以拟南芥野生型Col-4和蓝光受体突变体cry1,cry2和cry1cry2为材料在蓝光下进行缺K+处理,cry1cry2的下胚轴及根的伸长受抑制程度最大.经过对K+充足条件下的Col-4,cry1,cry2和cry1cry2的钾元素含量和持水性检测.以及采用定量PCR对K+转运栽体和离子通道相关基因如AKT1,AtKC1,AKUP1等表达水平的分析,发现cry1cry2的钾元素含量最高、持水性最低,且其K+转运载体和离子通道相关基因的表达量也最高.该结果说明蓝光下CRY1和CRY2的缺失对K+的吸收起促进作用.  相似文献

以拟南芥野生型(Col-4)和隐花素双突变体cry1cry2为材料,研究不同光照条件下不同浓度吲哚乙酸(IAA)和IAA极性运输抑制剂氨基酞氨酸(NPA)对幼苗下胚轴伸长的影响。结果显示,低浓度IAA(10-7mol/L)可促进连续白光和红光下cry1cry2幼苗下胚轴伸长,而连续蓝光下cry1cry2下胚轴的伸长则受到抑制。蓝光下相同浓度的NPA对cry1cry2幼苗下胚轴伸长的抑制程度比野生型要小。RT-PCR分析结果显示,瞬时蓝光处理时IAA合成关键酶基因IGPS以及生长素应答基因IAA1和IAA5在cry1cry2突变体中的转录水平比野生型中要高。这表明隐花素可能部分通过调节IAA合成和/或IAA极性运输,介导蓝光调控拟南芥下胚轴的伸长。  相似文献

以在20μmol m^-2s^-1白光下生长13d的拟南芥(Arabidopsis thaliana,Landsbcrg生态型)幼苗为材料,采用测定叶片花色素苷含量和Northern blot方法,研究蓝光与蔗糖在诱导植物花色素苷积累及相关基因表达中的作用。结果表明:蓝光处理后,叶片花色素苷积累随光强和照光时间的延长而增加,突变体hy4叶片的花色素苷含量明显低于野生型(WT),说明隐花色素1(cry1)是蓝光诱导花色素苷积累的主要光受体:WT中苯基苯乙烯酮合酶基因(CHS)的表达受蓝光诱导,处理4h即有表达,8h达到最高,之后逐渐下降;蓝光不能诱导突变体hy4中CHS基因的表达,说明cry1介导蓝光诱导CHS基因的表达。培养基中不含蔗糖,削弱了蓝光诱导的拟南芥叶片花色素苷的积累,CHS基因表达也受到抑制。蔗糖不仅作为碳源参与蓝光诱导的花色素苷积累,还可能作为信号分子参与蓝光诱导的CHS表达。  相似文献

通过构建表达光信号系统关键基因CRY1、CRY2和COP1启动子与GUS融合基因的拟南芥转基因植株,并对转基因植株进行GUS组织化学染色的结果表明,CRY1、CRY2和COP1的表达模式不受光条件的调控,并且在各器官有广泛的表达。分别分析CRY1基因启动子在cop1突变体以及COP1基因启动子在cry1突变体遗传背景中表达模式的结果表明,CRY1和COP1在转录水平上不存在明显的相互调控关系。  相似文献

光是控制植物生长发育十分重要的环境因子之一.隐花素是植物的蓝光受体,在植 物中调节多种光形态建成,包括抑制下胚轴的伸长、子叶的伸展和调节植物的开花时间 等,但隐花素依赖蓝光调节光形态建成的分子机制尚不清楚.本文采用比较蛋白质组学 方法研究了在持续蓝光和红光下生长的拟南芥隐花素双突变体cry1cry2和野生型幼苗的全蛋白图谱.采用基质辅助激光解吸飞行时间串联质谱(MALDI-TOF-TOF)进行肽质谱指纹图谱分析.在cry1cry2和野生型中鉴定了71个差异蛋白点.这些差异蛋白质反应光的变化可以形成6类,结果表明,光调节隐花素是通过控制许多相关基因的表达而实现的,为进一步研究拟南芥隐花素的光反应机制提供一些有用的信息.研究表明,蛋白质表达图谱可用于研究各种突变体在不同光照条件下光应答之间的关系.  相似文献

以拟南芥ceo1、突变体为材料,研究CEO1(clone eight-one)在镉胁迫条件下作用的结果表明,与野生型植株相比,150μmol·L^-1的CdCl2处理10d后,拟南芥ceo1突变体表现为植株生长矮小,叶片卷曲发黄,根系短小。镉处理后,拟南芥突变体幼苗叶中H2O2的积累较多;镉处理1h后的突变体中抗坏血酸过氧化物酶(APX)活性明显上升,至2h时又开始下降,而镉处理2h后,野生型APX活性才开始增加。镉处理2h后的野生型的谷胱甘肽还原酶(GR)显著增加,而突变体无明显变化。两种类型拟南芥的超氧化物歧化酶(SOD)与过氧化氢酶(CAT)的活性没有明显差异。  相似文献

