In the present study, the role of ethylene in nitric oxide (NO)-mediated protection by modulating ion homeostasis in Arabidopsis callus under salt stress was investigated. Results showed that the ethylene-insensitive mutant etr1-3 was more sensitive to salt stress than the wild type (WT). Under 100 mM NaCl, etr1-3 callus displayed a greater electrolyte leakage and Na+/K+ ratio but a lower plasma membrane (PM) H+-ATPase activity compared to WT callus. Application of exogenous 1-aminocyclopropane-1-carboxylic acid (ACC, an ethylene precursor) or sodium nitroprusside (SNP, a NO donor) alleviated NaCl-induced injury by maintaining a lower Na+/K+ ratio and an increased PM H+-ATPase activity in WT callus but not in etr1-3 callus. The SNP actions in NaCl stress were attenuated by a specific NO scavenger or an ethylene biosynthesis inhibitor in WT callus. Under 100 mM NaCl, the NO accumulation and ethylene emission appeared at early time, and NO production greatly stimulated ethylene emission in WT callus. In addition, ethylene induced the expression of PM H+-ATPase genes under salt stress. The recovery experiment showed that NaCl-induced injury was reversible, as signaled by the similar recovery of Na+/K+ ratio and PM H+-ATPase activity in WT callus. Taken together, the results indicate that ethylene and NO cooperate in stimulating PM H+-ATPase activity to modulate ion homeostasis for salt tolerance, and ethylene may be a part of the downstream signal molecular in NO action.  相似文献

The plasma membrane (PM) vesicles from Populus euphratica (P. euphratica) callus were isolated to investigate the properties of the PM H+-ATPase. An enrichment of sealed and oriented right-side-out PM vesicles was demonstrated by measurement of the purity and orientation of membrane vesicles in the upper phase fraction. Analysis of pH optimum, temperature effects and kinetic properties showed that the properties of the PM H+-ATPase from woody plant P. euphratica callus were consistent with those from herbaceous species. Application of various thiol reagents to the reaction revealed that reduced thiol groups were essential to maintain the PM H+-ATPase activity. In addition, there was increased H+-ATPase activity in the PM vesicles when callus was exposed to NaCl. Western blotting analysis demonstrated an enhancement of H+-ATPase content in NaCl-treated P. euphratica callus compared with the control.  相似文献

The influence of freezing treatment on plasma membrane (PM) H+-ATPase was investigated using plasma membrane vesicles isolated from calluses from Chorispora bungeana Fisch. & C.A. Mey. by the discontinuous sucrose gradient centrifugation. Freezing treatment (−4 °C) for 5 d resulted in significant increases in the ATPase activity and the activity of p-nitrophenyl phosphate (PNPP) hydrolysis, decreases in the Km for ATP hydrolysis and PNPP hydrolysis, and the shift of optimal pH from 6.5 to 7.0. Also, the activity PNPP hydrolysis was less sensitive to vanadate after freezing treatment compared to control, while the inhibition of ATP hydrolysis by hydroxylamine was more sensitive. In addition, freezing treatment also decreased the activation effects of trypsin on PNPP hydrolysis, but increased the activation effects of lysophosphatidylcholine on ATP hydrolysis. Taken together, these results suggested that PM H+-ATPase might play an important role during adaptation to freezing and enhancing the frost hardness in C. bungeana.  相似文献

Spartina patens, an intertidal C4 grass, grows in the upper salt marsh and tolerates coastal seawater salinity. The regulation of ion movement across the plasma membrane (PM) for plant salt tolerance is thought to be achieved by an electrochemical gradient generated by plasma membrane H+-ATPase. In this study, the change of PM H+-ATPase in response to NaCl was characterized for S. patens callus. Callus was cultured for 10 weeks under salinity levels of 0 mM, 170 mM, 340 mM, and 510 mM NaCl. Plasma membrane was isolated from a Dextran/PEG aqueous polymer two-phase system and the purity was demonstrated with membrane enzyme markers. There was a significant increase (up to 2-3 fold) of PM H+-ATPase activity when callus was grown on media containing NaCl. The incremental activation of PM H+-ATPase activity would enable the cell to tolerate higher cytoplasmic NaCl concentrations. PM H+-ATPase appeared to have a higher Vmax and a lower substrate concentration (Km to reach Vmax. When growth medium salinity increased from 0 mM to 170 and 340 mM, the Vmax of H+-ATPase increased from 0.64 to 1.00 and 1.73, respectively, while the Km decreased from 3.58 to 2.07 and 2.44 mM, respectively. In vitro NaCl inhibition kinetic data revealed a pattern of non-competitive inhibition by NaCl on PM H+-ATPase. The response of PM H+-ATPase in S. patens callus suggests that this species has evolved mechanisms that can regulate this important enzyme when cells are exposed to NaCl.  相似文献

