采用基因组DNA富集文库法FIASCO(Fast isolation by AFLP of sequences containing repeats)从掌叶木(Handeliodendron bodinieri)基因组中分离和筛选了10个新的微卫星位点,进而对掌叶木茂兰自然分布居群的遗传多样性进行了分析。结果表明, 每个位点在30株掌叶木个体上的平均等位基因数(A)为3.5(3~5个),平均观察杂合度(HO)为0.650(0.267~0.900),平均预期杂合度(HE)为0.494(0.224~0.652)。每个位点的第一排除概率值Pr(Ex1)为0.029~0.240,位点综合值为0.7496。单个位点的第二排除概率Pr(Ex2)为0.123~0.419,位点综合值为0.9517。这些信息预示着这些微卫星标记可以为研究喀斯特特有濒危树种掌叶木的基因流及居群遗传结构提供有效的遗传工具。  相似文献

垂珠花自然居群表型性状及遗传多样性分析   总被引:1,自引:0,他引:1  
为探索垂珠花自然居群间表型差异的内在因素,该研究用14个表型性状和微卫星分子标记法对6个垂珠花自然居群进行遗传多样性分析。结果表明:(1)14个表型性状在居群间和居群内皆存在极显著差异;居群间平均表型分化系数(V_(st))为22.64%,垂珠花自然居群内的表型变异大于居群间的表型变异,说明居群内变异是垂珠花居群的主要变异来源。(2)4对微卫星引物在87个个体上共检测到43个等位基因,平均每条引物10.63个,平均多态位点百分率(P)为100%;平均观察杂合度(H_o)和平均期望杂合度(H_e)分别为0.659和0.811,Shannon多样性指数(I)平均达1.894;居群内遗传多样性(H_s)高达0.811,居群间遗传多样性(D_(st))仅为0.110,居群间遗传分化系数(F_(st))为0.118,说明11.8%的遗传变异来自于居群间,88.2%的遗传变异来自于居群内,与表型变异研究所得结论大体相一致。表型性状和微卫星分子标记结果均表明垂珠花自然居群存在丰富的遗传变异,较高的基因流(Nm=2.050)使得垂珠花居群间的遗传变异小于居群内的遗传变异。  相似文献

大叶藻居群微卫星遗传多样性研究   总被引:1,自引:0,他引:1  
采用4对微卫星引物对大叶藻的7个地理居群进行了遗传多样性与遗传结构分析。扩增148株大叶藻得到57个等位基因, 每个位点平均等位基因数为6, 大叶藻居群的平均期望杂合度(He)为0.687, 平均观测杂合度(Ho)为0.417。青岛湾居群的遗传多样性最高(A=7.750, AR=7.043), 俚岛居群最低(A=4.750, AR=4.543)。从Fst值来看, 7个大叶藻居群间属于中度分化。UPGMA系统发育树显示, 中国4个大叶藻居群聚类到一起, 其遗传分化可能是由于历史大海草场的遗留小片段居群产生, 而中国、韩国、日本和爱尔兰居群间的遗传分化则主要是由于地理隔离造成的。自由交配估计结果支持海草的东亚起源说。青岛湾居群遗传多样性较高, 可优先作为大叶藻移植修复的材料和基因库, 并进行重点保护。  相似文献

东平湖麦穗鱼群体遗传结构的微卫星标记分析   总被引:5,自引:0,他引:5  
利用微卫星标记技术,采用26对鲤微卫星引物对山东东平湖麦穗鱼进行全基因组扫描.结果表明,有13对引物能获得稳定的特异性条带(占总数的50%),其中有6个微卫星位点具有多态性(占总数的23.1%).6个多态位点共检测到22个等位基因,每个位点的等位基因数从2个到7个不等,大小在80~406bp之间;平均多态信息含量(PIC)为0.5115,平均观测杂合度(H0)为0.6812,平均期望杂合度(HE)为0.5775.研究结果表明,东平湖麦穗鱼群体遗传多样性较丰富,种群结构合理,种质资源处于安全状态.  相似文献

血水草(Eomecon chionantha Hance)是中国亚热带地区特有的单种属植物。本研究采用(AG)15、(AC)15两种生物素探针及磁珠富集法从血水草基因组中开发并筛选出13个微卫星位点,且在该物种中能进行稳定的PCR扩增并表现出多态性。利用筛选出的13个SSR多态性标记对血水草4个野生居群24个个体进行PCR扩增,结果显示每个微卫星位点的等位基因数为2~7个,观测杂合度(HO)和期望杂合度(HE)分别为0.125~0.792和0.299~0.691。说明这些SSR标记可用于血水草遗传多样性的后续研究,并揭示该物种地理分布格局形成的原因,有助于保护和利用中国亚热带地区的血水草野生资源。  相似文献

