中国蚌科线粒体16S rRNA序列变异及系统发育   总被引:11,自引:0,他引:11  
首次测定了中国淡水贝类———蚌科 (Unionidae) 1 3个属代表种类的线粒体 1 6SrRNA部分序列。用Clustal X排序软件进行 1 6SrRNA序列的对位排列 ,序列总长度为 30 5— 32 0bp。通过Mega 2 0软件对所得线粒体 1 6SrRNA片段序列进行比较 ,共发现 1 0 8个碱基存在变异 ,其中包括 77个简约信息位点 ,并用“Pairwisedistance”计算了各属间的相对遗传距离。以贻贝为外类群 ,采用Mega 2 0软件中的“Neighbore Joining”法得到惟一一个分子系统树 ,系统树各分支的置信度由“Bootstrap”1 0 0 0循环检验。结果表明 :分布于中国的蚌科形成三个明显的类群 ,第一个类群 ,包括帆蚌属、蛏蚌属、丽蚌属和尖锄蚌属 ;第二个类群 ,由矛蚌属、扭蚌属、裂脊蚌属、鳞皮蚌属、尖嵴蚌属、楔蚌属和珠蚌属组成 ;第三个类群只包括无齿蚌和冠蚌两个属。由此可以看出 ,它们有可能分别隶属于三个不同的亚科 ,即小方蚌亚科 (帆蚌属、蛏蚌属、丽蚌属和尖锄蚌属 )、无齿蚌亚科 (无齿蚌属和冠蚌属 )和珠蚌亚科 (矛蚌属、扭蚌属、裂脊蚌属、鳞皮蚌属、尖嵴蚌属、楔蚌属和珠蚌属 ) ,由此证明小方蚌亚科在中国的存在 ,并且 ,对几个属的分类位置作了调整。其中鳞皮蚌属和蛏蚌属原来属于无齿蚌亚科 ,根据线粒体 1 6SrRNA的结果 ,前者应该放  相似文献

池蝶蚌(Hyriopsis schlegelii)和三角帆蚌(Hyriopsis cumingii)的亲缘关系一直存在争议, 为了更深入地对淡水珠蚌进行系统发育分析, 研究对池蝶蚌及其3个近缘物种三角帆蚌、褶纹冠蚌(Cristaria plicata)和背角无齿蚌(Anodonta woodiana)的转录组进行了比较分析。对它们共有单拷贝同源基因进行多序列联配后计算这4种淡水珠蚌之间的遗传距离, 结果表明池蝶蚌和三角帆蚌的遗传距离只有0.00746(小于1%), 而其他淡水珠蚌之间的遗传距离几乎是它的10倍(平均大于6%)。系统发育分析表明, 小方蚌亚科与无齿蚌亚科大约在5144万年前发生分歧, 而池蝶蚌和三角帆蚌发生分化的时间大约在424万年前。进一步的遗传差异分析鉴定了池蝶蚌转录组中的特有基因, 其中KEGG 通路富集的结果表明这些基因主要富集在免疫细胞膜分子和蛋白消化吸收等通路上。同时, GO注释结果表明有很多特有基因与池蝶蚌的优良性状相关, 如与发育相关的生长发育、系统发育和动物器官发育等生物学过程, 及与免疫相关的免疫系统过程、抗原处理和呈递等生物学过程。以上结果表明, 池蝶蚌和三角帆蚌具有更近的亲缘关系, 它们可能是同一物种的不同亚种或不同种群, 这对淡水珠蚌种质资源的保护和遗传育种具有重要参考意义。此外, 相比于三角帆蚌, 池蝶蚌可能具有一些与生长发育及免疫相关的特有基因, 这为探究相关的分子机制提供了线索。  相似文献

