紫斑牡丹栽培品种小孢子发育过程的细胞遗传学研究   总被引:4,自引:0,他引:4

本文对紫斑牡丹栽培品种的花平细胞减数分裂过程进行了系统研究,结果发现紫斑牡丹品种约68.96%的花平细胞在减数分裂过程表现正常,约有31.04%的花粉母细胞在减数分裂的比线期、中期Ⅰ、后期Ⅰ、中期Ⅱ、后期Ⅱ及四分体时期观察到染色体行为异常。本实验表明,在小孢子形成过程中,多数小孢子发育政党,但有约31.04%的花粉母细胞减数分裂异常,导致了花粉的败育。  相似文献

为深入了解鱼腥草花粉母细胞的减数分裂特征与花粉育性的关系,该研究采用卡宝品红染色法对2个鱼腥草居群花粉母细胞的减数分裂过程进行观察,并采用氯化三苯基四氮唑(TTC)染色法、I2-KI染色法、B-K培养基培养法及荧光显微镜观察法来检测鱼腥草花粉的活力及萌发率。结果发现:(1)鱼腥草减数分裂的进程与花序大小、花药颜色、花药长度均有密切的关系。(2)2个居群的鱼腥草中花粉母细胞减数分裂过程正常占88.2%,有11.8%的花粉母细胞减数分裂异常。(3)减数分裂异常表现在减数分裂过程中出现微核、落后染色体、染色体桥、不均等分离、多分体等现象,并发现在二分体阶段及单核花粉发育过程中存在细胞融合。(4)2个居群的鱼腥草花粉活力均不超过1.5%,花粉几乎不萌发。研究认为,鱼腥草花粉育性低的主要原因是单核花粉的发育过程异常,而非鱼腥草花粉母细胞减数分裂异常所致。  相似文献

该研究以3个紫斑牡丹品种(‘象牙白’、‘美人面’、‘紫金冠’)的单瓣与半重瓣花粉为材料,对其超微结构和生理生化特性进行观察分析,并对其花粉培养基进行筛选优化,探讨单瓣和半重瓣花粉的超微结构与花粉萌发率以及物质代谢与花粉萌发率之间的关系。结果表明:(1)紫斑牡丹花粉萌发的最适宜培养基为100 g·L^(-1)蔗糖+0.15 g·L^(-1)硼酸+10 g·L^(-1)琼脂。(2)花粉饱满率是影响花粉萌发的主要原因。(3)3个紫斑牡丹品种的单瓣花粉离体培养萌发率均显著高于半重瓣的花粉。(4)‘象牙白’、‘美人面’、‘紫金冠’单瓣花粉可溶性蛋白含量均高于半重瓣花粉,但三者单瓣花粉的MDA含量和3种保护酶(CAT、SOD、POD)活性均低于半重瓣花粉。研究认为,花粉体内蛋白质亏损和MDA、CAT、SOD、POD的积累可能是引起半重瓣紫斑牡丹花粉萌发率较低的生理原因之一。  相似文献

黄瓜同源三倍体创制及减数分裂行为观察   总被引:3,自引:1,他引:2  
采用常规杂交法研究黄瓜二、四倍体杂交过程中亲本育性、授粉组合及亲本基因型对杂交结实率的影响,并利用减数分裂制片法对获得的黄瓜同源三倍体进行了花粉母细胞减数分裂行为的观察.结果显示:(1)同源四倍体白交结实率比较低(13.0%~14.5%),可能与其花药内所包含的正常花粉粒比例小及花粉管萌发长度较短有关.(2)二、四倍体杂交组合的结实率很低(0.26%~0.02%),但在两种配组方式之间存在着明显差异,即以同源四倍体黄瓜为父本和二倍体黄瓜为母本的杂交结实率比较高,反之则杂交结实率比较低.(3)在二、四倍体杂交过程中,二、四倍体的基因型对杂交结实率的影响较大,以杂交双亲同属一个基因型的杂交效果较好.(4)同源三倍体花粉母细胞减数分裂过程与二倍体基本相同,但存在较高频率的染色体异常行为:中期I染色体构型复杂,在大多数花粉母细胞中可观察到单价体、二价体、三价体的存在;中期Ⅰ和Ⅱ有少数染色体游离于赤道板外;后期I和后期Ⅱ常出现落后染色体、染色体桥及细胞分裂不同步现象,其最终结果导致了不正常四分体和不可育配子的形成.(5)同源三倍体花粉粒的平均可染率和萌发率分别为18.8%和11.3%.研究结果表明,黄瓜二、四倍体正反交能直接获得同源三倍体材料;同源三倍体花粉母细胞减数分裂异常导致不育配子的形成是引起其育性低的细胞学原因;同源三倍体的部分育性为通过同源三倍体的回交来选育初级三体系奠定了基础.  相似文献

