目的:探讨E一选择素(E-selectin)基因多态性与新疆哈萨克族患者脑梗死(cebreral infarction,CI)的关系。方法:采用聚合酶链反应一限制性片段长度多态性(polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)和DNA序列测定法检测103例CI及110例对照组E-selectin基因第4外显子A561C(S128R)、第10外显子C1839T(L554F)多态性。结果:E-se-lectin基因S128R基因型频率和等位基因频率在CI组和对照组比较差异有显著性(P〈0.05),基因型频率的相对风险分析发现,SR基因型携带者患CI的风险是SS基因型的2.355倍(OR=2.355,95%CI:1.209~4.588);E-selectin L554F基因型在两组中的分布差异有显著性(X2=5.463,P〈0.05),基因型频率的相对风险分析,LF基因型患CI的风险是LL基因型的2.315倍(OR=2.315,95%CI:1.132~4.737)。结论:E-selectin S128R和L554F多态性与脑梗死易感性有关;R等位基因和F等位基因可能是新疆哈萨克族CI发病的遗传易感基因。  相似文献

为了研究人类自然抵抗相关巨噬细胞蛋白NRAMP1基因3′UTR多态性与新疆哈萨克族结核病易感性的关系,研究选取新疆哈萨克族活动性结核病患者213例,新疆哈萨克族正常对照者211人,用聚合酶链反应-限制性片断长度多态性(PCR-RFLP)分析的方法对NRAMP1基因3′UTR多态性进行基因分型,比较不同基因型及等位基因的频率,经统计学分析,探讨NRAMP1基因3′UTR多态性与新疆哈萨克族结核病易感性的关系.结果显示,在新疆哈萨克族活动性结核病患者组中NRAMP1基因3′UTR TGTG+/TGTG+基因型138例(64.8%),TGTG+/del基因型63例(29.6%),del/del基因型12例(5.6%);新疆哈萨克族正常对照组TGTG+/TGTG+基因型则为167例(79.1%),TGTG+/del基因型41例(19.5%),del/del基因型3例(1.4%).新疆哈萨克族结核病患者组TGTG+/del基因型和del/del基因型频率明显高于新疆哈萨克族正常对照组,差异有统计学意义(χ2=10.8,P0.01);在新疆哈萨克族结核病患者组的TGTG+等位基因频率为79.6%,del等位基因频率为20.4%,新疆哈萨克族正常对照组中TGTG+和del等位基因频率分别为88.9%和11.1%,del等位基因在新疆哈萨克族结核病患者组中的分布频率高,差异有统计学意义(χ2=13.7,P0.01).研究结果提示TGTG缺失等位基因可能是新疆哈萨克族结核病的易感基因,携带TGTG+/del和del/del基因型的新疆哈萨克族人群可能更易患结核病.  相似文献

目的:研究HLA-DRB1基因多态性与新疆哈萨克族人群结核病(TB)的相关性。方法:采用病例-对照的研究方法,应用聚合酶链反应-序列特异性引物(PCR-SSP)技术对231例新疆哈萨克族肺结核患者和230例新疆哈萨克族健康对照者的13个HLA-DRB1等位基因进行分型,比较其等位基因频率(GF)并计算其比值比(OR)。结果:与新疆哈萨克族人群对照组相比,新疆哈萨克族人群结核病例组中HLA-DRB1*04显著增高(11.72%比6.75%,p0.05,OR=1.889),HLA-DRB1*10也增高(2.86%比1.09%),但统计学上无显著性差异(Pc0.05)。结论:HLA-DRB1*04可能是新疆哈萨克族人群结核病的易感基因。  相似文献

