以酪蛋白和明胶为蛋白源,豆油为脂肪源配制团头鲂幼鱼基础饲料,制成维生素C水平为0.2、33.4、65.8、133.7、251.5和501.5 mg/kg 的6种等氮等能的试验饲料。对均重为(6.400.05)g的团头鲂幼鱼进行为期90d的饲养试验,以血清相关免疫指标、肝脏抗氧化指标、三种HSPs mRNA表达以及抗病原菌能力等指标为依据,研究Vc 对团头鲂幼鱼免疫力的影响。试验结果表明,与对照组相比,501.5 mg/kg Vc 试验组能显著提高血清补体3(C3)的浓度,133.7 mg/kg Vc 试验组能显著提高补体4(C4)的浓度;65.8、133.7和 251.5 mg/kg Vc 试验组能显著提高肝脏超氧化物歧化酶(SOD)的活性,各Vc 试验组均能显著提高肝脏抗超氧阴离子(ASAFR)的活性;133.7和251.7 mg/kg Vc 试验组能显著提高肝脏HSP60基因表达水平,251.5 mg/kg Vc 试验组能显著提高肝脏HSP70和HSP90基因表达水平(P0.05);各Vc 试验组鱼的成活率在感染嗜水气单胞菌后12h、24h均显著高于对照组(P0.05),其中以251.5 mg/kg Vc 试验组效果最佳。在日粮中添加Vc 对鱼体血清碱性磷酸酶(ALP)、总蛋白(TP)、白蛋白(Alb)、皮质醇(COR)以及肝脏丙二醛(MDA)的水平无显著影响(P0.05)。综上所述,在试验条件下,Vc 作为免疫刺激剂,其水平为133.7251.5 mg/kg时能有效地增强团头鲂幼鱼的免疫力。    相似文献

实验采用2×2双因子设计, 研究两种温度下在饲料中添加大黄素对团头鲂(Megalobrama amblycephala)生长、血液生理反应及肝脏HSP70 mRNA表达的影响。选取300尾健康的团头鲂初均重(7.29±0.01) g, 两种温度下(25℃、30℃)分别投喂不添加和添加25 mg/kg的大黄素配合饲料, 养殖时间为77d。结果显示: 在长期30℃高温下, 鱼体增重率(WGR)和特定生长率(SGR)、红细胞积压(HCT)、乳酸脱氢酶(LDH)显著升高, 饵料系数(FCR)和脏体比(VSI)、谷草转氨酶(AST)、总胆汁酸(TBA)显著下降(P0.05); 添加25 mg/kg大黄素, SGR和肝体比(HSI)、红细胞积压(HCT)、乳酸脱氢酶(LDH)显著升高, 饵料系数(FCR)和摄食率(FR)显著下降(P0.05)。温度和大黄素对谷丙转氨酶(ALT)和血糖(GLU)无显著影响(P0.05), 对尿素(UREA)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、补体3 (C3)、补体4 (C4)和肝脏HSP70 mRNA的相对含量存在显著交互作用(P0.05)。与常温对照组相比, 高温对照组UREA、TG、TC显著下降; 在高温下, 添加大黄素组UREA、TG、TC有升高趋势(P0.05), C3、C4和肝脏HSP70 mRNA的相对含量显著升高(P0.05)。以上结果表明, 在饲料中添加25 mg/kg大黄素有助于缓解长期高温对团头鲂血液生理的影响, 提高补体水平和肝脏HSP70 mRNA的相对含量, 促进鱼体的生长。    相似文献

