利用普通小麦(Triticum aestivum L.)“小偃6号”与黑麦(Secale cereale L.)品种“德国白粒”杂交,选育出“小偃6号”类型带有黑麦性状的种质材料。应用总基因组原位杂交(GISH)进行检测,在8份材料中探测到黑麦染色质的存在,其中附加系3个,代换系1个,易位系4个;进一步用荧光绿标记探针pSc119.2及荧光红标记探针pAs1的双色荧光原位杂交(FISH)技术,对其中部分品系的染色体组成进行分析鉴定,结果表明:易位系BC116-1是1RS/1BL小麦/黑麦易位系,BC152-1是涉及一条1B染色体的1RS/1BL易位系, 代换系BC97-2是2R(2D)二体代换系;附加系BC122-3附加了一条6R黑麦染色体,一条6B染色体的长臂缺失。同时,对连续的总基因组原位杂交和双色荧光原位杂交技术在小麦育种中的应用进行了讨论。  相似文献

加拿大披碱草与野大麦 2个四倍体多年生禾草杂交产生的属间杂种F1为三倍体 (2n =3x =2 1) ,丢失了与亲本染色体基数相同的 7条染色体。为查明该杂种F1的染色体构成 ,应用基因组分子原位杂交方法 ,将父本(♂ )野大麦基因组 (H1H1H2 H2 )DNA用荧光生物素标记作为探针 ,以母本 (♀ )加拿大披碱草基因组 (SSHCHC)DNA作为封组 ,对杂种F1根尖细胞有丝分裂中期的染色体DNA进行原位杂交。结果表明 :三倍体杂种F12 1条染色体中 ,有 14条出现偏黄色荧光信号 ,来自父本 (♂ )野大麦H1H2 基因组有 7条出现橙红色荧光信号 ,来自母本 (♀ )加拿大披碱草S基因组、加拿大披碱草HC 基因组的 7条染色体丢失。  相似文献

利用组成玉米异染色质钮的180-bp重复序列和TR-1元件以及45S rDNA对玉米自交系F107、GB57、二倍体多年生类玉米及其远缘杂交后代的染色体进行荧光原位杂交,确定了3种重复序列在亲本染色体上的分布;同时对远缘杂交后代进行了细胞学鉴定,通过荧光信号在染色体上的位置,证实远缘杂交后代中异源种质的染色体来源;讨论了异染色质钮重复序列对玉米和其野生种杂交后代外源染色体整合和染色体行为等方面研究的应用。  相似文献

基于掖478导入系的玉米百粒重QTL鉴定   总被引:2,自引:0,他引:2  
玉米百粒重是产量性状的主要组成因子,对其控制位点进行QTL鉴定或基因克隆,将有益于其遗传控制的研究和分子育种的实施。本研究以导入系SL19-41为材料,该导入系是以我国玉米育种中广泛应用的骨干自交系掖478(Ye478)为遗传背景导入QB80染色体片段的纯合系。使用该导入系与Ye478杂交构建分离群体(F2、F2:3家系和BC1F1),通过3个环境下的田间试验,利用Ici Mapping的逐步回归区间作图法进行百粒重QTL定位,以及进行QTL位点连锁标记的表型效应分析。结果表明:鉴定了2个百粒重QTL位点,其中位于第4染色体bnlg1784~umc1194区间QTL位点q KW4-1在3个环境下均被检测到,可解释的表型变异为6.74%~17.81%,阐明了导入系SL19-41百粒重性状的遗传机制,同时也获得了改良版的Ye478(Ye478QB80),为玉米百粒重的遗传改良提供有益的分子标记,也为克隆百粒重基因提供材料来源。  相似文献

选用抗玉米丝黑穗病自交系Mo17和SH15为供体,与受体感病自交系黄早四和昌7-2构建回交群体(BC3F1\BC4F2),通过田间人工接种玉米丝黑穗病原菌鉴定抗病性表现,评价群体抗病性。研究结果显示黄早四×(黄早四×Mo17)BC4F2群体发病率明显高于BC3F1群体;两个BC4F2黄早四×(黄早四×Mo17)和昌7-2×(昌7-2×SH15)群体的发病率差异较大。采用SSR标记分析抗病株的供体染色体导入片段,发现随着回交次数的增多,导入片段数量减少,但不同回交群体中供体导入片段数目明显不同。通过连锁不平衡分析,在染色体2.09和3.04区段发掘和验证2个抗玉米丝黑穗病主效QTL,连锁标记分别为umc2077和phio53或bnlg1965。本文研究结果为抗丝黑穗病基因精细定位和分子聚合育种提供了信息和材料。  相似文献