运用Meta分析的方法综合评价低密度脂蛋白受体相关蛋白基因(lowdensitylipoproteinreceptor-relatedprotein1gene,LPR-1)第三外显子C766T多态性与阿尔茨海默氏病(Alzheimer’sdisease,AD)发病风险的关系。通过检索Medline、Cochrane图书馆和中国生物医学文献数据库(CBM),纳入内容涉及LPR-1基因C766T多态性的基因型频率和(或)等位基因频率的独立病例对照研究。各文献满足研究方法相似,有综合的统计指标。研究年限为1997~2004年,语种不限。应用RevMan4.2软件进行统计分析。在AD组3764例和对照组3647例研究对象中,分别对基因型和等位基因频率做合并统计。各研究之间有显著的异质性,P<0.01,故采用随机效应模型分析合并结果。总体效应检验未发现CC基因型频率在病例对照之间统计学上的差异(Z=1.74,P=0.08,OR=1.17,95%CI:0.98~1.39),C等位基因频率在病例对照之间也未发现差异(Z=1.31,P=0.19,OR=1.11,95%CI:0.95~1.31)。Meta分析结果提示,LPR-1基因C766T多态性不是AD的主要风险因子,但尚不能完全排除其可能在AD发病中具有微弱的作用。  相似文献

目的:探讨亚甲基四氢叶酸还原酶(MTHFR)基因多态性与中国北方人群脑膜瘤发病的相关性。方法:选择2012年1月-2013年12月在黑龙江省哈尔滨医科大学附属第二医院的第一、三病房接受手术治疗的脑膜瘤患者317例(实验组)及320例非脑膜瘤患者(对照组)为研究对象,利用聚合酶链反应限制性多态性片段长度(PCR-RFLP)检测和比较两组MTHFR两个单核苷酸多态性位点(C677T、A1298C)各种基因型(CC、CT、TT)的分布情况及等位基因的频率。结果:两组MTHFR的C677T中CC基因型的频率和TT基因型的频率比较有显著性差异(CC:OR=2.012,95%CI=1.460-2.772;TT:OR=0.399,95%CI=0.254-0.628,P0.05),实验组MTHFR的(0.450)C677T中的T等位基因频率明显高于对照组(0.320)(OR=0.529,95%CI=0.420-0.666,P0.05)。两组A1298C的等位基因分布比较没有统计学差别(P0.05)。结论:MTHFR基因的C677T中TT等位基因提示潜在的易患脑膜瘤的风险,而CC等位基因会降低中国北方人群患脑膜瘤的风险。  相似文献

目的:探讨代谢酶CYP1A1基因MspI位点多态性与新疆汉族人群肺癌遗传易感性之间的相关性.方法:应用聚合酶链式反应(PCR)-限制性片段长度多态性(RFLP)技术检测59例新疆汉族肺癌和84例新疆汉族健康人的CYP1A1基因MspI位点多态性分布频率,并分析了CYP1A1基因MspI位点多态性与新疆汉族人群肺癌遗传易感性和患者性别之间的相关性.结果:(1)CYP1A1基因MspI位点3种多态基因型分布频率在两组间比较差异有统计学意义(χ2=6.682,P=0.035),CC基因型在病例组的分布频率显著高于正常对照组.(2)携带突变CC基因型的个体较携带TT基因型的个体患肺癌的危险性增加(OR=3.759.95%CI=1.228-11.494,P=0.035).(3)男女肺癌患者的CYP1A1基因MspI位点基因型及等位基因频率的差异均无显著性(P>0.05).结论:(1)CC突变基因型可能是新疆汉族人群的肺癌易感因素.(2)CYP1A1基因MspI位点多态性可能与新疆汉族肺癌患者的性别无关.  相似文献

目的:探讨多巴胺D4受体基因启动子区-1240L/S,-616C/G和-521C/T三个多态性与注意缺陷多动障碍(Attention deficit hyperactivity disorder,ADHD)的关系.方法:取无亲缘关系的ADHD患者及对照组各166名,采用等位基因特异性扩增技术(allele specific amplification,ASA)及聚合酶链式反应琼脂糖凝胶电泳技术,检测ADHD患者和对照组基因型和等住基因的频率分布.结果:DRD4基因-521C/T的基因型及等位基因频率分布在ADHD组与正常对照组存在显著性差异(p<0.05),ADHD组的T等位基因的频率显著高于正常对照组(x2=9.827,p=0.002,OR=1.639,95%CI=1.202-2.234).DRD4基因启动子区2个功能多态性位点-616C/G及-1240L/S的基因型及等位基因频率在正常组与ADHD组的分布无显著性差异(p>0.05).结论:-521C/T位点与ADHD的易感性存在关联,且-521C/T等位基因是决定ADHD个体易感性的重要因素,含有T等位基因的个体罹患注意缺陷多动障碍的相对风险增高.  相似文献

