细胞的种属不同,表达也不同。HA抗原在人体组织细胞中的高表达和广泛分布也说明HFRSV对人体有很强的侵袭力。  相似文献

三个狂犬病毒株,分别经地鼠肾细胞培养,将其抗原含量,病毒滴度和免疫原性进行比较。结果显示,三个病毒株的抗原含量有差异。但差异显著性,CTN-V10M3的毒力最高,CTN-BHK3的毒力最低,aG株的毒力虽然居中,但免疫原性最好,它仍是最适于地鼠肾细胞上培养的毒株。  相似文献

分析衣原体微病毒Vp3蛋白在分子重组、分子进化及病毒野生株筛查的作用并挖掘生物信息,揭示其临床研究价值。以ProteinBlast的Multriple alignment程序比对各株衣原体微病毒衣壳蛋白Vp3序列;以Distance tree(ProteinBlast)程序完成种系发生树。以比对获得的高保守区的氨基酸序列为目标,应用Karplus-schulz法进行柔性区域分析;通过Gamier-robson法和Chou-fasman法分析该序列的二级结构;使用Emini法完成表位的表面可及性分析,并用Kyte-Doolittle法和Hopp-woods法完成蛋白亲水性研究;应用Jameson-Wolf法完成表位的抗原指数分析。6株衣原体微病毒Vp3蛋白序列高度保守,差异主要在Chp1与其他5株微病毒的Vp3蛋白之间。各株微病毒的Vp3蛋白均有以α螺旋为主的结构,蛋白序列高保守区存在多个细胞表位。Vp3蛋白结构性质保守,也是衣原体噬菌体衣壳的重要组分。其蛋白分子结构复杂,高保守区有较强的免疫原性,在分子重组、沙眼衣原体微病毒野生株筛查研究中有实际研究价值。  相似文献

BTV-HbC株和蓝舌病毒标准株BTV-10分别接种在不同种系细胞如猴肾传代细胞(Vero)、人宫颈癌细胞(Hela)和小鼠神经胶质瘤细胞(C6)等细胞株上,比较研究了BTV-HbC在不同种系细胞上的增殖特征,BTV-HbC与BTV-10在相同细胞上的复制增殖特征,病毒与细胞相互作用的显微和超微结构特征.用免疫交叉反应研究了BTV-HbC株与BTV-10型标准株之间的血清学关系.本研究结合本室对BTV-HbC株基因组图谱分析和蓝舌病毒群特异性抗原编码基因S7的RT-PCR分析,进一步证实了BTV-HbC株可能是一个新的血清型蓝舌病毒.  相似文献

建立EB病毒(EBV)转化的B淋巴母细胞系(LCL),应用此细胞系作为抗原递呈细胞筛选抗原特异性的T细胞克隆。收集人乳头瘤病毒(HPV)感染者外周血20份,分离外周血单个核细胞(PBMC),利用Pan T细胞试剂盒去除PBMC中的T细胞,获得non-T淋巴细胞,EBV病毒转化non-T淋巴细胞。应用转化的LCL细胞作为抗原递呈细胞,ELISPOT筛选可能的HPV抗原特异性的T细胞克隆。成功建立18个LCLs,建株成功率为90%。转化成功的LCL细胞体积增大,聚集成团状或簇状。建株失败的两份标本,用于转化的non-T淋巴细胞数少于2×106。应用LCL细胞作为抗原递呈细胞筛选出64个HPV抗原特异性的T细胞克隆。成功建立LCLs,此细胞系细胞可作为体外研究HPV特异性免疫反应的刺激细胞。  相似文献