选择35个特征对我国蚌螨属15种的支序分析结果表明:蚌螨P-Ⅳ(胫节)端腹部的指状突是将它们聚集成两群的主要特征.今村蚌螨P-Ⅳ端腹部仅具有短矩,与外群最近,认为是我国的蚌螨属水螨中最原始的种类;雌螨前殖吸盘板的形态影响戊蚌螨亚属与多盘蚌螨亚属的种类聚类,由此推测可以将蚌螨P-Ⅳ端腹部的指状突与雌螨前殖吸盘板的形态也作为蚌螨亚属的区分特征.由于殖吸盘结构简单是祖征态之一,因此认为具有5对殖吸盘的邻近蚌螨比6对殖吸盘的厚蚌螨更原始.雌螨具有1对殖吸盘板的寄蚌螨亚属种类与沃蚌螨亚属种类在支序系统树中形成姊妹群,结果支持Vidrine 将沃蚌螨亚属从寄蚌螨亚属中分离的修订.  相似文献

利用PCR技术获得紫貂(Martes zibellina)和黄喉貂(Martes flavigula)线粒体DNA控制区全序列,并结合从GenBank中下载的9种鼬科动物相应序列,用ClustalX排序后对控制区结构进行分析,识别出延长终止序列区、中央区和保守序列区3个区域,指出了一个终止相关序列ETAS1及8个保守序列(CSB-F、E、D、C、B、1、2和3),并给出了序列通式,在CSB1和CSB2之间发现不同形式的短重复序列.此外,以狼为外类群,应用邻接法构建鼬科线粒体控制区全序列的系统进化树,结果表明:臭鼬亚科最先从鼬科中分化出来,随后剩余类群分为两大支系,即貂属种类与貂熊聚为一支,并与獾亚科的狗獾形成姐妹群;另一支为水獭亚科的物种与鼬属的林鼬形成姐妹群,再与虎鼬聚在一起,狗獾与貂属的紫貂亲缘关系最近,水獭亚科与鼬属亲缘关系最近.  相似文献

背瘤丽蚌稚蚌的生长与发育   总被引:1,自引:1,他引:0  
利用显微技术连续观察了背瘤丽蚌(Lamprotula leai)稚蚌发育、行为、生长和壳型变化等重要生物学特征。结果表明,刚脱落的稚蚌平均壳长208.5μm,在水温23.9~32.8℃条件下,经过75 d生长发育,进入幼蚌阶段,此时幼蚌平均壳长10.12 mm。壳长(L)、壳高(H)与日龄(t)的关系分别为L=0.220 4 e0.051 2t(R2=0.993 5)和H=0.227 7 e0.041 8t(R2=0.994 2),壳高(H)与壳长(L)的关系为H=0.781 5 L0.814(R2=0.998 1)。以稚蚌行为特征变化为依据,将背瘤丽蚌稚蚌发育划分为爬行期和埋栖期,再以稚蚌壳形状变化为依据,将爬行期稚蚌发育分为稚蚌脱落期、背部平直期、壳顶突出期,反映了背瘤丽蚌稚蚌生长与发育过程的动态变化。  相似文献

2009年5月31日-6月20日在广东省大沙河水库利用微型生态系统比较不同放养密度的褶纹冠蚌(Cristaria plicata)和背角无齿蚌(Anodonta woodiana)对水体氮、磷及浮游植物的影响,探讨两种蚌在控制南亚热带水库富营养化水体藻类水华上的可行性。实验结果表明,在褶纹冠蚌和背角无齿蚌处理组中,总氮、总磷和叶绿素a浓度显著增加,而铵氮的浓度显著下降;褶纹冠蚌和背角无齿蚌导致了浮游植物群落组成结构的改变和数量的增加,实验过程中绿藻所占的比例迅速上升。两种蚌之间没有显著的差异,只是在不同的作用强度下,时间上的响应不同。综合实验结果,褶纹冠蚌和背角无齿蚌难以有效地运用于我国华南地区水库的水质改善与富营养化控制。  相似文献

