【背景】假单胞菌是广泛存在于土壤、水体环境的微生物,其中Pseudomonas plecoglossicida NyZ12是一株能够以环己胺为唯一碳源和氮源生长的革兰氏阴性菌,其基因组达到7.0Mb左右。【目的】研究假单胞菌NyZ12的基因组是否具有可塑性和多变特征。【方法】以环己胺为唯一碳源和氮源生长的P. plecoglossicida NyZ12为研究对象,以琥珀酸或者代谢中间产物环己酮为碳源连续传代让其自然发生突变,然后筛选在以环己胺为唯一碳源和氮源的无机盐培养基上不能生长的突变体。将获得的突变体进行全基因组测序,并与野生型假单胞菌NyZ12的全基因组进行比对。【结果】以琥珀酸和环己酮为碳源分别筛选到一株突变体T1和T2,测序比对后发现假单胞菌突变体T1、T2的基因组发生大量的缺失和突变。对基因丢失的原因进行了分析,丢失的2个大片段中存在大量的重复序列、转座酶、转座子和原噬菌体。【结论】假单胞菌NyZ12的基因组具有可塑多变的特征。其可能的机制为进一步揭示微生物的适应和进化提供了参考。  相似文献

【目的】大量聚对苯二甲酸乙二醇酯(polyethylene terephthalate,PET)塑料作为废弃物被丢弃,严重危害生态健康。针对嗜热PET降解菌缺乏这一情况,本研究旨在获得能够降解PET的嗜热菌,并阐述其降解机制。【方法】采集云南腾冲热泉中的废弃PET瓶,分析其表面生物膜的微生物群落多样性,从中筛选能够以PET为营养源生长的嗜热菌,并基于16S rRNA基因序列加以鉴定;以菌株的定殖能力与生长曲线为指标,优选出降解能力较强的降解菌,并测定其最适pH、温度和NaCl浓度;降解能力较强的降解菌分别作用于PET及PET中间体双(羟乙基)对苯二甲酸酯[bis(hydroxyethyl)terephthalate,BHET]和对苯二甲酸单(2-羟乙基)酯[mono(2-hydroxyethyl)terephthalate,MHET],测定产物生成量与降解率;通过观察PET膜表面微观结构、活菌数、酯酶活性等探究降解菌与PET的互作过程。【结果】废弃PET瓶表面生物膜中的微生物群落多样性低;从生物膜中筛选出5株能够以PET为营养源生长的嗜热菌;其中,菌株JQ3以PET为唯一碳源生长最佳,作为降解能力较强的降解菌,被鉴定为嗜热淀粉芽孢杆菌(Bacillus thermoamylovorans),其最适生长pH为7.0、最适生长温度为50℃、最适生长NaCl浓度为0.5%;菌株JQ3以0.043 mg PET/d的速率降解PET,对苯二甲酸(terephthalic acid,TPA)产量在第7天达到峰值45.2 mmol/L;菌株JQ3对PET中间体降解效率显著,6 h可降解85.9%的BHET,60 h可降解50.1%的MHET。菌株JQ3能够定殖于PET表面并形成生物膜,侵蚀PET并造成开裂和剥落。【结论】B.thermoamylovorans JQ3作为一株嗜热PET降解菌,能够高温(60℃)降解PET及其中间体,为实现PET的有效降解提供了新策略。  相似文献

