为了阐明红色原鸡的群体遗传结构,以对其有效保护提供遗传学依据,采用33个微卫星标记对其群体中56个个体进行了PCR-聚丙烯酰胺多态性电泳检测。33个微卫星座位共检测到140个等位基因,所有座位都呈现出多态性,每个座位的等位基因数在2~8个之间,平均每个座位等位基因数4.24个,有效等位基因数3.30个。根据等位基因频率,计算出的群体表观杂合度、期望杂合度及多态信息含量分别为0.7980、0.6506和0.5948。结果表明,红色原鸡群体遗传多样性较丰富。  相似文献

银鲫是天然雌核发育的三倍体两性型种群, 因其遗传背景和生殖方式的特殊性, 已经成为研究单性和多倍体脊椎动物进化遗传学的理想模式鱼类。利用33个微卫星序列对雌核发育银鲫的突变速率和突变模式进行研究, 结果表明: (1) 22个子代个体中, 检测到1尾个体在15个微卫星位点具有18个突变等位基因; (2) 每个微卫星位点每代总体平均突变率是1.16×10-2, 95%置信区间是6.87×10-3~1.83×10-2, 与其他鱼类相比, 雌核发育银鲫的突变率明显偏高, 这与天然雌核发育鱼类处在单性生殖和两性生殖的过度阶段有密切关系; (3) 具有突变等位基因的13个位点的重复单元数目都在10次以上, 11个复合型微卫星位点的突变率(1.31×10-2)与21个完美型位点(1.00×10-2)的突变率没有明显差异(P = 0.67), 微卫星突变率受到重复单元数目的影响, 然而与重复结构类型和侧翼序列GC碱基含量无相关性; (4) 序列分析表明, 雌核发育银鲫的微卫星突变模式并不严格遵守逐步突变模型。  相似文献

茶花鸡是我国具有独特遗传特性的地方家禽品种,为了进一步阐明其群体遗传变异和遗传结构状况,采用了33个家鸡特异性的微卫星标记对该鸡种自然群体中30个个体进行了多态性电泳检测。33个微卫星座位共检测到105个等位基因,所有座位都呈现出多态性,每个座位的等位基因数在2~5个之间,平均每个座位等位基因数3.20个。群体平均杂合度和平均多态信息含量分别为0.612 9和0.527 6。结果表明,茶花鸡自然群体遗传多样性较丰富。  相似文献

大河猪是中国西南中海拔地区代表性猪种之一,长期以来在当地养猪生产中发挥了重要作用。为了阐明其群体遗传变异情况,为进一步有效保护和合理利用提供科学依据,采用分布在家猪19对染色体上的76个微卫星标记对该猪种60个随机抽样个体进行了微卫星PCR–聚丙烯酰胺凝胶电泳检测。共检测到347个等位基因,所有座位都呈现出多态性,每个座位的等位基因数在3—10个之间,平均每个座位等位基因数4.57个,有效等位基因数3.50个,群体平均杂合度及平均多态信息含量分别为0.696 2±0.071 6和0.644 1±0.091 4。结果表明,大河猪群体遗传多样性较丰富,选择潜力较大。  相似文献

利用47个鲤鱼微卫星标记, 对柏氏鲤和荷包红鲤抗寒品系自交F2代的92个个体基因组DNA进行检测, 得到162个等位基因, 各座位等位基因数2~6个, 片段长度100~444 bp, 有效等位基因数1.3069~4.2288, 杂合度0.2361~0.7677, 位点多态信息含量0.5368。利用统计软件SPSS的GLM程序对47个微卫星标记与鲤鱼体重、体长和体高相关性进行了分析, 结果表明HLJ695、HLJ716、HLJ739、HLJ759、HLJ774、K16与体重、体长和体高相关, HLJ776与体高相关。对同一标记不同基因型间进行多重比较, 找到与3种性状相关的基因型。  相似文献

利用微卫星分子标记对富钟水牛群体遗传结构和保种效果进行评估,旨在掌握保种区富钟水牛目前的保种现状,为下一步制定有效的保种措施提供参考依据。结果表明:15个微卫星座位中,共检测到86个等位基因,平均等位基因NA为5.733 3;平均有效等位基因NE为2.902 9;期望杂合度、观察杂合度、多态信息含量PIC分别为0.596 4、0.487 2、0.551 9;1个座位为低度多态,3个座位为中度多态,9个座位为高度多态,这些位点可用于遗传多样性分析。  相似文献

