昆明山海棠茎中化合物的分离及其抗炎活性   总被引:1,自引:0,他引:1  
从昆明山海棠茎的氯仿提取物中分离得到6个化合物,经鉴定为雷公藤内酯甲(Ⅰ)、昆明山海棠二萜内酯(Ⅱ)、雷公藤内酯乙(Ⅲ)、雷酚二萜酸(Ⅳ)、雷公藤碱(Ⅴ)及3-epikatonic acid(Ⅵ)。其中,化合物Ⅲ及Ⅵ为首次从该植物中分离得到,化合物Ⅴ为首次从该植物茎中分离得到。药理实验显示化合物Ⅰ和Ⅱ对大鼠足跖部角叉菜胶炎症模型具有明显的抑制作用。  相似文献

雷公藤植物的茎,叶在含激动素0.1mg/1,2.4-D 1mg/l的6.7-v培养基上诱导出愈伤组织。在22±2℃的条件下进行扩大培养后,收集培养物进行提取、分离。从薄层层析上显示出培养物中含有雷公藤甲素、雷公藤乙素等二萜内酯化合物。柱层析分离出5个萜类化合物,根据它们的mp、TLC、IR、~1HNMR、MS等图谱资料,其中4个分别鉴定为雷公藤酮,雷公藤内酯甲、3-Epikatonic acid,齐墩果酸,另一个化合物的结构有待进一步研究。  相似文献

雷公藤二萜内酯类化合物在结构上有一定的相似性:它们均含有3个六元环、1个五元内酯环和1~3个二元氧环,并且具有多种生物活性。本文选择了7个具有代表性的雷公藤二萜内酯化合物并通过X衍射分析获得其三维结构数据,以已有的药理实验数据为基础,对其精细立体结构变化和生物活性关系进行了探讨。研究结果表明:雷公藤萜内酯类化合物中的A、B、C三个六元环是其活性骨架的几何依托;18位上的羰基和7~8位上的三元氧环是其活性的可能结合部位;而该类化合物的活性部位则集中在C环的取代位置上;D环不是该类化合物具有生物活性的必需骨架部分。  相似文献

以雷公藤(Tripterygium wilfordii)不定根为材料,通过两相培养技术,研究有机溶剂邻苯二甲酸二丁酯(DBP)不同浓度及培养时间对雷公藤不定根生长及次生代谢产物含量的影响。结果显示,DBP浓度为6%、培养至第6 d时,不定根增长量达1.14 g/瓶,为对照(1.08 g/瓶)的1.06倍;DBP浓度为2%、培养至第8 d时,内酯醇含量达74.96μg/g,为对照(53.67μg/g)的1.40倍;DBP浓度为2%、培养至第2 d时,所收获的内酯醇总产量最高(0.40 mg/瓶),且为对照(0.27 mg/瓶)的1.48倍。本研究结果表明,在培养基中添加一定浓度DBP,虽然适合雷公藤内酯醇的形成,但不适合雷公藤生物碱的形成;不论添加DBP浓度大小、培养时间长短,不定根中吉碱和次碱含量及每瓶总产量均低于对照。  相似文献

由红根草(Salvia prionitis Hance)中分得三个abietane类二萜成分:红根草酮内酯(prioketolactone,1)、新红根草酮(neoprionitone,2)和二氢异丹参酮1(dihydroisotanshinone1,3)。经波谱分析确定了它们的化学结构。其中,1,2为新化合物,1是鼠尾草植物中首次发现的具有七元环内酯结构的二萜醌类化合物,3为首次从红根草中发现。  相似文献

采用双向发酵的原理,运用"发酵过程动态比较法"研究了灵芝接种于雷公藤药性基质上发酵不同时间所得菌质的化学成分、急性毒性和免疫功能的变化,以其确定雷公藤解毒持效双向发酵的最佳发酵终点。研究结果表明:发酵第30天所得的菌质(G30)总二萜的含量最低,为0.57%;与雷公藤生药比较,G30的LD50最高,且发酵第30天所得菌质的体液免疫和细胞免疫抑制作用最强。综合对成分含量、毒性及药效的动态数据联系比较、分析,确定了雷公藤解毒持效双向发酵的发酵终点与适宜的发酵周期是菌丝长满瓶后的第30天。  相似文献

