角质层是表皮细胞壁表面的一层不透水的脂肪性物质。角质层与表皮细胞紧密结合,植物表皮细胞形态和排列方式、气孔器的形态结构等微形态特征均能在角质层上反映出来。利用光学显微镜和扫描电镜对松属(Pinus)12种植物针叶角质层微形态特征进行观察和比较,详细描述20个性状,其中12个性状来自角质层内表面,8个性状来自角质层外表面。结果表明,这些特征可为该属属下分类和相似种的鉴别提供有用信息,具有重要的分类学意义:①表皮细胞长度、表皮毛长度、角质层外表面起伏程度、表皮细胞轮廓、有无气孔塞和针絮状物质等角质层微形态特征具有自身特异性,在属下可作为松属组级水平上的分类依据。角质层微形态 特征不支持将五针松组(P. Section Cembra)和白皮松组(P. Section Parrya)合并为P. Section Quinquefolius的观点,亦不支持将油松组(P. Section Pinus)分成P. Section PinusP. Section Trifolius的看法。②白皮松(P. bungeana)针叶角质层微形态特征既与五针松组有相同之处,又与油松组有相似之处,还有部分特征显示出不同于松属其他种类的独特性,可为白皮松亚属(P. Subgenus Parrya)的建立提供新依据。③扫描电镜下表皮细胞垂周壁纹路,气孔塞有无和外表面气孔形状等特征可为形态相似种火炬松(P. taeda)和湿地松(P. elliottii)提供种间界定依据。  相似文献

Summary The inner epidermis of the inner integument of cotton seed coats (fringe-layer) and the cuticles between this cell layer and the nucellus were examined in the light and electron microscope at different times of their development. The cells of the fringe-layer contain only small vacuoles and their cytoplasm is densely packed with organelles and free and membrane-bound polysomes. The lateral walls contain many plasmodesmata. At the time when the fruit capsules stop growing, the fringe-cells produce a cell wall labyrinth, resembling that of transfer cells. The cell wall labyrinth is restricted to the lateral walls. The differentiated state of the fringe-cells is short-lived. At about the time of elaboration of the cell wall labyrinth most of them become progressively vacuolated, lignify, and lose their cytoplasmic constituents. The development of the fringe-layer is well correlated with other developmental events in the inner integument, but not with the filling of embryo and endosperm with reserve substances.At anthesis, the fringe-layer and nucellus are covered by a thin cuticle proper of about 20 nm. After anthesis, the nucellar cells start to produce a cuticular layer of considerable, but variable, thickness (0.25–2.5 m), containing a polysaccharide network.In drying seeds the cells of the fringe-layer disrupt. The thin outer tangential wall remains attached to the seed coat. The rest of the cell, together with the cuticles and the collapsed cells of the nucellus, form a protective layer around embryo and endosperm, remaining attached to the seed coat at the chalazal end.  相似文献

Summary Black cuticles of larvae and pupae from various Lepidoptera were studied by light and electron microscopy. There are striking differences in the representation of black pigmentation, especially at the ultrastructural level. Two types may be described: 1. With the light microscope black melanin-like grana, electron-dense electron microscopically, are found in the distal parts of the exocuticle. This type is demonstrated in larvae of Celerio euphorbiae, Papilio machaon, and Phalera bucephala. 2. With the light microscope a dark homogeneous layer in the distal exocuticle can be recognized, however, electron microscopically no structures correlated with this dark pigment layer. This type of pigmentation was present in pupae of Pieris brassicae and Aglais urticae; in Pieris larvae the dark pigmented layer appeared to be limited to the epicuticle. In Celerio processes of the epidermal cells are involved in transporting precursors to the exocuticle. The conclusion was reached that black pigmentation in cuticles is based on different mechanisms as proposed by structural features. Termites, Coptotermes formosanus Shiraki, reared individually, were highly susceptible to entomopathogenic fungi, Paecilomyces fumosoroseus and Beauveria brongniartii and Metarhizium anisopliae, while termites reared in groups were highly resistant. Quantitative assays with an epifluoresent microscope revealed a significant difference in the number of conidia attachments among three entomopathogenic fungi. The conidia of B. brongniartii and P. fumosoroseus bound to termite cuticles more effectively than M. anisopliae conidia. Our results also suggested that self-grooming behavior is less effective, but mutual grooming is very effective in the removal of conidia from cuticles of their nestmates. Statistical analysis of removal rates indicated that conidia of P. fumosoroseus and B. brongniartii were removed more rapidly than M. anisopliae conidia from termite cuticles.  相似文献

