对利用斜面法和橡皮塞法保藏某些丝状真菌的效果进行了试验,共保藏丝状真菌29属,69种,128株。保藏时间2-17a,通过斜面转接,其存活率达89.8%。存活的菌株仍维持其原有特性。试验结果表明:利用斜面法与橡皮塞法保藏某些丝状真菌是简便可行的。  相似文献

野生鸡枞菌种质资源的有效保存是对野生鸡枞加以保护和利用的前提。以自行分离的5个野生鸡枞菌株作为研究对象,采用蒸馏水保藏法和-80°C冻结保藏法对野生鸡枞菌种长期保存的方法进行了实验研究,蒸馏水法分别保存于室温和4°C,-80°C冻结保藏同时采用程控降温法和泡沫盒降温法,保存20个月后对4种不同方法保存的5个菌株的保存效果进行比较。实验结果表明:蒸馏水室温保存法菌种存活率为100%,萌发期较短,为4-10 d,是一种简便、实用、有效而成本低廉的长期保存方法;-80°C冻结保藏法的存活率为56%-76%,萌发期7-16 d,泡沫盒降温法可以很好地控制降温速度,是一种简便有效的控温方法。  相似文献

定期转种法和低温冷冻保存法是临床实验室最常用的两种真菌保存方法,为比较两种方法保藏致病真菌活性的能力,本研究使用两种保藏方法对实验室689株致病真菌保藏5年后进行检测。定期转种法是将菌落接种于马铃薯斜面培养基并将其储存在4℃冰箱,每6个月转种1次。低温冷冻法是挑取马铃薯斜面培养基上生长良好的菌落于无菌10%甘油中,放置在-80℃储存。保藏5年后,将两种方法保藏的菌株转种复苏,比较菌株的复活率。对于念珠菌属Candida、新生隐球菌Cryptococcus neoformans、毛癣菌属Trichophyton、曲霉属Aspergillus和孢子丝菌属Sporothirix真菌,两种方法的菌株复活率无统计学差异;对于小孢子菌属Microsporum真菌和马尔尼菲蓝状菌Talaromyces marneffei,使用低温冷冻法保藏的菌株复活率高于定期转种法保藏的菌株复活率;对于着色霉属Fonsecaea真菌,低温冷冻法保藏的菌株复活率低于定期转种法保藏的菌株复活率。因此,我们认为对于常见致病真菌的长期保藏,使用10%甘油作为保护剂的低温冷冻法优于定期转种法,但其不适用于着色霉属Fonsecaea真菌的长期保藏。  相似文献

研究不同保藏介质、温度和时间的综合因素对B-298菌株活性的影响。为生产和研究提供能保持该菌株抑菌活性的适宜保藏方法。试验结果表明:介质、温度、时间的交互作用对菌株细胞存活率和代谢产物拮抗效应的影响都存在显著性差异;细胞存活率和拮抗效应与保藏时间呈反比;菌株保藏680 d,应用甘油超低温保藏法,菌株细胞存活率和抑菌活性最高,而采用斜面常温保藏,细胞存活率和抑菌活性最低。  相似文献

以芽孢杆菌(Bacillus)为材料,用冷冻真空干燥法和砂管法,在4℃条件下进行了六年的菌种保藏对比试验.结果表明,冷冻真空干燥法比砂管法保藏的菌种存活率高,存活率下降速率小;建立了菌种保藏过程中菌种相对存活率随时间变化规律的数学模型L′_f(t)=e~(-0.1323t)和L′s(t)=e~(-0.2900t).该模型的模拟结果能与试验结果较好地吻合.在室温条件下,砂管法的菌种存活率下降速率比4℃条件下大,三个月时存活率为零或接近零;而冷冻真空干燥法保藏菌种存活率与4℃的效果接近.因此冷冻真空干燥法是芽孢杆菌的比较有效的保藏方法.  相似文献

