硫酸铜对蚕豆根尖细胞有丝分裂的影响   总被引:14,自引:0,他引:14  
以蚕豆根尖为材料,研究硫酸铜对蚕豆根尖细胞的遗传毒性效应。采用蚕豆根尖细胞的微核试验方法和染色体畸变试验方法,以不同浓度的硫酸铜为诱变剂,测定蚕豆根尖细胞的有丝分裂指数、微核率和染色体畸变率。结果表明:不同浓度的硫酸铜均能使蚕豆根尖细胞有丝分裂指数明显增加,即5个实验组的分裂指数均明显高于对照组(P<0.01或P<0.001);不同浓度的硫酸铜对蚕豆根尖细胞有丝分裂各期百分数的影响有异;能诱发较高频率的微核率,即在一定浓度范围内,其微核率随硫酸铜处理浓度的升高而增加,但随着硫酸铜浓度的进一步升高而呈下降趋势;硫酸铜还能诱导染色体产生多种类型的畸变,染色体畸变率随硫酸铜处理浓度的升高而增加,随着硫酸铜浓度的进一步升高而呈下降趋势,但均明显高于对照组(P<0.001)。结论是硫酸铜对蚕豆根尖细胞具有明显的遗传毒性效应。  相似文献

蚕豆根尖细胞微核技术检测合成氨工业水的遗传毒性   总被引:10,自引:0,他引:10  
目的:探讨了合成氨工业水处理前后对蚕豆根尖细胞微核率的影响。方法:选取某化工厂生产流程中3个有代表性的水样,同时以蒸馏水(CK)为阴性对照,以不同浓度(0.5、5、50、500μg/L)的环磷酰胺处理为阳性对照,共5个处理组水平,蚕豆根尖细胞微核试验测定其MCN‰。结果:洗煤气水、洗甲醇水可诱导蚕豆根尖细胞产生较高的MCN‰与对照组相比,差异均具有统计学意义(P<0.01),并分别相当于50μg/L和5μg/L环磷酰胺的致突变效应。经处理后的生化出水致突变性显著降低。结论:合成氨工业水具有较强的致突变性,蚕豆根尖细胞微核技术可作为合成氨工业水的遗传毒性检测。  相似文献

采用微核实验技术,研究大气污染物SO2对蚕豆根尖和叶尖细胞的遗传损伤效应。结果表明2.80和28 mg.m-3的SO2熏气可以诱发蚕豆根尖和叶尖细胞损伤,导致根尖细胞有丝分裂指数下降,根尖和叶尖间期细胞微核率增高,并具有时间效应和浓度效应。经水恢复培养后,根尖分裂细胞数增多,微核率降低,说明恢复培养能够缓解高浓度SO2对根尖细胞的遗传损伤。用石蜡层隔断SO2在根部水中的溶解后,根尖细胞微核率低于叶尖细胞微核率,而在非隔断组中则相反,说明SO2在水中的溶解是产生毒性效应的重要原因。高浓度SO2熏气对蚕豆根尖和叶尖细胞具有遗传学毒性,由于根尖分生区具有较高的分裂指数和微核率,对环境SO2毒性的反应更灵敏,蚕豆根尖微核实验更适于对环境SO2的监测。  相似文献

应用蚕豆根尖微核试验,对SO2的遗传毒性效应进行了研究。结果表明:一定浓度范围内(0.108-14.00mg/m^3),蚕豆根尖微核细胞数与SO2浓度间呈正相关,太原市大气污染严重的冬季采暖期根尖细胞微核率明显高于非采暖期;SO2浓度0.604mg/m^3处理24h和48h或2.80-28.00mg/m^3熏气处理4h可使蚕豆根尖中具有微核的细胞数明显增加,结果表明,低浓度SO2较长时间接触或高浓度短期接触均可引起蚕豆根尖细胞遗传物质的损伤,应用蚕豆根尖细胞微核实验可对大气SO2污染进行生物监测。SO2(2.80-28.00mg/m^3)熏气实验中,接触时间延长能导致根尖细胞微核率下降,2.80mg/m^3熏气组下降较快,14.00mg/m^3熏气组下降较慢,研究结果提示,在运用蚕豆根尖微核实验监测环境SO2污染时要考虑蚕豆的染毒方式,避免假阴性结果的出现。  相似文献