Summary Tomato seedlings were exposed to 3 days of chilling at 5 °C in both the presence and absence of 5 mM ethanolamine. An array of pathological responses could be seen at both the light and electron microscope levels: apparent loss of cell turgor, vacuolization, reduction in the apparent volume of both the cytoplasm and the vacuolar protein bodies, apparent deposition of new material in the cell walls, general disorganization of organelles, especially plastids, and a general loss of cytoplasmic structure. Chilling injury was less pronounced in ethanolamine-treated cotyledonary tissues which appeared to have normal turgor and cell shape at the light microscope level and better preserved organelles at the ultrastructural level. Protection by ethanolamine at the ultrastructural level was most pronounced for the cytoplasm, the mitochondria, and the cell walls. Dead cells were seen with both treatments but were 2–3 times as numerous in the chilled-only tissue.  相似文献

In the stress fibers of two types of nonmuscle cells, epithelia (PtK2, bovine lens) and fibroblasts (Gerbil fibroma, WI-38, primary human) the spacing between sites of alpha-actinin localization differs by a factor of about 1.6 as determined by indirect immunofluorescence and ultrastructural localization with peroxidase-labeled antibody. Both methods reveal striations along the stress fibers with a center-to- center spacing in the range of 0.9 mum in epithelial cells and 1.5 mum in fibroblasts. Periodic densities spaced at comparable distances are seen in PtK2 and in gerbil fibroma cells when they are treated with tannic acid and examined in the electron microscope. In such cells, densities are found not only along stress fibers but also at cell-cell junctions, attachment plaques, and foci from which stress fibers radiate. These latter three sites all stain with alpha-actinin antibody on the light and electron microscope level. Stress fibers in the two cell types also vary in the periodicity produced by indirect immunofluorescence with tropomyosin antibodies. As is the case for alpha-actinin, the tropomyosin center-to-center banding is approximately 1.6 times as long in gerbil fibroma cells (1.7 mum) as it is in PtK2 cells (1.0 mum). These results suggest that the densities seen in the electron microscope are sites of alpha-actinin localization and that the proteins in stress fibers have an arrangement similar to that in striated muscle. We propose a sarcomeric model of stress fiber structure based on light and electron microscopic findings.  相似文献

Detailed cytological changes that accompany the rejuvenation of resting cells of Melosira granulata were studied with the electron microscope. Dormant and viable cells that we previously classified as the condensed state generally contain definable chloroplasts, mitochondria, a nucleus and other cytoplasmic remnants. However, there appears to be a continuous cytoplasmic degradation spectrum and some cells which appear intensely colored with the light microscope have discontinuous chloroplast membranes and few other cytoplasmic remnants. Rejuvenation of viable dormant cells is initially accompanied by the accumulation of both lipids and polyphosphates. In the earliest stages of expansion, these storage products are dispersed throughout the cell. In later stages of expansion, the lipids appear to be coalesced into larger droplets which are easily identified at the light microscope level. The fully expanded stage is characterized by the normal complement of organelles and their arrangement at the periphery of the cells and central cytoplasmic bridge. These cells appear both anabolically and catabolically active as evidenced by the abundance of endoplasmic reticulum, ribosomes and secretory and lytic vesicles. Prior to cell division, both lipids and polyphosphates a re reduced or absent in the cells. The ultrastructural features of the dormant, condensed state in resting cells of M, granulata are similar to those described for hypnospores. A rejuvenation sequence that produces cytological features common to resting state formation could provide a population of cells which could easily revert should environmental conditions become adverse.  相似文献

In order to improve understanding of the systematic relationships between taxa and of tepal-pollinator and tepal-habitat relationships, a detailed scanning electron microscopy (SEW study of the adaxial tepal surface of 135 taxa belonging to c. 14 genera in the tribes Persicarieae and Polygoneae (Polygonoideae-Polygonaceae) was carried out. The tepal surfaces have revealed some striking differences among the genera studied. Three main patterns of sculpturing and cell shape can be recognized. Type I: mostly rectangular to elongate cells with straight or undulating anticlinal walls; the cuticles smooth or striate in longitudinal direction and often continuous (Persicaria, Bistortu, Aconogonon, Koenigia and some species of Fagopyrum). Type II: irregular tesselated to elongated cells, rarely rectangular with mostly sinuate anticlinal walls; the cuticles rarely with longitudinal striation, but with strong orthogonal to reticulate ridges or striae, often without correlation between cells (Polygonum s.s., Polygonella, Fallopia, Parapteropyrum, Pteropyrum, Atraphaxis and Calligonum). Type III: mostly irregular, mammilliform, papillose cells with either longitudinal tightly pressed or randomly ridged cuticles (some species of Fagotyrum and Oxygonum). The present data generally support the division of Polygoneae and Persicarieae by Ronse Decraene & Akeroyd. The mammilliform, papillate epidermis of tepals in Oxygonum and some Fagopyrum is probably well-adapted to entomophily either as a foothold or even as a light-trap, and also shows some correlation with heterostyly. It is noteworthy that the tepal structure of Polygonella (tesselated form) is highly similar to that of Polygonum s. s. (especially P. sect. Duravia). Cuticular striation alone does not appear to have much systematic value, but has a mechanical function in affording extra strength to the cell walls. Some correlations between the degree/shape of cuticles and the pollination mechanisms or the habitat of each taxon are also briefly discussed. It is concluded that the studied tepal characters can be useful in improving the classification of tribes Persicarieae and Polygoneae, arid give additional support at the infrageneric level.  相似文献