Two complementary deoxyribonucleic acid (cDNA) clones encoding heat shock cognate 70 (HSC70) and inducible heat shock protein 70 (HSP70) were isolated from the liver of Wuchang bream (Megalobrama amblycephala Y.) using RT-PCR and rapid amplification of cDNA ends (RACE). They were named Ma-HSC70 and Ma-HSP70, respectively. The cDNAs were 2336 and 2224 bp in length [not including poly (A)] and contained 1950 and 1932 bp open reading frames (ORFs), respectively. The ORFs encoded proteins of 649 and 643 amino acids with predicted molecular weights of 71.24 and 70.52 kDa, and theoretical isoelectric points of 5.25 and 5.30, respectively. Genomic DNA structure analysis revealed that Ma-HSC70 gene contained seven introns with all introns conforming to the GT/AG rule whereas Ma-HSP70 gene did not contain any intron in the coding region. Amino acid sequence analysis indicated that both Ma-HSC70 and Ma-HSP70 contained three signature sequences of HSP70 family, two partial overlapping bipartite nuclear localization signal sequences (NLS) and cytoplasmic characteristic motif (EEVD). Homology analysis revealed that Ma-HSC70 shared more than 93.0% identity with the known HSC70s of other vertebrates, while Ma-HSP70 shared more than 85.0% identity with the known HSP70s of other vertebrates, and Ma-HSC70 and Ma-HSP70 shared 86.5% identity. Bioinformatics analysis indicated that the proteins encoded by Ma-HSC70 and Ma-HSP70 genes were hydrophilic, rich in B cells antigenic sites, without any signal peptide or transmembrane region. The two proteins also contained many protein kinase C phosphorylation sites, N-myristoylation sites, casein kinase II phosphorylation sites, and N-glycosylation sites, predicting that they could play essential roles in protein folding, translocation, intracellular localization, signal transduction and regulation. The predominant secondary structures of the two proteins were α-helix and random coil. Fluorescent real-time quantitative RT-PCR was used to study the effects of heat shock (34 °C), crowding stress (100 g L?1) and challenge with bacteria Aeromonas hydrophila on the mRNA expression of the two HSP70s in Wuchang bream liver. The results indicated that, during 24 h stress, Ma-HSC70 mRNA expression decreased at first and then rose to the level before stress under heat shock and crowding stress, but Ma-HSP70 mRNA expression increased at first and then decreased under heat stress, and appeared to increase continuously under crowding stress. After bacterial challenge, the mRNA levels of both Ma-HSC70 and Ma-HSP70 increased at first and then decreased. The cloning and expression analysis of the two HSP70s provide theoretical basis to further study the mechanism of anti-adverseness and expression characteristics under stress conditions of Wuchang bream.  相似文献

The aim of the present study was to investigate the effect of selenium (Se), polysaccharide of Atractylodes macrocephala Koidz. (PAMK), and the combination of Se and PAMK on the immune response, heat shock protein (HSP) levels under heat stress (HS) condition in chicken spleen. Two hundred chickens were randomly divided into two groups, the HS group and the control (Con) group. Then these chickens were treated with Se (0.3 mg/kg), PAMK (200 mg/kg) alone, and the combination of Se (0.3 mg/kg) and PAMK (200 mg/kg). The antioxidative enzymes, cytokines contents, and expression levels of HSP27 and HSP70 were examined in chicken spleen. The results indicated that HS induced higher levels of TNF-α, IL-4, HSP27, HSP70, and MDA levels but lower level of IFN-γ, IL-2, Gpx, and SOD in spleen (P?<?0.05). These responses were ameliorated by the treatment of Se, PAMK alone, and the combination of Se and PAMK (P?<?0.05 or not) The results showed that under common condition, Se and PAMK could improve the immune response by enhancing the levels of some cytokines to proper levels; however, under HS condition, Se and PAMK could change the abnormal levels of cytokines and oxidative damages to ameliorate the injury induced by HS. In addition, there existed synergistic effect on the modulation of these biomarkers in chicken spleen between Se and PAMK. So both Se and PAMK play important roles in regulating the immune function in chicken. Considering the synergistic effect on immune regulation of PAMK, this herb deserves further investigation to evaluate its role in improving the effect of traditional immune regulators.  相似文献