野猪(Sus scrofa)是家猪的祖先,也是野生动物遗传资源和生物多样性的重要组成部分.为了阐明其群体遗传变异,以对其进一步有效保护和合理利用提供科学依据,本研究采用分布在家猪19条染色体上的76个微卫星标记对云南南部地区野猪群体65只个体进行了群体遗传变异分析.共检测到349个等位基因,每个座位的等位基因数从3到9不等,有效等位基因数在1.683 5-8.066 7之间,平均每个座位等位基因数(4.592 1±1.145 2)个,有效等位基因数(3.709 9±0.990 4)个,群体平均表观杂合度、期望杂合度及平均多态信息含量分别为(0.949 2±0.113 5)、(0.711 6±0.078 7)和(0.663 4±0.092 1).结果表明,本实验检测到的野猪群体遗传多样性较丰富.  相似文献

目的探讨微卫星在转基因和基因突变小鼠中的变化,为基因修饰和遗传突变动物的遗传检测和表型分析提供理论依据和技术手段。方法根据文献报道,从GenBank中选取198个等位基因数量多、富含多态性的微卫星位点,以野生型动物为对照,对6种近交系遗传背景的转基因小鼠和5种自然基因突变的近交系小鼠进行微卫星多态性检测,选用1.5%琼脂糖凝胶电泳和STR扫描技术,比较分析微卫星不稳定性。结果共有40个微卫星位点在转基因和基因突变小鼠中表现出多态性。在基因突变小鼠中,微卫星不稳定性有55.6%(10/18)是由纯合变为杂合(Ⅰ型),有3个位点(16.6%,3/18)是纯合突变(Ⅱ型),有5个位点同时存在2种类型的突变。但是在转基因动物中,大多数的微卫星多态性为Ⅰ型突变(87.5%,28/32),只有2个位点(6.2%,2/32)是Ⅱ型突变。另外有2个位点同时存在2种类型的突变。结论基因修饰或基因突变可引起小鼠相关微卫星发生不稳定性,而且某些微卫星位点对基因改变敏感性较高。  相似文献

摘要: 利用30对微卫星分子标记对长江中下游5个鲢群体进行了遗传多样性分析。结果表明: 在30个基因座中, 共检测到144个等位基因, 每个座位检测到的等位基因数为1~10个, 其中有25个座位具有多态性, 多态位点百分率为83.33,5个群体的平均等位基因数A为4.0/4.1, 平均有效等位基因数Ne为2.4445~2.6332, 平均观察杂合度Ho为0.3233~0.3511, 平均期望杂合度He为0.4421~0.4704, 平均多态信息含量PIC为0.4068~0.4286。对数据进行F-检验, Fst值表明群体间的遗传分化程度中等, 并对基因型进行了基于Hardy-Weinberg平衡的卡方检验, 所得P值说明5个群体均一定程度上偏离了平衡。5个群体间的遗传相似系数为0.8466~0.9146,遗传距离为0.0893~0.1665, 并根据Nei氏标准遗传距离用UPGMA方法对5个鲢群体进行亲缘关系聚类。  相似文献

The patterns of mutation and evolution at 13 microsatellite loci were studied in the filamentous fungal genus Neurospora. First, a detailed investigation was performed on five microsatellite loci by sequencing each microsatellite, together with its nonrepetitive flanking regions, from a set of 147 individuals from eight species of Neurospora. To elucidate the genealogical relationships among microsatellite alleles, repeat number was mapped onto trees constructed from flanking-sequence data. This approach allowed the potentially convergent microsatellite mutations to be placed in the evolutionary context of the less rapidly evolving flanking regions, revealing the complexities of the mutational processes that have generated the allelic diversity conventionally assessed in population genetic studies. In addition to changes in repeat number, frequent substitution mutations within the microsatellites were detected, as were substitutions and insertion/deletions within the flanking regions. By comparing microsatellite and flanking-sequence divergence, clear evidence of interspecific allele length homoplasy and microsatellite mutational saturation was observed, suggesting that these loci are not appropriate for inferring phylogenetic relationships among species. In contrast, little evidence of intraspecific mutational saturation was observed, confirming the utility of these loci for population-level analyses. Frequency distributions of alleles within species were generally consistent with the stepwise mutational model. By comparing variation within species at the microsatellites and the flanking-sequence, estimated microsatellite mutation rates were approximately 2500 times greater than mutation rates of flanking DNA and were consistent with estimates from yeast and fruit flies. A positive relationship between repeat number and variance in repeat number was significant across three genealogical depths, suggesting that longer microsatellite alleles are more mutable than shorter alleles. To test if the observed patterns of microsatellite variation and mutation could be generalized, an additional eight microsatellite loci were characterized and sequenced from a subset of the same Neurospora individuals.  相似文献