Microscopic investigations over time were carried out to study and compare the pathogenesis of invasion of ticks and blowflies by Metarhizium anisopliae. The scanning electron microscope and stereo light microscope were used to observe and record processes on the arthropods' surfaces and the compound light microscope was used to observe and record processes within the body cavities. Two distinctly different patterns of invasion were found in ticks and blowflies. Fungal conidia germinated on the surface of ticks then hyphae simultaneously penetrated into the tick body and grew across the tick surface. There was extensive fungal degradation of the tick cuticle, particularly the outer endocuticle. Although large numbers of conidia adhered to the surface of blowflies, no conidia were seen to germinate on external surfaces. A single germinating conidium was seen in the entrance to the buccal cavity. Investigations of the fly interior revealed a higher density of hyphal bodies in the haemolymph surrounding the buccal cavity than in haemolymph from regions of the upper thorax. This pattern suggests that fungal invasion of the blowfly is primarily through the buccal cavity. Plentiful extracellular mucilage was seen around the hyphae on tick cuticles, and crystals of calcium oxalate were seen amongst the hyphae on the surface of ticks and in the haemolymph of blowflies killed by M. anisopliae isolate ARIM16.  相似文献

Abstract. The fate of [3H]thymidine ([3H]Tdr) pulse-labelled cells was followed in tracheal epithelium of young male rats. The time course for cell differentiation, and the relation of events to tissue composition were studied. In vivo labelling and light microscope autoradiography of epoxy embedded sections were used. Labelled and total nuclei for each cell type, and combinations of labelled cells which were adjacent to one another, were tallied. Hierarchical analyses of variance were performed on the several data sets. All cell types, except ciliated, were labelled at 1 hr. A few labelled ciliated cells were seen 24 hr post-label. The frequency of labelled intermediate cells peaked at day 2; goblet and ciliated cells at day 3. No significant changes occurred in the labelling index, but at 24 hr the frequency of adjacent labelled cells (ALC) had increased > 5-fold, and changes had occurred in patterns of ALC combinations. The labelled ciliated cells which were seen at 24 hr were adjacent to labelled intermediate cells. No labelled basal-ciliated cell combinations were seen at any time. Data indicated that ciliated cells can develop from S-phase intermediate cells within 24 hr, and neither basal nor superficial goblet cells are progenitors of ciliated cells. It is proposed that both superficial goblet cell and ciliated cell development is preceded by two divisions: a basal cell division followed by an intermediate cell division.  相似文献

The cotton aphid is one of the most serious pests of greenhouse vegetable crops worldwide. It is difficult to control because field populations usually include simultaneously several insect developmental stages. The current research evaluated an isolate (CS625) of Lecanicillium attenuatum, a fungal pathogen of aphids, as to its virulence against different developmental stages of cotton aphid, Aphis gossypii. The influence on mortality of several other factors also was examined: (a) insect moulting, (b) the number of conidia attached to insect cuticles and (c) germination rates of conidia on cuticles of aphids at various developmental stages. Mortality of cotton aphids treated with L. attenuatum conidia varied according to the developmental stage of the host, i.e. the LT50s with third-instar nymphs and adults was shorter than with first-instar nymphs. The number of spores attached to the surface of first-instar nymphs was approximately one-half of that on third-instar nymphs and adults. Also, the level of spore germination on the surface of first-instar nymphs was lower than on the surface of other stages of the aphid. After moulting, the numbers of conidia attached to new insect cuticles were less than on exuviae. These results suggest that early nymphal stages of cotton aphids may escape fungal disease due, at least in part, to a combination of three factors: low numbers of conidia attached to their cuticles; low levels of conidial germination and rapid ecdyses, which removed conidia before their germ tubes penetrated the host hemolymph.  相似文献