1990-1994年,从新疆地区的蚊、蜱和病人血清分离了多株病毒,为了明确这些病毒的分类地位,对其中的20株病毒进行了组织培养细胞感染实验和血清学检验,对部分毒株做了动物接种实验和理化性质鉴定。结果显示:20株病毒均可使BHK-21细胞病变(1-3天),主要表现为细胞圆宿、聚集,融合,破碎,脱落等;致Vero细胞病变为2-4天;15株病毒致C6/36细胞病变(2-4天),5株病毒对C6/36细胞连续观察7天未见细胞病变。11株病毒对乳鼠2-4天致死,对成年鼠2-5天致死。选取6株病毒进行理化性质鉴定,4株病毒(90260、91002、91004和91028)对5-氟脱氧尿苷耐受,对乙醚和酸敏感,提示为有膜RNA病毒;一株病毒(90265)对5-氟脱氧尿苷、乙醚和酸均敏感,提示为有膜DNA病毒;另一株病毒(9059)对5-氟脱氧尿苷耐受,对乙醚和酸也耐受,提示可能为无膜RNA肠道病毒。20株病毒中,17株病毒与甲病毒、乙型脑炎病毒和布尼亚病毒的特异性免疫腹水不反应,提示这些病毒中可能不存在甲病毒、黄病毒和布尼亚病毒;3株病毒(90260、91002和91004)只与甲病毒的特异性免疫腹水反应,与乙型脑炎病毒和布尼亚病毒的不反应,提示这三株病毒为甲病毒。  相似文献

选取不同代次的LR1毒株在Vero细胞上传代适应,不同天数连续动态观察细胞病变情况,同时用免疫荧光法和ELISA进行抗原检测,收毒前细胞反复冻融及超声波破碎,细胞破碎前后用ELISA检测抗原含量,半微量空斑法检测病毒滴度。结果证明:LR1株病毒能在Vero细胞上产生细胞病变,病变程度与其毒力明显相关;LR1株病毒在Vero细胞上繁殖的最佳收毒时间为11天左右;病毒释放性较差,冻融和超声波破碎细胞能明显提高其抗原含量和病毒滴度  相似文献

从我国分离到的一株单纯疱疹病毒Ⅰ型(HSV-1-168株)病毒基因组中,分离出含有糖蛋白D(gD)基因的1.2kb片段,插入带有痘苗病毒天坛株TK区的质粒pJSB1175P7.5k启动子下游,转染无白血病鸡胚原代细胞,获得带有HSV-1-168gD基因的重组痘苗病毒。此株重组病毒在感染细胞膜上表达HSV-1-168gD糖蛋白抗原,能与特异性单克隆抗体反应。在感染细胞中表达的膜抗原经SDS-PAGE分析,表达分子量为54kD糖蛋白。用Southern杂交分析了重组病毒DNA中特异的gD基因,对作为活疫苗的重组痘苗病毒株进行了一些微生物学活性、免疫原性和毒力等方面的研究。  相似文献

病毒学检测和监测是树鼩实验动物化和质量控制的重要标准和依据,而野生中缅树鼩是否携带人兽共患病毒鲜见报道。本研究采用酶联免疫吸附(ELISA)方法,对来源于云南昆明市城郊青龙峡地区的野生树鼩是否携带单纯疱疹病毒、轮状病毒、流感病毒、柯萨奇病毒、甲肝病毒、乙肝病毒、丙肝病毒、丁肝病毒、登革热病毒、出血热病毒和麻疹病毒等11种常见病毒进行筛查。结果表明,在已筛查的60只野生中缅树鼩中,可检测到单纯疱疹病毒和柯萨奇病毒,其血清抗体阳性比例分别为36.7%(22/60)和1.67%(1/60),而在粪便中仅检测到轮状病毒,其抗原阳性为6.7%(4/60),未检测到其他病毒,初步显示了野生树鼩自然状态下携带病毒的状况。为此,建议将单纯疱疹病毒、柯萨奇病毒和轮状病毒列为普通级树鼩病毒质量控制的首检项目,进一步大样本筛查将显示是否将其他病毒列为必检项目。  相似文献

肝炎的的大范围流行已成为国内外关注的重要公共卫生问题之一。甲肝、乙肝虽已有疫苗,但不能忽视未来由于病毒变异所带来的威胁。丙肝目前尚未发现非常有效的疫苗,且丙肝的病理学机制也尚未完全清晰,主要原因是缺乏理想的动物模型。树胸属于低等灵长类动物。研究发现,他对很多人类疾病易感,所以建立肝炎树胸动物模型成为现在肝炎研究的一个热点。本文介绍了各型肝炎主要是甲、乙、丙型肝炎研究中建立树胸动物模型的自内外进展情况及存在的问题。  相似文献