目的:探讨多囊卵巢综合征(PCOS)患者血清热休克蛋白70(HSP70)、缺血修饰白蛋白(IMA)水平的变化及与性激素指标和氧化应激指标的相关性。方法:选择2017年6月~2018年6月华中科技大学同济医学院附属同济医院收治的未经治疗的初诊PCOS患者87例为PCOS组,另随机选择同期在本院进行体检的健康育龄妇女87例为对照组,比较两组血清HSP70、IMA、性激素指标、氧化应激指标,分析血清HSP70、IMA水平与性激素、氧化应激的相关性。结果:PCOS组患者血清卵泡刺激素(FSH)、促黄体生成素(LH)、睾酮(T)、8-异前列腺素F2α(8-isoPGF2α)、丙二醛(MDA)、HSP70、IMA水平显著高于对照组,差异有统计学意义(P<0.05);PCOS组患者血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化酶(GSH-Px)水平低于对照组,差异有统计学意义(P<0.05);PCOS组血清雌二醇(E2)、泌乳素(PRL)水平与对照组比较差异无统计学意义(P>0.05)。PCOS患者血清HSP70、IMA水平与血清FSH、LH、T、8-iso PGF2α、MDA水平呈正相关关系,与SOD、GSH-Px水平呈负相关关系(P<0.05),与E2、PRL水平无显著相关性(P>0.05)。结论:血清HSP70、IMA水平在PCOS患者呈现异常升高,其水平改变与性激素及氧化应激水平密切相关,提示HSP70、IMA可能参与了PCOS的病理过程,临床检测HSP70、IMA的水平变化有可能为PCOS诊疗提供有效的预测作用。  相似文献

Heat shock proteins (HSPs) expression is commonly used as indicators of cellular stress in animals. However, very little is known about either the expression patterns of HSPs or their role in the stress-tolerance phenomenon in early life stages of fish. To this end, we examined the impact of food-deprivation (12 h), reduced oxygen levels (3.5 mg/L for 1 h) and heat shock (HS: + 5 °C for 1 h) on HSP70 and HSP90 protein expression in early life stages of the gilthead sea bream (Sparus aurata), a warm-water aquaculture species. Also, we investigated HSP70 and HSP90 response to food-deprivation (7 days) in early life stages of rainbow trout (Oncorhynchus mykiss), a cool-water aquaculture species, and the tolerance of this larvae to heat shock (either + 5 or + 10 °C for 1 h). Our results clearly demonstrate that food-deprivation enhances HSP70 and HSP90 protein expression in larvae of both species. In gilthead sea bream larvae, the stressors-induced HSP70 and HSP90 (only in the reduced oxygen group) protein expression returned to unstressed levels after 24 h recovery. In fed trout larvae, a + 5 °C heat shock did not elevate HSP70 and HSP90 expression, whereas 100% mortality was evident with a + 10 °C HS. However, food-deprived trout larvae, which had higher HSP70 and HSP90 protein content, survived HS and showed HS-dependent increases in HSP70, but not HSP90 expression. Overall, HSP70 and HSP90 protein expression in early life stages of fish have the potential to be used as markers of nutritional stress, while elevation of the tissue HSPs content may be used as a means to increase stress tolerance during larval rearing.  相似文献

The DNA fragmentation and failure in post-meiotic maturation of the spermatozoa because of testosterone withdrawal can affect the fertilization potential in varicocele (VCL) patients. To find out the exact mechanism of VCL-induced failure in histone-protamine replacement process and DNA fragmentation, the correlations between the levels of expression of HSP70-2a, HSP90, PCNA, TP1/2 and PCNA genes and the patterns of DNA methylation were investigated before and after testosterone administration in rats. In total, 40 mature male Wistar rats (10 in each group) were assigned between control (with no intervention), control-sham (undergone a simple laparotomy), VCL-induced (VCL-sole), and testosterone-treated VCL-induced (VCLT) groups. The HSP70-2a, HSP90, PCNA, TP1, and TP2 genes expressions and the patterns of global DNA methylation were determined in all groups. A statistically significant (p < 0.05) reduction were found in the HSP70-2a, HSP90, PCNA, TP1 and TP2 genes expressions in VCL-sole group. In VCLT group, testosterone was shown to significantly (p < 0.05) up-regulate the HSP70-2a, HSP90, PCNA, and TP2expression levels, but TP1 expression has not been changed. Furthermore, the VCLT group exhibited higher DNA methylation rates compared to VCL-sole animals. In conclusion, testosterone, by up-regulating the HSP70-2a and HSP90 expressions and maintaining the pre-existing HSP70-2a and HSP90 proteins levels, may be the reason for the significant increment in TP2 expression during post-meiotic stage and can boost the global methylation rates of DNA via up-regulating the PCNA expression, suggesting that administration of testosterone can mitigate the VCL-impaired histone-protamine replacement and DNA methylation rates and protect the cellular DNA content from VCL-induced oxidative stress.  相似文献

