棕种子萌发和幼苗生长十分缓慢 ,为了探索加快种子萌发和幼苗生长的方法 ,对棕种子进行不同处理、不同时间催芽和催芽后不同处理等试验 ,结果表明 :(1)生长调节剂 3号 ABT生根粉、吲哚丁酸和 2 ,4 - D可提高种子萌发率 ,促进幼苗生长。 (2 )播种前催芽可促使种子提前萌发 ,延长幼苗生长期 ,使当年生苗更粗壮 ;贮藏 3个月后催芽可提高 10 %~ 15%的发芽率。 (3)催芽后用 50 mg/ L和 10 0 mg/ L的 3号 ABT生根粉、或者 10 0 mg/ L的萘乙酸浸泡都可以提高幼苗出土率 ,但对幼苗生长没有明显影响。  相似文献

将不同胚龄的白及种子接种到不同培养基中进行培养,对白及种子萌发率、萌发时间、丛生芽增殖、生根等方面进行了研究。结果表明:白及种子萌发率与其胚龄及有胚率成正相关,萌发时间则与胚龄及有胚率成负相关;胚龄16周的种子在1 g/L花宝1号 2 g/L花宝2号的培养基上萌发率可达84%;胚龄等于或大于20周的种子萌发率不受培养基成分的影响,均可达100%,萌发时间只需7 d;丛生芽增殖的最佳培养基为1/2 MS 4.0 mg/L6-BA 0.2 mg/L NAA 100 g/L CM,其增殖倍数达4.41倍;诱导生根较好的培养基为1/2 MS 0.2 mg/L NAA,生根率达90%。  相似文献

对云南火焰兰进行无菌播种技术试验.结果表明,适宜萌动的培养基为1/4MS 椰子水20% 蛋白胨1 g L-1 蔗糖10 g L-1,播种30 d时萌动率可达93.1%;适宜萌发的培养基为VW 椰子水10%,培养75 d时萌发率达36.6%.盐浓度较高的MS培养基不适合种子萌发和原球茎生长,低盐的KC培养基上种子能较好地萌发和成苗;添加椰子水可明显提高种子萌动率,并有利于原球茎的生长和分化.光培养和暗培养对种子萌动的影响差异不明显,但暗培养3周会对种子后期发育不利.添加150 g L-1的苹果匀浆有利于幼苗生长.试管苗移栽成活率在95%以上.  相似文献

任木种子繁殖试验结果表明:(1)采自石灰岩土和酸性土上的种子萌发率没有明显差别。(2)种子用80℃热水浸泡处理效果最好,发芽率最高,达到80%~90%。IBA等生长素处理对种子萌发没有促进作用。(3)种子耐贮藏,干藏3年后与新鲜种子的发芽率相差不多。(4)全光照条件下,幼苗的生长速度比林荫下快。(5)任木幼苗在石灰岩土和园土上生长比酸性红壤上好。  相似文献

研究了培养基组成成分对中国石竹品种Diana F1 White组培程序中种子萌发、诱芽增殖、试管苗玻璃化及生根等重要环节的影响。结果表明:(1)种子适宜的萌发培养基为1/2MS,在此培养基中种子萌发率为83.33%,播种后30d时无菌苗高度达6.99cm,叶片数达26.7。(2)在芽诱导阶段玻璃化苗发生率最高可达53.85%。将光照强度提高至2000lx后,采用培养基MS+6-BA3.0mg/L+NAA0.3mg/L+琼脂8.0g/L+蔗糖8.0g/L进行诱芽培养,玻璃化苗发生率降至3.33%,外植体出芽率达到90%,单个外植体出芽数达到7.2个。(3)适宜的生根培养基为1/2MS+1.0mg/LIBA+琼脂8g/L+蔗糖40g/L,培养30d时生根率为100%,平均根条数达到24.7条,平均根长度达到4.7cm。试管苗在消毒后的腐殖土中移栽,成活率达95%。该研究结果为DianaF1试管苗的快速繁殖提供科学依据。  相似文献

以八角莲种子为外植体,MS为基本培养基,通过不同的激素种类和浓度配比,对八角莲进行组织培养研究。结果表明:种子在MS+BA1.0mg/L+IBA0.5mg/L+GA34.0mg/L培养基上容易萌芽,发芽率为72.4 %;培养基MS+BA10.0mg/L+GA30.5 mg/L可诱导种子幼苗形成丛生芽;继代繁殖在MS+BA(8.0~10 .0)mg/L+GA32 .0mg/L与低浓度BA或无BA的培养基上进行循环培养效果较好;MS+NAA1.0 mg/L+AC0.2g/L适宜诱导生根获得再生植株,生根率100%。带叶叶柄在MS+BA1.0mg/L+2-ip(0.5~1.0) mg/L+NAA0.02 mg/L培养基上可诱导愈伤及根,直接形成再生植株。生根苗移栽成活率90 %。  相似文献

