活性氧(reactive oxygen species,ROS)是植物体代谢所产生的小分子化合物,既是生长发育和防御途径的关键调节因子,又是需氧代谢的有毒副产物。植物细胞的生理过程受一个被活性氧调节的氧化还原网状途径的调控,本文从植物体内ROS产生的部位与时空特异性、ROS信号与NO和Ca2+波信号的互作等方面综述了ROS信号对植物抗性的调控作用研究进展。  相似文献

正常情况下植物细胞内活性氧(reactive oxygen species ROS)的产生和清除是平衡的,但是,一旦植物遭受环境胁迫,ROS的积累超过抗氧化剂防护系统清除能力,就会产生氧胁迫损伤细胞。由于叶绿体作为光合作用的场所与其他细胞器相比更易遭受氧化胁迫的伤害。因此,叶绿体进化了更强的防御机制调控电子传递链的氧化还原平衡及叶绿体基质中的氧化还原状态。活性氧具有双重效应.高浓度的活性氧对植物细胞有很强的毒害作用,低浓度时可充当信号分子参与植物的某些防卫反应过程,本文就叶绿体中活性氧的产生(三线态叶绿素、PSI和PSI I电子传递链)、网络清除(抗氧化剂,SOD,As—Glu循环系统,硫氧还蛋白)机制以及功能作用进行了综述。  相似文献

大量研究证明活性氧(ROS)在气孔运动中起信号分子的作用。保卫细胞中ROS的产生依赖于特定的酶, 其中NADPH氧化酶组分RBOH已得到深入研究, 并已证实其参与生物与非生物胁迫反应。植物激素包括脱落酸(ABA)、水杨酸(SA)、乙烯、生长素及细胞分裂素等, 它们均通过ROS的介导来调控气孔运动。生物胁迫(如毒性细菌和真菌)也会调控气孔运动。ROS参与这些调控过程。保卫细胞中存在多层次对ROS产生及其作用的调节, 抗氧化活性物质和ROS敏感蛋白(如蛋白激酶和磷酸酶)均可传递ROS信号并调节气孔运动。ROS对离子通道调节的证据也越来越多。保卫细胞由于可通过ROS整合复杂的信号途径, 已成为研究植物ROS信号转导过程的良好模式系统。  相似文献

过氧化物酶体是高度动态、代谢活跃的细胞器,主要参与脂肪酸等脂质的代谢及产生和清除不同的活性氧(reactive oxygen species, ROS)。ROS是细胞有氧代谢的副产物。当胁迫长期作用于植物,过量的ROS会引起氧胁迫,损害细胞结构和功能的完整性,导致细胞代谢减缓,活性降低,甚至死亡;但低浓度的ROS则作为分子信号,感应细胞ROS/氧化还原变化,从而触发由环境因素导致的过氧化物酶体动力学以及依赖ROS信号网络改变而产生快速、特异性的应答。ROS也可以通过直接或间接调节细胞生长来控制植物的发育,是植物发育的重要调节剂。此外,过氧化物酶体的动态平衡由ROS、过氧化物酶体蛋白酶及自噬过程调节,对于维持细胞的氧化还原平衡至关重要。本文就过氧化物酶体中ROS的产生和抗氧化剂的调控机制进行综述,以期为过氧化物酶体如何感知环境变化,以及在细胞应答中,ROS作为重要信号分子的研究提供参考。  相似文献

脱落酸(ABA)具有调节植物快速响应逆境的重要功能。植物细胞中ABA核心信号通路由ABA受体PYR1/PYLs/ RCARs、A类碱性蛋白磷酸酶PP2Cs和Snf1相关蛋白激酶SnRK2s组成。活性氧(ROS)和Ca2+是保卫细胞中的重要第二信使, 调控ABA诱导的气孔关闭。该文对保卫细胞中核心ABA信号蛋白的调控以及ROS和Ca2+介导的ABA信号转导等最新研究成果进行综述, 旨在阐明保卫细胞中ABA信号调控机制。  相似文献

活性氧(ROS)的产生是需氧代谢不可避免的结果。在植物细胞中,线粒体电子传递链(ETC)的复合物Ⅰ和Ⅱ是ROS产生的主要的部位。交替氧化酶和可能的内源鱼藤酮不敏感的NADH脱氢酶通过保持ETc的相对氧化状态限制线粒体产生ROS。线粒体基质中的抗氧化酶系统与小分子量的抗氧化剂一道起ROS的解毒作用。ROS除了引起细胞的伤害外,在植物中还能够作为一种普遍存在的信号分子起作用。在低浓度时,ROS能诱导防御基因的表达和引起适应反应;在高浓度时,引起细胞死亡。一氧化氮是植物合成和释放的一种气体,也可作为信号分子调节植物的生长和发育。  相似文献

