旨在检测嗜酸乳杆菌S-层蛋白以及S-层蛋白与抗生素联用对大肠杆菌(Escherichia coli)和金黄色葡萄球菌(Staphylococcus aureus)的抑制作用。采用液体发酵培养法获得嗜酸乳杆菌菌体,LiCl法提取S-层蛋白粗提物,凝胶过滤层析法纯化S-层蛋白,分别用E.coli和S.aureus处理Caco-2细胞2 h后,考察S-层蛋白在不同浓度和不同作用时间条件下对E.coli和S.aureus的抑制作用,并考察S-层蛋白联合抗生素对E.coli和S.aureus的抑制作用,实验分组:(1)空白对照;(2)嗜酸乳杆菌组;(3)S-层蛋白组;(4)抗生素组;(5)嗜酸乳杆菌+抗生素组;(6)S-层蛋白+抗生素组。结果显示,液体发酵得到嗜酸乳杆菌菌体,提取并纯化得到S-层蛋白;S-层蛋白对E.coli和S.aureus有很好的抑制效果,具有浓度依赖性,高浓度下抑制率达到42.2%和31.7%,差异极显著(P<0.01),且在E.coli和S.aureus作用的短时间内干预效果明显,0 h时的抑制率分别达到59.3%和48.4%;S-层蛋白联合抗生素的抑菌率分别达到81.7%和79.3%,差异极显著(P<0.01),效果优于单独使用抗生素。嗜酸乳杆菌S-层蛋白具有较强的抑菌作用,可以与抗生素联用,有望开发称为一种新型的抗菌药物。  相似文献

【目的】采用不同实验方法测定常用有机溶剂二甲基亚砜(DMSO)、丙酮和乙醇对细菌活性的影响,以指导抗菌类药物体外抑菌实验所用溶剂的选择和添加限量。【方法】采用常规体外抑菌实验方法(纸片扩散法、肉汤稀释法),并参照生长曲线法检测有机溶剂DMSO、丙酮和乙醇对大肠杆菌(Escherichia coli)及金黄色葡萄球菌(Staphylococcus aureus)的抑菌作用,采用扫描电子显微镜(SEM)观察溶剂作用后的细菌形态变化。【结果】3种溶剂对E.coli和S.aureus抑菌率达到20%时,在肉汤稀释法下,DMSO、丙酮、乙醇的浓度(体积比)分别为1.00%、0.25%、2.00%和1.00%、1.00%、0.50%;在生长曲线法下,溶剂浓度(体积比)分别为0.50%、1.00%、0.50%和1.00%、0.50%、0.50%;而在纸片扩散法下,32%(体积比)DMSO和32%(体积比)乙醇对E.coli产生明显抑菌圈,但3种溶剂对S.aureus均无抑菌圈出现。3种方法比较后得出:当3种溶剂的抑菌率达到20%时,溶剂浓度(体积比)均低于0.5%,对细菌整体生长活性影响较小。SEM结果表明控制溶剂使用限量可有效减少其对E.coli生长过程的影响。【结论】相对于DMSO和丙酮,乙醇对微生物生长繁殖能力的影响更加明显;采用相同浓度有机溶剂时,液态条件下(肉汤稀释法和生长曲线法)微生物受到有机溶剂的影响较大。  相似文献