Inhibition of glutathione synthesis reduces chilling tolerance in maize   总被引:8,自引:0,他引:8  
 The role of glutathione (GSH) in protecting plants from chilling injury was analyzed in seedlings of a chilling-tolerant maize (Zea mays L.) genotype using buthionine sulfoximine (BSO), a specific inhibitor of γ-glutamylcysteine (γEC) synthetase, the first enzyme of GSH synthesis. At 25 °C, 1 mM BSO significantly increased cysteine and reduced GSH content and GSH reductase (GR: EC 1.6.4.2) activity, but interestingly affected neither fresh weight nor dry weight nor relative injury. Application of BSO up to 1 mM during chilling at 5 °C reduced the fresh and dry weights of shoots and roots and increased relative injury from 10 to almost 40%. Buthionine sulfoximine also induced a decrease in GR activity of 90 and 40% in roots and shoots, respectively. Addition of GSH or γEC together with BSO to the nutrient solution protected the seedlings from the BSO effect by increasing the levels of GSH and GR activity in roots and shoots. During chilling, the level of abscisic acid increased both in controls and BSO-treated seedlings and decreased after chilling in roots and shoots of the controls and in the roots of BSO-treated seedlings, but increased in their shoots. Taken together, our results show that BSO did not reduce chilling tolerance of the maize genotype analyzed by inhibiting abscisic acid accumulation but by establishing a low level of GSH, which also induced a decrease in GR activity. Brassinosteroids (BRs), a class of plant steroid hormones, play a significant role in the amelioration of various biotic and abiotic stresses. In order to further explore and elaborate their roles in plants subjected to chilling stress, suspension cultured cells of Chorispora bungeana with or without 24-epibrassinolide (EBR) application were exposed to 4 and 0°C for 5 days. The EBR treated cells exhibited higher viability after exposure to low temperatures compared with the control. Under chilling stress, reactive oxygen species (ROS) levels and lipid peroxidation were increased in the cultured cells, which were significantly inhibited by EBR application. The activities of antioxidative enzymes such as ascorbate peroxidase (APX), catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD) were increased during chilling treatments, and these increases were more significant in the EBR applied suspension cells. The EBR treatment also greatly enhanced contents of ascorbic acid (AsA) and reduced glutathione (GSH) under chilling stress. From these results, it can be concluded that EBR could play the positive roles in the alleviation of oxidative damage caused by ROS overproduction through enhancing antioxidant defense system, resulting in improving the tolerance of C. bungeana suspension cultures to chilling stress.  相似文献

The objective of this study was to investigate whether abscisic acid (ABA), a second messenger in chilling stress responses, is involved in brassinosteroids (BRs)-induced chilling tolerance in suspension cultured cells from Chorispora bungeana. The suspension cells were treated with 24-epibrassinolide (EBR), ABA, ABA biosynthesis inhibitor fluridone (Flu) and EBR in combination with Flu. Their effects on chilling tolerance, reactive oxygen species (ROS) levels and antioxidant defense system were analyzed. The results showed that EBR treatment markedly alleviated the decrease of cell viability and the increases of ion leakage and lipid peroxidation induced by chilling stress, suggesting that application of EBR could improve the chilling tolerance of C. bungeana suspension cultures. In addition, similar results were observed when exogenous ABA was applied. Treatment with Flu alone and in combination with EBR significantly suppressed cell viability and increased ion leakage and lipid peroxidation under low temperature conditions, indicating that the inhibition of ABA biosynthesis could decrease the chilling tolerance of C. bungeana suspension cultures and the EBR-enhanced chilling tolerance. Further analyses showed that EBR and ABA enhanced antioxidant defense and slowed down the accumulation of ROS caused by chilling. However, Flu application differentially blocked these protective effects of EBR. Moreover, EBR was able to mimic the effect of ABA by markedly increasing ABA content in the suspension cells under chilling conditions, whereas the EBR-induced ABA accumulation was inhibited by the addition of Flu. Taken together, these results demonstrate that EBR may confer chilling tolerance to C. bungeana suspension cultured cells by enhancing the antioxidant defense system, which is partially mediated by ABA, resulting in preventing the overproduction of ROS to alleviate oxidative injury induced by chilling.  相似文献