活性氧(reactive oxygen species,ROS)是植物有氧代谢的副产物,同时环境胁迫也会使植物细胞中积累大量的活性氧。低浓度的活性氧可以作为信号分子存在,诱导防御基因的表达和植物对环境的适应反应。当逆境胁迫迫使植物细胞中产生大量活性氧时,就会导致细胞内的大分子物质及其他组分受损,阻碍植物的正常代谢和生长,甚至死亡。植物体内存在活性氧清除机制,可以在一定范围内维持活性氧的平衡。研究表明,利用植物体内自身的活性氧清除机制可以提高植物的抗逆性。对当前植物活性氧的研究动态进行概述,同时对植物活性氧清除机制在提高植物抗逆性方面的应用进行探讨。  相似文献

程序性细胞死亡(PCD)是生物体受遗传调控的自主细胞死亡现象, 在植物生长发育和抵抗环境胁迫中起重要作用。PCD的发生可受线粒体中活性氧(ROS)诱导。中国科学院遗传与发育生物学研究所李家洋研究组早期的研究发现了1个拟南芥(Arabidopsis thaliana)细胞死亡突变体mod1, 并暗示植物细胞中存在叶绿体与线粒体之间的信号交流调控PCD, 但其中的具体作用机制尚不清楚。最近, 他们通过大规模筛选mod1突变体的抑制突变体, 克隆了3个新的抑制基因plNAD- MDHDiT1mMDH1。此3个基因分别编码质体定位的NAD依赖的苹果酸脱氢酶、叶绿体被膜定位的二羧酸转运蛋白1和线粒体定位的苹果酸脱氢酶1, 突变后都可抑制mod1中ROS的积累及PCD的发生。通过对这些基因进行深入的功能分析, 他们论证了苹果酸从叶绿体到线粒体的转运对线粒体中ROS的产生及随后PCD的诱导起重要作用。该研究拓展了我们对植物细胞中细胞器间交流的认识, 为我们深入理解植物PCD发生机制提供了新线索, 是该领域的一项突破性进展。  相似文献

Reactive oxygen species (ROS) play a key signal transduction role in cells. They are involved in the regulation of growth, development, responses to environmental stimuli and cell death. The level of ROS in cells is determined by interplay between ROS producing pathways and ROS scavenging mechanisms, part of the ROS gene network of plants. Recent studies identified respiratory burst oxidase homologues (RBOHs) as key signaling nodes in the ROS gene network of plants integrating a multitude of signal transduction pathways with ROS signaling. The ability of RBOHs to integrate calcium signaling and protein phosphorylation with ROS production, coupled with genetic studies demonstrating their involvement in many different biological processes in cells, places RBOHs at the center of the ROS network of cells and demonstrate their important function in plants.  相似文献

越来越明显的证据表明,植物体十分活跃的产生着活性氧并将之作为信号分子、进而控制着诸如细胞程序性死亡、非生物胁迫响应、病原体防御和系统信号等生命过程,而不仅是传统意义上的活性氧是有氧代谢的附产物。日益增多的证据显示,由脱落酸、水杨酸、茉莉酸与乙烯以及活性氧所调节的激素信号途径,在生物和非生物胁迫信号的“交谈”中起重要作用。活性氧最初被认为是动物吞噬细胞在宿主防御反应时所释放的附产物,现在的研究清楚的表明,活性氧在动物和植物细胞信号途径中均起作用。活性氧可以诱导细胞程序性死亡或坏死、可以诱导或抑制许多基因的表达,也可以激活上述级联信号。近来生物化学与遗传学研究证实过氧化氢是介导植物生物胁迫与非生物胁迫的信号分子,过氧化氢的合成与作用似乎与一氧化氮有关系。过氧化氢所调节的下游信号包括钙“动员”、蛋白磷酸化和基因表达等。  相似文献

Stomata, the pores formed by a pair of guard cells, are the main gateways for water transpiration and photosynthetic CO_2 exchange, as well as pathogen invasion in land plants. Guard cell movement is regulated by a combination of environmental factors, including water status, light, CO_2 levels and pathogen attack, as well as endogenous signals, such as abscisic acid and apoplastic reactive oxygen species(ROS). Under abiotic and bioticstress conditions, extracellular ROS are mainly produced by plasma membrane-localized NADPH oxidases, whereas intracellular ROS are produced in multiple organelles. These ROS form a sophisticated cellular signaling network, with the accumulation of apoplastic ROS an early hallmark of stomatal movement. Here, we review recent progress in understanding the molecular mechanisms of the ROS signaling network,primarily during drought stress and pathogen attack. We summarize the roles of apoplastic ROS in regulating stomatal movement, ABA and CO_2 signaling, and immunity responses.Finally, we discuss ROS accumulation and communication between organelles and cells. This information provides a conceptual framework for understanding how ROS signaling is integrated with various signaling pathways during plant responses to abiotic and biotic stress stimuli.  相似文献