在水温27℃条件下, 分别对吉富罗非鱼(27.642.79) g投喂3组不同蛋白水平的饲料, 养殖周期56d。饲养结束后, 进行14℃低温应激实验, 研究低温应激后0-24h内, 鱼体血清生化指标与肝脏HSP70 mRNA表达量的变化。结果表明, 短期投喂50%蛋白水平的饲料可以提高罗非鱼血清中葡萄糖、总蛋白、甘油三酯与胆固醇水平以及血清谷丙转氨酶与谷草转氨酶活力。在低温应激后, 50%蛋白组血清葡萄糖、总蛋白、甘油三酯与胆固醇水平呈下降趋势, 而血清皮质醇水平与谷草转氨酶活力表现为先上升后下降变化。25%和38%蛋白组血清葡萄糖水平与谷丙转氨酶活力在应激期间呈上升趋势, 而谷草转氨酶与碱性磷酸酶活力和总蛋白与胆固醇水平呈先上升后下降变化。各实验组溶菌酶活力与肝脏HSP70 mRNA的表达量呈先上升后下降的变化。由此可知, 短期投喂高蛋白饲料可以提高罗非鱼血液中蛋白、血糖与脂肪含量, 增强鱼体抗应激能力, 同时, 高蛋白水平的饲料也会引起肝脏产生分解压力以及增加饲料成本。在生产中, 需要根据实际需要合理确定饲料的蛋白含量。    相似文献

目的:研究清胰利胆颗粒对重症急性胰腺炎患者血清高迁移率族蛋白1(HMGB1)、热休克蛋白70(HSP70)、热休克蛋白27(HSP27)、白介素-8(IL-8)水平的影响。方法:选取2015年8月至2016年7月我院收治的84例重症急性胰腺炎患者,根据随机数字法分为观察组和对照组,42例每组。对照组采取常规方案完成治疗,观察组在此基础上使用清胰利胆颗粒治疗。比较两组患者临床疗效,血淀粉酶恢复至正常时间、白细胞恢复至正常时间、胃肠功能恢复至正常时间、腹痛缓解时间及血清HMGB1、HSP70、HSP72、IL-8水平。结果:治疗后,观察组临床总有效率显著高于对照组[92.86%(39/42)比71.43%(30/42)](P0.05)。观察组的血淀粉酶恢复至正常时间、白细胞恢复至正常时间、胃肠功能恢复至正常时间及腹痛缓解时间显著短于对照组(P0.05)。治疗前,两组患者HMGB1、HSP70、HSP72、IL-8水平比较无显著差异(P0.05),治疗后,两组患者HMGB1、HSP70、HSP72、IL-8水平和治疗前相比显著下降(P0.05),和对照组相比,观察组的HMGB1、HSP70、HSP72、IL-8水平较低(P0.05)。结论:清胰利胆颗粒能有效降低重症急性胰腺炎患者血清HMGB1、HSP70、HSP27、IL-8水平,且可提高临床疗效。  相似文献

The expression patterns of different genes encoding antioxidant enzymes and heat shock proteins were investigated, in present study, by real-time quantitative PCR in the hepatopancreas of abalone Haliotis discus hannai fed with different levels of dietary zinc (6.69, 33.8, 710.6 and 3462.5 mg/kg) for 20 weeks. The antioxidant enzymes include Cu/Zn-superoxide dismutase (Cu/Zn-SOD), Mn-superoxide dismutase (Mn-SOD), catalase (CAT), mu-glutathione-s-transferase (mu-GST) and thioredoxin peroxidase (TPx). The results showed that the mRNA expression of these antioxidant enzymes increased and reached the maximum at the dietary zinc level of 33.8 mg/kg, and then dropped progressively. Expression levels of the heat shock proteins (HSP26, HSP70 and HSP90) firstly increased at 33.8 mg/kg dietary Zn level, and reached to the maximum at 710.6 mg/kg, then dropped at 3462.5 mg/kg (p<0.05). Excessive dietary Zn (710.6 and 3462.5 mg/kg) significantly increases the Zn content and significantly decreases the total antioxidant capacity (T-AOC) in hepatopancreas (p<0.05). These findings showed that dietary Zn (33.8 mg/kg) could highly trigger the expression levels of antioxidant enzymes and heat shock proteins, but excessive dietary Zn (710.6 and 3462.5 mg/kg) induces a high oxidative stress in abalone.  相似文献

