将ZmCI-1B全长启动子及其7个5'端缺失的启动子片段的表达载体分别转化农杆菌GV3101,经PCR鉴定正确之后,以拟南芥为遗传转化受体,利用花序侵染法将各个表达载体转化拟南芥。用PCR法检测转化后的拟南芥植株,取阳性植株的幼苗或花、角果进行GUS组织化学染色。结果表明,ZmCI-1B启动子在拟南芥中的调控特性与玉米中的不同;ZmCI-1B启动子及其7个5'端缺失启动子转基因拟南芥植株中GUS的染色部位各异,说明了不同长度的启动子功能特性不同,推测其可能与启动子上的顺式作用元件有关。  相似文献

拟南芥基因转移新方法一真空渗入法的研究   总被引:2,自引:1,他引:1  
以拟南芥(Arabidopsisthaliana)生态型Landsbengerecta为试材,在含有所构建的CaMVBari-1株系基因VI的质粒(pJO530Bari-1GVI)的根癌农杆菌(Agrobacteriumtumefaciens)菌种GV3101的介导下,研究了基因转移的新方法一真空渗入法。这种方法简便、快速、可靠且无需经过组织培养阶段即可获得大量转化植株。适宜的转化条件是将生长健壮,除去主苔后4-8d的成株连同营养钵倒置浸入被渗入培养基稀释的农杆菌孢子悬浮液,其光密度为0.8,在吸力为1.7m2/h的真空泵下断续处理2min/30s,置于24h连续光照下的气候室培养,待种子收获后在含有潮霉素的选择培养基选择转化植株.PCR分析及ELISA检测,该方法的转化效果高达0.71%。  相似文献

利用根癌农杆菌介导的遗传转化,将启动子诱捕(Promoter trapping)元件插入到棉花基因组,获得141个独立的转化子,其中97%的转化子经PCR扩增为阳性。不同组织中GUS基因的表达频率为:根部48%,茎的微管组织9.2%,叶5.2%,花51%;同时检测了不同植株中GUS基因的表达模式,发现GUS基因在不同株系的植株间的表达模式呈现较大的差异,有些植株中GUS基因是组织特异表达,有些则是器官特异表达,有些则在多个器官中均有表达。所建立的启动子诱捕系统中的GUS基因高频率、多模式和时空特异性表达为分离基因及其调节序列、开展功能基因组研究奠定了坚实的基础。  相似文献

农杆菌介导的玉米自交系愈伤组织转化条件的优化   总被引:13,自引:0,他引:13  
采用农杆菌介导法,将外源基因导入玉米骨干自交系掖515和掖502胚性愈伤组织并再生玉米植株。在农杆菌浸染玉米胚性愈伤组织过程中,采取真空渗入、部分酶解和超声波处理,可以显著提高转化率。农杆菌浸染玉米胚性愈伤组织时,在50kPa下真空渗入5min,掖515和掖502胚性愈伤组织的转化率分别从6.3%和4.5%提高到8.7%和7.8%。愈伤组织在农杆菌浸染前,以0.2%离析酶酶解10min,掖515和掖502胚性愈伤组织的转化率分别提高到8.3%和8.9%。在农杆菌浸染时,以功率100W超声波处理90-120s,掖515和掖502胚性愈伤组织的转化率最高可达9.1%和9.4%。  相似文献

本文利用发根农杆菌感染黄瓜植体得到的转化根,在不同激素配比的分分培养基上培养,进行再生植株的诱导试验,结果表明,NAA和BA的合适配比,有利于黄瓜植株的再生,转化根及其再生植株检测到冠瘿碱的存在,说明农杆菌Ri质粒的T-DNA已转移到黄瓜基因组中。  相似文献

遗传改良是人参育种的重要手段之一,而遗传转化和再生体系的建立是开展人参遗传改良工作的前提和基础。人参植株再生可以通过器官发生和体细胞胚发生,间接体细胞胚发生是人参植株再生的主要途径,从不同外植体,不同碳源,体细胞胚优化和无激素再生等方面进行了综述。在人参遗传转化方面,发根农杆菌和根癌农杆菌对人参的遗传转化均已成功,人参皂苷合成途径中的关键酶基因和抗除草剂基因也已陆续导入人参,得到了遗传改良的转化人参。发根培养系统可用于大量生产人参皂苷,讨论了rolC基因对人参发根诱导的作用,发根植株再生能力及生物反应器培养,最后指出了人参基因工程研究中存在的问题。  相似文献