【背景】萘并吡喃酮类化合物生物活性多样,是真菌Aspergillusniger中特征次生代谢产物。【目的】研究分离自海洋滩涂土壤的真菌Aspergillus niger XJJ-3中萘并吡喃酮类化合物结构及其抗菌和卤虫致死活性。【方法】以TLC分析为导向,综合运用多种色谱和光谱方法分离和鉴定萘并吡喃酮类化合物。采用微量稀释法测试化合物的抗菌和卤虫致死活性。【结果】从真菌A.niger大米发酵产物中共分离得到6个萘并吡喃酮类化合物,鉴定为RubrofusarinB(1)、Flavasperone (2)、Aurasperone A (3)、Asperpyrones C (4)、Asperpyrones B (5)和Fonsecinone A (6)。抗菌活性实验结果表明,化合物1-6对致病菌S. aureus ATCC33591、29213、E. faecium ATCC35667和V.parahemolyticus表现出不同程度的抑制活性,其中化合物2和4对S.aureus ATCC33591表现出较强抑制活性(MIC分别为43.7μmol/L和21.9μmol/L),化合物3对E.faecium ATCC35667抑制活性较强(MIC为21.9μmol/L)。卤虫致死活性实验结果表明,化合物1-6均表现出一定的卤虫致死活性,其中化合物2和3活性显著(LD50分别为35.0μmol/L和8.8μmol/L)。【结论】菌株XJJ-3可产生结构丰富的萘并吡喃酮类化合物,化合物1-6存在不同程度的抗菌和卤虫致死活性,该研究可为抗菌和细胞毒类药物的研发提供参考。  相似文献

目的探讨急性肾盂肾炎合并尿路感染危险因素与致病微生物的特点。方法选择2017年4月-2018年4月在本院肾内科就诊的急性肾盂肾炎合并尿路感染患者59例,无菌操作收集患者中段尿,导管收集患者膀胱尿。2 h内送检,采用定量接种的方法,将一定量混匀未离心尿液用接种环接种于血平板或麦康凯平板进行细菌培养。相关性分析采用Pearson相关性检验。结果糖尿病、肺部感染、尿路结石、脑血管疾病和肿瘤是常见的急性肾盂肾炎(APN)合并尿路感染的危险因素。男性中段尿检出率较高的菌种有大肠埃希菌(E. coli)、金黄色葡萄球菌(S. aureus)等;膀胱尿中检出率较高的菌种有大肠埃希菌(E. coli)、肺炎克雷伯菌(K. pneumoniae)。女性中段尿和膀胱尿中检出率较高的菌种均是大肠埃希菌(E. coli)和金黄色葡萄球菌(S. aureus)。女性总住院时间≥4 d的患者比例明显高于男性。APN患者中合并糖尿病与大肠埃希菌(E. coli)和粪肠球菌(E. faecalis)存在正相关,尿路结石与大肠埃希菌(E. coli)、肺炎克雷伯菌(K. pneumoniae)、屎肠球菌(E. faecium)存在正相关。结论患者尿液中大肠埃希菌(E. coli)最常见,女性APN患者更容易并发尿路感染,住院时间长于男性患者。合并糖尿病的患者更容易患有大肠埃希菌(E. coli)和粪肠球菌(E. faecalis)感染,合并尿路结石的患者更倾向于大肠埃希菌(E. coli)、肺炎克雷伯菌(K. pneumoniae)、屎肠球菌(E. faecium)感染。  相似文献

本文对太行菊叶片不同溶剂提取物进行抑菌作用研究。结果表明,太行菊乙醇提取液抑菌效果好于水提液。其中,80%乙醇提取液抑菌效果最显著,对S.aureus和E.coli的MIC分别为25 mg/m L和50 mg/m L,S.aureus对提取物更加敏感。  相似文献

研究谢氏丙酸杆菌(Propionibacterium shermanii,CICC10019)发酵过程中产生丙酸杆菌素对食源性致病菌及芽菜内生菌的抑菌效果。将丙酸杆菌素溶液发酵液用于绿豆芽保鲜,评价了其对绿豆芽菌落总数、pH和挥发性盐基氮的影响。结果表明:谢氏丙酸杆菌发酵时间为120 h时,产酸最多,pH为4.52,还原糖总量消耗大,发酵液抗菌活性最高;丙酸杆菌素溶液对金黄色葡萄球菌(Staphylococcus aureus,ATCC6538)、大肠杆菌(Escherichia coli,ATCC35401)、枯草芽孢杆菌(Bacillus subtilis,ATCC9372)、志贺氏菌(Shigella,ATCC12022)、沙门氏菌(Salmonella,ATCC14028)和副溶血性弧菌(Vibrio Parahemolyticus,ATCC17802)均产生显著的抑菌效应,对阪崎肠杆菌(E.sakazakii)没有抑制作用。用丙酸杆菌素溶液清洗绿豆芽,可将豆芽菌落总数降低1.20 lg CFU/g;4℃保存第10 d时处理过的豆芽pH为6.20,比对照组降低了1.60;挥发性盐基氮含量为28.70 mg/100 g,为对照组的50%。冷藏条件下芽菜保质期延长了2 d。  相似文献