采用PCR RFLP方法对蒙古绵羊和哈萨克绵羊MHC DRB3 基因第 2外显子 2 85bp的扩增产物进行多态性分析 ,共检测到 17种基因型 ,由A、B、C、D、E、F和H共 7个复等位基因控制。通过酶切图谱分析表明 ,蒙古绵羊和哈萨克绵羊的MHC DRB3 基因第 2外显子的第 15 4、16 8和 2 2 0位的碱基表现出多态性。统计分析表明 ,MHC DRB3 基因的部分基因型频率和等位基因频率在两个群体之间差异显著或极显著 (P <0 10、P <0 0 5或P <0 0 1)。χ2 适合性检验结果表明 ,蒙古绵羊和哈萨克绵羊的MHC DRB3 基因第 2外显子的HaeⅢ酶切位点均未达到Hardy Weinberg平衡状态 (P <0 0 1)。  相似文献

采用限制性内切核酸酶HaeⅢ对蒙古山羊和哈萨克山羊GOLA-DRB3基因外显子2的285bp扩增产物进行了PCR-RFLP多态性分析,共检测到17种基因型,由A、B、C、D、E、F和H等7个复等位基因控制;通过酶切图谱分析发现蒙古山羊和哈萨克山羊的GOLA-DRB3基因外显子2的154、168和220位碱基表现出多态性。并对基因型频率和等位基因频率进行了统计分析,结果表明,GOLA-DRB3基因的部分基因型频率和等位基因频率在两个群体之间差异显著(P<0.10或P<0.05)或极显著(P<0.01); χ2适合性检验结果表明,蒙古山羊和哈萨克山羊的GOLA-DRB3基因外显子2的HaeⅢ酶切位点均未达到Hardy-Weinberg平衡状态(P<0.01)。 Abstract:The exon2 of GOLA-DRB3 gene was amplified and a uniform fragment of 285bp was obtained in Mongolian Goat and Kazakh Goat.The 285bp PCR product was digested with restriction endomuclease HaeⅢ and genetic polymorphism was investigated by PCR-RFLP.Seventeen kinds of genotypes were found in two populations,which were controlled by seven alleles.There are significant differences in some genotypic frequencies and gene frequencies between the two populations(P<0.10,P<0.05,P<0.01);The results of χ2 test showed that genotypes of GOLA-DRB3 gene in two populations did not fit with Hardy-Weinberg equilibrium(P<0.01).  相似文献

Background

The α-adducin Gly460Trp (G460W) gene polymorphism may be associated with susceptibility to essential hypertension (EH), but this relationship remains controversial. In an attempt to resolve this issue, we conducted a meta-analysis.

Methods

Twenty-three separated studies involving 5939 EH patients and 5021 controls were retrieved and analyzed. Four ethnicities were included: Han, Kazakh, Mongolian, and She. Eighteen studies with 5087 EH patients and 4183 controls were included in the Han subgroup. Three studies with 636 EH patients and 462 controls were included in the Kazakh subgroup. The Mongolian subgroup was represented by only one study with 100 EH patients and 50 controls; similarly, only one study with 116 EH patients and 326 controls was available for the She subgroup. The pooled and ethnic group odds ratios (ORs) along with the corresponding 95% confidence intervals (95% CI) were assessed using a random effects model.