部分双壳贝类的线粒体遗传方式不同于标准的母系遗传(SMI),被称为双单性遗传现象(DUI)。池蝶蚌(Hyriopsis schlegelii)是淡水双壳贝类,是否存在双单性遗传现象?本文采用普通PCR扩增、SHOT-GUN测序及软件拼接获得了雄性池蝶蚌线粒体基因组(以下简称Hs-mtDNA)全序列,并与本实验室已报道的雌性池蝶蚌线粒体基因组全序列进行差异性分析。结果表明,雄性和雌性Hs-mtDNA全长分别为15961 bp和15939 bp,雄性比雌性长22 bp,雌雄线粒体基因组成与排列顺序一致。各蛋白编码基因的碱基数目均一致,碱基转换率为1.01%~7.34%,颠换率为0.00%~0.62%,氨基酸差异率为0.00%~9.35%;其中,COX1基因变异率为2.72%;COX2基因碱基变异率最高,达7.50%,雄性COX2的3'末端没有出现编码延伸区。雄性12S rRNA基因发生5 bp的碱基转换,差异率为0.6%;16S rRNA基因比雌性长9 bp,碱基差异率仅为1.2%。雌雄tRNA-His均位于H链上,介于COX2与ND3之间,没有出现位置的差异性。雌雄Hs-mtDNA的非编码区共有28个1~393 bp的片段,但未见控制区。在tRNA-Glu与tRNA-Tyr间有一段长393 bp的非编码区存在蛋白质翻译功能,但非雄性特异性蛋白。以COX1基因建立系统进化树,池蝶蚌和三角帆蚌(H.cumingii)聚在一起,而含有双单性遗传现象的无齿蚌属的Pyganodon grandis、小方蚌亚科的Venustaconcha ellipsiformis及小方形蚌属的Quadrula quadrula三者雄性聚为一支,雌性聚为一支。因此,雌雄池蝶蚌线粒体存在一定的差异性,但其差异要比其他具有双单性遗传现象的淡水双壳类小得多,且池蝶蚌线粒体遗传可能不存在双单性遗传现象。  相似文献

珠蚌科六种蚌的钩介幼虫形态比较研究   总被引:13,自引:4,他引:9  
本文观察了背角无齿蚌、沼纹冠蚌、剑状矛蚌、圆顶珠蚌、刻裂丽蚌和洞穴丽蚌等6种蚌的育儿囊类型;用光镜和扫描电镜观察和研究了幼虫的形态特征。结果表明,前4种蚌的育儿囊属外鳃类的同生型,后2种的育儿囊属外鳃类的四生型;6种幼虫均为钩介幼虫,其中前4种为有钩型,后2种为无钩型;6种幼虫的大小、形态有别,并证明种间幼虫的壳钧和壳饰的亚显微形态显然有异。本文还就6种蚌在Simpson分类系统中的地位与按它们的育儿囊和钩介幼虫类型得到的归类进行比较。结果表明,Simpson对6种蚌中3种的分类与归类不妥。本文为研究我国淡水贝类的分类提供一条新的途径,并为建立珠蚌类的自然分类系统积累资料。  相似文献

Abstract. The subfamily Ambleminae is the most diverse subfamily of fresh‐water mussels (order Unionoida), a globally diverse and ecologically prominent group of bivalves. About 250 amblemine species occur in North America; however, this diversity is highly imperiled, with the majority of species at risk. Assessing and protecting this diversity has been hampered by the uncertain systematics of this group. This study sought to provide an improved phylogenetic framework for the Ambleminae. Currently, 37 North American genera are recognized in Ambleminae. Previous phylogenetic studies of amblemines highlighted the need for more extensive sampling due to the uncertainties arising from polyphyly of many currently recognized taxa. The present study incorporated all amblemine genera occurring in North America north of the Rio Grande, with multiple species of most genera, including the type species for all but seven genera. A total of 192 new DNA sequences were obtained for three mitochondrial gene regions: COI, 16S, and ND1. In combination with published data, this produced a data matrix incorporating 357 gene sequences for 143 operational taxonomic units, representing 107 currently recognized species. Inclusion of published data provides additional taxa and a summary of present molecular evidence on amblemine phylogeny, if at the cost of increasing the amount of missing data. Parsimony and Bayesian analyses suggest that most amblemine genera, as currently defined, are polyphyletic. At higher taxonomic levels, the tribes Quadrulini, Lampsilini, and Pleurobemini were supported; the extent of Amblemini and the relationships of some genera previously assigned to that tribe remain unclear. The eastern North American amblemines appear monophyletic. Gonidea and some Eurasian taxa place as probable sister taxa for the eastern North American Ambleminae. The results also highlight problematic taxa of particular interest for further work.  相似文献

