翦股颖和高羊茅对铜、铅吸收及耐受性   总被引:15,自引:1,他引:14  
测定了高羊茅和翦股颖两种草坪植物在Cu、Pb污染下发芽率、叶绿素、净光合速率和SOD的变化,以及两种草对Cu、Pb的吸收.结果表明, 重金属污染对高羊茅种子的萌发产生显著影响.高浓度Pb污染以及Pb、Cu复合污染对高羊茅的叶绿素影响很大,Cu单独污染对高羊茅叶绿素的抑制作用较小.翦股颖的叶绿素变化不显著.在3个Pb处理及350 mg·kg-1 Cu处理下,翦股颖的SOD活性均高于对照,而只有在700 mg·kg-1Pb 和350 mg·kg-1 Cu+1 100 mg·kg-1 Pb处理下高羊茅的SOD活性才升高.高羊茅的净光合速率受Cu污染影响较小,而受Pb污染影响较大.在Cu、Pb单独污染条件下,翦股颖净光合速率均呈下降趋势.从生理角度来看,翦股颖对Cu、Pb污染的耐受性强于高羊茅,而高羊茅对Cu污染的耐受性强于Pb.高羊茅Cu吸收量大,但Cu吸收量不随污染浓度升高而升高.翦股颖对土壤Pb的吸收能力较强, 且Pb吸收量随污染浓度的增加而增加.翦股颖和高羊茅可分别用于Pb和Cu污染土壤的治理.  相似文献

The changes in nutrient content of grain tissues and seedling parts of two wheat genotypes (Triticum aestivum L., Excalibur and Janz) with low or high seed Zn content were followed from imbibition to early seedling development (12 days). The grains were separated into seed coat, endosperm and embryo, while the seedlings were separated into roots and shoots. The dry weight of the seed coat did not change throughout the experimental period, whereas the endosperm weight declined rapidly from day 4 onward. The weight of embryo did not show any difference between and within cultivars. About a half of seed Zn was remobilised into shoot and roots during 12 days of growth, regardless of the initial seed Zn content in both genotypes. The seed coat contained 55–77% of the total seed nutrients in the two wheat genotypes, except in the case of S (around 40%). Manganese, Fe, Ca, K, and P were remobilised effectively from the seed coat as well as from the endosperm, while remobilisation of Zn and Cu was relatively less from the seed coat than from the endosperm. After 10 days of growth, all nutrients monitored were completely remobilised from the endosperm. Remobilised K was directed primarily into shoots; an increase in K content in shoots was relatively higher than the accumulation of dry matter, with a consequent increase in K concentration in shoot tissue. The remobilisation of some nutrients (eg. Fe, Ca and Zn) from various grain tissues during inbibition, germination and early growth is different from the remobilisation in more mature plants.  相似文献

The present study was carried out to elucidate the mechanism of seed deterioration in two cotton (Gossypium hirsutum L.) cultivars (HS6 and H1098). The seeds were artificially aged at 40 +/- 1 degree C and 100% relative humidity for 4 days. In both cultivars, germinability decreased, whereas membrane deterioration, as assayed by electrical conductivity of the seed leachates, increased progressively with artificial ageing. The decrease in germinability was well correlated with increased accumulation of total peroxide and malondialdehyde content and decreased activities of antioxidant enzymes peroxidase, catalase, ascorbate peroxidase, glutathione reductase and superoxide dismutase. Hydropriming for 2 h and ascorbic acid priming for 12 h partially maintained germination and the activities of various antioxidant enzymes under artificial ageing and the accumulation of peroxide and MDA content was decreased. The results suggest that cotton seed deterioration during accelerated ageing is closely related to a decrease in activities of various peroxide scavenging enzymes and to lipid peroxidation.  相似文献