Results

There was a significant association between the α-adducin G460W gene polymorphism and EH in the pooled Chinese population under both an allelic genetic model (OR: 1.12, 95% CI: 1.04–1.20, P = 0.002) and a recessive genetic model (OR: 1.40, 95% CI: 1.16–1.70, P = 0.0005). In contrast, no significant association between the α-adducin G460W gene polymorphism and EH was observed in the dominant genetic model (OR: 0.88, 95% CI: 0.72–1.09, P = 0.24). In stratified analysis by ethnicity, significantly increased risk was detected in the Han subgroup under an allelic genetic model (OR: 1.13, 95% CI: 1.04–1.23, P = 0.003) and a recessive genetic model (OR: 1.43, 95% CI: 1.17–1.75, P = 0.0006).

Conclusions

In a Chinese population of mixed ethnicity, the α-adducin G460W gene polymorphism was linked to EH susceptibility, most strongly in Han Chinese.  相似文献

Objective

Epidemiological studies have shown that E-selectin gene polymorphisms (A561C and C1839T) may be associated with essential hypertension (EH), but the results are conflicting in different ethnic populations. Thus, we performed this meta-analysis to investigate a more authentic association between E-selectin gene polymorphisms and the risk of EH.

Methods

We searched the relevant studies for the present meta-analysis from the following electronic databases: PubMed, Embase, Cochrane Library, Google Scholar, Web of Science, Wanfang Data, and China National Knowledge Infrastructure (CNKI). Odds ratios (OR) with 95% confidence interval (CI) were used to evaluate the strength of the association between E-selectin gene polymorphisms and EH susceptibility. The pooled ORs were performed for dominant model, allelic model and recessive model. The publication bias was examined by Begg’s funnel plots and Egger’s test.

Results

A total of eleven studies met the inclusion criteria. All studies came from Asians. Ten studies (12 cohorts) evaluated the A561C polymorphism and EH risk, including 2,813 cases and 2,817 controls. The pooled OR was 2.280 (95%CI: 1.893–2.748, P<0.001) in dominant model, 5.284 (95%CI: 2.679–10.420, P<0.001) in recessive model and 2.359 (95%CI: 1.981–2.808, P = 0.001) in allelic model. Four studies (six cohorts) evaluated C1839T polymorphism and EH risk, including 1,700 cases and 1,681 controls. The pooled OR was 0.785 (95%CI: 0.627–0.983, P = 0.035) in dominant model, 1.250 (95%CI: 0.336–4.652, P = 0.739) in recessive model and 0.805 (95%CI: 0.649–0.999, P = 0.049) in allelic model.

Conclusion

The current meta-analysis concludes that the C allele of E-selectin A561C gene polymorphism might increase the EH risk in Asian population, whereas the T allele of E-selectin C1839T gene polymorphism might decrease the EH risk.  相似文献

The present study aimed at detecting the association of ovine major histocompatibility complex class II (Ovar II) DRB1 gene second exon and susceptibility or resistance to hydatidosis in three sheep breeds of Sinkiang. The MHC-DRB1 second exon was amplified by polymerase chain reaction (PCR) from DNA samples of healthy sheep and sheep with hydatidosis. PCR products were characterized by the restriction fragment length polymorphism (RFLP) technique. Five restriction enzymes, Mval, Haelll, Sacl, Sacll, Hin1l, were used, yielding 14 alleles and 31 restriction patterns. Frequencies of patterns Mvalbc, Hin1lab, Sacllab, Haelllde, Haellldf, Haellldd (P < 0.01) in Kazakh sheep, Saclab (P < 0.05) in Duolang sheep, and Haelllab, Haelllce, Haelllde, Haelllee (P < 0.01) in Chinese Merino (Sinkiang Junken type) sheep, were significantly higher in healthy sheep compared with infected sheep. These results indicated a strong association between these patterns and hydatidosis resistance. In contrast, the frequencies of Mvalbb, Saclaa, Hinl lbb, Haelllef (P < 0.01) and Haelllab (P < 0.05) in Kazakh sheep, Saclbb, Haelllae, Hin1lab (P < 0.05), Haelllaa, Haelllbe, Haelllef (P < 0.01) in Duolang sheep, Sacllaa (P < 0.05) and Haelllbd, Hin1lbb, Haelllcf, Haelllef (P < 0.01) in Chinese Merino sheep (Sinkiang Junken type) were significantly lower in healthy sheep compared with infected sheep. This indicated a strong association between these patterns and hydatidosis susceptibility. In addition, sheep with the pattern of Haelllef demonstrated a high hydatidosis susceptibility (P < 0.01) in all three breeds, while sheep with the pattern Haelllde demonstrated significant hydatidosis resistance (P < 0.01) in Kazakh and Chinese Merino sheep (Sinkiang Junken type). These results suggest that the Ovar-DRB1 gene plays a role in resistance to hydatidosis infection in the three sheep breeds.  相似文献