选用3株基孔肯雅病毒人工感染成年树鼩,进行了病毒血症、抗体动态变化、内脏组织病理改变和病毒在宿主体内定位的研究。结果表明,感染树鼩能产生2~6天的病毒血症。血凝抑制(Hi)抗体第6天产生,第30~50天达高峰:中和(NT)抗体在第10天产生,第30~40天达高峰,二者相关性非常显著(P<0.01)。补体结合(CF)抗体第14天产生,第40~50天为高峰,以后逐渐下降。第8~12天能在其脑、肺、肝、脾和肾等组织查到病毒,经病理检查这些内脏组织呈炎性改变和出血倾向,表明该病毒能侵袭树鼩各主要脏器。试验认为树鼩对基孔肯雅病毒敏感。  相似文献

Nine adult tree shrews, Tupaia glis, recently imported from West Malaysia were visually examined for ectoparasites while under general anesthesia. Three shrews were infested by the sucking louse, Sathrax durus , and six shrews had louse ova belonging to this species; two shrews had neither lice nor ova. A total of 20 adult female, 10 adult male, and three third instar nymphal lice was collected. Lice were located on the head, flanks, and dorsal body of shrews while ova were recorded mainly from the anterior flanks but also from some adjacent host sites. The tree shrews appeared to tolerate the lice well although louse vector capacity was not assessed. The last date that lice were recorded from shrews was 22 days after colony set-up, and the last date on which seemingly viable ova were recorded was 64 days after set-up showing that the infestations were ultimately lost.  相似文献

Diverse species of rodents and shrews, which are abundant worldwide, harbor a variety of viruses;some of these are closely related to human viruses and possess zoonotic potential. Previously studies have demonstrated that the mammarenavirus and hantavirus carried by rodents or shrews could cause diseases in human population. To determine the distribution of zoonotic viruses in Shenzhen city, the major city in southern China with a high population density, we analyzed 225 rodents(Rattus norvegicus and Rattus flavipectus) and 196 shrews(Suncus murinus) from urban and rural districts for the presence of mammarenavirus, hantavirus, and hepatitis E virus(HEV) by RT-PCR targeting the conserved regions. The infection rates for mammarenavirus, hantaviruses,and HEV in rodents and shrews were 3.56%, 6.89%, and 1.66%, respectively. Partial genome fragment analysis indicated that mammarenavirus and hantavirus strains had more than 90% and 99% nucleic acid identity with Cardamones virus and Seoul virus, respectively, which cause diseases in humans. Although the present HEV strains identified are typically found worldwide,phylogenetic analysis demonstrated a divergence of 16%. To our knowledge, the present work is the first report of the prevalence of mammarenavirus, hantaviruses, and rat HEV strains in rodents and shrews from Shenzhen city, China. Our findings highlight the zoonotic potential of rodent-and shrew-borne mammarenavirus and hantavirus, and the biodiversity of rat HEV isolates in Shenzhen city. The present work suggests that utilization of good hygiene habits is important to minimize the risk of zoonosis.  相似文献

The tree shrews are non-rodent, primate-like, small animals. There is increasing interest in using them to establish animal models for medical and biological research. This review focuses on the use of the tree shrews in in vivo studies on viral hepatitis, hepatocellular carcinoma (HCC), myopia, and psychosocial stress. Because of the susceptibility of the tree shrews (Tupaia belangeri) and their hepatocytes to infection with human hepatitis B virus (HBV) in vivo and in vitro, these animals have been used to establish human hepatitis virus-induced hepatitis and human HBV- and aflatoxin B1-associated HCC models. As these animals are phylogenetically close to primates in evolution and have a well-developed visual system and color vision in some species, they have been utilized to establish myopia models. Because dramatic behavioral, physiological, and neuroendocrine changes in subordinate male tree shrews are similar to those observed in depressed human patients, the tree shrews have been successfully employed to experimentally study psychosocial stress. However, the tree shrews holds significant promise as research models and great use could be made of these animals in biomedical research.  相似文献