采用农杆菌菌株GV3101感染子叶期苜蓿体细胞胚来研究苜蓿次级体细胞胚的遗传转化方法。农杆菌菌株GV3101双相载体pCAMBIA2301,此双相载体具有gus报告基因和nptⅡ抗卡那霉素筛选基因。感染的子叶期苜蓿体细胞在75 mg/L卡那霉素筛选压下,经过一系列诱导培养,最终获得转基因植株。然后,通过GUS组织化学定位分析来检测转基因植株不同器官中的GUS表达,并进一步通过PCR和Southern杂交确定转基因的稳定整合和转化率。结果表明转基因植株不同器官均有GUS表达,整合的nptⅡ基因的拷贝数是1~4,获得的转基因植株的转化率是65.82%。  相似文献

Pakchoi (Brassica rapa L. ssp. chinensis), a kind of Chinese cabbage, is an important vegetable in Asian countries. Agrobacterium mediated in planta vacuum infiltration transformation has been performed in pakchoi since 1998, but a detailed study on this technique was lacking. Pakchoi plants 40-50 days old with inflorescences were vacuum infiltrated with Agrobacterium tumefaciens strain C58C1 harboring the binary vector pBBBast-gus-intron. The transformation frequency in the harvested seeds mainly varied from 1 x 10(-4) to 3 x 10(-4) over several years, and it was lower than the frequency in Arabidopsis thaliana. Transformants were obtained from both the upper and the lower parts of the infiltrated plants with or without an elongated inflorescence. Stained ovules and pollen grains were found in the unopened flower 13 days post-infiltration, which was about 0.5-1 mm in diameter at infiltration time with an open ovary as revealed by paraffin sections. Histochemical assays revealed that Agrobacteria were more abundant in the flower tissue than in stem and leaf tissues at all times after infiltration despite the sharp decrease of live Agrobacteria in plant 14 days post infiltration as revealed by the colony forming units on the Agrobacteria culture medium. The results of vacuum infiltration transformation of pakchoi and Arabidopsis thaliana were compared and a strategy to optimize the transformation conditions to increase the transformation frequency in pakchoi was discussed.  相似文献

The visual marker GUS has been utilized in this study to understand the Arabidopsis thaliana vacuum infiltration transformation process by Agrobacterium tumefaciens. High transformation frequencies of up to 394 transgenic seeds per infiltrated plant were achieved. The results showed that the majority of the transgenic seeds from single infiltrated plants were from independent transformation events based on Southern analysis, progeny segregation, distribution of transgenic seeds throughout the infiltrated plants and the microscopic analysis of GUS expression in ovules of infiltrated plants. GUS expression in mature pollen and anthers was monitored daily from 0 to 12 days post-infiltration. In addition, all ovules from a single infiltrated plant were examined every other day. GUS expression frequencies of up to 1% of pollen were observed 3-5 days post-infiltration, whereas frequencies of up to 6% were detected with ovules of unopened flowers 5-11 days post-infiltration. Most importantly, transgenic seeds were obtained only from genetic crosses using infiltrated plants as the pollen recipient but not the pollen donor, demonstrating Agrobacterium transformation through the ovule pathway.  相似文献

Previously, we showed that transformation with the codA gene for choline oxidase allows plants to synthesize glycine betaine (GB) and enhances their ability to tolerate various kinds of stress during germination and vegetative growth. In this study, we examined the tolerance of transformed plants to salt stress at the reproductive stage, which is the stage at which plants are most sensitive to environmental stress. Salt-shock treatment of wild-type plants for 3 days resulted in the abortion of flower buds and decreased the number of seeds per silique. These deleterious effects were clearly visible 6 days after the termination of salt-shock treatment. Microscopic examination of floral structures revealed that salt stress inhibited the development of anthers, pistils, and petals. In particular, the production of pollen grains and ovules was dramatically inhibited. These effects of salt stress were significantly reduced by transformation with the codA gene, and our observations suggested that the enhanced tolerance of the transgenic plants was a result of the accumulation of GB in the reproductive organs. Indeed, levels of GB in flowers, siliques, and inflorescence apices were about five times higher than in leaves.  相似文献

The Caricaceae is a small family of tropical trees and herbs in which most species are dioecious. In the present study, we extend our previous work on dioecy in the Caricaceae, characterising the morphological variation in sexual expression in flowers of the dioecious tree Jacaratia mexicana . We found that, in J. mexicana , female plants produce only pistillate flowers, while male plants are sexually variable and can bear three different types of flowers: staminate, pistillate and perfect. To characterise the distinct types of flowers, we measured 26 morphological variables. Our results indicate that: (i) pistillate flowers from male trees carry healthy-looking ovules and are morphologically similar, although smaller than, pistillate flowers on female plants; (ii) staminate flowers have a rudimentary, non-functional pistil and are the only flowers capable of producing nectar; and (iii) perfect flowers produce healthy-looking ovules and pollen, but have smaller ovaries than pistillate flowers and fewer anthers than staminate flowers, and do not produce nectar. The restriction of sexual variation to male trees is consistent with the evolutionary path of dioecy from hermaphrodite ancestors through the initial invasion of male-sterile plants and a subsequent gradual reduction in female fertility in cosexual individuals (gynodioecy pathway), but further work is needed to confirm this hypothesis.  相似文献