Thyroid stimulating hormone receptor (TSHR) is thought to play a critical role in the pathogenesis of certain thyroid diseases, including Graves' disease (GD), multinodular thyroid goiter (MTG), and Hashimoto's thyroiditis (HT). In order to understand whether single nucleotide polymorphisms in the TSHR gene contribute to thyroid diseases, we have conducted a case-control study in which, we examined 8 TSHR gene single-nucleotide polymorphisms in introns 1, 4, 5, 6 and exons 7 and 8, respectively, among patients with thyroid diseases. These included one family with GD (3 patients and 9 healthy members); 60 patients with familiar thyroid diseases (30 with GD, 20 with MTG, and 10 with HT patients), 48 sporadic patients with GD and 96 healthy control individuals. Direct sequencing of all 10 exons and part of introns of TSHR gene, in these patients as well as healthy controls revealed eight polymorphisms. A novel polymorphism in exon 8 AGA(Arg) → CGA(Arg). However, there were no significant differences between patients and controls in the incidence of these polymorphisms. These results suggest that the polymorphisms (polymorphism in intron 1 at 81 bp upstream of exon 2; polymorphism in intron 4 at 135 bp upstream of exon 5; polymorphism in intron 4 at 365 bp upstream of exon 5; polymorphism in intron 5 at 69 bp upstream of exon 6; means polymorphism in intron 6 at 13 bp downstream of exon 6; polymorphism in intron 6 at 187 bp upstream of exon 7; E7+16: polymorphism in 16 bp of exon 7; polymorphism in 40 bp of exon 8) of the TSHR gene may not contribute to the pathogenesis of thyroid diseases.  相似文献

To investigate the role of microsomal epoxide hydrolase (mEH) polymorphisms in the aetiology of lung cancer and to assess the interaction between mEH polymorphisms and smoking, we performed a meta-analysis of seven published studies, which included 2078 cases and 3081 controls, and a pooled analysis of eight studies (four published and four unpublished at that time) with a total of 986 cases and 1633 controls. The combined metaanalysis odds ratios (ORs) were 0.98 (95% confidence interval [CI] = 0.72-1.35) for polymorphism at amino acid 113 in exon 3 (His/His versus Tyr/Tyr genotype) and 1.00 (95% CI= 0.71-1.41) for polymorphism at amino acid 139 in exon 4 (Arg/Arg versus His/ His genotype). In the pooled analysis, we observed a significant decrease in lung cancer risk (OR = 0.70, 95% CI = 0.51-0.96) for exon 3 His/His genotype after adjustment for age, sex, smoking and centre. The protective effect of exon 3 polymorphism seems stronger for adenocarcinoma of the lung than for other histological types. The OR for high predicted mEH activity, compared with low activity, was 1.54 (95% CI = 0.77-3.07) in the meta analysis and 1.18 (95% CI = 0.92-1.52) in the pooled analysis. We did not find a consistent modification of the carcinogenic effect of smoking according to mEH polymorphism, although the risk of lung cancer decreased among never smokers with high mEH activity and among heavy smokers with the exon 3 His/His genotype. In conclusion, this study suggests a possible effect of mEH polymorphisms at exon 3 in modulating lung cancer. If present, this effect may vary among different populations, possibly because of interaction with genetic or environmental factors.  相似文献
