模拟人体胃肠道环境筛选益生乳杆菌   总被引:7,自引:1,他引:6  
【目的】筛选具有益生特性的乳杆菌作为保健型酸奶的候选菌株。【方法】从健康人肠道和奶豆腐中分离筛选出耐受人工胃液的乳杆菌,对其进行体外益生特性(人工胃肠液耐受性、胆盐耐受性、抑菌活性及胆固醇降解能力)研究。【结果】从在乳杆菌分离培养基上有溶钙圈的41株菌株中筛选出5株耐酸、耐人工胃液较强的菌株,经16S rR NA基因测序鉴定,其中3株为乳杆菌,分别命名为植物乳杆菌Lp MT-3、植物乳杆菌Lp MT-5和唾液乳杆菌LsA F-7。在人工胃液中3株菌的耐受力均强于商品化的对照菌株LGG(鼠李糖乳杆菌GG);转入肠液4 h后直至26 h,Lp MT-5存活率基本稳定在45%左右,仅次于LGG。胆盐浓度为0.10%时,3株乳杆菌的耐胆盐能力均强于LGG;胆盐浓度为0.20%时,Lp MT-3和LsA F-7仍能存活。3株乳杆菌均具有抑菌活性,对粪肠球菌的抑制最明显,其次是金黄色葡萄球菌,对大肠杆菌、沙门氏菌的抑制作用较差。3株乳杆菌对胆固醇的清除效力依次为Lp MT-3LpM T-5Ls AF-7;清除率依次为Ls AF-7Lp MT-3LpM T-5。【结论】筛选出3株适应人体胃肠液环境、耐胆盐、抑菌及降胆固醇活力强的乳杆菌,可作为进一步开发新的益生菌产品和保健型酸奶的菌株。  相似文献

目的探究家蝇幼虫肠道细菌对幼虫取食与发育的影响,进而明确肠道细菌的部分功能。方法取3日龄家蝇幼虫,采用细菌传统分离法分离纯化其肠道内可培养细菌并进行分子鉴定,利用这些肠道细菌进行家蝇幼虫爬行选择实验,测定幼虫对不同细菌的选择性;利用这些肠道细菌发酵液与麦麸混合饲养家蝇幼虫,记录家蝇幼虫发育过程中的生物学参数,分别以LB培养原液与麦麸混合饲养的幼虫为阴性对照,以无菌水与麦麸混合饲养的幼虫为空白对照。结果在家蝇幼虫肠道中共分离出9个属10种细菌,分别为霍氏肠杆菌(Enterobacter hormaechei)、肺炎克雷伯菌(Klebsiella pneumoniae)、铜绿假单胞菌(Pseudomonas aeruginosa)、别雷斯不动杆菌(Acinetobacter bereziniae)、斯氏普罗威登斯菌(Providencia stuartii)、阴沟肠杆菌(Enterobacter cloacae)、乳酸乳球菌(Lactococcus lactis)、纺锤形赖氨酸芽胞杆菌(Lysinibacillus fusiformis)、居幼虫普罗威登斯菌(Providencia vermicola)和沙福芽胞杆菌(Bacillus safensis)。其中,除铜绿假单胞菌和斯氏普罗威登斯菌外,其余8种细菌对幼虫有显著吸引作用;除铜绿假单胞菌对家蝇生长有明显抑制作用以外,其余9种细菌对家蝇幼虫生长均有一定的促进作用,其中霍氏肠杆菌和别雷斯不动杆菌对其生长有显著促进作用。结论明确了家蝇幼虫肠道细菌的组成,确定了8种家蝇幼虫肠道细菌对其取食有吸引作用,9种细菌对其生长发育有促进作用。霍氏肠杆菌和别雷斯不动杆菌可作为添加剂用于家蝇的规模化饲养,以提高养殖效率。铜绿假单胞菌可作为生长抑制剂用于家蝇幼虫的防治。  相似文献

采用体外试验方法研究7株乳酸菌和双歧杆菌对人体消化道不良环境的抵抗能力、在肠道上皮的黏附定植能力及其降胆固醇潜力。结果表明:受试菌对CCL-187细胞粘附能力存在显著差异(P<0.05),粪肠球菌M1粘附能力最强;各菌对胃液和胰液都有较强耐受能力,在模拟胃液和胰液中分别培养3h和4h后仍能保证95%以上存活;在含胆汁培养基中受到不同程度抑制,但都能存活;受试菌在生长过程中能够使环境中胆固醇降低从25%~54%不等,粪肠球菌A30和A31和嗜酸乳杆菌A878和PB1能使胆固醇降低40%以上,是良好的降胆固醇备选菌株。  相似文献

