采用常规染色体制片方法,对雏蝗属的模式种白边雏蝗Chorthippus albomarginatus(De Geer)和异爪蝗属的模式种素色异爪蝗Euchorthippus unicolor(Ikonnikov)的染色体核型进行了分析比较。结果表明,两种蝗虫的染色体数目均为2n(♂)=17=16+XO;常染色体为中部着丝粒(m,6条)和端部着丝粒(t,10条)两种类型,性染色体类型为端部着丝粒。二者的相似性显示出2属具有较近的亲缘关系,并且在进化过程中处于较近的发展阶段。但2种蝗虫的核型公式和染色体的相对长度组成则不相同,白边雏蝗K(2n,♂)=6m+11t=8L+4M+4S+XO;素色异爪蝗K(2n,♂)=6m+11t=8L+6M+2S+XO;且2种蝗虫性染色体的位次也有明显差别,白边雏蝗的性染色体为第9位,而素色异爪蝗则为第8位。由此看出,该2属的蝗虫存在的不同遗传特征。  相似文献

采用常规染色体制片方法对雏蝗属的褐色雏蝗Chorthippusbrunneus(Thunb .) ,异色雏蝗C .big uttulus(Linnaeus) ,小翅雏蝗C .fallax(Zub .) ,青藏雏蝗C .qingzangensis(Ying)的染色体核型进行分析 ,结果 :染色体数目均为 2n(♂ ) =1 7=1 6+XO ;常染色体类型为两类 ,中着丝点染色体 (m ,6条 )和端着丝点染色体 (T ,1 0条 ) ;性染色体类型为端着丝点。褐色雏蝗、异色雏蝗和青藏雏蝗的核型公式和染色体的相对长度组成为K( 2n ,♂ ) =1 7=6m +1 1T =6L +6M +4S +XO ,K( 2n ,♀ ) =1 8=6m +1 2T =6L +6M +4S +XX ;小翅雏蝗的为K( 2n,♂ ) =1 7=6m +1 1T =6L +4M +6S +XO ,K( 2n ,♀ ) =1 8=6m +1 2T =6L +4M +6S+XX。褐色雏蝗性染色体中部有次缢痕。染色体臂数 4种均为NF =2 3(♂ ) ,2 4 (♀ )。  相似文献

本文研究了棉蝗(Chondracris roses rosea De Geer)的染色体组型和C带.结果表明:棉蝗的染色体数目:2n(♂)=22十XO=23,全部为端着丝粒染色体,NF=23,核型公式:2n(♂)=2X = 23t,按染色体长度顺序排列分成四组,但按相对程度也可分为三组;所有染色体都具有明显的着丝粒带;异染色质总量为15.94土0.26%;染色体组中最长与最短染色体之比为5.89 : l,臂比大于2:1的染色体比例为零,棉蝗的核型属"1C"核型.  相似文献

记述采自中国四川省网翅蝗科雏蝗属Chorthippus(Altichorthippus)1新种:周氏雏蝗Chorthippus(Altichorthippus)choui sp.nov.。本新种同昌都雏蝗Chorthippus(A.)changtunensis Yin,1984近似,其区别特征为:后足股节和胫节桔红色;雄性腹部末端桔红色;前翅中脉域宽为肘脉域宽的1.2倍;本新种同西藏雏蝗Ch.(A.)tibetanus Uvarov,1935也近似,其区别特征为前胸背板侧隆线在沟前区明显,呈角状弯曲,在前横沟和后横沟之间不消失。  相似文献

本文研究了贵州日本黄脊蝗 Patanga japonica Bolivar和印度黄脊蝗 P.Succincta Johansson的核型和c-带。结果表明:两种黄脊蝗的染色体数目均为2n()=22+x.=23,全部为端着丝粒染色体;核型公式2n(♂)=2X=231,NF=23. 但日本黄脊蝗的染色体绝对长度总和(79.75±2.22μm)比印度黄脊蝗的(71.4 7±0.63μm)长;异染色质总量(35.65±0.14μm)比印度黄脊蝗的(10.76±0.11μm多;日本黄脊蝗的核型属“1B”核型,印度黄脊蜱的核型属“lc”核型。  相似文献

报道中国两种波腿蝗的染色体C带核型,结果表明:红胫波腿蝗Asiotmethis zacharjini (Bei-Bienko, 1926) 2n ♂ =18, neo-X为亚中着丝粒染色体,其他均为近端着丝粒染色体,染色体除强染的着丝粒C带,S8染色体具强染端部C带带纹,neo-Y染色体还具有一条宽的弱染的近着丝粒端居间C带,性别决定机制是neo-XY ♂型,该种染色体组成和性别决定机制在我国癞蝗中为首次报道,蓝胫波腿蝗Asiotmethis jubatus (Uvarov, 1926) 2n=19♂,均为近端着丝粒染色体,仅具有明显强染的着丝粒C带,性别决定机制是XO ♂型;两种波腿蝗的异染色质含量存在显著性差异(α=0.05)。  相似文献

