Methyl jasmonate (MJ) and yeast extract (YE) induce protein expression and benzophenanthridine alkaloid accumulation in Eschscholtzia californica suspension cell cultures. One hundred microM MJ primarily induced dihydrosanguinarine 509.0+/-7.4 mg/l); 0.2 g/l YE induced sanguinarine (146.8+/- 3.8 mg/l) and an unknown compound. These results occur because dihydrobenzophenanthridine oxidase (DHBO) is induced by YE and not by MJ. YE and chitin (CHI) had similar effects on sanguinarine production and DHBO expression. Differential induction of secondary metabolites was shown in E. californica suspension cultures and the expression of proteins confirmed the metabolite results. Furthermore, treatment by various oligosaccharides helped us to understand the elicitation effect of YE in signal transduction pathways.  相似文献

(−)6aR,11aR-Dihydro-3-hydroxy-9,10-dimethoxy-6H-benzofuro[3,2c] [1]-benzopyran (10-methoxymedicarpin), (+)-(2,3,4,-trimethoxyphenyl)-2,3-dihydro-7-hydroxy-4H-1-benzopyran (7-hydroxy-2′,3′,4′-trimethoxyisoflavan) and (+)-(2,3,4-trimethoxy-5-hydroxyphenyl)-2,3-dihydro-7-hydroxy-4H-1-benzopyran (7,5′-dihydroxy-2′,3′,4′-trimethoxyisoflavan) were isolated for the first time from dried Medicago sativa hay. Structural assignments were based on 1H NMR and mass spectra, X-ray crystallography, and optical rotations.  相似文献

The quantitative carotenoid composition of the red flower petals of Adonis annua is reported. Optically pure (3S, 3′S)-astaxanthin occurs both as a diester (64% of total carotenoid) and as a monoester (11%). The optical purity was determined by hydrolysis of the natural esters in the absence of oxygen and subsequent HPLC analysis of the paren -ketol esterified with (−)-camphanic acid. All non-animal sources hitherto examined synthesize pure 3S,3′S- or 3R,3′R-isomers of astaxanthin, whereas marine animal sources contain mixtures of all three optical isomers, including the meso form.  相似文献

A crtD (1-HO carotenoid 3,4-dehydrogenase gene) homolog from marine bacterium strain P99-3 included in the gene cluster for the biosynthesis of myxol (3,4-didehydro-1,2-dihydro-β,ψ-carotene-3,1,2-triol) was functionally identified. The P99-3 CrtD was phylogenetically distant from the other CrtDs. A catalytic feature was its high activity for the monocyclic carotenoid conversion: 1-HO-torulene (3,4-didehydro-1,2-dihydro-β,ψ-caroten-1-ol) was prominently formed from 1-HO-γ-carotene (1,2-dihydro-β,ψ-caroten-1-ol) in Escherichia coli with P99-3 CrtD, indicating that this enzyme has been highly adapted to myxol biosynthesis. This unique type of crtD is a valuable tool for obtaining 1-HO-3,4-didehydro monocyclic carotenoids in a heterologous carotenoid production system.  相似文献

This study aimed to improve rosmarinic acid (RA) production in the whole plant culture of Solenostemon scutellarioides through elicitation. Amongst selected elicitors methyl jasmonate (MJ), salicylic acid (SA), and yeast extract (YE) caused significant elevation in RA accumulation. Elicitation with MJ (50 μM) and SA (50 μM) caused almost 1.7 and 1.4-fold increase in RA accumulation, respectively, within day 1. While YE (100 μg ml?1) elicitation showed highest RA content (~1.5-fold) in day 3. Preceding the elicitor-induced RA accumulation, there was a notable alteration in the specific activities of RA biosynthetic enzymes viz. phenylalanine ammonia lyase, tyrosine aminotransferase, hydroxyl-phenylpyruvate reductase and rosmarinic acid synthase up on MJ (50 μM), SA (50 μM) and YE (100 mg ml?1) elicitation. Based on differential responses of aforementioned enzymes, RA synthesis was further scaled up through combination of elicitors in pre-optimized doses. In synergy study, at a time exposure with MJ + SA + YE and MJ + SA followed by YE after 24 h has been found to produce significant elevation of RA (2.0 and 1.9-fold, respectively) within 24 h while later maintained a steady state increased level (~1.7 ± 0.2-fold) over control up to day 7.  相似文献