【目的】分析spoIIID基因突变对苏云金芽胞杆菌cry1、cry3、cry4和cry8基因启动子Pcry1Ac、Pcry3A、Pcry4A和Pcry8E的影响,比较以上启动子在无芽胞spoIIID基因突变体(HD-△SpoIIID)中的转录活性。【方法】分别构建了Pcry1Ac、Pcry3A、Pcry4A和Pcry8E与lacZ基因融合的转录分析载体,并导入HD-73野生型菌株和HD-△SpoIIID突变株中测定β-半乳糖苷酶活性;通过高温诱变方法在HD-△SpoIIID基础上筛选出缺失cry1Ac基因的HD--△SpoIIID突变体;构建了4种启动子与cry1Ac基因融合表达载体,分别将它们转入HD-ΔSpoIIID和HD--ΔSpoIIID中,分析Cry1Ac蛋白表达量及其生物活性。【结果】HD-73和HD-ΔSpoIIID菌株中四个启动子转录活性由高到低分别为:Pcry8E>Pcry1Ac>Pcry4A>Pcry3A;spoIIID基因的缺失未影响Pcry1Ac和Pcry8E转录活性,Pcry3A在HD-ΔSpoIIID菌株中转录活性略有升高,Pcry4A在HD-ΔSpoIIID菌株中转录活性在T5到T10略有降低。从翻译水平来看在HD-ΔSpoIIID中cry8E启动子略低于cry1Ac启动子,并高于Pcry4Aa,Pcry3A指导的Cry1Ac蛋白产量,生物活性测定结果与蛋白表达量相符。【结论】cry8E基因启动子Pcry8E在spoIIID突变体中在转录水平活性是最高的启动子,而cry1Ac启动子指导自身基因cry1Ac表达时,在翻译水平上略高于cry8E启动子指导的Cry1Ac产量。  相似文献

以拟南芥为材料,在红光和蓝光下对PRRs(pseudo-response regulators)突变体prr5p、rr7、prr9和toc1及其野生型的下胚轴表型进行比较观察,并采用实时定量PCR方法对突变体中光信号通路相关基因ZTL(zeitlupe)和CO(constans)的节律表达进行分析.结果表明:在红光下,prr5和toc1的下胚轴长度比野生型显著增长,在蓝光下,prr7p、rr9和toc1较野生型短,表明突变体降低了拟南芥对红光的敏感性,却增强了对蓝光的敏感性.红光和蓝光下,PRRs突变体中ZTL和CO的mRNA节律表达与野生型明显不同,其中红光下prr5和prr7、蓝光下prr5和toc1中的ZTLmRNA的表达显著下降且节律消失;红光下prr7和prr9以及蓝光下prr5突变体中的COmRNA表现基本无节律.因此推测,PRRs与ZTL的相互作用很可能在红光和蓝光信号转导途径中发挥作用,且PRRs基因极有可能参与了红光和蓝光对CO的调控.  相似文献

目的:检测TR3及其转录活性域缺失突变体在酵母双杂交系统中的转录自激活作用。方法:采用PCR方法扩增TR3全长序列及其缺失突变体,构建酵母双杂交系统的诱饵载体pGBKT7-TR3和pGBKT7-TR3/Δ1~690,将pGBKT7-TR3转化感受态酵母菌AH109后培养于含缺失培养基的平板上,检测β-半乳糖苷酶活性,判定其是否具有转录自激活作用。结果:构建了包含TR3全长序列和TR3/Δ1~690序列的诱饵载体,转化酵母菌AH109后在双缺和三缺培养基上未能使β-半乳糖苷酶活性滤纸片变蓝,说明β-半乳糖苷酶报告基因未被激活。结论:TR3及其转录活性域缺失突变体没有转录自激活作用,可用于酵母双杂交系统。  相似文献

植物蓝光调节的反应主要有向光性、抑制幼茎伸长、叶绿体迁移、刺激气孔张开和调节基因表达等。蓝光反应的有效波长是蓝光和近紫外光(320—400am),故蓝光受体也叫蓝光/近紫外光受体。植物蓝光受体研究近年来取得较大进展。以拟南芥为例,已得到确认的受体至少有隐花色素(CRY1、2)和向光蛋白(phototropin)两大类。转基因拟南芥对蓝光、紫外光和绿光敏感,并发现CRY1是一个可溶性蛋白。CRY2编码一个核蛋白,蓝光在转录水平对该蛋白进行调节,它的作用是增加拟南芥对蓝光的敏感性。CRY1和CRY2共同介导了拟南芥植物的向光性。隐花色素的蛋白与辅基之间以非共价键连接,可以吸收蓝光和近紫外光。CRY1和CRY2蛋白之间,尤其是N端相似性很高。向光蛋白目前只发现PHOT1和PHO12两种,向光蛋白作为丝/苏氨酸激酶蓝光受体含有两个光氧化结构域(LOV)并参与了植物向光性叶绿体运动、气孔开放等。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