对兰州百合(Lilium davidii var.unicolor)小孢子母细胞减数分裂异常进行了研究,发现存在不等二价体、同源染色体早分离、染色体桥、不均等分离、滞后染色体、核外染色体、微核等。分析了这些异常形成的可能机制及对正常小孢子形成的影响。人工花粉萌发实验表明:小孢子母细胞减数分裂异常是导致花粉败育的主要原因。认为兰州百合长期行无性繁殖引起染色体结构变异,导致减数分裂异常。  相似文献

采用染色体制片及爱氏苏木精染色-冬青油透明技术对青花菜花粉母细胞减数分裂及雄配子体发育过程进行了细胞学研究.结果表明:花粉母细胞减数分裂的细胞质分裂为同时型,四分体为正四面体型或十字交叉型;中期Ⅰ和中期Ⅱ,少数细胞可见赤道板外染色体;后期Ⅰ和后期Ⅱ部分细胞出现染色体桥及落后染色体;四分体时期可观察到二分体、三分体及含微核的异常四分体.雄配子体发育过程包括2次有丝分裂,成熟花粉为3细胞型,具3个萌发孔.减数分裂过程中染色体行为异常的花粉母细胞约占10.28%;雄配子体发育过程中异常频率约为3.2%,败育主要发生在单核期.  相似文献

以紫斑牡丹‘象牙白’单瓣和重瓣花的花粉为材料,对其超微结构及生理生化特性进行观察和分析,探讨单瓣和重瓣花花粉超微结构以及不同温度储存花粉活力与其可溶性蛋白含量、MDA含量和3种保护酶活性(POD、SOD、CAT)的关系,以确定紫斑牡丹单瓣和重瓣花花粉最佳储存温度。结果表明:(1)紫斑牡丹重瓣花花粉平均极轴长、平均赤道轴长、极轴长/赤道轴长均小于单瓣花花粉;室温(25℃)条件下,单瓣花花粉萌发率(83.62%)高于重瓣花(72.06%),单瓣花和重瓣花花粉的半衰期分别为40 d和60 d;随着储存温度降低和储存时间的延长,单瓣花和重瓣花的花粉萌发率较室温均显著提高,且单瓣花花粉萌发率始终高于重瓣花花粉。(2)紫斑牡丹单瓣和重瓣花花粉在短期储存(1~120 d)、较长时间储存和长期储存的适宜温度分别为4℃、-20℃,-80℃。(3)与室温相比,低温可提高储存期间紫斑牡丹花粉的可溶性蛋白质含量和保护酶活性,并降低MDA含量,从而有效延缓花粉萌发率的下降。(4)低温胁迫对紫斑牡丹单瓣花花粉的影响低于重瓣花花粉,在相同储存温度条件下,重瓣花花粉细胞受损程度更高、萌发率下降速率更快。研究认为,紫斑牡丹花粉在-80℃储存条件下基本处于代谢平衡状态,故低温能有效延长其花粉储存寿命;储存期间花粉萌发率下降的主要原因是细胞膜脂过氧化程度加剧,细胞损伤加重;紫斑牡丹重瓣花花粉粒小、瘪粒和畸形是导致其较单瓣花花粉萌发率低的主要原因。  相似文献

研究了矮牡丹Paeonia jishanensis Hong et W.Z.Zhao的小孢子发生及雄配子体的形成。矮 牡丹花药具4个小孢子囊,药壁结构属双子叶型,腺质绒毡层,小孢子母细胞减数分裂后胞质分裂为 同时型,四分体多为四面体形,少左右对称形,成熟花粉为2-细胞。对芍药属木本类型的雄性发育进行 了全面研究,还对小孢子母细胞减数分裂和单核小孢子发育时期的异常现象进行了观察,对能育花粉 与不育花粉的百分比进行了测定,结果表明,能育花粉为45.03%~84.18%,它们在不同花中,不同花 药中,甚至同一花药的不同花粉囊中表现都不完全一致。联系矮牡丹的致濒原因进行了讨论,认为雄配子体形成过程中的异常现象,并不是导致矮牡丹濒危的主要因素。  相似文献