考察保护剂、保藏温度及预冷冻方法对Schizochytrium sp.HX-308菌种存活率及发酵性能保持的影响。结果显示:在-80℃低温保藏6个月后,渗透性保护剂的细胞存活率均比非渗透性保护剂高了5%,其中用60%(质量分数)海藻糖的保护剂最终的株细胞存活率达到80.02%,明显优于其他保护剂。采用液氮-196℃保藏菌种(两步预冷冻法、60%海藻糖保护剂),存储6个月后存活率高达90.70%,生物量、油产量和二十二碳六烯酸(DHA)产量分别达到了61.65、26.41和11.10 g/L,为最优的保藏方法,为裂殖壶菌的实验室研究及工业化生产提供了一种长期安全的保藏法。  相似文献

以高产虾青素的雨生红球藻(Haematococcus pluvialis Flotow)FACHB-712藻株为材料,研究2种细胞形态(营养细胞和厚壁孢子)在低温保藏下的复苏率及其差异原因。结果显示,采用两步法(先预冻降温后再投入液氮中)冻存其营养细胞,在不同冻存条件下,其存活率均低于5%,以10%甘油作为保护剂、冻存速率为0.5℃/min、预冻温度为-40℃、保留30 min,然后再投入液氮罐(-196℃)中保藏,其存活率可达到13.3%。采用两步法冻存厚壁孢子,其复苏存活率高达66.13%,复苏萌发后细胞的生长特性、虾青素含量与液氮保藏前无明显差异(P > 0.05)。对液氮保藏前后藻细胞形态和超微结构观察结果表明,超低温保藏后,营养细胞的结构受到较大损伤,而厚壁孢子受到的损伤相对较小。当添加不同保护剂后,直接将厚壁孢子分别冻存在-20℃、-80℃低温及液氮中,发现-80℃低温冻存处理组的复苏存活率相对较高,可达27%。研究表明采用两步法先预冻降温后再投入液氮中冻存厚壁孢子,是长期保藏雨生红球藻FACHB-712的最佳方法,也可采用一步法将厚壁孢子冻存于-80℃冰箱中。  相似文献

我们测验了在室温(波动为5—36℃)贮藏4年半一8年,用脱脂牛奶作保护剂,真空冷冻干燥保存的559种20lo株细菌、放线菌、酵母菌、丝状真菌的存活力,以及其中部分菌株的主要生理特性。贮存在室温的真空冷冻干燥法保存的84种484株细菌,180种565株放线菌,59种459株酵母菌和236种502株丝状真菌的存活力分别是467(96·5%)、516(91.33%)、394(85.83%)和439(87·45%)。我们还观测了贮藏在5℃,1957年真空冷冻干燥法保存的细菌。在这种条件下保存16年的32种126株细菌,103株(81.74%)保持着活力,除个别菌株外,没有任何种的死亡。所测定的17株真空冷冻干燥保存的菌种的生理特性,与定期移植保存的同一菌株对比结果,除两株大肠杆菌,其中l株谷氨酸脱羧酶活力减退外,其他菌株都没有明显的变化。  相似文献

花蜜富含多种有机物和微量元素,不同植物花蜜的分泌量以及化学物质构成存在很大差异,不同花蜜采集和保存方法都会影响其成分分析结果。本研究参考相关文献,总结了常用的6种野外花蜜收集方式(注射器法、毛细管法、滤纸法、溶液稀释法、离心法和抽吸法),6种花蜜保存方式(4℃冷藏保存、-20℃冷冻保存、-80℃超低温保存、-196℃液氮保存、添加防腐剂保存和滤纸保存)以及3类花蜜化学特征检测方式(手持折光仪、比色法和色谱分析)。深入比较了各种方法的优缺点及其潜在的适用对象,在条件允许的情况下,毛细管法、-80℃超低温保存及色谱分析是花蜜收集、保存及分析最常使用的方法。  相似文献