The classification and evolutionary relationships are important issues in the study of the groupers. Cytochrome b gene fragment of twenty-eight grouper species within six genera of subfamily Epinephelinae was amplified using PCR techniques and the sequences were analyzed to derive the phylogenetic relationships of the groupers from the China Seas. Genetic information indexes, including Kimura-2 parameter genetic distance and Ts/Tv ratios, were generated by using a variety of biology softwares. With Niphon spinosus, Pagrus major and Pagrus auriga as the designated outgroups, phylogenetic trees, which invoke additional homologous sequences of other Epinephelus fishes from GenBank, were constructed based on the neighbor-joining (NJ), maximum-parsimony (MP), maximum-likelihood (ML) and minimum-evolution (ME) methods. Several conclusions were drawn from the DNA sequences analysis: (1) genus Plectropomus, which was early diverged, is the most primitive group in the subfamily Epinephelinae; (2) genus Variola is more closely related to genus Cephalopolis than the other four genera; (3) genus Cephalopolis is a monophyletic group and more primitive than genus Epinephelus; (4) Promicrops lanceolatus and Cromileptes altivelis should be included in genus Epinephelus; (5) there exist two sister groups in genus Epinephelus.  相似文献

基于线粒体Cytb基因部分序列,以波蛱蝶为外群,采用邻接法、最大简约法和贝叶斯法分别构建了中国线蛱蝶亚科10属25种蝶类的系统发育树,探讨了各主要类群间的系统发育关系。其结果表明,所有线蛱蝶亚科聚为两大枝:第一枝包括环蛱蝶属、菲蛱蝶属、蟠蛱蝶属和缕蛱蝶属,其中缕蛱蝶属与环蛱蝶族亲缘关系较近;第二枝包括丽蛱蝶属、穆蛱蝶属、线蛱蝶属、带蛱蝶属、律蛱蝶属和翠蛱蝶属,其中线蛱蝶属为非单系群,翠蛱蝶属和律蛱蝶属则为单系发生,并构成姐妹群。  相似文献

The Botiinae have traditionally represented a subfamily of the Cobitidae. At present, the classification and phylogenetic relationships of the Botiinae are controversial. To address systematic and phylogenetic questions concerning this group, we sequenced the complete cytochrome b gene from 34 samples, of which 24 represented 13 species of the East Asian botiine fishes, while the other 10 were non-botiine loach species. For the 1140 bp sequences determined, 494 sites were variable ones, of which 424 were parsimony informative. With Myxocyprinus asiaticus as an outgroup, molecular phylogenetic trees were constructed using the neighbor-joining, maximum parsimony, maximum likelihood and Bayesian methods. All molecular phylogenetic trees revealed that botiine fishes form a monophyletic group and are distantly related to other loaches, suggesting that the Botiinae should be placed in their own family. Within the Botiinae, there are three genera; Botia, Parabotia, andLeptobotia, each genus forming a monophyletic group, with the genus Botia as the most ancestral split. Our molecular results are in agreement with morphological analyses of botiines, suggesting that Botia is the ancestral genus, while Leptobotia and Parabotia were resolved as more derived sister groups.  相似文献