浊漳河水体污染物对蚕豆根尖细胞的遗传毒性研究   总被引:2,自引:0,他引:2  
运用蚕豆根尖细胞微核试验技术对浊漳河南段6个代表性样点水体中有机污染物的遗传毒性进行了研究,为浊漳河流域水体污染状况的有效监测提供理论依据.结果显示:(1)浊漳河南段各监测断面的水体有机污染物对蚕豆根尖细胞均产生了不同程度的损伤,表现出细胞微核、断片、染色体桥等多种异常形态,严重时导致细胞坏死和细胞凋亡.(2)6个样点水样处理的蚕豆根尖细胞微核率在21.47‰~64.77‰,极显著高于对照组(P<0.01).(3)各样点水样对蚕豆根尖细胞的遗传损伤程度依次为:王桥>店上>漳泽水库>北寨>黄碾>五阳;在有丝分裂后期,蚕豆根尖中异常细胞最高达70%,且异常细胞比率有随着污染程度增大而升高的趋势.研究表明,蚕豆根尖细胞微核是水质毒理检测的有效指标,其细胞有丝分裂后期异常细胞比率也可作为水质毒理检测的观察指标.  相似文献

利用蚕豆根尖微核技术监测化工铬渣堆场渗出液的遗传毒性,结果表明,化工铬渣渗出液含有较高浓度的Cr6+,能明显诱导蚕豆根尖细胞微核的形成,具有较强的遗传毒性。当铬渣渗出液中Cr6+浓度为186.6460 mg/L时,微核率达到最大。铬渣渗出液所诱导的微核率与阴性对照组相比有极显著差异。  相似文献

以蚕豆为材料 ,研究 SO2 体内衍生物 -亚硫酸钠与亚硫酸氢钠混合液 (3:1)对根尖细胞的遗传毒效应。结果表明 :0 .0 5~2 .0 0 mmol/ L的 SO2 衍生物处理可诱发两品种蚕豆根尖中的微核细胞明显增加 ;不同浓度 (0 .0 5~ 15 .0 0 mmol/ L ) SO2 衍生物处理 12、2 4、36 h后 ,根尖微核细胞率是对照组的 2 .5~ 5 .0倍 ,显著高于对照组。在一定浓度范围内 ,蚕豆根尖细胞微核率与SO2 衍生物浓度之间具有正的线性相关。研究结果表明 ,SO2 衍生物能够破坏蚕豆根尖细胞的遗传稳定性。  相似文献

采用蚕豆根尖细胞的微核试验和染色体畸变试验方法,测定不同浓度的褐藻寡糖对环磷酰胺(cyclophosphamide,CP)诱导的蚕豆根尖细胞的微核率、有丝分裂指数和染色体畸变率的影响。结果表明:褐藻寡糖能有效抑制环磷酰胺诱导的蚕豆根尖细胞微核的产生,即在一定浓度范围内,微核率随褐藻寡糖处理浓度的降低而减少,但低于一定浓度后反而呈上升趋势;不同浓度的褐藻寡糖均可使蚕豆根尖细胞有丝分裂指数增大;褐藻寡糖还能有效降低蚕豆根尖细胞染色体畸变率。因此,褐藻寡糖对蚕豆根尖细胞具有明显的诱抗活性和调节细胞分裂生长的效应。  相似文献