Suaeda salsa calli treated with different concentrations of NaCl were used to examine the response of the plasma membrane (PM) H+-ATPase to NaCl and its role in salt tolerance. The optimum concentration of NaCl for growth of the calli was 50 mM, while growth was significantly inhibited at 250 mM NaCl. The ion and organic solute contents of calli increased with increasing NaCl. Activity of the PM H+-ATPase increased when the calli were treated with NaCl over a certain concentration range (0–150 mM NaCl). However, the activity reached its maximum with 150 mM NaCl. Immunoblotting analysis of the PM H+-ATPase protein from calli cultures with anti-Zea mays H+-ATPase serum (monoclonal 46E5B11D5) identified a single polypeptide of ~90 kDa. The peptide levels increased in the calli treated with NaCl at 150 mM NaCl compared to control, but the increase at 50 mM NaCl was less pronounced. Northern blot analysis showed that the expression of the PM H+-ATPase also increased after the calli were treated with NaCl. These results suggest that the increase in PM H+-ATPase activity is due to both an increase in the amount of PM H+-ATPase protein and an up-regulation of the PM H+-ATPase gene, which is involved in the salt tolerance of S. salsa calli.  相似文献

We investigated the effects of silicon (Si) on time-dependent changes in root tonoplast H+-ATPase and H+-PPase activities, membrane fatty acid compositions and tonoplast fluidity in two barley (Hordeum vulgare L.) cultivars differing in salt tolerance. Plants were grown in NaCl-free (control) and NaCl-supplied (60 and 120 mM, respectively) nutrient solutions with or without 1.0 mM Si. Plant roots were harvested to isolate tonoplast vesicles for assay of H+-ATPase and H+-PPase activities at days 2, 4, and 6 after treatment in the first experiment and for analysis of membrane fatty acid composition and fluidity at day 4 after treatment in the second experiment. The results showed that tonoplast H+-ATPase and H+-PPase activities in roots of salt-treated plants increased at day 2, which was more obvious at 60 mM NaCl in the salt-tolerant cultivar than in the salt-sensitive cultivar, and then decreased at day 4 and onward. These enzyme activities decreased consistently from days 2 to 6 for treatment with 120 mM NaCl. However, inclusion of 1.0 mM Si significantly enhanced both H+-ATPase and H+-PPase activities in roots of salt stressed barley, which was irrespective of NaCl level or cultivar used. The ratio of unsaturated to saturated fatty acids (U/S) increased under salt stress for both cultivars. Addition of Si to salt treatment increased the ratio of U/S in salt-tolerant cultivar but it did not in salt-sensitive cultivar compared to non-Si-amended salt treatment. Salt treatment decreased tonoplast fluidity of roots of barley significantly compared with control treatment. However, root tonoplast fluidity was significantly lower in the Si-amended salt treatment than in the non-Si-amended salt treatment. These results were in line with the previous findings that Si could help increase antioxidative defense and reduce membrane lipid oxidative damage in barley under salt stress. The possible mechanisms involved in Si-enhanced salt tolerance were discussed with respect to cell membrane integrity, stability and function in barley.  相似文献