Kinetics and inhibition of Na(+)/K(+)-ATPase and Mg(2+)-ATPase activity from rat synaptic plasma membrane (SPM), by separate and simultaneous exposure to transition (Cu(2+), Zn(2+), Fe(2+) and Co(2+)) and heavy metals (Hg(2+) and Pb(2+)) ions were studied. All investigated metals produced a larger maximum inhibition of Na(+)/K(+)-ATPase than Mg(2+)-ATPase activity. The free concentrations of the key species (inhibitor, MgATP(2-), MeATP(2-)) in the medium assay were calculated and discussed. Simultaneous exposure to the combinations Cu(2+)/Fe(2+) or Hg(2+)/Pb(2+) caused additive inhibition, while Cu(2+)/Zn(2+) or Fe(2+)/Zn(2+) inhibited Na(+)/K(+)-ATPase activity synergistically (i.e., greater than the sum metal-induced inhibition assayed separately). Simultaneous exposure to Cu(2+)/Fe(2+) or Cu(2+)/Zn(2+) inhibited Mg(2+)-ATPase activity synergistically, while Hg(2+)/Pb(2+) or Fe(2+)/Zn(2+) induced antagonistic inhibition of this enzyme. Kinetic analysis showed that all investigated metals inhibited Na(+)/K(+)-ATPase activity by reducing the maximum velocities (V(max)) rather than the apparent affinity (Km) for substrate MgATP(2-), implying the noncompetitive nature of the inhibition. The incomplete inhibition of Mg(2+)-ATPase activity by Zn(2+), Fe(2+) and Co(2+) as well as kinetic analysis indicated two distinct Mg(2+)-ATPase subtypes activated in the presence of low and high MgATP(2-) concentration. EDTA, L-cysteine and gluthathione (GSH) prevented metal ion-induced inhibition of Na(+)/K(+)-ATPase with various potencies. Furthermore, these ligands also reversed Na(+)/K(+)-ATPase activity inhibited by transition metals in a concentration-dependent manner, but a recovery effect by any ligand on Hg(2+)-induced inhibition was not obtained.  相似文献

测定了Hg2+、Cd2+、Cu2+、Pb2+单一重金属胁迫对拟南芥种子发芽和幼苗生长的影响.结果表明,重金属对幼苗生长的毒性大于对种子发芽的毒性,以抑制种子发芽的IC50为指标,4种重金属的毒性顺序为Hg2+>Cd2+>Pb2+/Cu2+,以幼苗生长为指标,则毒性顺序为:Cu2+>Hg2+>Cd2+/Pb2+,并随着胁迫时间延长,种子萌发率下降.此外,不同重金属在不同发芽时段对种子的毒性也不尽相同,Cd2+的毒性在种子吸水后的0~12 h大于12~24 h,而Hg2+毒性在12~24 h大于0~12 h,其中,种皮对减轻重金属毒性起着十分重要的作用.通过非毒性离子(Ca2+、Mg2+、K+、Na+)与重金属离子(Hg2+、Cd2+、Cu2+、Pb2+)交互作用对拟南芥种子发芽及幼苗生长效应的研究发现, mmol·L-1的Ca2+、Mg2+、K+、Na+可以增强Hg2+对种子发芽的毒性,但对Cd2+的毒性却没有影响.对于幼苗来说,Ca2+、Mg2+、K+、Na+可以显著增强Hg2+的毒性,Ca2+可以缓解Cd2+的毒性,但却增加Cu2+的毒性,K+可以缓解Pb2+对幼苗的毒害作用.最后,本文对重金属的毒害机理进行了探讨.  相似文献

Cactus seeds have developed adaptations to survive with high temperatures and low soil moisture in their habitats. We studied the effect of the combination of four water potentials (0, −0.2, −0.4 and −0.6 MPa) and two temperatures (25°C and 35°C) on germination and seedling mass of four endemic Leptocereus species from Cuba. There were two semi-arid coast species (L. arboreus and L. santamarinae) and two species inhabiting dry inland karstic hills (L. ekmanii and L. scopulophilus). We hypothesized that: (a) a decrease in water potential and an increase in temperature would result in low and slow germination, as well as in low seedling mass, and (b) ungerminated cactus seeds after exposure to combined water and temperature stress would have a high recovery capacity. The minimum time required for the seeds to start germination (Tmin), mean germination time (MGT) and germinability were evaluated. In addition, seed mass, the recovery after treatments of non-germinated seeds, and the seedling fresh mass obtained under different treatments were compared between species. In general, germination was only obtained at 25°C and germinability and seedling mass were drastically affected by the reduction from 0 MPa to −0.2 MPa. Seeds showed thermoinhibition at 35°C at all water potentials. There was a tendency to increase the MGT with decreasing water potential in three species. Low seed recovery occurred at all combined treatments for three species. If the predictions of increased temperature and decreased rainfall for the Caribbean region occur, a reduction in the germination of the Leptocereus species studied is expected.  相似文献