目的:探讨热应激预处理诱导产生的热休克蛋白70对肝脏缺血/再灌注损伤的保护作用的机制.方法:应用pringle,s法制备肝脏缺血/再灌注损伤模型及热应激预处理模型.将实验大鼠随机分为热应激预处理(HP I/R)组与非预处理(I/R)组,对比观察两组动物肝脏缺血/再灌注后0、4、8、12、24 h时肝脏HSP70的表达、SOD活力和MDA的产生量及大鼠血清门冬氨酸转氨酶(aspartate transaminase,AST),丙氨酸转氨酶(alanine transaminase,ALT)的活性与肝脏病理组织学改变.结果:热应激预处理组各时间点肝脏HSP70的表达及SOD的活力均比非预处理组同一时间点高,而血清AST、ALT酶活性及MDA的产生量较非预处理组低,病理损伤也比非预处理组减轻.结论:热应激预处理诱导产生的热休克蛋白70可能通过促进SOD的产生,从而降低氧自由基对肝脏的损害,起到保护肝脏缺血/再灌注损伤的作用.  相似文献

摘要 目的:探讨帕金森病(PD)患者血清激肽释放酶6(klk6)、热休克蛋白70(HSP70)水平与病情严重程度以及PD轻度认知障碍(PD-MCI)的关系。方法:选择2021年2月至2022年2月徐州医科大学附属医院收治的165例PD患者(PD组),根据修订的Hoehn-Yahr分级将PD患者分为早期组(1.0~2.5级,59例)、中期组(3.0级,65例)和晚期组(4.0~5.0 级,41例),根据是否存在PD-MCI将PD患者分为PD-MCI组(66例)和非PD-MCI组(99例),另选择同期72例于我院门诊体检的健康志愿者为对照组。检测血清klk6、HSP70水平,比较不同分组血清klk6、HSP70水平差异,多因素Logistic回归分析影响PD患者发生PD-MCI的因素。结果:PD组血清klk6水平高于对照组(P<0.05),HSP70水平低于对照组(P<0.05),晚期组血清klk6水平高于早期组和中期组,且中期组血清klk6水平均高于早期组(P<0.05),晚期组血清HSP70水平低于早期组和中期组,且中期组血清HSP70水平低于早期组(P<0.05);PD-MCI组血清klk6水平高于非PD-MCI组(P<0.05),HSP70水平低于非PD-MCI组(P<0.05)。多因素Logistic回归分析结果显示年龄偏高、修订的Hoehn-Yahr分级晚期、高水平klk6是PD患者发生PD-MCI的危险因素(P<0.05),高水平HSP70是PD患者发生PD-MCI的保护因素(P<0.05)。结论:PD患者血清klk6水平增高,HSP70水平降低与修订的Hoehn- Yahr分级增加以及PD-MCI有关,检测血清klk6、HSP70水平有助于评估PD病情以及PD-MCI风险。  相似文献

Effects of heat stress on skeletal muscle mass in young and aged mice were investigated. Young (7-week) and aged (106-week) male C57BL/6J mice were randomly assigned to control and heat-stressed groups in each age. Mice in heat-stressed group were exposed to heat stress (41?°C for 60?min) in an incubator without anesthesia. Seven days after the exposure, soleus muscles were dissected from both hindlimbs. Protein content and the relative composition of Type II fibers in aged soleus were lower than those in young muscle. To investigate the effects of different carbohydrate (CHO) levels in the diet of Wuchang bream Megalobrama amblycephala, the fish were randomly divided into six treatment groups. Five groups were fed 19, 25, 31, 38 and 47% CHO, respectively, for 8 weeks, and a control group was fed a diet with no CHO. Growth performance and feed utilization were significantly (P < 0.05) affected by the dietary carbohydrate level. Maximum weight gain and specific growth rate occurred at the 31% dietary carbohydrate level. Compared to the control, the 31% CHO group had a significantly increased serum total protein content, respiratory burst activity of leucocytes, serum complement 3 (C3) levels, serum lysozyme activity, serum alkaline phosphatase (AKP) activity and hepatic total antioxidative capacity (T‐AOC), as well as a decrease in serum glutamic‐oxaloacetic transaminase (GOT) activity. Compared to the control, the 47% CHO group had significantly increased serum GOT activity, and a tendency toward an increase in serum cortisol content and a decrease in serum lysozyme activity, hepatic superoxide dismutase (SOD) activity and T‐AOC. The relative level of hepatic heat shock protein 70 (HSP70) mRNA in fish fed the 38% CHO diet was significantly higher than those of fish fed the 19, 25 and 31% CHO diets, respectively (P < 0.05). After challenge with Aeromonas hydrophila, fish fed the 47% CHO had the significantly lowest post‐challenge survival, and fish fed the 31% CHO had the significantly highest post‐challenge survival (P < 0.05). The results of this study suggest that ingestion of excessive dietary CHO can impact the non‐specific immune responses, decrease the hepatic antioxidant abilities, and thus affect the health status of M. amblycephala.  相似文献