基于GISH的甘蔗与斑茅F1染色体遗传与核型分析   总被引:1,自引:0,他引:1  
斑茅在甘蔗育种的利用是现代甘蔗育种种质创新的热点,育种者期望把斑茅中优异的特性通过杂交渗透到甘蔗中。甘蔗与斑茅的F1是斑茅利用研究的难点也是基础。本研究利用基因组原位杂交技术(GISH)分析甘蔗与斑茅的F1染色体构成和核型,探讨甘蔗与斑茅F1染色体的遗传行为。GISH结果表明,甘蔗与斑茅杂交F1的染色体众数68~69条,其中40条来自甘蔗热带种Badila,28~29条来自海南斑茅,未发现有染色体的交换或易位现象。参试材料大部分染色体都属于中部着丝点(m)的染色体,少数为近中部着丝点(sm),YCE95-41核型属2B型,其余的核型都为1B型。甘蔗与斑茅的染色体按n+n的方式传递给F1,本研究结果为斑茅种质在甘蔗育种中的利用及其杂交后代染色体细胞遗传研究提供参考依据。  相似文献

广西药用野生稻 (Oryza officinalis)具有多种优良特性 ,是水稻遗传育种的重要种质资源之一。本实验以 Biotin标记的药用野生稻总 DNA作探针 ,未标记的栽培稻 (Oryzasativa)总 DNA作封阻 ,以 HRP- DAB系统进行信号检测 ,对栽培稻与广西药用野生稻的杂种F1植株的根尖染色体制片进行基因组原位杂交。采用封阻比例 1∶ 2 0~ 30时 ,杂交效果较为理想 ,药用野生稻的 1 2条染色体显深棕色 ,而栽培稻的 1 2条染色体着色很浅。在有丝分裂间期的细胞核中 ,也检测到大量杂交信号分布于核的周边。  相似文献

通过将普通玉米与四倍体多年生玉米大量杂交,获得一株部分可育三倍体植株MT-71,为外源遗传物质在普通玉米的导入创造新种质。MT-71杂交结实率分别为3X/2X=7.84%,2X/3X=0.56%,3X/4X=0.52%,开放授粉结实率为1.18%;其花粉母细胞(PMC)染色体平均构型为2.30Ⅰ+3.72Ⅱ+6.49Ⅲ+0.23Ⅳ;3X/2X后代植株染色体几乎包括所有2n=20~30染色体数目,2X/3X和开放授粉后代中染色体数目仅有2n=20,21。结果表明,MT-71可产生有功能的雌雄配子,其中n=10,11的配子更易遗传;雌配子体对额外染色体的传递能力显著高于雄配子体,异源三倍体最有效的遗传方式为3X/2X。  相似文献

抗条锈病小偃麦双体异附加系山农87074-519的鉴定   总被引:7,自引:1,他引:6  
综合利用抗性接种鉴定、细胞学分析、SSR分子标记和基因组原位杂交(GISH)技术相结合的方法,对从长穗偃麦草与小麦复合杂交后代中选育的抗条锈病种质系山农87074-519进行了鉴定。结果表明,山农87074-519的根尖细胞染色体数目2n=44,花粉母细胞减数分裂中期I(PMCMI)绝大多数细胞内可观察到22个二价体,平均染色体构型2n=44=21.82Ⅱ 0.36Ⅰ,它与普通小麦中国春杂种F1的多数花粉母细胞内染色体构型为2n=21Ⅱ 1Ⅰ,因此它是1个附加了1对长穗偃麦草染色体的双体异附加系;以假鹅冠草St基因组总DNA作探针进行原位杂交发现山农87074-519的44条染色体中有2条出现黄绿色杂交信号,且杂交信号遍布整条染色体,证明其附加的长穗偃麦草染色体为St基组;利用SSR分子标记技术,在170对SSR引物中筛选出特异引物BARC165,它能稳定地在山农87074-519中扩增出长穗偃麦草特异标记BARC165268;将长穗偃麦草中BARC165的特异扩增片段克隆测序后制备成探针进行原位杂交,可在山农87074-519的间期染色体和有丝分裂中期染色体检测到杂交信号。山农87074-519综合农艺性状较好,对条锈病免疫,其抗性基因为显性,且位于附加的长穗偃麦草St基组染色体上,暂将其表示为YrSt。该种质系在小麦的遗传改良中具有重要利用价值。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.