利用微卫星DNA标记,对来自青海囊谦县、治多县以及甘肃阿克塞县3个地区的36份雪豹(Panthera uncia)粪便DNA样品进行了遗传多样性研究。结果显示,在8个微卫星位点上共检测到57个等位基因,有效等位基因数为2.190~5.488,平均每个位点的等位基因数为7.130,基因频率分布不均匀;期望杂合度为0.543~0.847,平均0.759;多态信息含量为0.458~0.829,平均0.722;表明这8个微卫星位点均为高度多态性位点,有较丰富的遗传多样性。3个样地雪豹居群之间的遗传距离与地理距离相关,地理距离最近的青海省囊谦县和治多县的雪豹居群遗传距离最小。根据雪豹平均遗传分化度Fst(0.053)、平均基因流(4.488)以及STRUCTURE聚类分析结果(当K=1时,ln P(D)值最大),推测3个居群间虽然有一定的遗传距离,但均来自同一个种群,暂无分化现象。  相似文献

利用10对SSR引物对濒危植物秦岭冷杉(Abies chensiensis)6个自然居群的120个个体进行了遗传多样性研究,旨在分析秦岭冷杉6个自然居群的遗传多样性、遗传结构及基因流变化.研究结果表明,120个个体在10个位点上共检测到149个等位基因,平均每个位点的等位基因数(A)为14.9,每个位点的有效等位基因数(e)为7.7,每个位点的平均预期杂合度(He)和平均观察杂合度(Ho)分别为0.841和0.243,Shannon多样性指数(Ⅰ)为2.13,自然居群杂合性基因多样度的比率(FsT)为6.7%,居群间的基因流(Nm)为3.45.利用Mantel检测到自然居群的遗传距离与地理距离间无显著相关性(r=0.490 6,P>0.05).秦岭冷杉自然居群的遗传多样性水平较低,遗传变异主要存在于居群内部.结合研究数据、实地调查及相关资料,推测秦岭冷杉自然居群间基因流较原来增大可能是因为居群间发生了远交衰退.  相似文献

为阐明布氏罗非(Tilapia buttikoferi)群体遗传变异和遗传结构状况,采用50个尼罗罗非鱼特异性的微卫星分子标记对45个布氏罗非鱼个体进行遗传检测.结果有27对引物能获得稳定的特异性条带,占总数的54%,其中16个多态性微卫星座位共检测出52个等位基因.每个座位的等位基因数为2~6之间,平均每个座位为3.24;平均观测杂合度(Ho)为0.5266,平均期望杂合度(He)为0.5237,平均多态信息含量为0.4652,表明布氏罗非鱼群体遗传多样性较丰富,种群结构处于合理状态.  相似文献

虾夷扇贝遗传结构及微卫星标记与经济性状相关分析   总被引:3,自引:0,他引:3  
选用30个多态性微卫星分子标记对大连大长山(96个)、大连獐子岛(96个)及日本青森县陆奥湾(96个)3个地区虾夷扇贝养殖群体(Patinopecten yessoensis)进行遗传多样性分析。结果显示,30个基因座共检测到198个等位基因(Na),平均等位基因数为6.63个,每个座位检测到的等位基因数为3-12,有效等位基因数(Ne)每个位点平均为4.70个,观测杂合度(Ho)平均值0.58,期望杂合度(He)的平均值为0.75,30个位点的平均多态信息含量为0.703,说明3个地区虾夷扇贝群体遗传多样性水平较高。运用统计软件SPSS11.5对30个微卫星标记与大长山群体生长性状相关性进行了分析。结果表明,位点DQ221714和FJ262378与壳长、壳高、壳宽和鲜重显著相关(P0.05)位点;位点FJ262372与壳宽、软体部重和闭壳肌重显著相关;位点FJ262369与软体部重及闭壳肌重显著相关。对差异显著的位点进行不同基因型间的多重比较,找到了与虾夷扇贝生长性状相关的基因型,进一步将所得显著性标记对大长山虾夷扇贝个体最大的17个和个体最小的13个基因组DNA进行检测,验证其准确性。结果表明,位点DQ221714在两组虾夷扇贝中存在特异性条带,可作为QTL定位候选标记。  相似文献