以OT百合‘Cocossa’为材料,采用醋酸洋红染色烤片法和石蜡切片法,对该品种花粉母细胞减数分裂过程以及绒毡层在花粉母细胞发育过程中的变化进行系统的细胞学观察。研究发现:(1)当百合花蕾长度在2.5~4.5 cm时,花粉母细胞处在减数分裂时期。(2)在花粉母细胞减数分裂的过程中,出现了较多染色体异常现象:中期Ⅰ出现染色体环;后期Ⅰ出现了滞后染色体和染色体桥;在末期Ⅰ和末期Ⅱ细胞出现不均等分裂和微核;处于同一花蕾和同一花药的花粉母细胞减数分裂具有不同步性,能同时观察到2个不同时期的分裂相;形成畸形、无核、多核的异常小孢子。(3)在末期Ⅱ和单核花粉粒时期,绒毡层延迟降解,引起多核花粉粒、畸形花粉粒以及花粉粒自溶现象。结果表明:OT百合‘Cocossa’小孢子败育是花粉母细胞异常减数分裂与绒毡层异常降解共同导致的。  相似文献

采用染色体制片技术对Cucumis属双二倍体种(Cucumis hytivus Chen et Kirkbride)小孢子发生和雄配子体发育进行了细胞学研究。结果显示:在小孢子发生过程中,约31%的花粉母细胞在减数分裂中期Ⅰ具有正常的19Ⅲ,约69%的花粉母细胞染色体构型复杂,平均构型0.41Ⅰ 14.69Ⅲ 0.06Ⅲ 0.93Ⅳ 0.62Ⅵ 0.07Ⅷ;在四分体时期,形成约8.78%的四分孢子,其余为各种异常的多分孢子;在雄配子体发育过程中,约10%的小孢子可进行有丝分裂,最终发育为正常的两细胞、三孔花粉,其余90%的小孢子最终成为败育花粉。此外,还观察到了减数分裂后期Ⅱ的染色体组分离和新种花粉形态的变异等特殊现象。  相似文献

采用压片法,对麦蓝菜的花粉母细胞减数分裂及体细胞核型进行了研究。结果表明,麦蓝菜的绝大多数花粉母细胞减数分裂中染色体的行为正常,在终变期同源染色体配对后可形成15个二价体;在少数花粉母细胞减数分裂中观察到落后染色体、染色体桥等异常行为;其减数分裂为同时型,其花粉粒育性为88.04%。麦蓝菜的染色体数目为2n=2X=30,核型公式为K(2n)=2X=30=22m(2SAT)+8 sm,染色体相对长度组成为2n=16M2+14M1,其核型为"1A"型。  相似文献

The low fertility of naked seed rice (NSR) was investigated by the following observations: somatic chromosome constitute, behavior of pollen mother cells (PMCs), the germination of mature pollen grains, the development of male and female gametes and the structure of the anther opening. The results indicated that somatic chromosomal number was 2n = 24, behavior of PMCs were normal and most of pollen grains could regularly develop further to mature male gametophytes in NSR. And dehiscence chamber and thickened endothecium cell (TEC) in numerous anthers of the NSR were developed abnormally after dicaryotic phase, result in few anthers complete opening and most partly opening or failure to opening, therefore much fewer of pollen grains attach on the stigma as compared with normal variety. Furthermore most of embryo sacs possessed abnormal structure and were sterile. All of above illustrated that the failure of the anther opening and the abortion of female gametophyte were main factors controlling the low seed-setting rate of the NSR.  相似文献

Radiobiological effects of a low-energy ion beam on wheat   总被引:14,自引:0,他引:14  
The radiobiological effects of a low-energy nitrogen ion (N+) beam on wheat were studied, particularly with regard to the induction of chromosome aberrations. The results demonstrated that the three test varieties showed different sensitivities to ion implantation, and a higher dose of ion implantation had a marked effect on the germination and survival rate of the seeds exposed. The germination rate and survival rate curve basically followed a similar trend in the same variety. Cytological analysis indicated that ion beams were effective in producing chromosome aberrations. The frequencies of mitotic or meiotic cells with chromosome aberrations increased linearly with increasing doses. The aberration types included, for example, acentric fragments, chromosome deletions, lagging chromosomes, chromosome bridges and micronuclei. In the root tip cells, aberrations chiefly consisted of acentric fragments and deletions. Chromosome bridges and lagging chromosomes were the main aberration phenomena observed in the pollen mother cells. The highest frequencies of root tip cells and pollen mother cells with chromosome aberrations were 15.2% and 39.8%, respectively. Pinellia ternata is an important traditional Chinese medicinal plant. Its different populations in China have various ploidy levels, based on x = 13, as well as extensive aneuploid series. The microsporogenesis process was observed in specimens from three populations from three regions of Hubei Province; they were characterized by normal and abnormal meiotic divisions in pollen mother cells (PMCs) at all stages simultaneously. Meiotic abnormalities including univalents/multivalents, chromosomal laggards/bridges and micronuclei appeared in about 50% of the PMCs, together with abnormal cytokinesis. Chromatin/chromosome transfer between meiocytes occurred only during the first division, at low frequency; this might contribute to these meiotic abnormalities. Although the remaining 50% of the PMCs presented normal cytological behavior, pollen fertility was only about 2%. These results provide cytological explanations for its low seed-set and the general use of asexual reproduction through tubers and bulbils; it also explains the wide variations in chromosome number.  相似文献