通过等渗的NaCl和PEG溶液模拟盐分和水分胁迫,设置渗透梯度,在控制条件下对杠柳种子的萌发过程中总萌发率、幼苗鲜重、胚根生长、种子活力、发芽值等指标系统研究,对种子萌发率与渗透势之间关系进行回归分析,主要结果包括:(1)杠柳种子萌发过程中总萌发率、幼苗鲜重、活力指数和发芽值四项指标均随NaCl和PEG溶液的渗透势降低逐渐下降,综合研究活力指数和发芽值表明渗透势≥-0.5 MPa和≤-1.4 MPa时,PEG对种子萌发抑制作用大于NaCl,其他情况相反;(2)杠柳种子逐日萌发率和胚根日变化研究表明,与NaCl相比,PEG推迟杠柳种子萌发,并且对胚根增长抑制作用较大;(3)建立盐分和水分胁迫条件下种子萌发率与渗透势回归方程,发现杠柳种子在PEG胁迫下的萌发临界值和极限值为-1.0和-1.4 MPa,在NaCl胁迫下是-0.9和-1.3 MPa,解除胁迫条件,不同处理的杠柳种子复水萌发率均达到100%。说明杠柳具有良好的耐盐抗旱的特性。  相似文献

花生果针入土后16天(16 DAP),种子干重和鲜重开始迅速增加。整个发育阶段可分为5个时期:组织分化期(0~20 DAP)、成熟前期(21~28 DAP)、成熟中期(29~40DAP)、成熟中后期(41~62 DAP)和成熟后期(63~88DAP)。种子发芽率在成熟前期和中期迅速提高并到达最大值,而苗成活率在成熟中后期达到最大值,苗鲜重则以88 DAP种子的为最大。种子发育过程中,贮藏蛋白质的合成与积累模式与种子干重变化相似。SDS-PAGE分析表明,种子发育初期(16 DAP)子叶中已积累花生球蛋白和伴花生球蛋白I。双向凝胶电泳显示花生球蛋白各个亚基在20DAP时均已存在,伴花生球蛋白I的主要亚基在整个发育过程中其等电点有所变化,含量也逐渐增加。其他蛋白质在种子发芽力形成阶段(20~40 DAP)的变化较为显著。  相似文献

以小麦品种‘德抗961'为材料,用NO供体硝普钠(SNP)浸种研究外源NO对盐胁迫下小麦种子萌发的影响.结果表明:0.06 mmol/L的SNP浸种24 h后对盐胁迫下小麦种子发芽率、发芽指数、活力指数和吸胀速率的下调都有显著缓解作用;SNP浸种对盐胁迫下α-淀粉酶的活性无明显影响,但能显著提高盐胁迫下β-淀粉酶的活性;进一步研究表明,SNP浸种预处理对盐胁迫下的α-淀粉酶同工酶变浅的条带有所恢复(尤其是条带3),同时使盐胁迫下变浅的β-淀粉酶同工酶的条带有明显的恢复(尤其是d、e、f、g).并且SNP能显著降低盐胁迫下小麦地上部分和根中的Na^+含量,提高其K+含量,从而使K^+/Na^+显著提高.以上结果表明:SNP浸种预处理提高盐胁迫下小麦种子的萌发,主要是通过提高β-淀粉酶的活性来实现的.  相似文献

Arisaema dracontium (green dragon) is a perennial herb that is widely distributed in eastern North America. However, in Canada, at the northern edge of its distribution, the species is designated as “vulnerable” with respect to conservation status. In natural populations, seedlings are uncommon; the present study was undertaken in order to characterize seed and seedling properties in green dragon. Seeds were sampled from five sites, ranging from Ontario at the northern limit of the distribution range, to Louisiana in the south. Seed germinability ranged from 25 to 55%, depending upon source. Experiments indicated that neither the hard seed coat nor a water-soluble exudate from the seed was responsible for inducing or maintaining dormancy. Patterns of seed germination appear to reflect general climatic conditions at the sites where seeds had originated. Cold stratification at 3°C produced significantly greater relative germinability in all seed collections except the most southerly one, from Baton Rouge. These seeds also had a slower overall speed of germination. In contrast, germination of seeds from the most northerly site was promoted by cold stratification and occurred over a relatively brief period. Germination in alternating light and dark conditions decreased the speed of germination compared to germination in the dark, however exposure to light changed the phenology of germination by promoting development of adventitious roots and primary leaves in these seedlings.  相似文献

The developmental changes in morphology and germinability of Jatropha curcas seeds, effects of temperature and light on seed germination , and changes in desiccation tolerance of mature seeds were studied in this paper. The results indicated that J. curcas seeds reached a physiological maturation at 58 days after anthesis, and that seed germination percentage reached a peak at physiological maturation, and then decreased. The optimal germination temperature was 25 to 30℃. J. curcas seeds were insensitive to dehydration at physiological maturation . There was not a notable effect of light on seed germination. Therefore, J. curcas seed was a non-photoblastic and orthodox seeds .  相似文献