【目的】比较高效矿物风化固氮假单胞菌(Pseudomonas azotoformans) F77及其亲缘关系较近的假单胞菌(Pseudomonas paracarnis) P1风化黑云母的效应和机制。【方法】通过检测两株菌在不同时间点的发酵液中细胞数量、pH值、葡萄糖剩余量、葡萄糖酸浓度和可溶性Fe、Al释放量,比较它们对黑云母的风化效果与生理机制。采用RNA-seq技术研究这两株菌风化黑云母过程中出现差异的分子机制。【结果】在持续5 d的风化试验中,菌株F77发酵液中的细胞数量和pH值低于菌株P1,葡萄糖酸浓度是菌株P1的27.3-53.9倍,Fe和Al元素的释放量是菌株P1的3.3-23.3倍。比较转录组数据表明,菌株F77特有的基因数量(2 872)和差异基因数量(1 832)均多于菌株P1 (分别为1 903和1 258)。菌株F77在胞内物质跨膜转运与碳代谢、细胞运动、趋化与信号诱导等途径中基因数量也高于菌株P1。此外,菌株F77的超氧化物歧化酶和过氧化氢酶基因差异表达倍数、葡萄糖酸合成基因数量和差异表达倍数也明显高于菌株P1。【结论】菌株F77风化黑云母以及合成葡萄糖酸的能力显著高于菌株P1。菌株F77通过产生葡萄糖酸来促进黑云母的风化。添加黑云母显著促进了菌株F77胞内与矿物风化相关基因的表达,如物质跨膜转运、细胞运动与趋化、信号诱导、碳代谢及能量代谢等途径基因。此外,葡萄糖酸合成途径基因、超氧化物歧化酶基因以及过氧化氢酶基因在矿物风化中可能发挥重要作用。  相似文献

【目的】为挖掘优良促生菌(PGPR)菌株,分析和定位功能基因,发现其科学价值和工业化开发,为农业生产服务。【方法】以Bacillus mycoides Gnyt1菌株为材料,采用二代和三代测序技术相结合的研究体系,对菌株进行全基因组测序研究,分析菌株核基因组可能存在的功能基因和菌株分泌铁载体相关的基因。【结果】本研究基因组序列拼接后总长度为5597907 bp,GC百分含量为35.57%,该菌株中与铁载体分泌相关的基因共9条,其中2条基因主要存在于Porphyrin metabolism途径,与细胞内铁的运输代谢有关。【结论】通过全基因组测序,最终确定了与铁载体分泌相关的功能基因为GYT1和GYT2,为下一步基因功能验证奠定了基础。  相似文献

【目的】原核生物有两条代谢途径N-甲基化途径(Pmt途径)和磷脂酰胆碱合酶途径(Pcs途径)合成磷脂酰胆碱(PC)。本文对土壤细菌Pseudomonas sp.593的磷脂酰胆碱合成进行了研究,试图弄清楚假单胞菌的磷脂酰胆碱的合成途径。【方法】通过氨基酸序列比较,获得已报道的磷脂酰胆碱合酶(Pcs)的氨基酸保守序列,并设计简并引物,从Pseudomonas sp.593总DNA中PCR扩增出磷脂酰胆碱合酶基因(pcs)的片段,然后用扩增的DNA片段作探针,对Pseudomonas sp.593基因组DNA亚克隆文库进行菌落原位杂交,获得pcs全基因序列;利用同源重组原理进行活体突变,获得Pseudomonas sp.593 pcs-突变体;采用薄层层析(TLC)法分析细菌总磷脂,检测PC含量以及pcs基因活性。【结果】TLC分析显示Pseudomonas sp.593细菌仅在添加外源胆碱的M9或LB培养基中生长时才合成PC;从Pseudomonas sp.593细菌中克隆出894 bp的DNA序列,编码的蛋白具有磷脂酰胆碱合酶活性;活体缺失pcs基因后,Pseudomonas sp.593 pcs-突变体在添加或不添加胆碱的条件下都不能合成PC。【结论】Pcs途径是土壤Pseudomonas sp.593乃至其它假单胞菌合成磷脂酰胆碱的唯一途径。  相似文献

