目的:构建小麦细胞分裂素氧化酶基因TaCKXI原核表达载体并进行表达,以期得到大量的His标签融合蛋白.方法:根据GenBank中的TaCKXI序列以及pET-24a载体中的多克隆位点设计引物,以含有TaCKXl编码基因的批pMD-QRCKXI重组质粒为模板,经PCR扩增得到TaCKXI基因的DNA片段.将所得的片段与pET-24a载体连接,转化DH5α大肠杆菌,筛选阳性克隆,其测序结果与原序列一致,表明原核表达载体pET-TaCKXI已构建成功.提取per-TaCKXI质粒转化到BL21(DE3)pLysS表达菌株中,经IPTG诱导后收集菌体进行SDS-PAGE电泳鉴定,并优化其表达条件.结果:在大肠杆菌中获得TaCKXI基因融合表达,主要以包涵体形式存在;融合蛋白的分子量为58.91kD;IPTG终浓度为0.5、1.0、1.5.2.0mmol/L时,诱导融合蛋白产量相差不大.选用0.5mmol/L诱导15h获得大量的融合蛋白.经用原核表达蛋白纯化试剂盒纯化,得到了单一的融合蛋白.结论:小麦TaCKXI基因在大肠杆菌中获得了高效表达,为今后TaCKXI蛋白多克隆抗体的制备奠定了基础.  相似文献

稀有密码子影响人RNaseH-Apaf1融合蛋白原核表达   总被引:1,自引:0,他引:1  
通过限制性内切酶克隆法构建两个pET-28a-TAT-RNaseH-Apaf1重组质粒,其中一个质粒经过稀有密码子的优化。将两个重组质粒分别转化原核表达宿主菌BL21(DE3)和Transetta(DE3),通过SDS-PAGE观察相应融合蛋白诱导表达的情况,并用His标签抗体和Apaf1抗体通过Western Blotting鉴定诱导蛋白。菌液PCR和测序证明经稀有密码子优化和未经稀有密码子优化的人源RNaseH-Apaf1序列成功重组到pET-28a-TAT载体中;经稀有密码子优化的重组质粒成功地在大肠杆菌宿主菌中表达RNaseH-Apaf1融合蛋白,并用Western Blotting检测到该融合蛋白。结果表明,稀有密码子可影响人RNaseH-Apaf1融合蛋白的原核表达。  相似文献

目的:构建pET-42a(+)-HPV58E6E7原核表达质粒,诱导表达人乳头瘤病毒(HPV)58型E6E7融合蛋白。方法:采用PCR方法扩增出HPV58 E6E7融合基因的全长序列,利用DNA重组技术将其定向插入原核表达载体pET-42a(+)中,构建pET-42a(+)-HPV58E6E7原核表达质粒,用限制性内切酶酶切和核酸序列检测对重组质粒进行鉴定;将其转入宿主菌大肠杆菌BL21进行诱导以表达HPV58E6E7融合蛋白,并用谷胱甘肽琼脂糖树脂纯化回收HPV58E6E7融合蛋白,用SDS-PAGE及Western印迹鉴定表达蛋白的相对分子质量及抗原性。结果:PCR、限制性内切酶酶切和核酸序列检测证实重组质粒中插入的目的基因大小、方向正确;HPV58E6E7融合蛋白得到高效原核表达及纯化,表达蛋白的分子大小正确,抗原性良好。结论:pET-42a(+)-HPV58E6E7原核表达质粒构建成功,HPV58E6E7融合蛋白得到高效表达及有效纯化,为检测HPV58型治疗性疫苗的免疫效果提供了抗原。  相似文献

目的:克隆、表达人vasorin(VASN)蛋白。方法:利用PCR方法从HepG2细胞的cDNA中扩增获得目的基因,并插入带有6×His标签的原核高效可溶性表达载体pET28a中,构建重组表达质粒pET28a-VASN,将重组表达质粒转化大肠杆菌BL21(DE3),经IPTG诱导后目的基因获得表达,对融合目的蛋白进行Ni2+金属螯合柱纯化。结果:内切酶鉴定及基因序列测定证实重组表达质粒构建成功;对目的蛋白进行了原核表达,SDS-PAGE显示相对分子质量为61×103的特异表达条带;Western印迹证实目的蛋白为VASN,且主要以包涵体形式存在;对经尿素变性的表达产物进行了亲和层析纯化,有利于以后的变性、复性过程。结论:获得了人VASN融合蛋白,为其进一步的生物学功能研究奠定了基础。  相似文献

目的构建SAP 2重组原核表达载体并表达、纯化出可溶性的蛋白,为抗体制备及Sap2抗原检测奠定基础。方法提取白念珠菌基因组DNA为模板,经PCR方法获取SAP 2目的基因。双酶切SAP 2基因与原核表达载体pMAL-c2x(+),连接酶切产物,转化大肠杆菌TOP10感受态细菌,筛选菌落和测序鉴定。将pMAL-c2x/SAP2重组质粒转化大肠杆菌BL21(DE3)感受态细胞,经异丙基硫代-β-D-半乳糖苷(IPTG)诱导表达出可溶性的融合蛋白,经直链淀粉树脂亲和层析、蛋白酶Factor Xa切割标签获得纯化的Sap2蛋白。结果经PCR扩增获得正确的SAP 2序列并定向插入原核表达载体pMAL-c2x(+)中。重组原核表达载体pMAL-c2x/SAP2经IPTG诱导14 h后表达出可溶性的融合蛋白,并经纯化、切除标签后得到目的蛋白。结论成功构建了白念珠菌天冬氨酸蛋白酶原核表达质粒pMAL-c2x/SAP2,该质粒在BL21(DE3)中可获得高效融合表达,通过亲和层析纯化及标签切割得到了氨基酸序列同天然蛋白一致的目的蛋白。  相似文献

