用染色体特异的卫星DNA探针进行染色体荧光原位杂交(FISH)分析恶性血液病,发现着丝粒和着丝粒周染色体的重排并非罕见.分子生物学技术和基因组计划的发展,促进了对异染色质分子本质的研究.本文就着丝粒和着丝粒周异染色质的分子结构及其重排机制作一综述。 Abstract:Fluorescence in situ hybridization(FISH) analysis with chromosome-specific satellite DNA probes of various hematopoietie malignancies have revealed that centric and pericentric chromosome rearrangements occurred more frequently than hitherto admitted.The development of molecular biology and genome project have fostered much interest in the structure and function of centromeric heterochromatin.In this paper,recent advances in molecular structure and rearrangement mechanisms of centric and pericentric heterochromatin are reviewed and future study is also discussed.  相似文献

着丝粒是真核生物有丝分裂和减数分裂染色体正确分离和传递所必需的染色体区域。十多年来, 已对包括拟南芥、水稻、玉米在内的一些植物的着丝粒进行了较深入的分子生物学研究。在不同的植物间, 着丝粒DNA的保守性很低, 呈现快速进化, 但着丝粒的DNA序列类型和组织方式基本相似, 一般是由夹杂排列着的卫星DNA串联重复阵列和着丝粒专一的反转录转座子构成。与着丝粒DNA相反, 着丝粒/着丝点的结构性和瞬时蛋白质在包括植物在内的真核生物中保守。与其他真核生物的情况一样, 拥有含着丝粒组蛋白H3(CENH3)的核小体是植物功能着丝粒染色质最基本的特征, CENH3在着丝粒染色质的识别和保持中起着关键作用。  相似文献

采用限制酶AluI显带、CA_(?)/DA/DAPI荧光染色和常规C带技术研究了家猪染色体着丝粒结构异染色质,结果表明:着丝粒结构异染色质至少可被区分为3类,并且在染色体组内各有其特异的染色体分布。将家猪染色体DA/DAPI荧光带和限制酶AluI显带与人类染色体比较,发现家猪13—18号端着丝粒染色体显带特征与人染色体1,9、16、Y一致。提示家猪13—18号端着丝粒区结构异染色质存在与人类随体DNA相似的DNA组成。  相似文献

本文首次报道虫草蝠蛾(鳞翅目,蝙蝠蛾科,蝠蛾属)的有丝分裂染色体核型。应用醋酸分离和热干燥技术,研究了云南的两种虫草蝠蛾Hepialus zhayuensis Chu et Wang和Hepialus sp.的有丝分裂染色体,它们的染色体数目为2n=64。在有丝分裂的早中期染色体上清晰地呈现出散漫着丝粒。然而,分裂中期和较晚的中期阶段,每条染色体都具显著的初级着丝粒(即主缢痕)。它们的雄性中期核型中都有一对典型的异形性染色体,X染色体着色稍淡,且都具中或亚中着丝粒;Y染色体比X染色体长,染色很深。 在雄性的分裂间期细胞中,观察到异固缩性染色质体,此异固缩体是Y染色体。  相似文献

采用复制带、C带和硝酸银染色等分带技术研究了水貂的核型和带型。结果表明,2n=30,枝型为10(M)+16(SM)+2(A),XX(M)。C-带显示该水貂的一些染色体的结构异染色质比较丰富,从着丝粒区域延伸到两臂上,No.5染色体着丝粒结构异染色质有些弱化;X染色体的结构异染色质较常染色体的丰富。Ag-NORs有3个,分布在No.8染色体的次缢痕区域和一条No.2染色体长臂接近着丝粒的区域。  相似文献

采用原代和传代培养方法对8头毛冠鹿(Elaphodus cephalophus)的皮肤细胞进行了染色体研究,发现了一种核型与以前所报道的几种核型不一致,确定为一新核型。在该核型中,染色体众数2n=47,2条X染色体异型,一条为端着丝粒,另一条为近端着丝粒。C-带显示该核型中异染色质除了分布在2条X染色体长臂中之外,在第一对大的端着丝粒染色体中的一条近着丝粒区出现一异染色质“柄”。结合C-带及薄层扫描结果对毛冠鹿种内常染色体、性染色体中异染色质的含量和分布与染色体多态的关系进行了探讨。  相似文献

植物着丝粒是染色体重要结构域,介导动粒装配。不同物种间着丝粒重复序列快速趋异进化,着丝粒功能保守,确保有丝分裂和减数分裂过程中染色体正确分离和准确传递。伴随染色质免疫共沉淀技术(Chromatin immunoprecipitation, ChIP)、ChIP 与高密度芯片相结合技术(ChIP-chip)、ChIP 与高通量测序相结合技术(ChIP-seq)的应用,植物着丝粒研究获得里程碑式进展:某些模式植物着丝粒DNA 序列、蛋白质结构、功能获得大量新认识;着丝粒基本蛋白质组蛋白H3 被用来界定着丝粒大小和边界;某些非着丝粒区域被激活为新着丝粒,在世代传递中保持稳定性。本文对植物着丝粒结构、功能、进化研究进行了综述,并探讨了植物着丝粒研究存在的问题。  相似文献