【目的】筛选固态发酵苦荞高产多肽及发酵产物液具有抗菌、抗氧化活性的菌株。【方法】采用米曲霉、酱油曲霉、雅致放射毛霉和少孢根霉分别对苦荞进行固态发酵,以蛋白酶活力、水解度、可溶性肽得率、抑菌率和体外自由基清除率作为筛菌指标。【结果】米曲霉固态发酵苦荞的可溶性肽得率最高达38.83%±1.18%,发酵产物液对大肠杆菌和金黄色葡萄球菌的抑菌率分别为96.62%±1.66%和97.54%±0.54%,同时羟自由基(·OH)清除率和二苯基苦味酰基苯肼自由基(DPPH·)清除率分别为55.65%±1.25%和10.84%±1.03%。对米曲霉发酵2 d发酵产物液的不同分子量分布及活性分析表明,分子量大小对抗菌及抗氧化活性有一定的影响。【结论】米曲霉可作为固态发酵苦荞制备多肽且发酵产物液具有抗菌及抗氧化活性的最佳菌株,并在多肽产量提升及抗菌、抗氧化活性的研究上具有巨大空间。  相似文献

本研究选用肉桂(Cinnamomum cassia)的桂皮、桂叶、桂枝、果实、花萼5种植物部位为研究对象,通过气质联用对各部位精油的主要成分组成进行了分析比较。在此基础上,从抑菌圈、最低抑菌浓度(MIC)、最低杀菌浓度(MBC)等方面比较各部位精油对金黄色葡萄球菌(Staphylococcus aureus)、大肠杆菌(Escherichia coli)抑制活性。结果表明,各部位精油中的主要成分均为反式肉桂醛、邻甲氧基肉桂醛等,但各成分的含量存在明显差异,以花萼中的反式肉桂醛含量最高(87.68%)。各部位精油对两种供试微生物均具有显著抑菌效果,以桂枝中所含精油的抑菌效果最佳,对E.coli和S.aureus的MIC分别为0.05%和0.025%,MBC同样也分别为0.05%和0.025%。  相似文献

以采集自四个红树林地点的16份混合土壤为研究材料,选用7种选择性培养基,共分离获得330株放线菌。其中217株菌经16SrRNA基因序列分析,发现近75%菌株属于小单孢菌属(Micromonospora),其他还包括多形态孢菌属(Polymorphospora),疣孢菌属(Verrucosispora)等小单孢菌科的2个属和非小单孢菌科的9个属。采用美蓝酶标仪法对所分离到的放线菌进行抗菌活性检测,共50株菌表现出对金黄色葡萄球菌(Staphylococcus aureus ATCC 51650)、大肠杆菌(Escherichia coli ATCC 25922)和白色念珠菌(Candida albicans ATCC 10231)有不同程度抗性。然后利用高效液相色谱(HPLC)和液质联用技术(LC-MS)对有生物活性的菌株进行化学筛选,最后确定了5株可能产新颖化合物的小单孢菌。  相似文献

2004年4月采集大连金州湾沿岸畜禽养殖场猪、牛、鸡和某中学人粪便样品。经改良的生化鉴定方法鉴定,得大肠埃希菌(Escherichia coli)猪源75株、牛源78株、鸡源69株、人源68株,占粪大肠菌比率分别为85.23%、92.86%、80.23%、80.00%。为验证改良的生化鉴定方法准确性,从中随机选取8株E.coli,扩增16S rRNA序列并与Genebank中E.coli16S序列比对,确定这8株细菌与E.coli同源相似率达99%以上,进而验证改良的生化鉴定方法的可行性。  相似文献