应用微卫星多态分析四个鲤鱼群体的遗传多样性   总被引:28,自引:1,他引:27  
选择12个斑马鱼功能基因的微卫星标记和12个鲤鱼微卫星标记,检测黑龙江鲤(Cyprinus carpiohaematopterusTemminck et Schlegel)、散鳞镜鲤(C.carpio)、荷包红鲤抗寒品系(C.carpiovar.wuyuanensis)、松浦鲤(C.carpioSongpu carp)的群体遗传多样性。共检测到3882个扩增片段,长度为126~489bp,在群体间扩增出1~5个等位基因不等,共计59个等位基因,平均等位基因2.46个。数据经PHYLIP V3.6软件估算和MEGA3软件作图,确立4个群体间的亲缘关系。并应用Bootstrap检验估计系统树中节点的自引导值,并进行了系统发生分析。结果表明①4个群体检测的有效等位基因数都在55个以上,平均观测杂合度为0.36~0.43,平均期望杂合度为0.49~0.53,平均多态信息含量为0.21~0.25,说明这几个群体多态性属于中度偏高水平,遗传多样性较高;②群体间相似系数都在0.84以上,遗传距离较近,为0.067~0.170,与前人研究结果一致。聚类分析显示,松浦鲤与散鳞镜鲤亲缘关系最近,荷包红鲤抗寒品系与它们的亲缘关系较黑龙江鲤更近;③在3个斑马鱼功能基因相关的微卫星位点上,黑龙江鲤缺失特异扩增条带,这可能与几种鱼类的育成史有关。  相似文献

鲤的微卫星引物对草鱼基因组分析适用性的初步研究   总被引:27,自引:0,他引:27  
运用微卫星DNA-聚合酶链反应(STR—PCR)基因分型技术,选取已发表的28对鲤的微卫星引物,探讨鲤的引物用于草鱼基因组微卫星分析的可能性。通过优化PCR反应条件,消除了影子带和异源核酸双链分子两类微卫星相关假阳性带对STR—PCR分析的干扰。在此基础上,筛选出7对引物可在湘江野生草鱼基因组中扩增出特异性条带,占总数的25%;其中的4对引物(约占总数的14.3%)在8尾湘江野生草鱼小群体中即检测到了个体间等位基因的多态性。这些初步的结果表明鲤的微卫星引物可以用于草鱼基因组的分析。  相似文献

The genome of the social amoeba Dictyostelium discoideum is known to have a very high density of microsatellite repeats, including thousands of triplet microsatellite repeats in coding regions that apparently code for long runs of single amino acids. We used a mutation accumulation study to see if unusually high microsatellite mutation rates contribute to this pattern. There was a modest bias toward mutations that increase repeat number, but because upward mutations were smaller than downward ones, this did not lead to a net average increase in size. Longer microsatellites had higher mutation rates than shorter ones, but did not show greater directional bias. The most striking finding is that the overall mutation rate is the lowest reported for microsatellites: approximately 1 x 10(-6) for 10 dinucleotide loci and 6 x 10(-6) for 52 trinucleotide loci (which were longer). High microsatellite mutation rates therefore do not explain the high incidence of microsatellites. The causal relation may in fact be reversed, with low mutation rates evolving to protect against deleterious fitness effects of mutation at the numerous microsatellites.  相似文献

Ten tetranucleotide microsatellite loci were isolated from GATA-enriched library of rock carp (Procypris rabaudi). The loci were tested in 25 individuals from four populations. The number of alleles per locus ranged from 3 to 6. Polymorphism information content (PIC) ranged from 0.460 to 0.788. Observed and expected heterozygosities ranged from 0.375 to 0.739 and from 0.531 to 0.833, respectively. These microsatellites will facilitate the study of population genetics of rock carp.  相似文献