In different plant species, secondary metabolite biosynthesis is regulated by the phytohormone jasmonic acid (JA), which is derived by the action of lipoxygenase. In this study, we examined mono- and sesquiterpenoid accumulation and the related signal transduction pathways and biosynthetic genes in adventitious root cultures of Panax ginseng C.A. Meyer as induced by yeast extract (YE, 3 g/L), a biotic elicitor, and salicylic acid (SA, 200 μM), a signaling elicitor. The lipoxygenase (LOX) gene was highly expressed in 24 and 12 h after treatment with SA and YE. JA content was significantly increased in 24 h after SA treatment. The H2O2 content was the highest in 24 and 72 h after the onset of SA and YE treatment, respectively. RNA blot analysis showed that farnesyl diphosphate synthase (FPS) and isopentenyl pyrophosphate isomerase (IPPI) genes encoding enzymes of the biosynthesis of mono- and sesquiterpenoids were up-regulated by both elicitors. Farensol, isochiapin B sesquiterpenoids, champhor, and cineole monoterpenoids were highly accumulated after 24 h of SA treatment, while YE treatment induced bacchotricuneatin C, guaiazulene, isochiapin B, and p-benzoquinone sesquiterpenoid production. These results suggest that mono- and sesquiterpenoid accumulation induced by SA and YE occurs due to the IPPI and FPS expression and may be mediated by reactive oxygen species signaling and jasmonic acid signal transduction.  相似文献

A new neolignan, 5,8-epoxy-6,7-dimethyl 2′,3′,2″,3″-dimethylene dioxy-4′,1″-dimethoxy-1,2:3,4-dibenzo-1,3-cyclooctadiene, from the petrol extract of Clerodendron inerme seeds, was characterized by spectroscopic and X-ray crystallographic methods. The heteroditopic ligand 4′-(4,7,10-trioxadec-1-yn-10-yl)-2,2′:6′,2″-terpyridine, 2, contains an N,N′,N″-donor metal-binding domain that recognizes iron(II), and a terminal alkyne site that selectively couples to platinum(II). A screening of microalgae strains is described, with the objective to discover more species besides the known cyanobacterium Nodularia harveyana which excrete the manifold biologically active and co-mutagenic indole alkaloid norharmane (9H-pyrido(3,4-b)indole) into their environment. Seven more cyanobacterial species, Anabaena cylindrica, Anabaena inaequalis, Anabaenopsis siamensis, Chroococcus minutus, Nostoc carneum, Nostoc commune and Phormidium foveolarum, were newly discovered. The norharmane concentrations detected for cyanobacterial culture media varied in a species-dependent manner from less than 1 up to 525 μg l−1. The risk for humans and livestock, resulting from the natural appearance of norharmane as an extracellular metabolite of various cyanobacteria, is discussed.  相似文献

Toddalidimerine, a new dimeric benzophenanthridine alkaloid, has been isolated from the roots of Toddalia asiatica. On the basis of spectral analysis it has been characterized as 1,3-(8-hydrochelerythrinyl-8′-hydro-N-norchelerythrinyl) acetone. The presence of dihydrochelerythrine and 8-acetonyldihydrochelerythrine has been confirmed in this plant.  相似文献

The biotransformation of the fungistatic agent (R)-(+)-1-(4′-chlorophenyl)propan-1-ol (1) by the phytopathogen Botrytis cinerea has been studied. The main reaction pathways involved hydroxylations on several positions as well as condensations with secondary metabolites of the fungus. Concentrations of two iridoid glucosides (2′-cinnamoyl mussaenosidic acid and 2′-caffeoyl mussaenosidic acid) were measured in different populations of several Avicennia species. In vitro cultures of Argemone mexicana (Papaveraceae) were induced from leaves of mature plants. Sanguinarine, a benzophenanthridine, was the main alkaloid in the cultures, even in the absence of inducers of secondary metabolism. The accumulation of this metabolite was increased by adding methyl jasmonate and fungal elicitors, although in a limited fashion in comparison to other sanguinarine-producing species. Evidence of a transport mechanism, which may be related to the magnitude of the response, was obtained based on the fluorescent properties of bezophenathridines in the elicited cultures.  相似文献

Valeriana amurensis Smir. ex Kom. is a traditional Chinese medicinal plant as its roots serve as a rich source of valtrate, a compound with antifungal cytotoxic sedation and tranquilizing activities. In this study, an adventitious root culture of V. amurensis was established, and the effects of different elicitors, including salicylic acid (SA), chitosan (CH), jasmonic acid (JA), methyl jasmonate (MJ), yeast extract (YE), and yeast extract carbohydrate fraction (YECF) along with the precursor citronellal (CI) on production of valepotriates were assayed. MJ and JA highly promoted valtrate production; while, CH moderately promoted valtrate production. In contrast, CI, SA, YE, and YECF were ineffective in promoting valtrate production. Using a response surface methodology, optimal combinations of MJ, JA, and CH were shown to have strong synergistic effects on valtrate production, and this was subsequently confirmed in experimental studies.  相似文献

Alanine racemase of Bacillus stearothermophilus catalyzes transamination as a side reaction. Stereospecificity for the hydrogen abstraction from C-4′ of pyridoxamine 5′-phosphate occurring in the latter half transamination was examined. Both apo-wild-type and apo-fragmentary alanine racemases abstracted approximately 20 and 80% of tritium from the stereospecifically-labeled (4′S)- and (4′R)-[4′-3H]PMP, respectively, in the presence of pyruvate. Alanine racemase catalyzes the abstraction of both 4′S- and 4′R-hydrogen like amino acid racemase with broad substrate specificity. However, R-isomer preference is a characteristic property of alanine racemase.  相似文献