In this paper we assess the relative contributions to total pod yield of cohorts of flowers of known age. Bean plant (Phaseolus vulgaris L.) growth was monitored and 'births' of individual flowers were recorded and their fates followed. For every cohort of flowers an array of yield measures was calculated, including the mean and total cohort dry weight of fruits, and the numbers of viable and aborted seeds and completely undeveloped ovules. When mature, pods were removed from half of the plants. Plants whose mature pods were removed produced significantly greater numbers of flowers, ovules and viable seeds and a greater total weight of pods than did untreated plants. However, the proportion of ovules giving rise to viable seeds and to aborted seeds and undeveloped ovules was the same in plants of both groups. Approximately 50% of ovules of marketable pods, in all plants, yielded viable seeds. The 12 cohorts of flowers contributed markedly different amounts to plant yield. Early and later cohorts contributed much less than did middle cohorts in terms of marketable pod dry weight and numbers of fully developed seeds, aborted seeds, and undeveloped ovules. Peak productivity was therefore obtained from the middle phase of flowering in these annual bean plants. The lower pod yield of later cohorts is interpreted as a result of competition for limited resources between maturing pods and new flowers. We consider possible causes for the various fates of flowers and ovules and discuss flowers as plant modules suitable for demographic examination.  相似文献

种子的选择性败育在被子植物中普遍存在,开展结籽格局及其影响因素的研究有助于深入了解种子的形成机制及多样化的生殖对策。以刺叶锦鸡儿Caragana acanthophylla为材料,通过传粉过程、胚珠发育动态和资源分配状况的研究,以探讨种子的选择性败育格局及相关影响因素。结果显示:(1)刺叶锦鸡儿具高度自交不亲和性,为泛化的传粉系统,蜂类是主要的传粉者。自然状态的结实率为(86.00±4.96)%,不存在传粉限制,但受精具明显的时间效应。(2)刺叶锦鸡儿单花期4—5d,每朵花有(14.00±0.14)粒胚珠。胚珠的发育从荚果顶部开始,其中,开花后第3天荚果顶部胚珠开始膨大;第11天绝大多数胚珠出现了膨大,此时,在荚果基部的胚珠开始败育,随后荚果顶端受精后的胚珠也出现败育,最终仅荚果中部形成2—3粒种子。其结籽格局为成熟荚果中部胚珠,而败育荚果顶部和基部胚珠的选择性败育类型。(3)受微地形影响,居群内开花植株具斑块分布,对较少开花植株通过添加水肥进行资源调控后,结籽率显著提高,说明在种子形成过程中存在资源限制。综上所述,受精顺序和资源限制两个互逆的资源梯度决定了刺叶锦鸡儿的结籽格局。其中,...  相似文献

In angiosperms, ovules are "packaged" within individual flowers, and an optimal strategy should occur depending on pollination and resource conditions. In animal-pollinated species, wide variation in ovule number per flower occurs, and this contrasts with wind-pollinated plants, where most species possess uniovulate flowers. This pattern is usually explained as an adaptive response to low pollen receipt in wind-pollinated species. Here, we develop a phenotypic model for the evolution of ovule number per flower that incorporates the aerodynamics of pollen capture and a fixed resource pool for provisioning of flowers, ovules, and seeds. Our results challenge the prevailing explanation for the association between uniovulate flowers and wind pollination. We demonstrate that when flowers are small and inexpensive, as they are in wind-pollinated species, ovule number should be minimized and lower than the average number of pollen tubes per style, even under stochastic pollination and fertilization regimes. The model predicts that plants benefit from producing many small inexpensive flowers, even though some flowers capture too few pollen grains to fertilize their ovules. Wind-pollinated plants with numerous flowers distributed throughout the inflorescence, each with a single ovule or a few ovules, sample more of the airstream, and this should maximize pollen capture and seed production.  相似文献

Twenty plants with various phenotypic abnormalities to the flowerswere selected from very large populations of Thryptomene calycinain the Grampian and Black Ranges. Most of these had impairedreproductive function. Normal flowers were epigynous with fivesepals, five petals, five anthers, a single style and two anatropousovules.