Summary. As the outermost boundary of the cell, the plasma membrane plays an important role in determining the stress resistance of organisms. To test this concept in a cryophyte, we analyzed alterations of several components in plasma membranes isolated from suspension-cultured cells of Chorispora bungeana Fisch. & C.A. Mey in response to treatment at 0 and −4 °C for 192 h. When compared with the controls growing at 25 °C, both the membrane permeability and fluidity showed recovery after the initial impairment. Linolenic acid and membrane lipid unsaturation increased by about 0.8-fold following cold treatments, although the kinetics of the increase varied with the temperatures examined. During the treatments, the plasma membrane H+-ATPase (EC 3.6.1.3) activity increased by 78.06% at 0 °C and 100.47% at −4 °C. However, the plasma membrane NADH oxidase (EC 1.6.99.3) activity only decreased when exposed to a lower temperature (−4 °C), and remained at 63.93% after being treated for 192 h. After the treatments, the physical properties of the plasma membranes of suspension-cultured cells, especially the −4 °C treated cells, were similar to those in the wild plants. These findings indicate that the specific mechanism of cold resistance of C. bungeana is tightly linked with the rapid and flexible regulation of membrane lipids and membrane-associated enzymes, which ensure the structural and functional integrity of the plasma membrane that is essential for withstanding low temperature. Summary The effects of temperature and pressure on Na+/K+-adenosine triphosphatases (Na+/K+-ATPases) from gills of marine teleost fishes were examined over a range of temperatures (10–25°C) and pressures (1–680 atm). The relationship between gill membrane fluidity and Na+/K+-ATPase activity was studied using the fluorescent probe 1,6-diphenyl-1,3,5-hexatriene (DPH). The increase in temperature required to offset the membrane ordering effects of high pressure was 0.015–0.025°C·atm-1, the same coefficient that applied to Na+/K+-ATPase activities. Thus, temperature-pressure combinations yielding the same Na+/K+-ATPase activity also gave similar estimates of membrane fluidity. Substituion of endogenous lipids with lipids of different composition altered the pressure responses of Na+/K+-ATPase. Na+/K+-adenosine triphosphatase became more sensitive to pressure in the presence of chicken egg phosphatidylcholine, but phospholipids isolated from fish gills reduced the inhibition by pressure of Na+/K+-ATPase. Cholesterol increased enzyme pressure sensitivity. Membrane fluidity and pressure sensitivity of Na+/K+-ATPase were correlated, but the effects of pressure also dependent on the source of the enzyme. The role of the plasma membrane (PM) H+-ATPase (E.C. 3.6.1.3) in the plants response to salt stress was studied in the perennial leguminosae forage Medicago arborea L. and its close relative Medicago citrina (Font-Quer) Greuter, a species exposed to saline conditions in its original habitat. Plants were solution cultured for 8 days in 1 or 100 mM NaCl. Leaf growth and CO2 assimilation were more inhibited by salt in M. arborea than in M. citrina. Both species were able to osmoregulate, and salt-treated plants maintained turgor potentials, with no differences between species. Contrasting ion distribution patterns showed that M. citrina was able to exclude Na+ from the leaves more selectively, while M. arborea had a greater buildup of leaf blade Na+. Isolation of purified PM and quantification of H+-ATPase protein by Western blot analysis against the 46E5B11D5 or AHA3 antibodies showed an increase in response to salt stress in the expanding (92%) and expanded leaves (87%) of M. citrina, while no differences were found in the corresponding leaves of M. arborea. The assay of H+-ATPase specific activity of the two leaf types in salinized M. citrina confirmed this increase, as activities increased with 55% and 104% for the expanded and expanding leaves, respectively, while no significant differences were found for either leaf type of salinized M. arborea. A possible role of the increased expression of the PM H+-ATPase for leaf expansion and ion exclusion in salt-stressed plants is discussed.  相似文献

The aim of our work is to show the importance of the role of hydrophobic bonds in maintaining Mg2+-ATPase or sucrase activity and Na+-coupled d-glucose uptake normal for the brush border of rat enterocytes. The activity of the two enzymes and the d-glucose uptake were therefore measured under the action of n-aliphatic alcohols and related to the fluidity determined by ESR. Three concentrations were used for the first eight alcohols, those of octanol being about 1500-times lower than those of methanol. For each alcohol the d-glucose uptake and the fluidity were linear functions of the logarithm of the concentration, the linear regressions being practically parallel and equidistant. The concentrations (C) of the eight alcohols inhibiting the d-glucose uptake by 80% were similar to those increasing the membrane fluidity by 3%. The linear relationship which existed in both cases between log 1 / C and log P, P being octanol / water partition coefficients of the alcohols, was evidence of great sensitivity to the hydrophobic effect of the alcohols. Only the first alcohols, however, produced any notable inhibition of Mg2+-ATPase and sucrase. Hydrophobic bonds are thus shown to have little influence in maintaining the activity of Mg2+-ATPase and sucrase, but they modulate the Na+-coupled d-glucose uptake.  相似文献