以授粉后不同发育时期的同色兜兰种子为材料,观察其形态特征和萌发过程,并探讨建立同色兜兰高效快繁体系的最佳条件。结果表明,种子发育中后期即授粉后210~240d为较适宜的采收期,授粉后210d的种子萌发率最高(达77.79%);1/4 MS和1/2MS为同色兜兰适宜的基本培养基,添加100mL/L椰乳或1g/L蛋白胨对种子萌发及原球茎生长和分化有明显的促进作用;添加1g/L活性炭对原球茎褐化有一定的抑制作用,但添加剂量不宜过大;添加香蕉汁和苹果汁对同色兜兰种子萌发和原球茎生长分化有抑制作用;暗处理对同色兜兰种子萌发无影响;分化后的原球茎在壮苗和生根培养基上培养120d即可得到4~5片叶、高3~5cm的同色兜兰健壮试管苗。  相似文献

随着白芨胚龄增大,胚成熟度不断增加,有胚率和萌发率逐渐提高,种子萌发时间逐渐变短。胚龄为20周后采收最好,有胚率最高,萌发率为100%,萌发时间只需1周。种子萌发时胚先发育成原球茎,原球茎再分化出叶片和根系,形成完整的小植株;花宝一号1g/L+花宝二号2g/L+10%椰子汁培养基可促进种子萌发及幼苗生长,比常用的基本培养基加生长调节剂更简捷,幼苗生长健壮。  相似文献

研究了影响金线兰种子非共生萌发的因素,并应用正交试验研究基本培养基、6-BA、ZT、NAA四种因素对原球茎增殖的作用。结果表明:授粉类型对金线兰种子非共生萌发影响较大,异株异花、同株异花以及同株同花授粉所得的种子的萌发率分别为78.53%、69.62%、39.87%;蒴果成熟度以生长150d为宜,采收后萌发率可达78.59%;冷藏影响种子的活力,种子的萌发率随冷藏时间的延长而降低;使用次氯酸钠浸泡后的种子与对照相比,其萌发率无明显差异;NAA对原球茎增殖作用显著,适宜于原球茎增殖的培养基为1/2MS+ZT0.5mg/L+NAA1.0mg/L。  相似文献

Asymbiotic seed germination of Coelogyne flaccida varied with the capsule stage and the culture medium used for germinating seeds. The capsules were harvested at two different stages of development. The seeds were cultured on three asymbiotic orchid seed germination defined and undefined media, i.e. Mitra (M) medium, Murashige and Skoog (MS) medium and potato dextrose agar (PDA) medium. The seeds obtained from undehisced green capsules germinated with a maximum germination percentage (84.50 ± 0.33%) on M medium followed by MS and PDA medium. The effect of cytokinins, such as 6-benzylaminopurine and furfurylaminopurine and the synthetic auxin α-naphthalene acetic acid, on seed germination was also assessed. Simultaneously, in vitro multiplication using protocorms as explants was also studied. The effect of organic growth supplements, such as banana homogenate (BH, 25, 50, 75 g l? 1) and peptone (P, 1.0, 1.5, 2.0 g l? 1), was tested on the de novo formation of protocorm-like bodies (PLBs), development of the maximum number of shoots and early formation of plantlets using the M medium. Among the treatments, the highest regeneration frequency (87.50 ± 0.20%) and the highest number of PLBs per explant (10.25 ± 0.50) were obtained in P (1.5 g l? 1)-supplemented cultures, and the plantlets were formed within 18 weeks of culture. BH favoured the development of healthy plantlets, with a maximum fresh weight of 1.02 ± 0.04 g per plantlet.  相似文献

分别以三种催芽处理10、20、40 d的暴马丁香种子进行离体培养试验,对种子做三种切割处理,中间切、两端切和完整种子。结果表明:采用切开种子的方法进行离体培养可提高种子发芽率,中间切种子的萌发最好,且在培养10 d后发芽率即达到最高值;经过40 d催芽处理的种子优于未经催芽和只进行20 d催芽处理的种子;切开种子以MS培养基附加5 mg/L BA或5 mg/LBA+0.1 mg/L IBA为最适培养基。  相似文献