It is well established that liver ischemia-reperfusion induces the expression of heat shock protein (HSP) 70. However, the biological function of HSP70 in this injury is unclear. In this study, we sought to determine the role of HSP70 in hepatic ischemia-reperfusion injury in mice. Male mice were subjected to 90 min of partial hepatic ischemia followed by up to 8 h of reperfusion. HSP70 was rapidly upregulated after reperfusion. To explore the function of HSP70, sodium arsenite (8 mg/kg iv) was injected before surgery. We found that this dose induced HSP70 expression within 6 h of treatment. Induction of HSP70 with arsenite resulted in a >50% reduction in liver injury as determined by serum transaminases and histology. In addition, arsenite similarly reduced liver neutrophil recruitment and liver nuclear factor-kappaB activation, and attenuated serum levels of tumor necrosis factor-alpha and macrophage inflammatory protein-2, but increased levels of interleukin (IL)-6. In HSP70 knockout mice, arsenite did not protect against liver injury but did reduce liver neutrophil accumulation. Arsenite-induced reductions in neutrophil accumulation in HSP70 knockout mice were found to be mediated by IL-6. To determine whether extracellular HSP70 contributed to the injury, recombinant HSP70 was injected before surgery. Intravenous injection of 10 microg of recombinant HSP70 had no effect on liver injury after ischemia-reperfusion. The data suggest that intracellular HSP70 is directly hepatoprotective during ischemia-reperfusion injury and that extracellular HSP70 is not a significant contributor to the injury response in this model. Targeted induction of HSP70 may represent a potential therapeutic option for postischemic liver injury.  相似文献

The present study was conducted to evaluate the effect of various feeding patterns of emodin on growth, non-specific immune response, and disease resistance to Aeromonas hydrophila in juvenile Wuchang bream. Healthy Megalobrama amblycephala (initial weight: 3.47 ± 0.032) were grown in a circulating water system for 8 weeks. Five groups were studied: one control group was fed with a basal diet for eight weeks (Pattern 1, P1), and three treatment groups were fed with a trial diet of 30 mg emodin kg−1 at one-week (Pattern 2, P2), two-week (Pattern 3, P3), four-week (Pattern 4, P4) intervals. The final treatment group maintained the trial diet for the entire eight-week study duration (Pattern 5, P5). Results indicated that different feeding patterns of emodin significantly influenced the weight gain rate of Wuchang bream (P < 0.05). Fish in the P4 treatment group had significantly higher rates of weight gain (WG) than those in other treatment groups. There were no significant differences in survival rates or feed conversion ratios (FCR) between treatment groups and the control group. White blood cell count (WBC), respiratory burst activity, superoxide dismutase (SOD) activity, myeloperoxidase (MPO) activity and tumor necrosis factor-α (TNF-α) activity were shown to increase at first and then decrease from P3 condition to P5 condition. Fish under P4 treatment showed the most significant improvement of all tested parameters compared to control. Significantly higher levels (P < 0.05) of plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity were observed in P2 and P4 treatment groups when compared with the control group, while no significant differences were observed in the AST and ALT activity of fish in P2, P3, P4 and P5 treatment groups. In a bacterial challenge experiment with A. hydrophila, fish under P4 and P5 treatment showed lower cumulative mortality than the control group. The results of this study suggest that an initial 4-week feeding interval is recommended for the economic and practical culture of M. amblycephala.  相似文献