The karyotype of the dolly vardenSalvelinus malma from Alaska was analysed. A pair of huge acrocentrics, which could have resulted from either tandem fusion or centric fusion followed by pericentric inversion, was observed. Ag-NORs were observed at the terminal regions of the second largest chromosome pair, and individual size-differences in Ag-NORs and in chromosomes with Ag-bands were found. From a comparison of the karyotypes of the dolly varden from Alaska and Hokkaido (Japan), karyotype differentiation by pericentric inversion and translocation was discussed.  相似文献

Centromere positions on 7 maize chromosomes were compared on the basis of data from 4 to 6 mapping techniques per chromosome. Centromere positions were first located relative to molecular markers by means of radiation hybrid lines and centric fission lines recovered from oat-maize chromosome addition lines. These centromere positions were then compared with new data from centric fission lines recovered from maize plants, half-tetrad mapping, and fluorescence in situ hybridizations and to data from earlier studies. Surprisingly, the choice of mapping technique was not the critical determining factor. Instead, on 4 chromosomes, results from all techniques were consistent with a single centromere position. On chromosomes 1, 3, and 6, centromere positions were not consistent even in studies using the same technique. The conflicting centromere map positions on chromosomes 1, 3, and 6 could be explained by pericentric inversions or alternative centromere positions on these chromosomes.  相似文献

A centric shift in the S11 chromosome of Aiolopus strepens, previously described as a pericentric inversion (Cabrero & Camacho, 1982), was analyzed by using the C-banding procedure. The location of breakage points as well as the posibilities of pericentric inversion and three-break transposition are discussed. The relationship between C-bands and NOR location as revealed by both silver staining and a double procedure Ag-NOR C-banding is also discussed.  相似文献

百合族具有非常一致的二型性核型,由4条长染色体以及20条短染色体组成。目前有两个假说解释其二型性核型的来源,着丝粒横裂和多倍化。但是,具体是哪一种机制起主要作用仍然不清楚。根据文献以及自己的实验结果,我们整理并重新分析了百合亚科和美德兰亚科所有属的核型资料。比较核型分析结果表明,来自单条染色体的特征、染色体臂数、核型不对称性以及染色体的相对长度诸方面的证据都支持着丝粒横列是百合族核型进化的主要机制,但不能排除其它的机制也在起着作用,如臂间倒位和易位。臂间倒位和易位可能在郁金香族的核型进化中起着主要的作用。另外,本研究还报道了三个种的核型,粗茎贝母 (Fritillaria crassicaulis)、准格尔郁金香 (Tulipa suaveolens) 和尖果洼瓣花 (Gagea oxycarpa)。  相似文献

观察了新近发现于我国云南的果蝇属暗果蝇种组(Drosophila obscura species group)种类D.luguensis、D.dianensis和D.limingi的有丝分裂中期核型,并将3个种的核型与各自的近缘种类进行了比较。D.luguensis具2n=12条染色体,包括3对中央着丝粒(V形)染色体、2对近端着丝粒(棒状)染色体以及1对微小(点状)染色体。其中X和Y染色体均为中央着丝粒染色体。D.dianensis和D.limingi具2n=10条染色体,包括1对大的V形常染色体,1对小的V形常染色体,2对J形(亚中着丝粒型)常染色体和1对点状染色体。其中X染色体为J形,Y染色体为短棒状。基于核型比较的结果以及D.sinobscura亚组地理分布的资料,结合种间系统发育关系研究结果,认为D.luguensis可能保留了该亚组祖先种类的核型。D.sinobscum的核型(2n=12:2V,1J,2R,1D)可能由一个pre-“sinobscura-hubeiensis”谱系的一个分支通过臂间倒位演化而来,而D.hubeiensis的核型(2n=10:4V,1D)可能由该谱系的另一分支通过着丝粒融合(2对近端着丝粒常染色体的融合)而形成。推测在D.dianensis和近缘欧洲种D.subsilvestris(2n=12:3V,2R,1D)间、D.limingi和东亚近缘种D.tsukubaensis(2n=12:3V,2R,1D)间的物种分化过程中,可能有相似的染色体变异类型发生。  相似文献