【目的】对3株罗伊氏乳杆菌的生物学特性进行分析比较,为后期生产应用提供一定的参考。【方法】对实验室保藏的3株罗伊氏乳杆菌的生长曲线、pH曲线、耐受人工胃液能力、耐受猪胆盐能力、黏附能力、抑菌能力和对抗生素的耐药性等特性进行了分析比较。【结果】3株菌生长趋势大致相同;3株菌对人工胃液均具有良好的耐受性,且可以有效地抑制大肠杆菌和金黄色葡萄球菌的生长;菌株L0和L2对高胆盐的环境耐受性较差,菌株L1则对高胆盐环境具有极强的耐受性;菌株L1和L2具有很强的黏附能力;3株菌对20种抗生素表现出不同的耐受性。【结论】菌株L1的生物学特性明显优于其他两株菌株,有利于后期的生产应用。  相似文献

采用半选择性培养基MRS从传统发酵食品中筛选出2株细菌,经鉴定一株为双歧杆菌,另一株为乳酸杆菌.按照益生菌标准对上述2株菌的耐酸、耐胆盐、抗药性、抑菌等性能进行评价,结果表明其均可作为益生菌用于降胆固醇产品.  相似文献

为探究秦岭地区野生细鳞鲑(Brachymystax lenok)肠道细菌组成多样性,筛选出产胞外酶菌株,利用传统分离培养并分子鉴定的方法和基于16S r RNA基因克隆的现代分子生物技术相结合测定秦岭野生细鳞鲑肠道细菌菌群多样性并构建系统发育树,利用淀粉酶、蛋白酶、纤维素酶及脂肪酶4种胞外酶筛选培养基筛选出产上述酶的细菌。细菌传统分离培养并分子鉴定法从细鳞鲑肠道获得18个属的细菌类群,分别归属于变形菌门、拟杆菌门和厚壁菌门,其中,气单胞菌属(Aeromonas)为优势菌群。基于16S r RNA基因克隆的现代分子方法获得22个属的细菌类群,分别归属于变形菌门、拟杆菌门、厚壁菌门和放线菌门,其中,鞘氨醇杆菌属(Sphingomonas)为优势菌群。4种胞外酶筛选获得53株细菌产胞外酶,其中21株可在低温(10℃)环境下产胞外酶。结果表明,传统分离培养法与基于16S r RNA基因克隆的现代分子生物技术相结合能够更有效全面地分析细鳞鲑鱼肠道微生物的多样性,并且细鳞鲑肠道微生物具有一定的产酶活性。  相似文献

摘要:目的 分离小菌虫肠道可培养细菌,并研究其产消化酶活性,探讨肠道细菌对小菌虫消化食物的影响。方法 采用传统细菌分离培养方法分离小菌虫肠道细菌,利用16S rDNA序列进行细菌分子鉴定;利用筛选培养基鉴别各细菌的产蛋白酶、脂肪酶、淀粉酶和纤维素酶活性。结果 在小菌虫肠道中分离到4种可培养细菌,分别是枯草芽胞杆菌(Bacillus subtilis)、肺炎克雷伯菌(Klebsiella pneumoniae)、Pseudocitrobacter faecalis和芽胞杆菌(Bacillus sp.)。其中,2种芽胞杆菌属细菌有产消化酶活性。枯草芽胞杆菌有产纤维素酶、淀粉酶和蛋白酶活性;芽胞杆菌仅有产蛋白酶活性,但产酶能力低于枯草芽胞杆菌。结论 小菌虫肠道细菌中可培养细菌结构简单,但其中的芽胞杆菌属细菌有产纤维素酶、淀粉酶和蛋白酶能力,说明小菌虫肠道中的2种芽胞杆菌属细菌可能有协助小菌虫进行食物消化的功能。  相似文献