核基因组微卫星标记揭示大理茶参与了普洱茶的驯化过程   总被引:1,自引:0,他引:1  
利用11个核基因组微卫星标记对普洱茶3个居群、大理茶3个居群及过渡型大理茶2个居群共104株古茶树进行了遗传学分析。研究表明,普洱茶、大理茶和过渡型大理茶居群的遗传多样性相对较低,平均等位基因(Na)为4.852,平均香农多样性指数(I)为1.17,平均期望杂合度(He)和观测杂合度(Ho)分别为0.59和0.52,其中大理茶的遗传多样性水平低于普洱茶和过渡型大理茶。AMOVA分析表明,普洱茶和大理茶之间遗传分化显著(FST=0.305),遗传变异主要在居群内(分别为93.51%和89.41%),而居群间的遗传变异较低(分别为6.49%和10.59%)。主成分分析和STRUCTURE聚类分析均支持大理茶和普洱茶为不同的组,过渡型大理茶主要由大理茶驯化而来,并在栽培过程中与大理茶产生了遗传分化。在混栽的大理茶和普洱茶居群间存在由大理茶向普洱茶的明显基因渐渗,证实了大理茶参与了普洱茶的驯化过程。最后,讨论并提出了对大理茶和普洱茶古茶树资源保护的相关建议。  相似文献

Manilkara maxima is an endemic tree species of the Atlantic Forest in southern Bahia, Brazil. It is considered important for forest conservation due to its mutualistic interactions with endemic and endangered animals. Our aim was to develop microsatellite markers to estimate genetic diversity in order to provide information for effectiveness of future conservation programs. We used next generation sequencing technology to develop the first specific microsatellite markers for M. maxima. Seventeen new microsatellite loci were applied in 72 individuals sampled in three natural populations. On average, the number of alleles per loci was 8.8. The expected heterozygosity varied between 0.72 and 0.77, indicating that the developed set of molecular markers is useful for genetic diversity studies. Additionally, the estimated value for the combined probability of exclusion (Q) was greater than 0.999, which indicates the powerful of these molecular tools for paternity and kinship analysis. Our results demonstrate that the set of microsatellites developed in this work is a powerful tool for population genetics, molecular ecology and conservation biology purposes.  相似文献

We investigated reproduction in a semi‐free‐ranging population of a polygynous primate, the mandrill, in relation to genetic relatedness and male genetic characteristics, using neutral microsatellite and major histocompatibility complex (MHC) genotyping. We compared genetic dissimilarity to the mother and genetic characteristics of the sire with all other potential sires present at the conception of each offspring (193 offspring for microsatellite genetics, 180 for MHC). The probability that a given male sired increased as pedigree relatedness with the mother decreased, and overall genetic dissimilarity and MHC dissimilarity with the mother increased. Reproductive success also increased with male microsatellite heterozygosity and MHC diversity. These effects were apparent despite the strong influence of dominance rank on male reproductive success. The closed nature of our study population is comparable to human populations for which MHC‐associated mate choice has been reported, suggesting that such mate choice may be especially important in relatively isolated populations with little migration to introduce genetic variation.  相似文献

The black muntjac (Muntiacus crinifrons) is a vulnerable species found in the mountains of eastern China, about which little is known. Here we develop 11 polymorphic microsatellite loci from a (CA)(n) -enrichment library for the animal. In the analyses of 25 individuals sampled, unbiased expected heterozygosity levels varied from 0.686 to 0.876 and the number of alleles per locus varied from five to nine. Results that 11 microsatellite loci were highly polymorphic indicated that these markers are sufficiently powerful to address such questions as parentage, mating system and population genetic structure of M. crinifrons.  相似文献

Four microsatellites were used to examine the genetic variability of the spawning stocks of Chinese sturgeon, Acipenser sinensis, from the Yangtze River sampled over a 3‐year period (1999–2001). Within 60 individuals, a total of 28 alleles were detected over four polymorphic microsatellite loci. The number of alleles per locus ranged from 4 to 15, with an average allele number of 7. The number of genotypes per locus ranged from 6 to 41. The genetic diversity of four microsatellite loci varied from 0.34 to 0.67, with an average value of 0.54. For the four microsatellite loci, the deviation from the Hardy–Weinberg equilibrium was mainly due to null alleles. The mean number of alleles per locus and the mean heterozygosity were lower than the average values known for anadromous fishes. Fish were clustered according to their microsatellite characteristics using an unsupervised ‘Artificial Neural Networks’ method entitled ‘Self‐organizing Map’. The results revealed no significant genetic differentiation considering genetic distance among samples collected during different years. Lack of heterogeneity among different annual groups of spawning stocks was explained by the complex age structure (from 8 to 27 years for males and 12 to 35 years for females) of Chinese sturgeon, leading to formulate an hypothesis about the maintenance of genetic diversity and stability in long‐lived animals.  相似文献