Twenty-four enamines were synthesized and reported for the first time. Their chemical structures were confirmed by means of 1H NMR, ESI mass spectra, and elemental analyses, and four of them were determined by single crystal X-ray diffraction analysis. All of the compounds were assayed for antibacterial (Bacillus subtilis ATCC 6633, Escherichia coli ATCC 35218, Pseudomonas fluorescens ATCC 13525, and Staphylococcus aureus ATCC 6538) and antifungal (Aspergillus niger ATCC 16404, Candida albicans ATCC 10231, and Trichophyton rubrum ATCC 10218) activities by MTT method. Compounds (E)-ethyl 3-(4-hydroxyphenylamino)-2-(4-methoxyphenyl)acrylate (9b), (E)-ethyl 3-(3,5-difluorophenylamino)-2-(4-chlorophenyl)acrylate (11b), (E)-ethyl 3-(3,5-dichlorophenylamino)-2-(4-chlorophenyl)acrylate (12b), and (E)-ethyl 3-(4-methylphenylamino)-2-(4-chlorophenyl)acrylate (15b) showed considerable antibacterial activities against S. aureus ATCC 6538 with MICs of 3.8, 1.9, 1.1, and 0.9 microg/mL, respectively. Structure-activity relationship (SAR) analysis disclosed, generally, an E-isomer exhibited higher antibacterial activity than the corresponding Z-isomer. An electron-withdrawing group on A-ring led to some decrease in activity, while on B-ring, a similar substitution provided higher activity.  相似文献

The antibacterial activity of the methanolic extract and its fractions of aerial parts of Aniheinis tinctoria (Asteraceae) was investigated against representative gram-positive Staphylococcus aureus (ATCC 25923) and Enterococcus faecalis (ATCC 29212) and gram-negative strains Escherichia coli (ATCC 25922) and Pseudomonas aeruginosa (ATCC 27853). The activity was concentrated mainly in the dichloromethane (DCM) and hexane fractions of crude methanolic extract. The 5 mg of DCM extract per disk produced 15-16 mm of inhibition zone against S. aureus and P. aeruginosa, however, no activity was found against E. faecalis and E. coli. The hexane fraction showed activity against S. aureus, P. aeruginosa and E. faecalis. As DCM fraction showed the highest antibacterial activity in the disk diffusion assay, the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values of only this fraction was determined against S. aureus and P. aeruginosa. These values were found to be in the range of 1.25 to 10 mg/ml.  相似文献

The purpose of the present study was to investigate the antibacterial activity of seven ethanolic extracts and three aqueous extracts from various parts (leaves, stems and flowers) of A. aroma against 163 strains of antibiotic multi-resistant bacteria. The disc diffusion assay was performed to evaluate antibacterial activity of the A. aroma crude extracts, against several Gram-positive bacteria (E. faecalis, S. aureus, coagulase-negative stahylococci, S. pyogenes, S. agalactiae, S. aureus ATCC 29213, E. faecalis ATCC 29212) and Gram-negative bacteria (E. coli., K. pneumoniae, P. mirabilis, E. cloacae, S. marcescens, M morganii, A. baumannii, P. aeruginosa, S. maltophilia, E. coli ATCC 35218, P. aeruginosa ATCC 27853, E. coli ATCC 25922). All ethanolic extracts showed activity against gram-positive bacteria. Among all obtained extracts, only leaf and flower fluid extracts showed activity against Gram-negative bacteria. Based on this bioassay, leaf fluid extracts tended to be the most potent, followed by flower fluid extracts. Minimal inhibitory concentration (MIC) values of extracts and antibiotics were comparatively determined by agar and broth dilution methods. Both extracts were active against S. aureus, coagulase-negative stahylococci, E. faecalis and E. faecium and all tested Gram-negative bacteria with MIC values from 0.067 to 0.308 mg/ml. In this study the minimal bactericidal concentration (MBC) values were identical or twice as high than the corresponding MIC for leaf extracts and four or eight times higher than MIC values for flower extracts. This may indicate a bactericidal effect. Stored extracts have similar antibacterial activity as recently obtained extracts. The A. aroma extracts of leaves and flowers may be useful as antibacterial agents against Gram- negative and Gram-positive antibiotic multi-resistant microorganisms.  相似文献