该文观察了雷公藤内酯醇阳离子聚合物(TPL-PEI-Cy D)对乳腺癌干细胞的影响。采用CCK-8测定TPL-PEI-Cy D、雷公藤内酯醇(TPL)对HL-7702细胞的毒性,用TGF-β1孵育培养MCF-7细胞,流式细胞仪检测其中CD_(44)~+CD_(24)~–标志的细胞比例;免疫磁珠法分选其中CD_(44)~+CD_(24)~–细胞群;TPL-PEI-Cy D作用于MCF-7干细胞后,流式细胞术检测其中CD_(44)~+CD_(24)~–细胞比例的变化。雷公藤内酯醇在接入聚乙烯亚胺–环糊精后,对肝细胞的毒性较雷公藤内酯醇单体显著降低(P0.05),用TGF-β1培养能富集高比例的乳腺癌干细胞;免疫磁珠分选法从中分离出肿瘤干细胞;TPL-PEI-Cy D较TPL更高效地抑制MCF-7干细胞后中CD_(44)~+CD_(24)~–细胞的比例(P0.05)。结果说明,TPL-PEI-Cy D的肝细胞毒性降低且能高效地抑制乳腺癌干细胞的性能,有望开发成为治疗乳腺癌的中药新制剂。  相似文献

雷公藤内酯醇对 PC12细胞增殖的抑制作用及机制初探   总被引:3,自引:0,他引:3  
目的:研究雷公藤内酯醇(triptolide)对PC12细胞增殖的影响及其作用的机制,为其在临床上治疗肿瘤提供实验依据.方法:利用形态学观察、四甲基偶氮唑(MTT)比色分析、流式细胞术和逆转录聚合酶链式反应(RTPCR)检测雷公藤内酯醇对体外培养的嗜铬细胞瘤细胞(PC12 cell)增殖的影响.结果:雷公藤内酯醇(5×103、25×103 g/L)与PC12细胞作用24 h、48 h或72 h均可抑制PC12细胞的增殖,并且这种抑制作用可随着雷公藤内酯醇浓度的增加而增强.但低浓度的雷公藤内酯醇(1×103g/L)对PC12细胞增殖无明显影响.5×103 g/L雷公藤内酯醇与PC12细胞作用24 h后,可使细胞周期中的G0~G1期比例增加,S期比例下降.PC12细胞与雷公藤内酯醇作用后,细胞的翻译延伸因子2A3-2的表达减弱,而且作用48 h与作用24 h相比,2A3-2的表达减弱更为明显.结论:雷公藤内酯醇可抑制PC12细胞的增殖,该抑制可能是通过改变2A3-2基因的表达从而阻止细胞的G0~G1期向S期过渡来实现的.  相似文献

自花后46d到88d果实成熟.黄皮种子的发芽率由0升至100%.而活力指数逐渐上升,到花后74d达到最大值,之后略有下降.每粒种子的呼吸强度在花后46-67d持续增加,此后则渐渐减弱,但湿藏2d后又回升.黄皮种子的发育明显超前于果实.花后74d时.每粒种子的干重已接近最大值,这时种子活力最大.而果实的鲜重虽然已接近最大值.但其干重却只有成熟时的73%。花后46-53d的种子,其发芽率小于100%,轻微脱水能提高种子的发芽率及活力指数,花后60d至果实成熟.种子发芽率均为100%.这时任何程度的脱水都会引起活力指数的下降,但不同发育时期的黄皮种子耐脱水力有差别.其中以花后67d的耐脱水力最强.花后88d果实成熟时种子的耐脱水力最弱。  相似文献

用薄层层析对三七、人参和西洋参愈伤组织进行的初步鉴定表明,三种愈伤组织都含有皂甙和主要皂甙成分Rb_1、Rg_1,三七愈伤组织还含有一种抗癌皂甙Rh_1。对愈伤组织的生长,三七低于人参高于西洋参;对愈伤组织中总皂甙含量,三七均高于人参和西洋参。三种植物细胞悬浮培养结果类似于他们的愈伤组织培养,但生长又进一步提高。三七细胞悬浮培养中皂甙产生的时间进程几乎与生长平行,合适的收获期为培养30天。寡糖素不仅增强三七培养细胞的皂甙形成而且促进细胞生长,较合适的浓度为1.25 ppm。通过以上研究,使三七悬浮培养细胞的生长(干重增加178毫克)为最初培养愈伤组织的4倍以上,总皂甙产率高达20.6毫克,为最初培养愈伤组织的8.5倍。  相似文献