Summary The culture fluids from two Cellvibrio strains, in the stationary phase of growth, are shown to contain heat-resistant, low molecular weight substances with antibiotic-like effects on blue-green algae. Morphological changes and lysis of cells were observed in various species of blue-green algae; ultrastructural changes were noted in the cell walls of growing vegetative cells of Anabaena inaequalis. The % G+C content of DNA from four species of blue-green algae has been determined by melting temperature measurement. The whole cells of blue-gree algae and lipopolysaccharides isolated from these cells were shown to stimulate the production of macro-(mainly) and microglobulin antibodies in rabbits. The macro- and microphage indices in rabbits increased significantly after the injection of LPS isolated from blue-green algae 24--48 hours before infecting the animals with a virulent Y. pseudotuberculosis strain. Besides, the inhibiting action of this strain on the migration of phagocytes to the site of infection was abolished immediately after the injection. The use of the indirect hemagglutination test allowed to prove the absence of close antigenic interrelations between blue-green algae and the following organisms: Spirulina platensis, Microcystis aeruginosa, Phormidium africanum and P. uncinatum.  相似文献

Summary Known nitrogen-fixing species of blue-green algae are capable of reducing acetylene to ethylene, but acetylene is not reduced by Anacystis nidulans, which does not fix nitrogen. Cycad root nodules which contain blue-green algae as endophytes reduce acetylene. Acetylene reduction is inhibited by carbon monoxide. Nitrate or ammonium-nitrogen has no immediate effect on algae reducing acetylene, but algae grown on nitrate-nitrogen gradually lose their capacity to reduce acetylene. Nitrate-nitrogen also inhibits heterocyst formation in these algae and there is a fairly direct correlation between the abundance of heterocysts in a particular sample and its capacity to reduce acetylene. Aphanizomenon flosaquae reduces acetylene and fixes nitrogen in unialgal culture and there is strong presumptive evidence that these reductions are carried out by the alga rather than by associated bacteria. The molar ratios of ethylene: ammonia produced vary within the range 1.4–1.8.  相似文献

Dianchi Lake covers about a 300 km2 area. Kunming city on the edge of the lake is surrounded by industrial establishments. Farm land surrounds the remaining areas of the lake. The lake water is polluted by the Kunming city municipal sewage from 3 million inhabitants, the industrial effluent and farm runoff. Water samples were collected from 12 sites along the shore of the lake during the dry (May) and rainy (August) seasons for genotoxicity testing with the Vicia micronucleus assay during the year 1995. Genotoxicity in terms of micronuclei (MCN) frequencies in the root tip cells of Vicia showed a consistent elevated frequency of MCN over the control sample at the 0.05-0.01 levels of significance in both seasons. The MCN frequencies of sites A (Daguanhe), B (Gaoqiao) and C (Xiyuan) were 3.5-4 times as high as the control values (5.25/1000 cells) in the dry season. In the rainy season, the MCN frequencies of water samples from most of the 12 sites were relatively lower than those of the dry season except sites J (Haikou), and K (Kunyang). The average MCN frequency of the dry season samples was 14.97 per 1000 cells and that of the rainy season samples was 12.24 per 1000 cells while the average control value was around 5.00/1000 cells.  相似文献

Nutrient loadings to the nearshore of southeastern Lake Michigan have undergone a remarkable reduction. This reduction can affect the nutrient supply and result in biological changes. Changes in phytoplankton community, particularly the blue-green algae, can be related to nutrient changes. After thermal stratification, sudden increases in the blue-green algae population were significantly correlated to soluble reactive phosphorus concentrations. Phosphorus-stimulated low dissolved silica and phosphorus limitations after stratification appear to be primary factors contributing to the success of these algae.  相似文献

Summary Mesosome-like, unit-membrane structures are clearly defined in the blue-green algae, Spirulina and three strains of Synechococcus, after osmium or potassium permanganate fixation and observation with the electron microscope. A study was undertaken to determine the effects of passage through the cooling system of the Zion Nuclear Plant, Illinois, U.S.A., on the blue-green algae present in lake water. The blue-green algae were isolated by means of membrane filtration, enumerated and identified to genera. As a result of cooling system passage there was a definite reduction in the numbers of blue-green algae. This reduction appeared to be due to mechanical, as well as elevated temperature, damage.