目的 了解不同分离来源铜绿假单胞菌的全基因组基本特征,以此分析基因组多态性及其遗传进化关系。方法 选择10株医源性和食源性来源的铜绿假单胞菌代表性菌株,应用Solexa高通量测序技术对其进行全基因组测序,以此进行多位点序列分型(multilocus sequence typing, MLST),比较各菌株基因组中携带的耐药基因、毒力基因及插入序列(insertion sequence, IS)元件,并通过比较基因组学分析方法拟合泛基因组和核心基因组积累曲线,筛选核心基因SNP构建系统发育分子进化树。结果 10株菌的基因组从6.3~7.0 Mbp大小不一,包含5 868~6 598个基因,平均G+C含量为67.1%;发现10个菌株各具不同的ST型。在这10个菌株的基因组中,共检测到75种耐药基因,包括抗β-内酰胺酶类、抗氨基糖苷类、抗氟喹诺酮类等;共发现188种毒力基因,不同来源菌株间无明显差异;各菌株之间IS元件种类和数量差异较大。分析发现,铜绿假单胞菌具有开放型泛基因组和稳定型核心基因组;10株菌可分为3个进化分支,且不同分离时间和来源无明显相关性。结论 本研究获得10株不同分离来源的铜绿假单胞菌的全基因组序列,初步证实食品及患者分离来源菌株基因组数据无明显相关性,为后续铜绿假单胞菌的分子流行病学和致病性机制研究提供数据参考。  相似文献

【目的】获得高产聚β-羟基丁酸酯(Poly-β-hydroxybutyricacid,PHB)菌株。【方法】以假单胞菌属Pseudomonas koreensis PK3菌株为出发菌株,采用硫酸二乙酯和紫外线相结合的诱变方法进行多轮诱变。【结果】经过初筛和复筛得到一株高产PHB突变菌株,命名为Pseudomonas koreensis UVCN-18。连续传代9次后,发酵28 h条件下,PHB产量达到15.94 g/L,占细胞干重69.54%,较原出发菌株Pseudomonas koreensis PK3(4.42 g/L)提高了2.61倍,并且该菌株具有良好的遗传稳定性。【结论】采用硫酸二乙酯和紫外线相结合的诱变方法,成功获得了一株高产PHB突变菌株。  相似文献

解淀粉芽孢杆菌生防菌BS-3全基因组测序及生物信息分析   总被引:1,自引:0,他引:1  
【背景】解淀粉芽孢杆菌BS-3是从健康橡胶树树根中分离获得的一株对真菌具有较强抗菌活性的内生细菌,有作为生物农药的潜力。【目的】解析菌株BS-3的基因组序列信息,以深入研究该菌株防病促生机制及挖掘次级代谢产物基因资源。【方法】采用第二代BGISEQ与第三代Pac Bio平台相结合的测序技术,对生防菌BS-3进行全基因组测序,并对测序数据进行基因组组装、基因预测与功能注释、共线性分析及次级代谢产物合成基因簇预测等。【结果】BS-3全基因组大小为3 870 130 bp,平均GC含量为46.88%,共编码4 161个基因;含有92个t RNA基因、28个r RNA、10个sRNA;含有122个串联重复序列、98个小卫星DNA、2个微卫星。在COG、GO、KEGG、NR和Swiss-Prot数据库分别注释到基因2 875、2 620、1 885、4 040和3 328个。同时,预测到BS-3中有10个次级代谢产物合成基因簇,编码表面活性素、丰原素、多烯类、儿茶酚型嗜铁素等抑菌物质。基因组测序数据提交至NCBI获得GenBank登录号为CP060384。【结论】为基因组层面上解析菌株BS-3具有良好防病效果的内在原因提供基础数据,为深入了解解淀粉芽孢杆菌次级代谢合成途径提供参考信息,对菌株BS-3后续相关研究具有重要意义。  相似文献