The cuticles of the crayfish Austropotamobius pallipes (Lereboullet), and the crabs Cancer pagurus L. and Carcinus maenas (L.), have been examined with the scanning electron microscope. Laminae are composed of sheets of horizontal fibres which subsequently are into the inter-laminae. The inter-laminae contain sheets of fibres which are at a slight angle from the horizontal between sucessive laminae. The disposition of the pore canals is clearly related to fibre pattern. The sinuous macrofibres described by Dennell (1973) are discrete from the pore canals.  相似文献

The periostracum and cells lining the periostracal groove of Anodonta cygnea L. have been studied at the electron microscope level. The cells lining the inner face of the outer fold differ in fine structural details, five cell types being recognized. Along the length of the outer surface of the middle fold, to which the periostracum is closely applied, only two cell types are evident. At the base of the periostracal groove the two epithelia are separated by a bulbous region containing a group of basal cells which initiate the periostracum. The periostracum, which is homogenously electron-lucid, originates in the intercellular space between a basal cell and the first cell of the middle fold. It increases in thickness in the periostracal groove due to the secretory activity of the different outer fold cells. The cells of the middle fold do not appear to be involved in periostracum formation.  相似文献

When leaves of the Taxodiaceae are placed in 10% chromium trioxide at 40o C for 6–24 hours, a macerated cuticle is obtained. Examination of the inner surface of these cuticles by the scanning electron microscope reveals a consistent steady state structure, which we show to be usable for comparative study. The variations of the sculpture, both within the same leaf and throughout the family, are presented. Both the structural and the evolutionary significance of the features are discussed.  相似文献

An electron microscope study of sheep myocardial cells has demonstrated the presence of a transverse tubular system, apparently forming a network across the cell at each Z band level. The walls of these tubules resemble the sarcolemma in consisting of two dense layers—plasma membrane and basement menbrane; continuity of the tubule walls with the sarcolemma can be seen when longitudinal sections of a cell are obtained between two subsarcolemmal myofibrils and at the same time perpendicular to the cell surface. The demonstration of communication between the lumen of the transverse tubular system and the extracellular space appears to be more definite in this study than in any work hitherto published. It provides anatomical evidence of a possible direct pathway for transmission of the activating impulse from the sarcolemma to the myofibril Z bands.  相似文献

Temperature-sensitive (ts) mutants of Staphylococcus aureus with defective cell wall biosynthesis have been differentiated from other ts mutants by their ability to grow at the restrictive temperature (43 C) in the presence of 1 m NaCl. Under all conditions they possess normal colonial and cellular morphology at the level of resolution of the light microscope and are, therefore, not protoplasts. However, differences between mutant and wild-type cells can be seen by scanning electron microscopy. Many of the mutants contained concentrations of nucleotide precursors of peptidoglycan synthesis in excess of those present in wild-type cells, at both 30 and 43 C. The types of peptidoglycan precursors accumulated by six of the mutants have been determined, and specific enzymatic defects in three of these have been identified.  相似文献

The fine structure of conspicuous extracellular materials during the life history of a sea star (Patiria miniata) is described. The outer surface of the developing sea star is covered by two morphologically different cuticles that appear sequentially during ontogeny. The primary cuticle, which is about 120 nm thick and two-layered, is present from mid-blastula through the end of the larval stage. The secondary cuticle, which is about 1 micron thick and three-layered, first appears on the epidermis of the rudiment region of the larva and, after metamorphosis, covers the entire epidermis of the juvenile and adult stages. During ontogeny, there are only two conspicuous gut cuticles: the first lines the newly invaginated archenteron at the start of the gastrula stage, and the second lines the esophagus during the larval stage. A blastocoelic basal lamina first appears at mid-blastula and persists as subectodermal and subendodermal basal laminae. Ruthenium red-positive granules are detectable between the lateral surfaces of adjacent ectodermal cells during part of the gastrula stage; this transient intercellular material may possibly aid in lateral adhesion between cells.  相似文献

Summary A simple and reliable method has been developed for the in situ LR White embedding of cell monolayers grown on glass cover-slips. Combined with cytochemical or immunological procedures, this technique allows light and/or electron microscopy investigations of a large number of cells in the same horizontal plane within a relatively short period of time. It can be applied to cells grown on microgrid finder cover-slips which allows a distinct site of even an individual cell of a monolayer to be studied at first at the light microscope level and subsequently at the electron microscope level. Hence, it is also suitable for controlling manipulation of single cells, followed by their serial sectioning after relocation in the electron microscope.  相似文献