The purpose of this study was to investigate the effects of various presowing treatments on the germinability (final germination percentage) and germination rate of loquat seeds in order to increase seedling production in nurseries (applied research) as well as provide answers for important physiological issues related to loquat seeds and their seed coat (basic research). Three experiments were carried out with various pre-sowing treatments. These treatments included full or partial removal of seed coat (perisperm), partial cutting of cotyledons as well as moist chilling at 5°C for 13 days and/or soaking the seeds in water or 250 ppm gibberellic acid (GA3) solution for 24 h. According to the results, cotyledons excision resulted in delayed germination, regardless of the presence or absence of the seed coat in comparison with the decoated seeds that demonstrated the highest germination rate amongst them. In addition, even the partial excision of seed coats affected positively both the germinability and the germination rate, compared to the control-intact seeds. Furthermore, control-intact seeds had a higher germination percentage when exposed to moist chilling independently of the application or not of gibberellin; while the combination of gibberellin application and moist chilling improved both the percentage and the rate of germination of decoated seeds. In conclusion, the role of perisperm (seed coat) in the germination procedure of loquat seeds seems to be important, indicating the existence of seed coat-imposed dormancy on loquat seeds. Finally, the existence of a mild endogenous embryo-dormancy on loquat is also discussed.  相似文献

9个高羊茅品种初期生长对镉胁迫的响应   总被引:1,自引:0,他引:1  
用Cd2+浓度0(对照)、25、50、75 mg/L分别处理9个高羊茅品种种子,测定供试品种种子发芽势、发芽率、幼苗苗高、根长、鲜重、叶绿素含量、丙二醛含量、质膜透性等8项指标,比较9个品种耐镉性。结果表明:与对照相比,在不同浓度Cd2+胁迫下,家园的发芽势和发芽率以及麦哲伦鲜重呈先略上升后下降趋势,其余品种的均呈下降趋势;9个高羊茅品种幼苗根长、苗高、叶绿素含量均呈下降趋势;9个高羊茅品种丙二醛含量和质膜透性均呈上升趋势。对测定的8项指标进行综合评价结果显示:在Cd2+25 mg/L胁迫下,9个品种耐Cd2+性由强到弱的顺序为热浪>翠碧A>里园2号>缤狗>麦哲伦>家园>宇宙星>回报>爱瑞3号;在Cd2+50 mg/L胁迫下,耐Cd2+性的顺序为热浪>翠碧A>缤狗>麦哲伦>里园2号>家园>宇宙星>爱瑞3号>回报;在Cd2+ 75 mg/L胁迫下,耐Cd2+性的顺序为热浪>缤狗>里园2号>翠碧A>家园>麦哲伦>爱瑞3号>宇宙星>回报。所以,在治理Cd2+污染土壤时,要根据污染严重程度,合理选用高羊茅品种。  相似文献

Methyl jasmonate (MeJA) is a naturally occurring plant growth regulator and play vital roles in plant defense and many developmental processes such as root growth and seed germination. This study was undertaken to study the possible role of using methyl jasmonate to alleviate the adverse effect of water stress on soybean genotypes (Giza 22 and 35). The results showed that water stress reduced shoot length, fresh and dry weights of shoot and root, photosynthetic pigments, relative water content and oil content in the shoots of all soybean genotypes. On the other hand, there was a considerable increase in cell wall fractionation, saturated and unsaturated fatty acids, flavonoids, phenolic acid and sugar fraction content in the shoots of the soybean genotypes in response to the water stress. Foliar spray with methyl jasmonate increased all the above parameters as compared to stressed plants. The results investigate the important role of MeJA in alleviation of water stress in soybean plants and suggest that MeJA could be used for improving plant growth under water stress as a potential growth regulator. The soybean genotypes Giza 22 was found to be more resistant to water stress than Giza 35.  相似文献

The K(+) ionophore nigericin is shown to be highly effective as an ionophore for Pb(2+) but not other divalent cations, including Cu(2+), Zn(2+), Cd(2+), Mn(2+), Co(2+), Ca(2+), Ni(2+), and Sr(2+). Among this group a minor activity for Cu(2+) transport is seen, while for the others activity is near or below the limit of detection. The selectivity of nigericin for Pb(2+) exceeds that of ionomycin or monensin and arises, at least in part, from a high stability of nigericin-Pb(2+) complexes. Plots of log rate vs log Pb(2+) or log ionophore concentration, together with the pH dependency, indicate that nigericin transports Pb(2+) via the species NigPbOH and by a mechanism that is predominately electroneutral. As with monensin and ionomycin, a minor fraction of activity may be electrogenic, based upon a stimulation of rate that is produced by agents which prevent the formation of transmembrane electrical potentials. Nigericin-catalyzed Pb(2+) transport is not inhibited by physiological concentrations of Ca(2+) or Mg(2+) and is only modestly affected by K(+) and Na(+) concentrations in the range of 0-100 mM. These characteristics, together with higher selectivity and efficiency, suggest that nigericin may be more useful than monensin in the treatment of Pb intoxication.  相似文献