【背景】枯草芽孢杆菌YN145是一株从湖南省桃江县的健康稻株中分离的细菌,前期研究中该菌对稻瘟病菌拮抗效果显著,在生物防治方面有很大的应用潜力。【目的】深入研究该菌株的生防机制并挖掘次级代谢产物基因资源。【方法】在4株稻瘟病菌生防菌中,选择其胞外抗菌物质抑制稻瘟病菌黑色素合成效果最佳的菌株YN145,采用紫外-可见分光光度计在波长400 nm处测定胞外和菌丝体内黑色素液的吸光度值,采用菌丝生长抑制平板法和分生孢子萌发抑制法测定抑菌活性。采用PacBio第三代测序和IlluminaHiSeq第二代测序相结合的技术对菌株YN145进行全基因组测序,并对测序数据进行组装,注释预测基因的功能,分析次级代谢产物合成基因簇。【结果】菌株YN145的胞外抗菌物质能较好地抑制稻瘟病菌黑色素合成、分生孢子萌发和菌丝生长。菌株YN145全基因组大小为4 167 871 bp,GC含量为43.86%,编码序列(coding sequence, CDS)数量为4 294个;共找到85个tRNA、30个rRNA和92个sRNA。同时预测到5个已知的次级代谢产物合成基因簇,分别编码合成bacillaene、bac...  相似文献

【目的】从湖南长沙市采集到的土样中分离假单胞菌并进行归类,研究各菌株抑菌和抗肿瘤生物活性,以丰富假单胞菌菌种资源并为微生物次级代谢物的挖掘奠定基础。【方法】采用大蜡螟诱集法诱集分离假单胞菌,结合形态观察、生理生化特征和16S rRNA基因序列同源性分析,鉴定并归类各细菌,通过平板扩散法、对峙培养法和肿瘤细胞毒性试验分别研究各菌株抑制细菌、拮抗真菌和抗肿瘤细胞等生物活性。【结果】从湖南长沙市郊区菜地、林地中分离得到5株假单胞菌,归类并命名为Pseudomonas protegens CY01、绿针假单胞菌CY02(Pseudomonas chlororaphis CY02)、栖稻假单胞菌CY04(Pseudomonas oryzihabitans CY04)、Pseudomonas sp.CY05和恶臭假单胞菌CY06(Pseudomonas putida CY06)。P.protegens CY01和P.chlororaphis CY02对枯草芽胞杆菌(Bacillus subtillis)和金黄色葡萄球菌(Staphylococcus aureus)具有较好的抑菌效果,P.chlororaphis CY02对水稻稻瘟病菌(Pyricularia oryzae)具有良好的拮抗作用,对小鼠黑色素瘤细胞B16具有较强的细胞毒性。【结论】分离得到的P.chlororaphis CY02,在抑制病原细菌、拮抗水稻稻瘟病菌和抗肿瘤细胞等方面具有显著效果。  相似文献

Scirtothrips perseae Nakahara was discovered attacking avocados in California, USA, in 1996. Host plant surveys in California indicated that S. perseae has a highly restricted host range with larvae being found only on avocados, while adults were collected from 11 different plant species. As part of a management program for this pest, a “classical” biological control program was initiated and foreign exploration was conducted to delineate the home range of S. perseae, to survey for associated natural enemies and inventory other species of phytophagous thrips on avocados grown in Mexico, Guatemala, Costa Rica, the Dominican Republic, Trinidad, and Brazil. Foreign exploration efforts indicate that S. perseae occurs on avocados grown at high altitudes (>1500 m) from Uruapan in Mexico south to areas around Guatemala City in Guatemala. In Costa Rica, S. perseae is replaced by an undescribed congener as the dominant phytophagous thrips on avocados grown at high altitudes (>1300 m). No species of Scirtothrips were found on avocados in the Dominican Republic, Trinidad, or Brazil. In total, 2136 phytophagous thrips were collected and identified, representing over 47 identified species from at least 19 genera. The significance of these species records is discussed. Of collected material 4% were potential thrips biological control agents. Natural enemies were dominated by six genera of predatory thrips (Aeolothrips, Aleurodothrips, Franklinothrips, Leptothrips, Scolothrips, and Karnyothrips). One genus each of parasitoid (Ceranisus) and predatory mite (Balaustium) were found. Based on the results of our sampling techniques, prospects for the importation of thrips natural enemies for use in a “classical” biological control program in California against S. perseae are not promising.  相似文献