Rapid in vitro propagation of the terrestrial orchid, M. khasiana through immature seed culture was achieved. Immature seeds of 8-9 week after pollination (WAP) cultured on MS medium (2% sucrose) supplemented with 500 mgl(-1) casein-hydrolysate and 1 microM N6-benzyladenine (BA) exhibited germination of 75% seeds after 107 days of culture and subsequently supported the development of PLBs. Subsequent culture on MS medium enriched with 6 microM of indole-3-acetic acid (IAA), 18 microM each of BA and kinetin induced multiple shoots and plantlets. Transfer of PLBs to MS medium with 0.1% activated charcoal (AC) facilitated rapid proliferation of PLBs, while AC at 0.2% favored shoot bud induction and rhizome enlargement. The plantlets, developed on medium with IAA, BA and kinetin, after hardening in vitro for 8-10 weeks were planted in community pots and transferred to poly-house. The plantlets showed 65% survival under field conditions.  相似文献

Epipactis flava Seidenf., is an endangered Thai rheophytic orchid that has decreased rapidly throughout its natural habitat and urgently requires conservation using in vitro techniques. Effect of pollination types (self- and cross-pollination) and seed from different capsule ages (2, 4, 6 and 8 weeks after pollination) on asymbiotic seed germination efficiency was performed on semi-solid VW medium supplemented with 150 mL/L coconut water and 50 g/L potato extract. Highest germination rate was 79.2% with definite rate of protocorm developmental stage 5 at 24.0% using seeds from 6 week old cross-pollination capsules. Anatomical investigation revealed young seed has been formed at 6 weeks old capsules. Identical seeds were then cultured on semi-solid and liquid systems of five different media including modified-VW, MS, BM, MM and KC for 2 months. Results showed that semi-solid and liquid VW medium promoted the highest seed germination rates at 70.2% and 70.4%, respectively, with the highest definite rate of protocorm developmental stage 5 at 54% found on semi-solid BM medium. In vitro seedlings were transferred to culture on both semi-solid and liquid VW and MS media for 2 months. Higher growth parameters, as indicated by shoot number and fresh weight, were obtained on liquid MS medium. Clumps of plantlets containing 5–8 shoots were acclimatized and transplanted into different potting media for 3 months. Survival percentages in all tested substrates were recorded at nearly 100% with no significant differences, while 63–68% of the living plantlets produced new shoots.  相似文献

为建立新疆狭叶薰衣草(Lavandula angustifolia)的快速繁殖体系,以种子、茎、叶为外植体,对种子萌发、愈伤组织诱导、丛芽分化和生根的最适培养条件进行了研究;用水蒸气蒸馏法提取狭叶薰衣草挥发油,采用气相色谱-质谱法测定挥发油成分。结果表明,种子浸泡的适宜时间为6 h,切开种皮培养,出芽时间最少为6 d;诱导种子出芽的适宜培养基为MS+6-BA2 mg/L;以茎为外植体诱导愈伤组织效果较好,适宜培养基为MS+6-BA 2 mg/L+2,4-D 1 mg/L;诱导分化丛芽的适宜培养基为MS+6-BA 1 mg/L+NAA 0.5 mg/L;生根的适宜培养基为1/2MS+NAA 1 mg/L+6-BA 0.5 mg/L;盆栽薰衣草和无菌苗薰衣草的挥发油主要成分相差较大,离体培养的薰衣草的主要挥发性成分有叶绿醇、丁香油烃、氧化石竹烯等。  相似文献

During germination in the siliquae of developing seeds of a precociously germinating (PG) line of Chinese cabbage (Brassica rapa spp. Pekinensis), the synthesis of cruciferin and oleosins was maintained and the activities of isocitrate lyase and malate synthase increased. Thus, both developmental and germinative/post-germinative events coexisted within the same PG seeds. Drying of the seeds resulted in the developmental events being switched off and germinative/post-germinative ones being initiated. Only about 18% of the seeds of the PG line completed germination in the siliquae, so the potential for germination of the remaining non-germinating seeds was tested during their development. Isolation of these developing seeds from the siliquae and imbibition in water induced PG, but developmental events were terminated and the seeds entered a germinative/post-germinative programme. This termination of developmental events in the induced PG seeds was not permanent, however, and they could be re-induced by incubating the seeds in ABA on Murashige and Skoog (MS) medium. If incubated on MS medium plus a low concentration of ABA, both developmental and germinative/post-germinative events could be induced to coexist in the PG seeds up to the time of establishment of the seedlings. Control of the developmental and germinative/post-germinative events in the seeds was influenced by the environment in which they were developing, which could be mimicked by manipulating the nutritional status and ABA content in which the seeds were in contact.Keywords: Abscisic acid, ABA, Brassica rapa, Chinese cabbage, stored reserves, precocious germination, seed development   相似文献