A callus isolated from Jerusalem artichoke tuber was found tohave much more cell wall material and tyrosinase activity thanthe original tuber, and its dry weight was dependent on thesucrose concentration in the culture medium. All calluses anda crown gall tested contained no detectable inulin. Even thoughdifferent cultures were widely different in their dry weight,this was closely related to their total hexose content, andthe latter in turn was proportional to their free hexose content. Summary The effects of DNA hypomethylating drugs (azacytidine and ethionine) on induction of microspore-derived calluses and embryos were studied in barley (Hordeum vulgare L.) ev. Igri. The results were as follows: (1) Yield of calluses and embryos pretreated with the different concentrations of azacytidine for 3 d was several-fold higher than that of the control. The highest yield of calluses and embryos in all treatments appeared at a concentration of 3 mg l−1, which reached 11.03 per anther. It was 110-fold higher than the control. (2) There was a significant difference in yield of calluses and embryos between the different days of pretreatment. The highest yield was obtained at a 3-d pretreatment. If the period of pretreatment was shorter or longer than 3 d, yield of calluses and embryos was reduced sharply, and was similar to that of the control. (3) The data obtained with ethionine pretreatment were very similar to those obtained with azacytidine. (4) Tests on the different methods of pretreatment showed that yield of calluses and embryos pretreated with distilled H2O, mannitol, azacytidine, and ethionine was much higher than other pretreatments and the control, and reached 6.53–11.39 per anther. The yield of calluses and embryos pretreated with DNA hypomethylating drugs was higher than with mannitol. However, pretreatment with hypomethylation drugs supplemented with induction medium was not effective.  相似文献

In this study, two cultivars of Medicago sativa (cv. Yazdi and cv. Hamedani) were used for callus production. Calluses were transferred to MS medium containing 0, 30, 60, 90, and 120 mM NaCl and 0, 5, 10 mM proline. After 4–5 weeks dry weight and intracellular free proline of the calluses were measured. The growth of callus in both cultivars decreased with increasing salt concentration. Addition of exogenous proline to the culture medium increased the dry weight and free proline content of callus. The difference between control and treated calluses with 10 mM exogenous proline in the medium was significant. The data obtained from experiments indicated that the responses of two Medicago cultivars was genotype dependent.  相似文献

The seasonal variation of the periphytic community attached to an artificial substratum (glass tubes) was studied during two different periods in a lagoon connected to the Paranapanema River, the main tributary of the Jurumirim Reservoir (São Paulo–Brazil). An analysis of dry weight, ash free dry weight, chlorophyll a, phaeophytin and primary productivity of periphyton was carried out. The first experiment lasted from August to December 1993, the second from February to June 1994. Tubes were removed after 7, 14, 21, 28, 60, 90 and 119 days of incubation. In the 1st experiment, the periphytic community reached a higher biomass after the 4th week of colonization (28th day), in the 2nd experiment after the 2nd week (14th day). This discrepancy is related to seasonal differences in environmental factors (water temperature, nutrients concentrations and water discharge) that determine initial colonization. After the first stages of colonization, the biomass and primary productivity of periphyton reached their maximum values after the 60th day of incubation. In both experiments, three developmental phases could be discerned. In the initial phase, an exponential growth was observed. In the second phase, the bioderm reached its maximum biomass and productivity. In the third phase, a decrease of biomass and productivity occurred.  相似文献

Responses of sunflower tissues to NaCl stress were studied in control (C), salt-stressed (S) and salt-adapted (T) calluses in terms of proline, polyamines and ethylene content for a period of 21 days. Salt-adapted calluses showed their adaptation to salinity by growing in the medium with 175 mM NaCl, at a similar rate than C calluses on medium without salt. Proline concentration was 27 times higher in salt-adapted calluses compared to control calluses at time 0, but salt stressed calluses (S calluses) were able to increase proline by day 21, demonstrating that proline was not just an osmoregulator but might be involved in other responses in sunflower salt-stressed calluses. Putrescine (Put) was the most abundant polyamine in C calluses at time 0, while spermidine (Spd) was the main polyamine in salt tolerant (T) calluses. Ethylene increased in C calluses until day 14, decreasing thereafter. In salt-adapted calluses, ethylene increased significantly over the concentration in C and S calluses by the end of the experiment. In control calluses, the highest level of total polyamines and the lowest of ethylene was found on day 21, while T calluses synthesized the highest ethylene level and had the lower polyamines level by this time. It seems that in salt-adapted calluses ethylene was related to stress tolerance and in salt sensitive tissues (S calluses), ethylene formation was related to senescence. The present data suggests a close relationship between proline, polyamines, ethylene and salt-stress tolerance in sunflower dedifferentiated tissues.  相似文献