丝状真菌不仅是致病菌,而且在异源表达工业酶、化学制品以及药物活性物质中发挥着越来越重要的作用。随着人类基因组计划的实施和推进,生命科学研究已进入了功能基因组时代,特别是蛋白质组学,在蛋白质水平对丝状真菌细胞生命过程中蛋白质功能和蛋白质之间的相互作用以及特殊条件下的变化机制进行研究,对生命的复杂活动进行深入而又全面的认识也为丝状真菌工业酶制剂和重组药物的开发提供广阔的创新空间。本文综述了蛋白质组学的研究内容和方法,总结了其在丝状真菌致病菌、抗生素产生菌和纤维素酶产生菌中的应用现状。不同层次的功能基因组学分析可以从各个角度掌握生物体的代谢网络和调控机制,本文还对蛋白质组学以及功能基因组学各部分内容的整合运用进行了展望。  相似文献

虫霉目真菌的活力对超低温储存较为敏感。储存法在大范围应用前,需对储存效果进行详细评估。将蚜科专化菌努利虫疠霉以初级分生孢子形式(2-3′105个孢子/mL)在-80℃超低温存储12个月。结果显示日常用于培养该真菌的含0.1%乳化芝麻油的萨氏培养基作为超低温保护基质能有效地储存努利虫疠霉孢子,比常见的冷冻保护剂如二甲亚砜和甘油的效果好。孢子悬液经解冻和培养后可获得最多的生物量,而且菌种保持了较高的生长速率。更重要的是,萨氏培养基的主要成分4%葡萄糖、1%蛋白胨和1%酵母粉在低温存储过程中发挥了协同作用,能保  相似文献

Lyophilization, desiccation on pumice stone and storage in dilute glycerol at-20C were compared for the preservation of Lactobacillus cultures. After storage for two years, 73% of the lyophilized cultures were viable. Desiccation on pumice stone gave erratic results and is not recommended. Storage in 20% (v/v) glycerol at-20C is simple and convenient and after 24 months, 75% of the cultures were viable. It is a useful method of maintaining routinely accessed cultures.  相似文献

The galactomannans (GMs) that are produced by filamentous fungi belonging to Pezizomycotina, many of which are pathogenic for animals and plants, are polysaccharides consisting of α-(1→2)-/α-(1→6)-mannosyl and β-(1→5)-/β-(1→6)-galactofuranosyl residues. GMs are located at the outermost layer of the cell wall. When a pathogenic fungus infects a host, its cell surface must be in contact with the host. The GMs on the cell surface may be involved in the infection mechanism of a pathogenic fungus or the defense mechanism of a host. There are two types of GMs in filamentous fungi, fungal-type galactomannans and O-mannose type galactomannans. Recent biochemical and genetic advances have facilitated a better understanding of the biosynthesis of both types. This review summarizes our current information on their biosynthesis.  相似文献

We have evaluated the survival and potential morphological alterations of 45 species of pathogenic filamentous fungi that had been stored in sterile water following Castellani’s method in the National Collection of Pathogenic Fungi (NCPF). Storage duration varied from 2 months to over 21 years. Ninety percent of stored organisms were shown to be viable. Viability was largely independent of the duration of storage, but did apparently vary to some degree in an organism-specific manner. In addition, certain fungi were shown to have undergone morphological alterations during storage, and exhibited significant degrees of pleomorphism upon revival. This was especially marked for several isolates of dermatophytes, where storage resulted in loss of recognisable colonial features, and overproduction of sterile mycelium with aberrant or no conidia. These findings suggest that while Castellani’s method remains an easy and inexpensive method for long-term preservation of most fungi, water storage should be supplemented by a second storage method to increase the chances of retaining both viability and morphological stability over long periods.  相似文献