Rate and pattern of mutation at microsatellite loci in maize   总被引:30,自引:0,他引:30  
Microsatellites are important tools for plant breeding, genetics, and evolution, but few studies have analyzed their mutation pattern in plants. In this study, we estimated the mutation rate for 142 microsatellite loci in maize (Zea mays subsp. mays) in two different experiments of mutation accumulation. The mutation rate per generation was estimated to be 7.7 x 10(-4) for microsatellites with dinucleotide repeat motifs, with a 95% confidence interval from 5.2 x 10(-4) to 1.1 x 10(-3). For microsatellites with repeat motifs of more than 2 bp in length, no mutations were detected; so we could only estimate the upper 95% confidence limit of 5.1 x 10(-5) for the mutation rate. For dinucleotide repeat microsatellites, we also determined that the variance of change in the number of repeats (sigma(m)2) is 3.2. We sequenced 55 of the 73 observed mutations, and all mutations proved to be changes in the number of repeats in the microsatellite or in mononucleotide tracts flanking the microsatellite. There is a higher probability to mutate to an allele of larger size. There is heterogeneity in the mutation rate among dinucleotide microsatellites and a positive correlation between the number of repeats in the progenitor allele and the mutation rate. The microsatellite-based estimate of the effective population size of maize is more than an order of magnitude less than previously reported values based on nucleotide sequence variation.  相似文献

Fifteen lines each of Drosophila melanogaster, D. simulans, and D. sechellia were scored for 19 microsatellite loci. One to four alleles of each locus in each species were sequenced, and microsatellite variability was compared with sequence structure. Only 7 loci had their size variation among species consistent with the occurrence of strictly stepwise mutations in the repeat array, the others showing extensive variability in the flanking region compared to that within the microsatellite itself. Polymorphisms apparently resulting from complex nonstepwise mutations involving the microsatellite were also observed, both within and between species. Maximum number of perfect repeats and variance of repeat count were found to be strongly correlated in microsatellites showing an apparently stepwise mutation pattern. These data indicate that many microsatellite mutation events are more complex than represented even by generalized stepwise mutation models. Care should therefore be taken in inferring population or phylogenetic relationships from microsatellite size data alone. The analysis also indicates, however, that evaluation of sequence structure may allow selection of microsatellites that more closely match the assumptions of stepwise models.  相似文献

The rate of microsatellite mutation is dependent upon both the allele length and the repeat motif, but the exact nature of this relationship is still unknown. We analyzed data on the inheritance of human Y-chromosomal microsatellites in father-son duos, taken from 24 published reports and comprising 15,285 directly observable meioses. At the six microsatellites analyzed (DYS19, DYS389I, DYS390, DYS391, DYS392, and DYS393), a total of 162 mutations were observed. For each locus, we employed a maximum-likelihood approach to evaluate one of several single-step mutation models on the basis of the data. For five of the six loci considered, a novel logistic mutation model was found to provide the best fit according to Akaike's information criterion. This implies that the mutation probability at the loci increases (nonlinearly) with allele length at a rate that differs between upward and downward mutations. For DYS392, the best fit was provided by a linear model in which upward and downward mutation probabilities increase equally with allele length. This is the first study to empirically compare different microsatellite mutation models in a locus-specific fashion.  相似文献

The hypermutable nature of some microsatellite loci implies realistic possibilities for the large-scale detection of germline mutations by pedigree analysis. We have developed a model system for mutation analysis by the characterisation of patterns of mutation at three hypervariable microsatellites (two tetranucleotide and one pentanucleotide repeat loci) in barn swallows, all three markers having mutation rates at the percentage level. Here, we study how the mutation rate varies between individual birds of a Spanish population of barn swallows. A total of 53 mutations were identified from 2920 germline transmissions in 90 families with a total of 694 offspring. Mutations were not randomly distributed among individuals (P = 0.020). Attempts to correlate mutation rate with allele size, degree of inbreeding, immunocompetence and male age only revealed a strong effect of allele size. The mean mutation rate differed between colonies of breeding swallows which was probably due to a corresponding variation in allele size between colonies. There was no difference in the mean mutation rate between the Spanish and an Italian population. These results corroborate earlier findings, at the population level, of an allele size effect on the microsatellite mutation rate.  相似文献