A single bout of exercise increases production of heat shock protein 70 (HSP70), which protects cells against various stresses. In this study, we investigated whether endurance exercise training enhances liver level of HSP70 and, if so, whether HSP70 contributes to hepatic protection against stress in vivo. Mice of an exercise-training group performed 60 min of treadmill running 5 days/wk for 4 wk. The resting level of liver HSP70 was 4.5 times higher in the trained than in sedentary mice. After 4 wk of exercise training, both groups of mice were exposed to the following stresses: 1) heat stress, 2) cold stress, 3) oxidative stress, 4) ethanol stress, and 5) exercise stress by compelling the mice to run on a treadmill until exhausted. After exposure to the stresses, the liver was immediately isolated. Elevation of liver HSP70 in the trained mice was evident, whereas no elevation was found in the sedentary mice. On exposure to heat, diethyldithiocarbamate and ethanol, activities of glutanic oxalacetic transaminase in plasma, and liver caspase-3, a key enzyme of apoptotic processing, were elevated in the sedentary mice but not in the trained mice. These results suggest that exercise training enhanced the resting level of liver HSP70 and hepatic protection against various stresses, at least partly attributing to the suppression of caspase-3 activity by the increase in HSP70.  相似文献

A study was conducted to assess the impact of summer season-related heat stress and nutritional stress individually as well as simultaneously on the adaptive capability of goats. Twenty four adult Osmanabadi bucks (average body weight (BW) 16.0 kg) were divided into four groups, C (n = 6; control), HS (n = 6; heat stress), NS (n = 6; nutritional stress) and CS (n = 6; combined stress). The study was conducted for a period of 45 days. The NS and CS bucks were under restricted feed to induce nutritional stress. The HS and CS bucks were exposed to summer heat stress. The animals exhibited different physiological adaptive behavior in the morning and afternoon. Further, the higher plasma cortisol (p < 0.01) and aldosterone (p < 0.05) was recorded in CS group as compared to other groups. The highest degree of degenerative changes and hyperactivity of endocrine cells was recorded in CS group liver and adrenal gland respectively. The higher expression of adrenal and hepatic Heat Shock Protein 70 (HSP70)messenger ribonucleic acid (mRNA) was reported in CS and HS goats respectively. It can be concluded from this study that plasma cortisol and adrenal HSP70 gene expression may be considered as ideal biological markers for combined stresses in Osmanabadi bucks.  相似文献

目的通过研究冷拘束应激中大鼠心脏和肝脏组织热休克蛋白(HSP70)表达量以及血清中肌酸激酶(CK)、谷丙转氨酶(ALT)含量的变化,探讨冷拘束应激对心脏和肝脏的影响和差异。方法选取50日龄清洁级大鼠77只,随机分为7组,即0h,0.5h,1.0h,1.5h,2.0h,2.5h,3.0h。采用拘束冷应激法,应激相应时间后,用免疫组织化学方法观察大鼠心脏和肝脏组织HSP70的表达,并检测血清中CK和ALT含量。结果 (1)心脏中HSP70表达量随着应激时间的延长而增加,1.5h组较0h组有明显的增加(P〈0.05),而2.0h后有极明显的增加(P〈0.01)。(2)肝脏中HSP70表达量随应激时间的延长呈缓慢增加,仅3.0h组有较明显的增加,与0h组、0.5h组及1.0h组比较有显著差异(P〈0.05)。(3)应激开始阶段大鼠血清中的CK含量迅速上升,之后平缓升高;大鼠血清中的ALT含量呈现上升趋势,2.5h以前上升较平缓,2.5h以后急骤升高。结论冷拘束应激可使大鼠心脏中HSP70的表达量和血清中CK含量在较短的时间内显著上升,且随着时间的增加而递增,而肝脏中HSP70的表达量和血清中ALT含量在较短的时间内无明显变化,只有在3.0h时才显著增加,提示心脏对冷拘束应激较敏感,而肝脏对冷拘束应激有较强的耐受性。  相似文献