The peripheral leg nerves of grasshoppers are initially formed by a set of pioneer neurons and guidepost cells. These cells are used as guiding structures for later-arising axons of sensory neurons. The development of the central projections of the pioneer cells, the guidepost cells and some sensory cells is shown with Lucifer Yellow injection or with DiI application. The axons of the pioneer cells Ti1 enter the central nervous system at 38% of embryonic development. They turn anteriorly close to the midline and ascend with no major branching to the brain. The axons of the guidepost cells Fe1 and Tr1 follow the same path but do not ascend to the brain. Sensory axons of the subgenual organ and the femoral organ probably do not follow the central path pioneered by the former neurons. They end ipsilaterally in the respective thoracic neuromere, as is found in the adult.  相似文献

在水/有机溶剂双相反应体系中,研究了来源于西梅的(R)-醇腈酶催化酮与丙酮醇腈合成(R)-酮醇腈的立体选择性转氰反应.系统探讨了不同酶源、酶粉颗粒大小、底物浓度、两底物配比、酶浓度和底物结构对转氰反应的影响.结果发现西梅醇腈酶能高效催化三甲基硅酮与丙酮醇腈的立体选择性转氰.酶粉颗粒大小以直径0.3~0.45 mm为优,底物浓度以21 mmol/L左右为佳,底物丙酮醇腈与三甲基硅酮摩尔浓度比以2∶1为宜,酶浓度以60.9 g/L左右为好.西梅醇腈酶对3, 3-二甲基-2-丁酮几乎没有催化活性,而对其硅结构类似物三甲基硅酮却具有非常高的立体选择性和催化活性,在上述优化反应条件下反应24 h的底物转化率和产物光学纯度均高达99%以上,表明底物中的硅原子对西梅醇腈酶的催化活性有非常显著的促进作用.  相似文献

An isolate, designated GC-29, that could incompletely oxidize glucose to acetate and carbon dioxide with Fe(III) serving as the electron acceptor was recovered from freshwater sediments of the Potomac River, Maryland. This metabolism yielded energy to support cell growth. Strain GC-29 is a facultatively anaerobic, gram-negative motile rod which, in addition to glucose, also used sucrose, lactate, pyruvate, yeast extract, casamino acids or H2 as alternative electron donors for Fe(III) reduction. Stain GC-29 could reduce NO3(-), Mn(IV), U(VI), fumarate, malate, S2O3(2-), and colloidal S0 as well as the humics analog, 2,6-anthraquinone disulfonate. Analysis of the almost complete 16S rRNA sequence indicated that strain GC-29 belongs in the Shewanella genus in the epsilon subdivision of the Proteobacteria. The name Shewanella saccharophilia is proposed. Shewanella saccharophilia differs from previously described fermentative microorganisms that metabolize glucose with the reduction of Fe(III) because it transfers significantly more electron equivalents to Fe(III); acetate and carbon dioxide are the only products of glucose metabolism; energy is conserved from Fe(III) reduction; and glucose is not metabolized in the absence of Fe(III). The metabolism of organisms like S. saccharophilia may account for the fact that glucose is metabolized primarily to acetate and carbon dioxide in a variety of sediments in which Fe(III) reduction is the terminal electron accepting process.  相似文献