将薇甘菊Mmchi1基因cDNA序列的编码区插入到原核表达载体pET-32a(+)中,构建融合表达质粒,并在大肠杆菌中进行了融合蛋白6×His-Mmchi1的诱导表达、Western blot和酶活性分析.结果表明,0.1 mmol/L IPTG、25℃诱导4 h,在大肠杆菌Rosetta-gami(DE3)中能获得可溶性的6×His-Mmchi1;Western blot证实表达的6×His-Mmchi1能与抗6×His的单抗发生特异性反应,分子量约为55 kD,与预测的融合蛋白分子量相符;纯化后的6×His-Mmchi1最佳酶活性pH和温度分别为5.5~6.5和35~40℃.为进一步研究融合蛋白6×His-Mmchi1的功能奠定了基础.  相似文献

为获得大量高纯度的GhCOMT2蛋白以便研究其功能和性质,以pMD18-GhCOMT2质粒为模板,PCR扩增GhCOMT2基因的cDNA编码区,构建原核表达载体pET-28a-GhCOMT2,经酶切鉴定并测序后转化到大肠杆菌BL21 (DE3)中进行诱导表达,并采用Western blotting方法鉴定表达产物.结果表明:在大肠杆菌BL21(DE3)菌株中成功表达了与标签蛋白融合的GhCOMT2蛋白,大小约为40.062 kD,浓度为0.62 mg/mL.重组蛋白的最佳诱导条件为:0.2 mmol/L IPTG在16℃诱导12 h.重组蛋白以可溶形式高效表达,用蛋白标签亲和层析柱(His TrapTM HP)获得纯化重组蛋白,Western blotting分析表明其能与His多克隆抗体起特异性反应.  相似文献

根据基因库中查到的金黄色葡萄球菌肠毒素A(SEA)基因序列和人体表皮生长因子(EGF)基因序列进行密码子优化,以适于大肠杆菌表达.人工合成SEA基因与EGF基因.将两目的基因克隆至原核表达栽体pFT22b中,经测序验证表明成功构建了重组表达质粒pET22b-EGF-SEA.将构建好的pET22b-EGF-SEA质粒转化大肠杆菌BL21(DE3),经IPTG诱导进行表达;SDS-PAGE分析表明融合基因EGF-SEA在大肠杆菌BL21(DE3)中以包涵体的形式得到了高效表达,产物相对分子质量约为44kDa,与理论值大小一致.包涵体经洗涤,变性、复性后用His Bind Kit进行分离纯化,所得蛋白纯度≥95%.高纯度EGF-SEA融合蛋白的获得为进一步研究其生物学活性及肿瘤治疗奠定了基础.  相似文献

目的:表达和纯化人肿瘤坏死因子α抑制肽-抗炎酸性尾巴融合蛋白。方法:利用PCR搭接方法及基因合成方法获得目的基因,插入带有6×His标签的原核高效可溶性表达载体pET32a中,构建重组表达质粒pET32a-T9-ac-9,将重组表达质粒转化大肠杆菌BL21(DE3),经IPTG诱导目的基因表达;对融合蛋白进行Ni2+金属螯合柱纯化。结果:构建的重组表达质粒经PCR、内切酶鉴定及基因序列测定证实;目的蛋白在大肠杆菌中获得表达,SDS-PAGE显示相对分子质量为22.917×103;对表达产物进行了亲和层析纯化,从上清中获得了纯度较高的人肿瘤坏死因子α抑制肽-抗炎酸性尾巴融合蛋白。结论:获得了可溶性的人肿瘤坏死因子α抑制肽-抗炎酸性尾巴融合蛋白,为其生物学功能研究奠定了基础。  相似文献

猪繁殖与呼吸综合征(PRRS)是严重危害全球养猪业的重要病毒性传染病。通过PCR方法从高致病性PRRSV WUH1株中扩增得到结构蛋白ORF7的基因片段,并将目的基因插入原核表达载体pET-30a中,得到重组表达质粒pET-30a-ORF7。重组表达质粒转化大肠杆菌BL21(DE3)细胞,经IPTG诱导,SDS-PAGE聚丙烯酰胺凝胶电泳分析表明,融合蛋白得到了高效表达,表达的融合蛋白分子量约20kD。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. 传染性法氏囊病(Infection bursal disease, IBD)是由鸡传染性法氏囊病毒(Infectious bursal disease virus, IBDV)引起的鸡和火鸡的一种高度接触性传染病,给世界各国的禽养殖业带来了巨大损失.自IBDV发现至今新的变异株不断出现,分子结构的改变导致病毒致病力的改变及宿主对疫苗应答的改变,使得传统的疫苗已不能控制其流行,因此各国学者对其基因组结构和功能进行了广泛深入的研究,并积极研制新型有效的疫苗以达到防治的目的.  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献