啄木鸟群体共包含254种,但早期对啄木鸟的分类学研究局限于形态学、行为学和鸣唱法等方向,分子研究结果又彼此存在差异,因此啄木鸟分类系统目前还有争议。分子数据的大量累积为啄木鸟系统发育的研究提供了条件。利用从GenBank获取的ND2基因序列,对啄木鸟科179种啄木鸟采用NJ法、MP法以及ML法分别进行了分子进化树构建,同时用NJ法重建了Picumnus属分子进化树。此外又利用cytb基因序列采用NJ法对Dryobates属和Picoides属进行分子进化树重建。我们的结果显示,姬啄木亚科Picumnus属新大陆物种彼此的进化关系仍需进一步研究,而该属唯一一种旧大陆种P.innominatus,种内遗传差异在种间水平依然显著,这说明其种内遗传差异一直被低估了。在啄木鸟亚科内,Hemicircus属和Nesoctites属为姐妹关系,而Geocolaptes olivaceus插入了Campethera属支系中,并非与其构成姐妹群体。此外,一些姐妹群在新旧大陆上的分布,推测这种分布可能与第四纪冰川有关。啄木鸟从旧大陆向新大陆迁移,在第四纪冰川作用下不断发生分布范围的扩张与收缩,最终形成现存的分布格局。  相似文献

The Chinese oriental vole (Eothenomys chinensis) belongs to subfamily Arvicolinae, which is endemic to the mountains in southwest China. E. chinensis and other Arvicoline species display a number of features that make them ideal for evolutionary studies of speciation and the role of Quaternary glacial cycles on diversification. In this study, the complete mitochondrial genome of E. chinensis was sequenced. It was determined to be 16,362 bases. The nucleotide sequence data of 12 heavy-strand protein-coding genes of E. chinensis and other 19 rodents were used for phylogenetic analyses. Trees constructed using three different phylogenetic methods (Bayesian, maximum parsimony, and maximum likelihood) showed a similar topology demonstrating that E. chinensis was clustered in subfamily arvicolinae--formed a solid monophyletic group being sister to the subfamily Cricetinae. And the trees also suggested that E. chinensis is a sister to the genus Microtus and Proedromys.  相似文献

The evolutionary relationships of species of Danio and the monophyly and phylogenetic placement of the genus within the family Cyprinidae and subfamily Rasborinae provide fundamentally important phyloinformatics necessary for direct evaluations of an array of pertinent questions in modern comparative biology. Although the genus Danio is not one of the most diverse within the family, Danio rerio is one of the most important model species in biology. Many investigations have used this species or presumed close relatives to address specific questions that have lasting impact on the hypothesis and theory of development in vertebrates. Largely lacking from this approach has been a holistic picture of the exact phylogenetic or evolutionary relationships of this species and its close relatives. One thing that has been learned over the previous century is that many organismal attributes (e.g., developmental pathways, ecologies, behaviors, speciation) are historically constrained and their origins and functions are best explained via a phylogenetic approach. Herein, we provide a molecular evaluation of the phylogenetic placement of the model species Danio rerio within the genus Danio and among hypothesized closely related species and genera. Our analysis is derived from data using two nuclear genes (RAG1, rhodopsin) and five mitochondrial genes (ND4, ND4L, ND5, COI, cyt b) evaluated using parsimony, maximum likelihood, and Bayesian analyses. The family Cyprinidae is resolved as monophyletic but the subfamily Rasborinae (priority over Danioinae) is an unnatural assemblage. Danio is identified as a monophyletic group sister to a clade inclusive of the genera Chela, Microrasbora, Devario, and Inlecypris, not Devario nor Esomus as hypothesized in previous studies. Danio rerio is sister to D. kyathit among the species of Danio evaluated in this analysis. Microrasbora and Rasbora are non-monophyletic assemblages; however, Boraras is monophyletic.  相似文献