The intracellular localisation of phytochrome in oat (Avena sativa L. cv. Garry Oat) coleoptiles was analysed by electron microscopy. Serial ultrathin sections of resin-embedded material were indirectly immunolabeled with polyclonal antibodies against phytochrome together with a gold-coupled second antibody. The limits of detectability of sequestered areas of phytochrome (SAPs) were analysed as a function of light pretreatments and amounts of the far-red absorbing form of phytochrome (Pfr) established. In 5-d-old dark-grownAvena coleoptiles SAPs were not detectable if less than 13 units of Pfr — compared with 100 units total phytochrome of 5-d-old dark-grown seedlings — were established by a red light pulse. In other sets of experiments, seedlings were preirradiated either with a non-saturating red light pulse to allow destruction to occur or with a saturating red followed by a far-red light pulse to induce first SAP formation and then its disaggregation. These preirradiations resulted in an increase of the limit of detectability of SAP formation after a second red light pulse to 38–41 and 19–23 units Pfr, respectively. The interaction of photo- and gravitropic stimulation was studied by analysing the curvature of maize (Zea mays L.) coleoptiles subjected to rotation on horizontal clinostats. Gravitropic curvature in different directions with respect to the stimulation plane was found to be transient. This instability was caused by an increasing deviation of response direction from the stimulation plane towards the caryopsis. The bending angle as such, however, increased steadily. This reorientation of the gravitropic response towards the caryopsis is thought to be caused by the clinostat-elicited nastic curvature found in maize coleoptiles. In contrast, the response to phototropic stimulation was stable, in both, orientation and curving. Although stimulation by gravity was not capable of inducing a stable tropistic response, it could inhibit the response to opposing phototropic stimulation, if the counterstimulation was given more than 90 min after the onset of gravistimulation. For shorter time intervals the influence of the phototropic stimulus obscured the response to the first, gravitropic stimulation. For time intervals exceeding 90 min, however, the phototropic effects disappeared and the response was identical to that for gravity stimulation alone. This gravity-induced inhibition of the phototropic response was confined to the plane of gravity stimulation, because a phototropic stimulation in the perpendicular direction remained unaffected, irrespective of the time interval between the stimulations. This concerned not only the stable phototropic curving, but also the capacity of the phototropic induction to elicit a stable directional memory as described earlier (P. Nick and F. Schäfer, 1988b, Planta 175, 380–388). This was tested by a second bluelight pulse opposing the first. It is suggested that gravity, too, can induce a directional memory differing from the blue-light elicited memory. The mechanisms mediating gravi- and phototropic directional memories are thought to branch off the respective tropistic signal chains at a stage where photo- and gravitropic transduction are still separate.This work was supported by the Deutsche Forschungsgemeinschaft and a grant of the Studienstiftung des Deutschen Volkes to P. Growth parameters were determined for tall (rht3) and dwarf (Rht3) seedlings of wheat (Triticum aestivum L.). Plant statures and leaf length were reduced by 50% in dwarfs but root and shoot dry weights were less affected. Leaves of dwarf seedlings had shorter epidermal cells and the numbers of cells per rank in talls and dwarfs matched the observed relationships in overall length. Talls grew at twice the rate of dwarfs (2.3 compared with 1.2 mm h-1). [3H]Gibberellin A1 ([3H]GA1) was fed to seedlings via the third leaf and metabolism was followed over 12 h. Immature leaves of tall seedlings transferred radioactivity rapidly to compounds co-chromatographing with [3H]gibberellin A8 ([3H]GA8) and a conjugate of [3H]GA8, whereas leaves of dwarf seedlings metabolised [3H]GA1 more slowly. Roots of both genotypes produced [3H]GA8-like material at similar rates. Isotopic dilution studies indicated a reduced 2-hydroxylation capacity in dwarfs, but parallel estimates of the endogenous GA pool size, obtained by radioimmunoassay, indicated a 12–15 times higher level of GA in the dwarf immature leaves. Dwarfing by the Rht3 gene does not appear to operate through enhanced, or abnormal metabolism of active gibberellins and the act of GA metabolism does not bear an obligate relationship to the growth response.Abbreviations GAn gibberellin An - HPLC high-performance liquid chromatography  相似文献

该研究建立了同时测定檬苦素和诺米林含量的高效液相色谱法,并采用该方法对8种不同柠檬样品中柠檬苦素和诺米林含量进行了测定。色谱条件如下:安捷伦Eclipse XDB-C18(250 mm×4.6 mm,5μm)色谱柱,流动相为0.1%磷酸水溶液(A)-乙腈(B),梯度洗脱,流速为1.0 mL·min~(-1),柱温为30℃,波长为210 nm。结果表明:3个产地尤力克果皮中柠檬苦素含量依次为云南瑞丽(EUR_D)、四川安岳(EUR_C)和广东河源(EUR_Y),诺米林含量依次为EUR_D、EUR_C和EUR_Y;不同品种柠檬果皮中柠檬苦素含量依次为北京柠檬(M_NM)、粗柠檬(R_NM)、无核柠檬(S_NM)、热那亚(RNY)、里斯本(LSB),诺米林含量依次为S_NM、R_NM、LSB、RNY和M_NM。综上结果表明,不同产地和品种柠檬果皮中柠檬苦素和诺米林含量均存在不同程度差异,该HPLC法可为不同产地和不同品种柠檬果实质量鉴别、资源评价及深度开发提供理论依据。  相似文献

Sequestered particles of phytochrome (SAPs) were partially purified from red-light-irradiated oat coleoptiles. Phytochrome pelletability was enhanced by using buffers containing 10 mM Mg2+ or high concentrations (0.6–0.8 M) of orthophosphate (Pi). Combining the pelletability of phytochrome in the presence of Mg2+ with that in the presence of 0.6 Pi resulted in a strong enrichment (about 100-fold) of pelletable phytochrome. Antisera were raised against Mg2+-Pi-pellets from darkgrown seedlings. Using these antisera, no evidence was found by Western blotting and immunocytochemistry that SAPs contain major proteins other than phytochrome. The major contamination of these enriched SAP preparations consisted of protein crystals which are probably catalase. The preparations contained methyltransferase and protein-kinase activities which were not associated with SAPs. Phytochrome purified from SAPs served as a substrate for protein-kinase activity but not for the methyltransferase activity.