太湖日本沼虾野生群体遗传结构的微卫星分析   总被引:3,自引:0,他引:3  
利用8个高度多态性的微卫星位点分析了太湖日本沼虾野生群体的遗传结构.结果表明:在15个群体中至少有3个位点经Bonferroni校正后显示杂合不足,显著偏离了HardyWeinberg平衡;15个群体中观测杂合度均大于0.683,显示出较高的遗传多样性水平,但其波动明显,如太湖东、南部的渡口和陆巷等群体的遗传多样性高于西、北部的华庄和洋渚等群体;突变-漂移平衡分析结果显示,15个群体中部分位点杂合显著过剩,偏离了突变-漂移平衡,且近期曾经历过瓶颈效应,群体数量曾经下降;群体间AMOVA分析表明,太湖日本沼虾群体间遗传分化程度较低(FST=0.011),98.9%的遗传变异来自群体内,1.1%来自群体间,并没有形成显著的遗传结构,在种质资源保护和管理上可视作一个单元;华庄与吴塘门群体间DA遗传距离达到0.206,已接近种间分类界限,故太湖日本沼虾种质资源可持续利用工作仍须深入的研究.  相似文献

The genetic variability of 22 heterologous microsatellite markers was analyzed in two Indian goat breeds, namely Bengal and Chegu. The heterozygosity, polymorphism information content (PIC), and probability of identity of two individuals were calculated for all microsatellite loci in both the breeds. The observed number of alleles varied between 4 and 13 at the studied microsatellite loci. The evaluated microsatellite loci exhibited high mean heterozygosity of 0.69 +/- 0.11 and 0.66 +/- 0.07 in Bengal and Chegu goats, respectively. The mean PIC values of the studied loci in these breeds were 0.79 +/- 0.08 and 0.78 +/- 0.05, respectively. The probability of identity of two random individuals from different breeds, taking into account, all the 22 microsatellite loci was as low as 5.523 x 10(-40). On the basis of these results, we propose that these microsatellite markers may be used with reliability for studying genetic diversity and for identification of individuals in Indian goat breeds.  相似文献

Microsatellite loci were isolated from enriched partial genomic libraries of Acyrthosiphon loti and Acyrthosiphon pisum. Twenty of those loci were characterized in A. pisum. Fifteen of those loci were polymorphic. Genetic diversity varied across loci, allele repeat number ranging from two to 15, and observed heterozygosity from 0.1 and 0.96. An additional eight microsatellite loci originally isolated from other aphids but cross‐priming with A. pisum showed polymorphism as well. Allele size ranged from three to 9 and observed heterozygosity from 0.43 to 0.84. Overall, we present 23 microsatellite loci that can be used to reveal polymorphism in pea aphids.  相似文献

利用磁珠富集法, 以生物素标记的(CA)15为探针, 构建了蛇鳄龟(Chelydra serpentina L.)微卫星富集文库。通过PCR法从富集文库中共筛选出70条微卫星序列, 一共设计了48对微卫星引物, 采用PCR扩增的方法从中筛选出36对引物, 对一个蛇鳄龟养殖群体进行遗传多样性分析。通过分析, 36个位点获得的等位基因数从29不等, 平均为4.361, 有效等位基因为1.4617.767, 平均为3.498。等位基因片段大小为56342 bp, 观测杂合度为0.0671.000, 平均为0.725; 期望杂合度为0.3160.850, 平均0.600; 多态信息含量为0.26550.8359, 平均为0.5573; 结果表明此蛇鳄龟养殖群体存在较高的遗传多样性水平。群体内固定系数-0.6880.856, 平均为-0.214, 说明蛇鳄龟群体中杂合子过剩。    相似文献

Eleven highly polymorphic microsatellite loci were isolated from the North African gundi (Ctenodactylus gundi), a gregarious rock‐dwelling rodent found in northern Africa. These loci exhibited high genetic diversity (eight to 19 alleles per locus) and high levels of heterozygosity (62–98%). Collectively, these microsatellite markers are sufficiently variable to estimate levels of relatedness and population genetic structure for C. gundi.  相似文献

Although it is a major freshwater gastropod species, genetic diversity of Bellamya aeruginosa was completely unknown. Eighteen microsatellite loci were isolated and characterized from (AC)(15)-enriched genomic libraries of the freshwater snail B. aeruginosa. Most of the 18 loci were successfully amplified and high polymorphic information content values were found, ranging from 0.244 to 0.792 (mean 0.541). The number of alleles per locus ranged from 5 to 13 (mean 8.8), the expected heterozygosity varied from 0.347 to 0.950 (mean 0.815) and the observed heterozygosity varied from 0.087 to 0.782 (mean 0.431). Eight loci showed significant deviation from Hardy-Weinberg equilibrium after Bonferroni's correction and no significant genotypic linkage disequilibrium was detected between most locus pairs, except for TXH79-TXH97 and TXH113-TXH121. These 18 polymorphic microsatellite loci should be useful for population genetics analysis and species identification of Bellamya.  相似文献