We report on a comparative molecular cytogenetic and in silico study on evolutionary changes in human chromosome 7 homologs in all major primate lineages. The ancestral mammalian homologs comprise two chromosomes (7a and 7b/16p) and are conserved in carnivores. The subchromosomal organization of the ancestral primate segment 7a shared by a lemur and higher Old World monkeys is the result of a paracentric inversion. The ancestral higher primate chromosome form was then derived by a fission of 7b/16p, followed by a centric fusion of 7a/7b as observed in the orangutan. In hominoids two further inversions with four distinct breakpoints were described in detail: the pericentric inversion in the human/African ape ancestor and the paracentric inversion in the common ancestor of human and chimpanzee. FISH analysis employing BAC probes confined the 7p22.1 breakpoint of the pericentric inversion to 6.8 Mb on the human reference sequence map and the 7q22.1 breakpoint to 97.1 Mb. For the paracentric inversion the breakpoints were found in 7q11.23 between 76.1 and 76.3 Mb and in 7q22.1 at 101.9 Mb. All four breakpoints were flanked by large segmental duplications. Hybridization patterns of breakpoint-flanking BACs and the distribution of duplicons suggest their presence before the origin of both inversions. We propose a scenario by which segmental duplications may have been the cause rather than the result of these chromosome rearrangements.  相似文献

The chromosomes of subterranean rodents of the South American genus Ctenomys are highly variable with diploid numbers ranging from 10 to 70. The phylogenetic relationships of this group have been analysed cladistically using G-banded karyotypes as have the chromosomal rearrangements involved in its karyotypic differentiation. One group, called the 'Corrientes group', has very variable chromosomes but low allozymic and morphological differentiation among its members. This group has been analysed with respect to chromosomal speciation. Using a member of another subfamily (Octodontomys gliroides) as an outgroup, the results indicate that karyotypes with low diploid and fundamental numbers are plesiomorphic. The range of diploid numbers studied here is between 22 and 70, while the fundamental numbers are between 40 and 86. It was found that the main chromosomal rearrangement that transforms karyotypes towards higher diploid and fundamental numbers is the acquisition of new chromosomal material via unknown mechanisms, followed by pericentric inversions that generate new chromosomal arms, centric fusions and centric fissions. In spite of their low differentiation regarding allozymic and morphological features, it was found that the karyomorphs of the Corrientes group have enough chromosomal differentiation to consider them as distinct species. Beside the range of diploid and fundamental numbers of this group (42–70 and 80–84 respectively), their pairwise chromosomal differences are high. The most closely related of them differ in one nonhomologous arm, one Robertsonian change and a whole chromosome duplication. The most differentiated taxa differ in 20 arms with lack of homology, 12 Robertsonian changes (one with monobrachial homology), six pericentric inversions and the above mentioned probable arm duplication. For these reasons, it is probable that some kind of chromosomal speciation has occurred in the Corrientes group.  相似文献

The chromosomes of five Crocidura species have been studied by means of routine, G-, C- and Ag NOR-banding techniques. The karyotypes of the species with 2n = 40, i.e. C. gueldenstaedtii, C. suaveolens and C. sibirica, are identical. Short arms of the Y-chromosome of C. sibirica are larger. The karyotypes of C. leucodon (2n = 28) and C. zarudnyi (2n = 22) could be obtained as a result of tandem and centric fusion and pericentric inversions. Small amounts of heterochromatin were detected in autosomes of the species investigated. The Y-chromosomes are completely heterochromatic. Four NOR-bearing pairs are detected in the species investigated.  相似文献

从二倍体细胞大染色体组(Macrochromosomes)的G带特征比较分析,鹤形目的黑水鸡与鸡形目的家鸡带型相似程度很大,标志着这两种鸟类的亲缘关系较近,同时也充分说明了不同目鸟类在进化上染色体变异的保守性。其带型差异仅在于两次臂间倒位、两次臂内倒位和一次着丝点融合。此外,我们采用QM荧光——银染的相继染色法发现黑水鸡的W染色体上有NOR存在,这种核仁组织者区域与性染色质发生连锁的现象在鸟类是首次发现。  相似文献

The B1 cell line of rat cerebral endothelium origin exhibits several dicentric and multicentric chromosomes. These chromosomes, unlike multicentrics in mouse (Vig and Zinkowski 1986) do not show premature centromere separation. All centromeres deposit kinetochore proteins and appear to be functional. Even the centromeres which fail to migrate to the poles during anaphase and make side arm bridges bind to spindle microtubules. Some multicentric chromosomes show kinetochores spaced apart with intervening stretches of euchromatin while others are located adjacent to each other thus exhibiting tandem repeats and forming a compound kinetochore (Brinkeley et al. 1984). Also, unlike mouse multicentric chromosomes in which different pericentric regions and the centromeres replicate at different times, the rat chromosomes appear to replicate all pericentric and centric regions in a given multicentric simultaneously. The present studies indicate that centromeres in rat and mouse replicate during the last part of the S-phase and in continuation with the pericentric heterochromatin.  相似文献