We have studied for the first time the taxonomic composition of filamentous fungi associated with the bivalve mollusks Crenomytilus grayanus (Bunker, 1853) and Modiolus modiolus (Linnaeus, 1758), collected in polluted biotopes of Ussuriiskii Bay (Peter the Great Bay, Sea of Japan). One hundred fifty-five strains of higher fungi were isolated, and 35 species of filamentous fungi were identified. Out of these, 28 species were anamorphous fungi of the genera Aspergillus (12), Penicillium (10), Acremonium (1), Alternaria (1), Aureobasidium (1), and Cladosporium (3) (Deuteromycota), and 7 were species of the genera Chaetomium (6) and Myxotrichum (1) (Ascomycota). Twenty-four species of filamentous fungi were isolated from the internal organs of C. grayanus: 8 from the muscles, 8 from the hepatopancreas, 6 from the female gonads, 4 from the mantle, 3 from the kidneys and male gonads, and 1 from the gills. Twenty-one species of filamentous fungi were isolated from the internal organs of Modiolus modiolus: 10 from the muscles, 6 from the hepatopancreas, 5 from the female gonads, 3 from the kidneys, 3 from the mantle, and 1 from the gills. The bulk of the isolated species of the genera Aspergillus, Penicillium, Cladosporium, and Chaetomium were pathogenic and toxigenic fungi.  相似文献

Secretome represents a main target for understanding the mechanisms of fungal adaptation. In the present study, we focus on the secretomes of fungi associated with infections in humans and other mammals in order to explore relationships between the diverse morphological and phylogenetic groups. Almost all the mammalian pathogenic fungi analyzed have secretome sizes smaller than 1000 proteins and, secreted proteins comprise between 5% and 10% of the total proteome. As expected, the correlation pattern between the secretome size and the total proteome was similar to that described in previous secretome studies of fungi. With regard to the morphological groups, minimum secretome sizes of less than 250 secreted proteins and low values for the fraction of secreted proteins are shown in mammalian pathogenic fungi with reduced proteomes such as microsporidia, atypical fungi and some species of yeasts and yeast-like fungi (Malassezia). On the other hand, filamentous fungi have significantly more secreted proteins and the highest numbers are present in species of filamentous fungi that also are plant or insect pathogens (Fusarium verticilloides, Fusarium oxysporum and Basidiobolus meristosporus). With respect to phylogeny, there are also variations in secretome size across fungal subphyla: Microsporidia, Taphrinomycotina, Ustilagomycotina and Saccharomycotina contain small secretomes; whereas larger secretomes are found in Agaricomycotina, Pezizomycotina, Mucoromycotina and Entomophthoromycotina. Finally, principal component analysis (PCA) was conducted on the complete secretomes. The PCA results revealed that, in general, secretomes of fungi belonging to the same morphological group or subphyla cluster together. In conclusion, our results point out that in medically important fungi there is a relationship between the secretome and the morphological group or phylogenetic classification.  相似文献

[目的] 分泌蛋白在植物病原真菌致病过程中发挥着重要的作用,前人多以单种菌株分泌蛋白预测分析,尚未见多种类型真菌中分泌蛋白的预测及对比研究报道。[方法] 本研究基于多种不同营养方式真菌的全基因组序列,根据分泌蛋白所具有的基本特征,采用在线分析程序(主要包括SignalP v5.0、ProtComp v9.0等)对模式生物、死体营养型真菌、半活体营养型真菌及活体营养型真菌共19种菌株的分泌蛋白开展预测及功能分析。[结果] 在上述真菌约13万条蛋白序列中,分泌蛋白所占比例为0.74%-4.83%,其中,活体营养型真菌所具有的分泌蛋白数量占比最高,平均为3.51%,而死体营养型真菌和模式生物平均比例最低,平均为1.36%。同时,活体营养型真菌具有的功能种类最多,为433种,其次为半活体营养型真菌,为266种,而模式生物具有的功能种类最少,为100种,其中,功能为假设蛋白和非特征蛋白的蛋白数量最多。[结论] 该研究为深入解析分泌蛋白在实现不同营养方式真菌的致病机制方面提供理论依据。  相似文献