目的比较7株乳酸菌各项生理特性,筛选高效、安全降解胆固醇的益生菌株,为后期开发应用奠定基础。方法对选定菌株依次进行体外降胆固醇、胆盐水解酶(BSH)活力、人工胃肠液及胆盐耐受性、细胞粘附性能的测定和比较,并分析抗生素耐药性和抑制病原菌活性,最后利用16S rRNA基因测序确定细菌种属。结果不同菌株各项生理指标存在差异。其中菌株ZL-2、JQI-7和PC-26的BSH酶活分别为0.010、0.015和0.030 U,可高效清除体外胆固醇,3株菌体外降解胆固醇率在58%以上;并且耐酸、耐胆盐,具备较好的HT--29细胞粘附性,不存在抗生素耐药性,可有效抑制大肠埃希菌、金黄色葡萄球菌、铜绿假单胞菌和阴沟肠杆菌生长。结论菌株ZL-2、JQI-7和PC-26满足降胆固醇益生菌株的生物学特性,可用于进一步研究和开发。  相似文献

目的通过体外实验从发酵制品及人体肠道内分离筛选出安全高效的降胆固醇乳酸菌,并研究其生理生化特性,为今后功能性乳制品的开发提供优良菌种。方法应用MRS.胆固醇培养基液体发酵法进行目的菌株筛选,并测定其耐酸及耐胆盐等能力。结果筛选出4株具有降胆固醇能力的乳酸菌,其降解率分别为DM8403163.1%、DM8403470.3%、DM850381.1%、DM8605663.1%;同时4株乳酸菌均表现出不同程度的耐酸及耐胆盐能力,其中DM86056能力最强,其他3株乳酸菌次之;经鉴定,DM84031为保加利亚乳杆菌、DM84034为嗜热链球菌、DM8503为詹氏乳杆菌、DN86056为屎肠球菌。结论经上述实验综合比较,DM86056降胆固醇能力比较强且耐酸及耐胆盐等能力也较强,因此有待于将其应用于动物体内进行深入研究。  相似文献

目的研究3株益生菌株的粘附性质,初步探讨细菌与肠细胞的粘附机制。方法选取已被深入研究的LGG作为阳性对照,应用显微观察和平板计数的方法测定3株乳杆菌和肠上皮细胞Caco-2的粘附,选择疏水性、表面电荷和自聚合能力3项指标来描述细菌的表面性质,应用粘附抑制试验和酵母凝集试验来测定糖类专一性粘附。结果无论是用显微观察还是平板计数,ST-Ⅲ均是所测试的4株菌中粘附能力最强的,当加入细菌和细胞比约为60∶1时,ST-Ⅲ在Caco-2上的粘附数为(7.43±0.65)CFU/细胞,强于对照菌LGG[(3.99±0.57)CFU/细胞]。在所测试的4株菌中,粘附能力和疏水性具有很好的相关性,同时自聚合能力对粘附也表现出一定的促进作用。除LGG外,D-甘露糖和甲基-α-D-甘露糖苷均能抑制另外3株乳杆菌的粘附,同时这3株乳杆菌也能不同程度地凝集酵母。结论3株益生菌的粘附均涉及到非特异性的疏水结合和甘露糖特异性粘附两个过程;ST-Ⅲ是1株具有良好粘附能力的益生菌,有待进一步的研究。  相似文献

One hundred and six strains from the Proteus-Providence group and 27 other strains from the rest of the Enterobacteriaceae were subjected to 178 morphological. physiological and biochemical tests and the results analysed by computer. Most of the Proteus-Providence strains grouped into six main clusters; (1) Proteus mirabilis , (2) Proteus vulgaris , (3) Proteus morganii , (4) Providencia alcalifaciens, Shigella dysenteriae , (5) Proteus rettgeri , (6) Providencia stuartii . On the basis of these groupings a scheme has been drawn up for distinguishing between the different taxa in the Proteus-Providence group.  相似文献

To screen the lactic acid bacteria with cholesterol-lowering and triglyceride-lowering activity in vitro and evaluate their probiotic function. By plate separating, cholesterol-lowering and triglyceride-lowering activity in vitro were determined; and by evaluating the probiotic functions, including tolerances to simulated gastric and intestinal juice, the antibacterial spectrum, and the adhesion ability to Caco-2 cells, the probiotic strains with cholesterol-lowering and triglyceride-lowering activity in vitro were screened, and then were identified by phenotypical and physiological tests and 16Sr DNA. Finally, the cholesterol-lowering and triglyceride-lowering activity in vivo of the strains were evaluated using male Sprague-Dawley rats. Two strains L2-16 and L2-73 with stronger cholesterol-lowering and triglyceride-lowering activity in vitro, stronger tolerance to simulated gastric and intestinal juice and adhesion ability to Caco-2 cells, and wider antibacterial spectrum were screened from traditional Chinese fermented cucumber and were identified as Lactobacillus acidophilus and Enterococcus faecalis, respectively. Compared with a hyperlipidemia diet without lactic acid bacteria, the diet supplemented with Lactobacillus acidophilus L2-16 and Enterococcus faecalis L2-73 significantly reduced serum total cholesterol, triglycerides, and low-density lipoprotein cholesterol levels, and liver total cholesterol and triglyceride levels in rats (P?<?0.05). Moreover, the diet supplemented with Lactobacillus acidophilus L2-16 and Enterococcus faecalis L2-73 significantly increased the fecal elimination of bile acids (P?<?0.05). Lactobacillus acidophilus L2-16 and Enterococcus faecalis L2-73 may have application prospect in the production of some fermented foods such as fermented vegetables, milk, or meat, and probiotic preparations with the function to lower the serum lipid and liver lipid levels.  相似文献