Lead toxicity is a major environmental health problem in the United States. Bone is the major reservoir for body lead. Although lead has been shown to impair bone metabolism in animals and at the cellular level, the effect of Pb(2+) at the molecular level is largely unknown. We have used circular dichroism (CD), and a hydroxyapatite binding assay to investigate the effect of Pb(2+) on the structure and mineral binding properties of osteocalcin, a noncollagenous bone protein. The CD data indicate Pb(2+) induces a similar structure in osteocalcin as Ca(2+) but at 2 orders of magnitude lower concentration. These results were explained by the more than 4 orders of magnitude tighter binding of Pb(2+) to osteocalcin (K(d)=0.085 microM) than Ca(2+) (K(d)=1.25 mM). The hydroxyapatite binding assays show that Pb(2+) causes an increased adsorption to hydroxyapatite, similar to Ca(2+), but at 2-3 orders of magnitude lower concentration. Low Pb(2+) levels (1 microM) in addition to physiological Ca(2+) levels (1 mM) caused a significant (40%) increase in the amount of mineral bound osteocalcin as compared to 1 mM Ca(2+) alone. These results suggest a molecular mechanism of Pb(2+) toxicity where low Pb(2+) levels can inappropriately perturb Ca(2+) regulated processes. In-vivo, the increased mineral bound osteocalcin could play a role in the observed low bone formation rates and decreased bone density observed in Pb(2+)-intoxicated animals.  相似文献

ALA-D (EC 4.2.1.24) is the first cytosolic enzyme in the haem metabolic pathway. Some metals compete with its major cofactor Zn(2+), modifying both enzyme structure and function. Our purpose was to contribute to the understanding of the biochemical role of metals such as Pb(2+), Cd(2+), Cu(2+), Mg(2+), Zn(2+), Na(+), K(+) and Li(+) on ALA-D, using chicken embryos as experimental model. Mg(2+) and Zn(2+) showed enzyme activation in yolk sac membrane (YSM) (113% at 10(-5) M Mg(2+) and from 10(-4) M Zn(2+)), and slight inactivation in liver. Cd(2+) and Cu(2+) caused a non allosteric inhibition in both tissues (100% from 10(-4) M). Surprisingly Pb(2+) was not such a strong inhibitor. Interference of cations during the Schiff base formation in enzymatic catalysis process is explained considering their Lewis acid-base capacity, coordination geometry and electron configuration of valence. Interactions among monovalent cations and biochemical substances are governed chiefly by its electrostatic potential. 0.1 M K(+) and 0.4 M Na(+) produced 30% of enzymatic inhibition by the interference on interactions among the functional subunits. Li(+) activated the YSM enzyme (130% at 10(-5) M) due to a more specific interaction. This study may contribute to elucidate for the first time the possible structural differences between the YSM and liver enzymes from chicken embryo.  相似文献

The focus of this study is to investigate the regulatory role of K(+) influx in Arabidopsis pollen germination and pollen tube growth. Using agar-containing media, in vitro methods for Arabidopsis pollen germination have been successfully established for the first time. The pollen germination percentage was nearly 75% and the average pollen tube length reached 135 microm after a 6 h incubation. A decrease in external K(+) concentration from 1 mM to 35 microM resulted in 30% inhibition of pollen germination and 40% inhibition of pollen tube growth. An increase in external K(+) concentration from 1 mM to 30 mM stimulated pollen tube growth but inhibited pollen germination. To study how K(+) influx is associated with pollen germination and tube growth, regulation of the inward K(+) channels in the pollen plasma membrane was investigated by conducting patch-clamp whole-cell recording with pollen protoplasts. K(+) currents were first identified in Arabidopsis pollen protoplasts. The inward K(+) currents were insensitive to changes in cytoplasmic Ca(2+) but were inhibited by a high concentration of external Ca(2+). A decrease of external Ca(2+) concentration from 10 mM (control) to 1 mM had no significant effect on the inward K(+) currents, while an increase of external Ca(2+) concentration from 10 mM to 50 mM inhibited the inward K(+) currents by 46%. Changes in external pH significantly affected the magnitude, conductance, voltage-independent maximal conductance, and activation kinetics of the inward K(+) currents. The physiological importance of potassium influx mediated by the inward K(+)-channels during Arabidopsis pollen germination and tube growth is discussed.  相似文献