Arabidopsis thaliana plants with null mutations in the genes encoding the alpha and beta subunits of the single heterotrimeric G protein are less and more sensitive, respectively, to O3 damage than wild-type Columbia-0 plants. The first peak of the bimodal oxidative burst elicited by O3 in wild-type plants is almost entirely missing in both mutants. The late peak is normal in plants lacking the Gbeta protein but missing in plants lacking the Galpha protein. Endogenous reactive oxygen species (ROS) are first detectable in chloroplasts of leaf epidermal guard cells. ROS production in adjacent cells is triggered by extracellular ROS signals produced by guard cell membrane-associated NADPH oxidases encoded by the AtrbohD and AtrbohF genes. The late, tissue damage-associated component of the oxidative burst requires only the Galpha protein and arises from multiple cellular sources. The early component of the oxidative burst, arising primarily from chloroplasts, requires signaling through the heterotrimer (or the Gbetagamma complex) and is separable from Galpha-mediated activation of membrane-bound NADPH oxidases necessary for both intercellular signaling and cell death.  相似文献

Reactive oxygen species (ROS) are known as toxic metabolic products in plants and other aerobic organisms. An elaborate and highly redundant plant ROS network, composed of antioxidant enzymes, antioxidants and ROS-producing enzymes, is responsible for maintaining ROS levels under tight control. This allows ROS to serve as signaling molecules that coordinate an astonishing range of diverse plant processes. The specificity of the biological response to ROS depends on the chemical identity of ROS, intensity of the signal, sites of production, plant developmental stage, previous stresses encountered and interactions with other signaling molecules such as nitric oxide, lipid messengers and plant hormones. Although many components of the ROS signaling network have recently been identified, the challenge remains to understand how ROS-derived signals are integrated to eventually regulate such biological processes as plant growth, development, stress adaptation and programmed cell death.  相似文献

Reactive oxygen species (ROS) and reactive nitrogen species (RNS) constitute key features underpinning the dynamic nature of cell signaling systems in plants. Despite their importance in many aspects of cell biology, our understanding of oxidative and especially of nitrosative signaling and their regulation remains poorly understood. Early reports have established that ROS and RNS coordinately regulate plant defense responses to biotic stress. In addition, evidence has accumulated demonstrating that there is a strong cross-talk between oxidative and nitrosative signaling upon abiotic stress conditions. The goal of this mini-review is to provide latest findings showing how both ROS and RNS comprise a coordinated oxidative and nitrosative signaling network that modulates cellular responses in response to environmental stimuli.Key words: abiotic stress, nitrosative stress, oxidative stress, reactive nitrogen species, reactive oxygen species, signaling  相似文献

Several reactive oxygen species (ROS) are continuously produced in plants as byproducts of many metabolic reactions, such as photosynthesis, photo respiration and respiration, Depending on the nature of the ROS species, some are highly toxic and rapidly detoxified by various cellular enzymatic and nonenzymatic mechanisms. Oxidative stress occurs when there is a serious imbalance between the production of ROS and antioxidative defence. ROS participate in signal transduction, but also modify cellular components and cause damage. ROS is highly reactive molecules and can oxidize all types of cellular components. Various enzymes involved in ROS-scavenging have been manipulated and over expressed or down regulated. An overview of the literature is presented in terms of primary antioxidant free radical scavenging and redox signaling in plant cells. Special attention is given to ROS and ROS-anioxidant interaction as a metabolic interface for different types of signals derived from metabolisms and from the changing environment.  相似文献

活性氧(ROS)是一类由O2转化而来的自由基或具有高反应活性的离子或分子。植物消耗的O2约有1%在叶绿体、线粒体、过氧化物酶体等多种亚细胞单位中被转化成了ROS。ROS有益或有害取决于它在植物体内的浓度。低浓度的ROS作为第二信使能在植物细胞信号转导途径中介导多种应答反应,高浓度的ROS则引起生物大分子的氧化损伤甚至细胞死亡。植物体内ROS产生和清除之间的平衡十分重要,并由一套有效的酶促和非酶促抗氧化系统来监控。该文主要系统介绍了植物ROS的种类、产生部位、在信号转导中的作用及其对植物细胞造成的主要伤害等方面的研究进展,为利用基因工程手段来提高植物对环境胁迫的抗性提供信息和思路。  相似文献