ABSTRACT: BACKGROUND: Hepatitis B virus (HBV) infection continues to be an escalating global health problem. Feasible and effective animal models for HBV infection are the prerequisite for developing novel therapies for this disease. The tree shrew (Tupaia) is a small animal species evolutionary closely related to humans, and thus is permissive to certain human viral pathogens. Whether tree shrews could be chronically infected with HBV in vivo has been controversial for decades. Most published research has been reported on adult tree shrews, and only small numbers of HBV infected newborn tree shrews had been observed over short time periods. We investigated susceptibility of newborn tree shrews to experimental HBV infection as well as viral clearance over a protracted time period. RESULTS: Forty-six newborn tree shrews were inoculated with the sera from HBV-infected patients or tree shrews. Serum and liver samples of the inoculated animals were periodically collected and analyzed using fluorescence quantitative polymerase chain reaction, enzyme-linked immunosorbent assay, Southern blot, and immunohistochemistry. Six tree shrews were confirmed and four were suspected as chronically HBV-infected for more than 48 (up to 228) weeks after inoculation, including three that had been inoculated with serum from a confirmed HBV-infected tree shrew. CONCLUSIONS: Outbred neonatal tree shrews can be long-term chronically infected with HBV at a frequency comparable to humans. The model resembles human disease where also a smaller proportion of infected individuals develop chronic HBV related disease. This model might enable genetic and immunologic investigations which would allow determination of underlying molecular causes favoring susceptibility for chronic HBV infection and disease establishment vs. viral clearance.  相似文献

目的对树[鼠句]抓取和保定、采血、灌胃基本实验技术方法进行探讨,逐步规范树[鼠句]实验技术。方法选用成年树[鼠句]进行抓取和保定,分徒于法和器具法(自制捕捉保定袋),对130只树[鼠句]采用尾静脉、股动(静)脉两种采血方法;采取人用8号胃管可对树[鼠句]绎口灌胃给药。结果所采用徒手、器具的方法抓取和保定树[鼠句]均能有效地控制动物,不会发生动物死亡或很少逃逸;两种方法都顺利采集到所需血量,股动(静)脉单次最大采血量可达2mL而不损伤动物;12只树[鼠句]连续灌胃10d,成功率100%。结论自制的捕捉保定袋经济实用,摸索的几种树[鼠句]实验技术方法具有操作简便、安全、快捷等优点。  相似文献

目的调查滇西亚种树鼩体外寄生虫的自然感染状况,为建立树鼩质量控制标准提供依据。方法对野生的滇西亚种树鼩和自繁F1代树鼩各60只分别用尸检法和活体法检测,随即采用拔毛或用透明胶带粘取腋下、耳根、腹股沟、肛门附近等处的毛发制片,置于体视镜和显微镜下观察虫体和虫卵。结果野生滇西亚种树鼩和自繁F1代树鼩体外寄生虫自然感染率分别为:虱子25%/6%、蚤5%/0、蜱1.6%/0、水蛭1.6%/0、虱子和蚤混合感染1.6%/0。结论野生树鼩体外寄生虫的自然感染多为虱子和蚤,其次还有蝉、水蛭,总感染率明显高于自繁F1代。使用灭虱灵对于驱除树鼩体外寄生虫具有较好的疗效。  相似文献

Tupaias, or tree shrews, are small mammals that are similar in appearance to squirrels. The morphological and behavioral characteristics of the group have been extensively characterized, and despite previously being classified as primates, recent studies have placed the group in its own family, the Tupaiidae. Genomic analysis has revealed that the genus Tupaia is closer to humans than it is to rodents. In addition, tupaias are susceptible to hepatitis B virus and hepatitis C virus. The only other experimental animal that has been demonstrated to be sensitive to both of these viruses is the chimpanzee, but restrictions on animal testing have meant that experiments using chimpanzees have become almost impossible. Consequently, the development of the tupaia for use as an animal infection model could become a powerful tool for hepatitis virus research and in preclinical studies on drug development.  相似文献