该试验以高山离子芥试管苗(Chorispora bungeana)为试材,采用固液培养法,设置对照(不添加PEG-6000,CK),轻度干旱胁迫(5%PEG-6000)、中度干旱胁迫(20%PEG-6000)、重度干旱胁迫(40%PEG-6000)4个干旱处理水平,分析干旱胁迫对高山离子芥幼苗抗氧化系统、活性氧代谢等部分生理特征的影响,以揭示高山离子芥在干旱胁迫下的生理响应特征,为进一步探讨其对干旱环境的适应机制奠定基础。结果显示:(1)随着干旱胁迫程度的增加以及在各时间胁迫处理下,抗氧化酶SOD活性及可溶性糖含量显著升高,POD活性、丙二醛含量、CAT活性和APX活性均经历了先升后降的过程。(2)超氧阴离子(O-·2)的产生速率和过氧化氢(H2O2)的含量均显著升高;高山离子芥试管苗叶片相对电导率呈现出升-降-升的变化趋势。(3)相关分析结果显示,MDA与相对电导率、可溶性糖、SOD、APX、O-·2及H2O2呈极显著正相关关系,可溶性糖与SOD、POD、O-·2及H2O2呈极显著正相关关系;相对电导率以及保护酶系均与O-·2、H2O2呈极显著正相关关系。研究表明,高山离子芥具有较强的耐旱性,高山离子芥试管苗在响应干旱胁迫过程中,抗氧化酶系、活性氧代谢、脂质过氧化及渗透调节物等共同参与了高山离子芥试管苗对干旱胁迫的综合抗逆性形成,从而积极启动应对外界干旱环境的耐旱响应机制。  相似文献

In situ plasma membrane H+-ATPase activity was monitored during pH-regulated dimorphism of Candida albicans using permeabilized cells. ATPase activity was found to increase in both the bud and germ tube forming populations at 135 min which coincides with the time of evagination. Upon reaching the terminal phenotype the mycelial form exhibited higher H+-ATPase activity as compared to the yeast form. At the time of evagination H+-efflux exhibited an increase. K+ depletion resulted in attenuated ATPase activity and glucose induced H+-efflux. Alternative respiration pathway (AP) is an important pathway which can be induced by environment stresses in plants. In the present study, we show a new mechanism involving the AP in nitrogen deprivation-induced tolerance of Poa annua callus to salt stress. The AP capacity markedly increased under a 600 mM NaCl treatment or nitrogen deprivation pretreatment and reached a maximum under the nitrogen deprivation pretreatment combined with the NaCl treatment (–N+NaCl). Malondialdehyde (MDA) and H2O2 content and Na+/K+ ratio significantly increased under the 600 mM NaCl treatment but less under the–N+NaCl treatment. Moreover, both the nitrogen deprivation and the NaCl stress stimulated the plasma membrane (PM) H+-ATPase activity and increased pyruvate content. The maximal stimulating effect was found under the–N+NaCl treatment. When the AP capacity was reduced by salicylhydroxamic acid (SHAM, an inhibitor of AP), content of MDA and H2O2 and Na+/K+ ratio dramatically increased, whereas PM H+-ATPase activity decreased. Moreover, exogenous application of pyruvate produced a similar effect as the nitrogen deprivation pretreatment. The effects of SHAM on the Poa annua callus were counteracted by catalase (a H2O2 scavenger) and diphenylene iodonium (a plasma membrane NADPH oxidase inhibitor). Taken together, our results suggest that the nitrogen deprivation enhanced the capacity of AP by increasing pyruvate content, which in turn prevented the Poa annua callus from salt-induced oxidative damages and Na+ over-uptake.  相似文献
