红掌品种亲缘关系SRAP分析   总被引:1,自引:0,他引:1  
利用相关序列扩增多态性(SRAP)分子标记,从100对引物组合中筛选出 26对多态性高、条带清晰的SRAP引物,对33个红掌品种进行遗传多样性和亲缘关系分析。结果如下:(1)26对引物共扩增出366条条带,其中有314条多态性条带,多态性比率为85.79%。引物组合产生的条带数在9~23之间,平均每对引物组合扩增出14.1条和12.1条多态性条带。(2)根据SRAP扩增结果,利用UPGMA法进行聚类分析,33份材料的遗传相似系数在0.55~0.94之间,在遗传相似系数0.786处可将33个红掌品种分为5个类群。结果表明,供试品种遗传多样性丰富,本研究为品种鉴定和杂交育种提供了参考信息。  相似文献

利用SRAP和SSR各23对引物对20个中国主要黑芝麻品种进行了遗传多样性分析。结果显示,23对SRAP引物共扩增出DNA带672条,其中多态性带152条,比率为22.62%,平均每对引物扩增总带数和多态性条带分别为29.22条和6.61条。23对SSR多态性引物共扩增出DNA带92条,每对引物扩增出3~6条,平均4.00条;每对引物扩增出多态性带1~5条,平均3.09条,多态性带比率平均为77.17%。20个黑芝麻品种间的遗传相似系数为0.8547~0.9804,遗传距离为0.0159~0.0921,遗传多样性匮乏,遗传基础狭窄。聚类结果表明,来自主产区江西的11个品种明显聚在一起,且江西黑芝麻品种的遗传相似系数高于其他省份品种,遗传距离低于其他省份品种,与其他省份品种的差异均达到极显著水平。加强资源引进和利用是拓宽中国黑芝麻品种遗传基础的迫切要求。  相似文献

以26种冬青属植物种质资源为研究材料,利用RAPD和AFLP技术对基因组DNA进行扩增,以研究其物种间遗传多样性以及亲缘关系.结果表明:在RAPD分析中,从400条10个碱基的寡核苷酸引物中筛选出反应稳定、扩增性强、重复性好的引物20个,共扩增出312条多态性条带,多态率为95.41%;聚类分析显示26种冬青属植物间,布利奥特夫人枸骨叶冬青和黄果在AFLP分析中,10对选择性引物组合均扩增出了丰富的多态性片段,共扩增出350条谱带,其中336条具有多态性,占95.96%.综合RAPD和AFLP聚类结果,枸骨、无刺枸骨和日拉斯纳尔逊枸骨的亲缘关系较近,钝齿冬青、金宝石钝齿冬青和龟甲冬青三者的亲缘关系较近,可为冬青属植物的杂交育种与种质创新提供理论依据.  相似文献

利用RAPD、ISSR和SRAP 3种分子标记对我国南方地区松花型花椰菜主栽品种进行鉴定,分析了品种间的遗传多样性。3种标记共产生370条扩增带,238条为多态性条带,其多态率为64.32%。其中只有SRAP标记的引物m e1/em1可将20个品种全部鉴别。遗传相似系数分析表明,松花型花椰菜品种之间的亲缘关系较近,遗传背景比较狭窄。聚类分析表明品种间的亲缘关系与熟性、地理分布相关。研究表明,分子标记能有效地应用于花椰菜品种鉴定,且综合多种分子标记分析品种间的遗传多样性将更加准确可靠。  相似文献

利用SSR与RAPD分子标记评估甘蔗品种的遗传多样性   总被引:1,自引:0,他引:1  
利用SSR与RAPD两种分子标记对美国、中国台湾以及中国大陆不同甘蔗育种单位选育的甘蔗品种或亲本材料的遗传多样性进行评估。其中19对SSR引物共扩增出87条带,多态性带为84条,多态性比例为96.55%,扩增出的条带数范围为2~8条,平均每对引物扩增出4.58条带,引物的PIC值范围为0.34~0.93,平均0.64。21条RAPD引物共扩增出184条带,扩增条带数范围为3~16,平均每条引物扩增8.76条带,其中多态性带为184,多态性比例为100%,引物PIC范围为0.53~0.97,平均0.86。结果表明,两种分子标记都能较好的评估甘蔗品种的遗传多样性。  相似文献