Parsimony analyses of the internal transcribed spacer regions of nuclear ribosomal DNA (ITS 1 & ITS 2) for 38 taxa sampled from the Phebalium group (Rutaceae: Boronieae) and two outgroups confirm that, with the exception of Phebalium sensu stricto and Rhadinothamnus, six of the currently recognised genera within the group are monophyletic. The data indicate that Phebaliums. str. is paraphyletic with respect to Microcybe, and Rhadinothamnus is paraphyletic with respect to Chorilaena. Rhadinothamnus and Chorilaena together are the sister group to Nematolepis. Drummondita, included as an outgroup taxon, clustered within the ingroup as sister to Muiriantha and related to Asterolasia.The phylogeny suggests that the evolution of major clades within a number of these genera (e.g. Phebalium) relates to vicariance events between eastern and south-western Australia. Leionema is an eastern genus, with the most basal taxon being the morphologically distinct Leionema ellipticum from northern Queensland. Leionema also includes one species from New Zealand, but this species (as with some others) proved difficult to sequence and its phylogenetic position remains unknown. Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.

Komárek has recently reviewed the various species assigned to the green algal genusNeochloris Starr (Chlorococcales, Chlorococcaceae) and removed those with uninucleate vegetative cells to a new genus,Ettlia. Watanabe & Floyd, unaware ofKomárek's work, also reviewed the species ofNeochloris and distributed them among three genera—Neochloris, Chlorococcopsis gen. nov., andParietochloris gen. nov.—on the basis of details of the covering of the zoospore and the arrangement of the basal bodies of the flagellar apparatus. This paper reconciles these two treatments and makes additional recommendations at the ranks of genus, family, order, and class.  相似文献

Little is known about how tropical land-use systems contribute to the conservation of functionally important insect groups, including dung beetles. In a study at the margin of Lore Lindu National Park (a biodiversity hotspot in Central Sulawesi, Indonesia) dung-beetle communities were sampled in natural forest, young secondary forest, agroforestry systems (cacao plantations with shade trees) and annual cultures (maize fields), each with four replicates (n = 16 sites). At each site we used 10 pitfall traps, baited with cattle dung, along a 100 m transect for six 3-day periods. The number of trapped specimens and species richness at the natural forest sites was higher than in all land-use systems, which did not significantly differ. Each land-use system contained, on average, 75% of the species richness of the natural forest, thereby indicating their importance for conservation. However, a two-dimensional scaling plot based on NESS indices (m = 6) indicated distinct dung beetle communities for both forest types, while agroforestry systems and annual cultures exhibited a pronounced overlap. Mean body size of dung beetles was not significantly influenced by land-use intensity. Five of the six most abundant dung beetle species were recorded in all habitats, whereas the abundance of five other species was significantly related to habitat type. Mean local abundance and number of occupied sites were closely correlated, further indicating little habitat specialisation. The low dung beetle diversity (total of 18 recorded species) may be due to the absence of larger mammals in Sulawesi during historical times, even though Sulawesi is the largest island of Wallacea. In conclusion, the dung beetle fauna of the lower montane forest zone in Central Sulawesi appears to be relatively robust to man-made habitat changes and the majority of species did not exhibit strong habitat preferences.  相似文献

The genusKarschia, in the earlier sense, including saprophytes and parasites on lichens, has been thought to be a non-lichenized parallel genus of the lichen genusBuellia. Modern workers included it on the one hand inBuellia, on the other hand combined it with bitunicate ascomycetes. It is now proved thatKarschia is heterogeneous and contains but superficially similar members both of the genusBuellia of theLecanorales and of typical or masked bitunicateAscomycetes. Therefore, it can not be regarded as a link betweenLecanorales andDothideales. The type species ofKarschia belongs to theDothideales.