Low-density polyethylene (LDPE) belongs to commodity polymer materials applied in biomedical applications due to its favorable mechanical and chemical properties. The main disadvantage of LDPE in biomedical applications is low resistance to bacterial infections. An antibacterial modification of LDPE appears to be a solution to this problem. In this paper, the chitosan and chitosan/pectin multilayer was immobilized via polyacrylic acid (PAA) brushes grafted on the LDPE surface. The grafting was initiated by a low-temperature plasma treatment of the LDPE surface. Surface and adhesive properties of the samples prepared were investigated by surface analysis techniques. An antibacterial effect was confirmed by inhibition zone measurements of Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). The chitosan treatment of LDPE led to the highest and most clear inhibition zones (35mm(2) for E. coli and 275mm(2) for S. aureus).  相似文献

Complex physalin metabolites present in the capsules of the fruit of Physalis angulata L. have been isolated and submitted to a series of assays of antimicrobial activity against Pseudomonas aeruginosa ATCC 27853, Staphylococcus aureus ATCC 29213, S. aureus ATCC 25923, S. aureus ATCC 6538P, Neisseria gonorrhoeae ATCC 49226, Escherichia coli ATCC 8739; E. coli ATCC 25922, Candida albicans ATCC 10231 applying different methodologies such as: bioautography, dilution broth, dilution agar, and agar diffusion techniques. A mixture of physalins (pool) containing physalins B, D, F, G inhibit S. aureus ATCC 29213, S. aureus ATCC 25923, S. aureus ATCC 6538P, and N. gonorrhoeae ATCC 49226 at a concentration of 200 mg/microl, using agar dilution assays. The mixture was inactive against P. aeruginosa ATCC27853, E. coli ATCC 8739; E. coli ATCC 25922, C. albicans ATCC 10231 when applying bioautography assays. Physalin B (200 microg/ml) by the agar diffusion assay inhibited S. aureus ATCC 6538P by +/- 85%; and may be considered responsible for the antimicrobial activity.  相似文献

Phanquone is active against a wide range of Gram-positive and Gram-negative organisms. Its activity is affected by the nature of the suspending fluid, pH and anaerobic growth conditions. Its ability to chelate metal ions was examined and found to be related to its antibacterial activity, which was reduced by the presence of added metal ions, e.g. Co (II), Cu(II), Fe(II) and Fe(III) in nutrient media for both E. coli and S. aureus. When antibacterial activity was examined in dis-nutrient media for both E. coli and S. aureus. When antibacterial activity was examined in distilled water, then certain added metal ions, whilst antagonizing activity was examined in distilled water, then certain added metal ions, whilst antagonizing the activity of Phanquone against E. coli, exerted a co-operative effect in the case of S. aureus. The addition of EDTA and NTA lowered the activity of Phanquone against S. aureus, but not E. coli, while the addition of thiol-containing compounds lowered its activity against E. coli but not S. aureus. concentration quenching was observed for S. aureus but not for E. coli, while overnight pre-incubation at 4 degrees C resulted in the appearance of a growth zone inside the zone of inhibition in the case of S. aureus but not E. coli. Phanquone may have a different mode of action against the two organisms.  相似文献