采用RAPD和AFLP技术,对10种冬青属植物基因组进行DNA片段扩增,以研究该属种间遗传多样性.结果表明:在RAPD分析中,通过对100种10个碱基随机引物的筛选,发现11种引物能得到多态性较高扩增产物,11种引物共扩增出301条多态性条带,多态率为98.63%.在AFLP分析中,3对选择性引物组合均扩增出了丰富的多态性片段.利用RAPD和AFLP技术分析,结果按UPGMA类平均法进行聚类,聚类结果显示冬青和代茶冬青,木姜冬青和浙江冬青以及光枝刺缘冬青与毛枝三花冬青之间的亲缘关系最近.  相似文献

利用SRAP标记对中国22个番木瓜主要栽培品种(系)进行亲缘关系和遗传多样性研究。结果表明:(1)筛选出的20对SRAP引物共扩增249条谱带,其中多态性条带110条,多态性比率为43.20%,平均每对引物扩增的条带数和多态性条带数分别为12.45条和5.50条。(2)引物多态性信息含量(PIC)值变化范围为0.15~0.79,平均值为0.49。(3)聚类分析和主坐标分析显示,供试材料间的遗传相似系数为0.72~0.96,遗传多样性水平较低,在相似性系数为0.87时,可将所有试材分为3个类群。该研究结果有效地揭示了中国番木瓜主要栽培品种(系)资源的遗传背景和亲缘关系,可为番木瓜种质资源的分类、保护和有效利用以及新品种选育提供理论依据。  相似文献

利用ISSR分子标记技术对5个地区6个麦冬居群进行遗传多样性分析。选用10条扩增带型清晰且重复性好的引物进行扩增,共获得115条带,其中89条具有多态性,多态性比例为77.39%。当遗传相似系数(GS)为0.61时,可将6个麦冬居群分为2大类群。居群的GS平均值为0.643,表明供试居群之间存在较近的亲缘关系,且地域越近的其亲缘关系关系越近。研究表明ISSR分子标记技术能够很好地用于不同地域同种物种的亲缘关系分析。  相似文献

用SSR和AFLP技术分析花生抗青枯病种质遗传多样性的比较   总被引:10,自引:0,他引:10  
由Ralstonia solanacearum E.F.Smith引起的青枯病是若干亚洲和非洲国家花生生产的重要限制因子,利用抗病品种是防治这一病害最好的措施。虽然一大批抗青枯病花生种质资源材料已被鉴定出来,但对其遗传多样性没有足够的研究,限制了在育种中的有效利用。本研究以31份对青枯病具有不同抗性的栽培种花生种质为材料,通过简单序列重复(SSR)和扩增片段长度多态性(AFLP)技术分析了它们的遗传多样性。通过78对SSR引物和126对AFLP引物的鉴定,筛选出能显示抗青枯病种质多态性的SSR引物29对和AFLP引物32对。所选用的29对多态性SSR引物共扩增91条多态性带,平均每对引物扩增3.14条多态性带;32对多态性AFLP引物共扩增72条多态性带,平均扩增2.25条多态性带。在所筛选引物中,4对SSR引物(14H06,7G02,3A8,16C6)和1对AFLP引物(P1M62)检测花生多态性的效果优于其他引物。SSR分析获得的31个花生种质的遗传距离为0.12-0.94,平均为0.53,而AFLP分析获得的遗传距离为0.06~0.57,平均为0.25,基于SSR分析的遗传距离大于基于AFLP分析的遗传距离,疏枝亚种组的遗传分化相对大于密枝亚种组。基于两种分析方法所获得的聚类结果基本一致,但SSR数据聚类结果与栽培种花生的形态分类系统更为吻合。根据分析结果,对构建青枯病抗性遗传图谱群体的核心亲本和抗性育种策略提出了建议。  相似文献

利用ISSR分子标记对9个观叶福禄桐品种进行遗传多样性和亲缘关系分析,从100条引物中筛选出9条稳定、多态性高的引物用于PCR扩增,共获得70条带,其中多态性条带61条,多态百分率为87.14%。聚类结果显示,品种间相似性系数为0.346 9-0.816 3,聚类结果与品种间的地理来源紧密相关,从外观形态上比较,亲缘性较近的叶形和株型相似性较高;观叶福禄桐各品种间基因型差异较小,亲缘关系较近,遗传基础相对较窄。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