Two original mechanisms of nuclear restitution related to different processes of meiotic division of pollen mother cells (PMCs) have been found in male meiosis of the lines of maize haploids no. 2903 and no. 2904. The first mechanism, which is characteristic of haploid no. 2903, consists in spindle deformation (bend) in the conventional metaphase-anaphase I. This leads to asymmetric incomplete cytokinesis with daughter cell membranes in the form of incisions on the mother cell membrane. As a result, the chromosomes of the daughter nuclei are combined into a common spindle during the second meiotic division, and a dyad of haploid microspores is formed at the tetrad stage. The frequency of this abnormality is about 50%. The second restitution mechanism, which has been observed in PMCs of haploid no. 2904, results from disturbance of the fusion of membrane vesicles (plastosomes) at the moment of formation of daughter cell membranes and completion of cytokinesis in the first meiotic division. This type of cell division yields a binuclear monad. In the second meiotic division, the chromosomes of the daughter nuclei form a common spindle, and meiosis results in a dyad of haploid microspores. The frequency of this abnormality is as high as 15%. As a result, haploid lines no. 2903 and no. 2904 partly restore fertility.  相似文献

Houttuynia cordata (Saururaceae) is a leaf vegetable and a medicinal herb througout much of Asia. Cytomixis and meiotic abnormalities during microsporogenesis were found in two populations of H. cordata with different ploidy levels (2n = 38, 96). Cytomixis occurred in pollen mother cells during meiosis at high frequencies and with variable degrees of chromatin/chromosome transfer. Meiotic abnormalities, such as chromosome laggards, asymmetric segregation and polyads, also prevailed in pollen mother cells at metaphase of the first division and later stages. They were caused by cytomixis and resulted in very low pollen viability and male sterility. Pollen mother cells from the population with 2n = 38 showed only simultaneous cytokinesis, but most pollen mother cells from the population with 2n = 96 showed successive cytokinesis; a minority underwent simultaneous cytokinesis. Cytomixis and irregular meiotic divisions appear to be the origin of the intraspecific polyploidy in this species, which has large variations in chromosome numbers.  相似文献

Our study of cytological phenotype of meiotic mutation pam resulted in detecting a failure of cytokinesis in mutant pollen mother cells in the form of a block of fusion of membrane vesicles of the cell plate, and an impossibility of formation of daughter cell membranes. The mutation does not disturb the division spindle structure and function. Asynchrony of meiosis in pam is the result of arrest of pollen mother cells at metaphase 1 and metaphase 2.  相似文献

The pathways of micro- and megagametophyte development in Agave fourcroydes (henequén) and A. angustifolia were studied. We used histology and light microscopy to observe anther ontogeny and ovary differentiation in relation to flower bud size. Both species have the same sexual reproductive strategies and gametophyte development that may be divided into three phases: (1) premeiotic, which includes the establishment of the megaspore mother cell and the pollen mother cell; (2) meiotic, the formation of mature microspores and functional megaspores; (3) postmeiotic, which encompasses the development of mature pollen grains and the formation of the embryo sac. A successive type microsporogenesis was found in both species with formation of T-shaped tetrads and binuclear pollen grains. In vitro germination tests revealed very low pollen fertility. The female gametophyte is formed from two micropylar megaspore cells after the first meiotic division (bisporic type). Male and female gametogenesis occur asynchronously with microsporogenesis finishing before macrosporogenesis. The results so far show that the formation of male and female gametophytes in henequén is affected at different stages and that these alterations might be responsible for the low fertility shown by this species.  相似文献

采用压片法,对黄芩花粉母细胞减数分裂及核型进行了研究。结果表明:黄芩的大多数花粉母细胞减数分裂中染色体的行为正常,在终变期同源染色体配对后可形成9个二价体,后期Ⅰ染色体以9∶9的方式向细胞两极分离,其减数分裂为同时型;在少数花粉母细胞减数分裂中观察到落后染色体、染色体桥等异常行为;其花粉粒育性为76.49%。黄芩的染色体数目为2n=2X=18,核型公式为K(2n)=2X=18=16m+2 sm,染色体相对长度组成为2n=1 s+4M1+3M2+1L,其核型为"1A"型。  相似文献