The currative properties of aromatic and medicinal plants have been recognized since ancient times and, more recently, the antimicrobial activity of plant essential oils has been used in several applications, including food preservation. The purpose of this study was to create directly comparable, quantitative data on the antimicrobial activity of some plant essential oils prepared in the National Institute of Research-Development for Chemistry and Petrochemistry, Bucharest to be used for the further development of food packaging technology, based on their antibacterial and antifungal activity. The essential oils extracted from thyme (Thymus vulgaris L.), basil (Ocimum basilicum L.), coriander (Coriandrum sativum L.), rosemary (Rosmarinus officinalis L.), sage (Salvia officinalis L.), fennel (Foeniculum vulgare L.), spearmint (Mentha spicata L.) and carraway (Carum carvi L.) were investigated for their antimicrobial activity against eleven different bacterial and three fungal strains belonging to species reported to be involved in food poisoning and/or food decay: S. aureus ATCC 25923, S. aureus ATCC 6538, S. aureus ATCC 25913, E. coli ATCC 25922, E. coli ATCC 35218, Salmonella enterica serovar Enteritidis Cantacuzino Institute Culture Collection (CICC) 10878, Listeria monocytogenes ATCC 19112, Bacillus cereus CIP 5127, Bacillus cereus ATCC 11778, Candida albicans ATCC 10231, Aspergillus niger ATCC 16404, Penicillium spp. CICC 251 and two E. coli and Salmonella enterica serovar Enteritidis clinical isolates. The majority of the tested essential oils exibited considerable inhibitory capacity against all the organisms tested, as supported by growth inhibition zone diameters, MICs and MBC's. Thyme, coriander and basil oils proved the best antibacterial activity, while thyme and spearmint oils better inhibited the fungal species.  相似文献

为了确定莳萝蒿精油的化学成分,并探究其抑菌活性及抑菌机理。该研究采用水蒸气蒸馏法提取莳萝蒿精油,并通过气相色谱-质谱联用法测定其化学成分。采用抑菌圈法、二倍稀释法和生长曲线法测定精油的抑菌活性,采用电导率法和扫描电镜法探究精油的抑菌机理。结果表明:(1)莳萝蒿精油的主要化学成分包括醇类(47.12%)和萜烯类(19.90%),在所有成分中桉油精(12.39%)含量最高,其次为松油醇(8.70%)。(2)精油对金黄色葡萄球菌和大肠杆菌的抑菌圈直径分别为(22.57±1.68)mm和(15.36±0.71)mm。(3)精油对金黄色葡萄球菌和大肠杆菌的最小抑菌浓度分别为3.25和7.5μL/mL,最小杀菌浓度分别为7.5和15μL/mL。(4)当精油浓度为1.625和3.25μL/mL时,其分别能够延缓金黄色葡萄球菌和大肠杆菌的生长;当精油浓度为3.25和7.5μL/mL时,其能够完全抑制金黄色葡萄球菌的生长;当精油浓度为7.5和15μL/mL时,其能够完全抑制大肠杆菌的生长。(5)经精油处理之后的细菌,其相对电导率明显增大,且随精油浓度的增加而增大,同时其细胞膜发生了萎缩和破裂的现象。研究发现,莳萝蒿精油富含醇类和萜烯类等多种活性物质,对金黄色葡萄球菌和大肠杆菌具有良好的抑菌活性,且莳萝蒿精油能够改变细胞的膜结构,导致细菌中的内溶物发生泄漏,从而抑制细菌生长。  相似文献

In an attempt to isolate bioactive constituents, ethyl acetate cone extract of Metasequoia glyptostroboides was subjected to a column chromatographic analysis that resulted in isolation of an abietane-type diterpenoid, taxodone. Its structure was elucidated by spectroscopic means. Further, taxodone showed potential antibacterial effect as diameters of zones of inhibition against foodborne pathogenic bacteria, such as Listeria monocytogenes ATCC 19166, Salmonella typhimurium KCTC 2515, S. enteritidis KCTC 2021, Escherichia coli ATCC 8739, E. coli O157:H7 ATCC 43888, Enterobacter aerogenes KCTC 2190, Staphylococcus aureus ATCC 6538 and S. aureus KCTC 1916, were found in the range of 9.4 to 14.2 mm. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values of taxodone against the employed bacterial pathogens were found in the range of 250 to 1000 and 250 to less than 2000 microg/ml, respectively. Also the compound had a strong antibacterial effect on the viable counts of the tested bacteria. These findings indicate that the bioactive compound taxodone present in M. glyptostroboides could be used as an antibacterial agent in food industry to inhibit the growth of certain important foodborne pathogens.  相似文献