In Proteus morganii, P. mirabilis, and Providencia stuartii the ability to ferment trehalose and resistance to tetracycline were associated in 90%-97% of the strains. The same was true of at least 78% of the strains of Serratia marcescens. Proteus vulgaris showed a more quantitative association of the two traits. As the characters occur independently in 3-10% of the strains, the association is considered to be due to simultaneous selection in some natural niche. The trehalose fermenting, tetracycline and polymyxin resistant species ferment few other carbohydrates, fewer than the remainder of the Serratia species.  相似文献

The lipid composition of Providencia stuartii has been shown to resemble closely that of Proteus mirabilis. The ability of some Pv, stuartii strains to survive exposure to high concentrations of the antiseptic chlorhexidine could not be explained in terms of differences in lipid content between sensitive and resistant strains. In addition, resistance could not be attributed to either reduced adsorption of the antiseptic or to its gross enzymic degradation.  相似文献

目的观察新疆传统发酵乳品中分离的14种菌株的生长特点及产酸能力,筛选出具有较强耐胆盐能力,并能在人工胃肠液中存活的菌株。方法对10株乳酸菌和4株酵母菌进行生长曲线、pH、耐胆盐能力和耐人工胃肠液检测。结果 10株乳酸菌和4株酵母菌具有良好的生长曲线和产酸能力;马乳酒样乳杆菌具有较强的耐胆盐能力;希氏乳杆菌、马乳酒样乳杆菌、乙醇假丝酵母和东方伊萨酵母具有较强的耐人工胃液能力;乳酸乳球菌、哈尔滨乳杆菌、瑞士乳杆菌、马乳酒样乳杆菌、乙醇假丝酵母和东方伊萨酵母具有较强的耐人工肠液能力。结论 10株乳酸菌和4株酵母菌具有优良的益生特性,有望成为益生菌制剂的备用菌株。  相似文献

Plasmid-encoded urease gene clusters found in uropathogenic isolates of Escherichia coli, Providencia stuartii, and Salmonella cubana demonstrated DNA homology, similar positions of restriction endonuclease cleavage sites, and manners of urease expression and therefore represent the same locus. DNA sequence analysis indicated that the plasmid-encoded urease genes are closely related to the Proteus mirabilis urease genes.  相似文献

Outer membrane fractions were prepared from 11 bacteria in the family Enterobacteriaceae: Escherichia coli serotypes O1K-, O4K2, O26K60, O75K-, and O111K58, Shigella flexneri, Salmonella typhimurium, Klebsiella pneumonia, Serratia marcescens, Proteus vulgaris, Proteus mirabilis, and Providencia stuartii. All strains studied were found to contain one non-peptidoglycan-bound, heat-modifiable outer membrane protein, and one or two peptidoglycan-associated major outer membrane proteins in the 27,000- to 40,000-dalton range. Crossed immunoelectrophoresis using sodium dodecyl sulfate-polyacarylamide gel electrophoresis for separation of the antigens in the first dimension of the procedure was shown to provide a useful model system for studying the antigenic relationships of the major outer membrane proteins in Enterobacteriaceae species. Peptidoglycan-bound major outer membrane proteins of all bacteria studied reacted with antiserum against the purified peptidogylcan-bound matrix protein I of E. coli O26K60 in this system. Non-peptidoglycan-associated proteins of all strains cross-reacted with protein II of E. coli O26K60 in both their unmodified and their heat-modified forms. These results indicate that the genes coding for the major outer membrane proteins in the family Enterobacteriaceae have been well enough conserved during the course of evolution to allow significant antigenic cross-reactivity between the corresponding proteins in different enterobacterial species.  相似文献