Body weight, body length, and other developmental parameters were studied in the European ferret, Mustela putorius, from birth to 26 weeks of age. Males and females did not differ in weight until the seventh week of live, and they did not differ in length until the ninth week of life, when the males began to grow faster. The fungus Sclerotium rolfsii is presently under development as a bioherbicide for broadleaf weed species using fungus-infested substrates as application material in this laboratory. The effect of environmental factors and three precursors (citric acid, ascorbic acid, and sodium succinate) on mycelial growth, oxalic acid production, and virulence by SC64 in liquid culture were investigated. The results showed that for mycelia growth the optimum liquid medium was Modified Richard's solution (MRS) among the five tested media, but potato dextrose broth (PDB) produced the maximum oxalic acid production and virulence on detached Solidago canadensis leaves. When PDB was used as the basic medium, the oxalic acid/mycelial dry weight (mg g–1) ratio reached the peak 4 days after inoculation. The optimum temperature for oxalic acid production was at 27°C, but increased mycelial dry weight and virulence were observed at 30°C. The optimum range of initial pH value for oxalic acid accumulation was 4.0–6.0, with the optimal pH 5.0; highest mycelial growth was with an initial pH 3.5–6.0 (optimum pH 5.0) and subsequently pH 3.5–5.5 (maximum at pH 3.5). Both mycelial dry weight and oxalic acid production showed a decreasing trend as a result of the precursor of oxalic acid being added to PDB. Among the three precursors, the greatest decrease in mycelial dry weight, and oxalic acid production was caused by sodium succinate. This clarification of optimal conditions for production of mycelial biomass while insuring high concentrations of oxalic acid and high virulence should be useful for further development of this fungus as biocontrol agent.  相似文献

Phaseolus vulgaris BBL-290 plants were grown in growth chambers in the Southeastern Plant Environment Laboratory and exposed to either single (at seedling, flower, or podfill) or multiple (biweekly or weekly) treatments of ferulic acid (FA). In the first experiment, plants were harvested one week after FA treatment (0, 1.0, 2.0 mM) and at final harvest (56 days old). FA delayed leaf expansion during the seedling and flowering stages. The total plant leaf area and the plant dry weight of plants treated with 1.0 and 2.0 mM FA as seedlings were reduced one week after treatment by 38–48%. The total plant leaf area and the plant dry weight of plants treated at flowering with 2.0 mM FA were reduced by 25% one week after treatment. Treatment with 2.0 mM FA at podfill caused the senescence and abscission of older leaves and reduced total plant leaf area, plant dry weight and mean pod dry weight by 54, 40, and 48%, respectively, one week after treatment. The plants treated at the seedling and flowering stages recovered by final harvest. In a subsequent experiment, FA (0, 0.50, 1.0, 1.5 mM) reduced total plant leaf area at the seedling and flowering stages but not at podfill. The youngest expanding leaves were most sensitive to FA at flowering. The leaf area of these leaves was reduced by 35 and 25%, one and two weeks after treatment, respectively. Their absolute growth rates were reduced from 31 to 56% one week after treatment at flowering. Their relative growth rates were reduced by 50% one week after treatment. Growth rates then recovered within two weeks after treatment. In the final experiment, biweekly exposures of FA (0.25, 0.50, 0.75, 1.0) reduced total plant leaf area but did not affect any other growth parameters. Weekly exposures of FA (0.25, 0.50, 0.75, 1.0) reduced total plant leaf area up to 34%, absolute growth rate up to 58%, leaf number up to 31% and pod number up to 58%. As the frequency of exposure to FA increased, the concentration necessary to affect bean plant growth and development decreased.  相似文献
